This study compared the heat-shock response and metabolic energy status in hatchery-raised and two groups of wild-caught steelhead trout parr collected from the Navarro River watershed, California. Wild parr were from coastal and inland sites with different thermal regimes. Fish were exposed in the laboratory to 25 ± 0.2°C for 2-h and the heat-shock response was assessed in muscle tissue via induction of heat-shock proteins (hsps) 63, 72, 78, and 89. Metabolic measurements included muscle phosphocreatine (PCr), ATP, ADP, and AMP, and hepatic glycogen. Inland and coastal fish overlapped considerably with regard to their hsp responses and energetic endpoints, but hatchery fish were distinct in the biochemical patterns they exhibited. Hsp expression levels after temperature shock were significantly lower in hatchery than in wild fish. Hatchery fish also had significantly lower hepatic glycogen and higher muscle ADP, ATP, and PCr concentrations than wild fish. Coastal and inland steelhead did not differ significantly with regard to peak hsp72 and hsp89 levels or to concentrations of energy metabolites. However, fish from the warm-water, inland site expressed significantly less hsp63, maintained higher basal levels of hsp72, and induced hsp89 more slowly than fish from the cold-water, coastal site. Discriminant function analysis revealed that hatchery fish can be distinguished from wild Navarro River fish with 84.9% certainty using the following function: f(x) = − 43.6 + 0.14(Gly) + 4.1(PCr) + 186.4(AMP) + 80.8(ADP) − 0.14(hsp63) + 0.005(hsp72). This study demonstrates that within a single species, rearing conditions or genetic variation can influence an organism’s disposition and cellular response to thermal stress. Extrapolation of results from laboratory studies on hatchery fish to wild fish may therefore not be possible, and caution must be used when interpreting hsp data obtained for wild fish with different thermal histories.  相似文献

The culture of the mulloway (Argyrosomus japonicus), like many other Sciaenidae fishes, is rapidly growing. However there is no information on their metabolic physiology. In this study, the effects of various hypoxia levels on the swimming performance and metabolic scope of juvenile mulloway (0.34 ± 0.01 kg, mean ± SE, n = 30) was investigated (water temperature = 22 °C). In normoxic conditions (dissolved oxygen = 6.85 mg l− 1), mulloway oxygen consumption rate (M·o2) increased exponentially with swimming speed to a maximum velocity (Ucrit) of 1.7 ± < 0.1 body lengths s− 1 (BL s− 1) (n = 6). Mulloway standard metabolic rate (SMR) was typical for non-tuna fishes (73 ± 8 mg kg− 1 h− 1) and they had a moderate scope for aerobic metabolism (5 times the SMR). Mulloway minimum gross cost of transport (GCOTmin, 0.14 ± 0.01 mg kg− 1 m− 1) and optimum swimming velocity (Uopt, 1.3 ± 0.2 BL s− 1) were comparable to many other body and caudal fin swimming fish species. Energy expenditure was minimum when swimming between 0.3 and 0.5 BL s− 1. The critical dissolved oxygen level was 1.80 mg l− 1 for mulloway swimming at 0.9 BL s− 1. This reveals that mulloway are well adapted to hypoxia, which is probably adaptive from their natural early life history within estuaries. In all levels of hypoxia (75% saturation = 5.23, 50% = 3.64, and 25% = 1 .86 mg l− 1), M·o2 increased linearly with swimming speed and active metabolic rate (AMR) was reduced (218 ± 17, 202 ± 14 and 175 ± 10 mg kg− 1 h− 1 for 75%, 50% and 25% saturation respectively). However, Ucrit was only reduced at 50% and 25% saturation (1.4 ± < 0.1 and 1.4 ± < 0.1 BL s− 1 respectively). This demonstrates that although the metabolic capacity of mulloway is reduced in mild hypoxia (75% saturation) they are able to compensate to maintain swimming performance. GCOTmin (0.09 ± 0.01 mg kg− 1 m− 1) and Uopt (0.8 ± 0.1 BL s− 1) were significantly reduced at 25% dissolved oxygen saturation. As mulloway metabolic scope was significantly reduced at all hypoxia levels, it suggests that even mild hypoxia may reduce growth productivity.  相似文献

Abstract

Elemental deficiencies of nitrogen, phosphorus, potassium, calcium, magnesium, sulfur, iron, manganese, copper, zinc, or boron (N, P, K, Ca, Mg, S, Fe, Mn, Cu, Zn, or B) were induced in plants of Florabella Pink strawflower [Bracteantha bracteata (Vent.) A. A. Anderberg]. Rooted stem cuttings were planted in 4.87‐L plastic containers and fertilized with a complete modified Hoagland's solution or this solution minus the element that was to be investigated. Plants were harvested for tissue analyses as well as dry weights when initial foliar symptoms were expressed and later under advanced deficiency symptoms. Deficiency symptoms for all treatments were observed within 7 weeks. The most dramatic expression of foliar symptoms occurred with N (chlorotic lower foliage leading to necrotic margins on the mature leaves), Ca (black necrotic spots on the tips of the young leaves), S (uniform chlorosis of young leaves and recently mature leaves), B (thick, leathery, and deformed young leaves), Fe (uniform yellowish‐green chlorosis on the young leaves), and Zn (brownish‐gray necrosis on the tips of the mature leaves). At the initial stage, only Fe‐deficient plants weighed less than the control, whereas K‐, Ca‐, and Mg‐deficient plants had greater dry weights than plants receiving the complete modified Hoagland's solution (control plants). Dry weights of plants treated with solutions not containing N, P, Ca, S, Cu, or Mn were significantly lower when compared with the control plants under an advanced deficiency. Foliar‐tissue concentration data will assist plant‐tissue analysis laboratories in establishing foliar symptom standards for growers.  相似文献

The marine habitat provides a large number of structurally-diverse bioactive compounds for drug development. Marine sponges have been studied over many years and are found to be a rich source of these bioactive chemicals. This study is focused on the evaluation of the activity of six diterpene derivatives isolated from Spongionella sp. on mitochondrial function using an oxidative in vitro stress model. The test compounds include the Gracilins (A, H, K, J and L) and tetrahydroaplysulphurin-1. Compounds were co-incubated with hydrogen peroxide for 12 hours to determine their protective capacities and their effect on markers of apoptosis and Nrf2/ARE pathways was evaluated. Results conclude that Gracilins preserve neurons against oxidative damage, and that in particular, tetrahydroaplysulphurin-1 shows a complete neuroprotective activity. Oxidative stress is linked to mitochondrial dysfunction and consequently to neurodegenerative disorders like Parkinson and Alzheimer diseases, Friedreich ataxia or Amyotrophic lateral sclerosis. This neuroprotection against oxidation conditions suggest that these metabolites could be interesting lead candidates in drug development for neurodegenerative diseases.  相似文献

研究了从苦皮藤根皮中分离的一株编号为2K3内生真菌代谢产物的杀菌活性。生物测定结果表明,2K3菌株菌丝体的甲醇和乙酸乙酯提取物对多种植物病原真菌具有较强的抑菌活性。其中,甲醇提取物的抑菌活性比乙酸乙酯提取物强,在1 000μg/mL浓度下对烟草赤星病菌、马铃薯干腐病菌、小麦赤霉病菌、苹果炭疽病菌、玉米大斑病菌的菌丝生长抑制率均大于90%;抑制孢子萌发法试验结果表明,2K3菌株菌丝体的甲醇提取物对烟草赤星病菌玉米大斑病菌和马铃薯干腐病菌的EC50分别为59.4μg/mL、96.5μg/mL和132.5μg/mL;管碟法生测结果表明,2K3菌株菌丝体的甲醇提取物对枯草芽孢杆菌和蜡状芽孢杆菌的抑菌圈直径分别为20 mm和27 mm;盆栽试验结果表明,2K3菌株菌丝体的甲醇提取物在2 000μg/mL浓度下对小麦白粉病的治疗和保护作用均在70%以上。  相似文献
