We report the development of quantitative competitive (QC) PCR assays for quantifying the 16S, 18S ribosomal and atzC genes in nucleic acids directly extracted from soil. QC-PCR assays were standardised, calibrated and evaluated with an experimental study aiming to evaluate the impact of atrazine application on soil microflora. Comparison of QC-PCR 16S and 18S results with those of soil microbial biomass showed that, following atrazine application, the microbial biomass was not affected and that the amount of 16S rDNA gene representing 'bacteria' increased transitorily, while the amount of 18S rDNA gene representing fungi decreased in soil. In addition, comparison of atzC QC-PCR results with those of atrazine mineralisation revealed that, in response to atrazine treatment, the amount of atzC gene increased transitorily in soil pre-treated with atrazine, suggesting that accelerated atrazine biodegradation in soil could be due to a transient increase in the size of the atrazine mineralising community. 相似文献

- 1. Dabry's sturgeon, a large, long‐lived migratory fish is endemic to the Yangtze River. Over‐fishing and habitat destruction have caused large‐scale declines in natural stocks in the last two decades.
- 2. Examining patterns of genetic diversity has become an integral component of many management plans for endangered species. DNA fingerprinting was applied to detect genetic diversity in Dabry's sturgeon collected in 1958–1959, 1980–1981 and 1998–1999.
- 3. Studies on direct genetic parameters (genetic variability, hypervariable loci and heterozygosity) and indirect parameters (band‐sharing coefficient and allelic frequency) showed that the continuous decline in wild populations has caused the loss of genetic diversity in present‐day sturgeon.
- 4. The present‐day populations have the lowest genetic variability; thus, effective management is needed to preserve genetic diversity.
- 5. A conservation strategy is urgently required, comprising artificial rearing facilities coupled with breeding management plans.

Many chlorine-containing pesticides, for example 2-chloro-s-triazines, are of great concern both environmentally and toxicologically. As a result, ascertaining or predicting the fate and transport of these compounds in soils and water is of current interest. Transformation pathways for 2-chloro-s-triazines in the environment include dealkylation, dechlorination (hydrolysis), and ring cleavage. This study explored the feasibility of using computational chemistry, specifically the hybrid density functional theory method, B3LYP, to predict hydrolysis trends of atrazine (2-chloro-N4-ethyl-N6-isopropyl-1,3,5-triazine-2,4-diamine) and related 2-chloro-s-triazines to the corresponding 2-hydroxy-s-triazines. Gas-phase energetics are described on the basis of calculations performed at the B3LYP/6-311++G(d,p)//B3LYP/6-31G* level of theory. Calculated free energies of hydrolysis (delta h G298) are nearly the same for simazine (2-chloro-N4,N6-diethyl-1,3,5-triazine-2,4-diamine), atrazine, and propazine (2-chloro-N4,N6-di-isopropyl-1,3,5-triazine-2,4-diamine), suggesting that hydrolysis is not significantly affected by the side-chain amine-nitrogen alkyl substituents. High-energy barriers also suggest that the reactions are not likely to be observed in the gas phase. Aqueous solvation effects were examined by means of self-consistent reaction field methods (SCRF). Molecular structures were optimized at the B3LYP/6-31G* level using the Onsager model, and solvation energies were calculated at the B3LYP/6-311++G(d,p) level using the isodensity surface polarizable continuum model (IPCM). Although the extent of solvent stabilization was greater for cationic species than neutral ones, the full extent of solvation is underestimated, especially for the transition state structures. As a consequence, the calculated hydrolysis barrier for protonated atrazine is exaggerated compared with the experimentally determined one. 以pcDNA3.1-pIL-18为模板,采用PCR技术扩增到了猪白细胞介素18(IL-18)的成熟蛋白基因,通过KpnⅠ+SacⅠ双酶切及连接反应,构建了pET32c—pIL—18原核表达质粒。经过限制性内切酶分析、PCR鉴定及DNA序列测定证实,重组质粒中的基因片段连接正确。之后,重组质粒转化大肠杆菌BL21(DE3),于37℃、1.0mmol/L IPTG条件下诱导表达。菌体裂解产物经SDS—PAGE分析,在分子质量约为33ku处出现了预期的目的蛋白。用8mol/L脲对表达产物变性,经Ni^2+NTA柱纯化,透析复性,得到了纯化的IL-18蛋白。Western—blot分析证实,纯化的重组IL-18蛋白具有反应活性。上述研究结果为重组IL-18的应用奠定了基础。 相似文献

A primary lung tumor in a dog treated with intensity-modulated radiation therapy was imaged approximately 6 weeks and 1-year posttreatment with combined positron emission tomography (PET) and computed tomography, utilizing the radiotracers 18F-fluorodeoxyglucose and 18F-fluorothymidine. These two tracers allowed discrimination of tumor from inflammation, and demonstrated spread of tumor along airways over time after treatment. Fusion of functional imaging with anatomic imaging is a useful tool, particularly in the field of oncology, with the potential for PET markers that delineate tumor from normal or reactive tissue, and potential or actual response to therapy. 相似文献

从发生在四川、重庆等省市的斑点叉尾(鮰)急性流行性传染病病鱼的肝脏、肾脏内分离到1株高致病性菌株(CCF00024),人工感染健康鱼表现出与自然病鱼相同的症状,并从中分离获得同种细菌,证实其为斑点叉尾(鮰)急性流行性传染病的病原菌.形态、生理生化检测表明,该菌为非发酵型直杆菌,严格需氧,革兰氏阴性,极生多鞭毛,对除麦芽糖和甘露糖以外的多种糖类不利用产酸,氧化酶阴性,DNA酶、蛋白酶、脲酶、赖氨酸脱羧酶阳性,MR、VP阴性.在以该菌16S rDNA序列(GenBank登录号AY970826)和GenBank及RDP数据库内同源性较高的细菌16S rDNA序列构建的系统发育树中,分离菌CCF00024与嗜麦芽寡养单胞菌(Stenotrophomonas maltophilia)聚在一簇,特别是与S.maltophilia M5-1的同源性最高,序列相似性达99.6%,结合形态和生理生化特点将其鉴定为嗜麦芽寡养单胞菌(Stenotrophomonas maltophilia).药敏试验结果表明,对磺胺甲(口恶)唑、磺胺异(口恶)唑、阿齐霉素、洛美沙星高度敏感,而对新霉素、卡那霉素、氨苄青霉素,头孢唑啉、先锋霉素V和链霉素不敏感. 相似文献
