进行了日本晚樱的组培快繁试验。试验结果表明,日本晚樱最佳的增殖培养基是MS/ML + 6-BA 0.5 mg/L + NAA0.05 mg/L +白糖40g/L +琼脂4.5g/L,增殖系数为2.47~2.56;最佳生根培养基为ML+IBA0.05mg/L +NAA0.05 mg/L +白糖20g/L +琼脂4.5g/L,生根率为100%;炼苗12~17d后移栽至草炭土中,成活率达90%。  相似文献

The objective of this study was to broaden the genetic base in oleiferous Brassica juncea by resynthesis, using 10 diverse parental lines of oleiferous B. rapa and two lines of B. nigra of both Indian and exotic origin. Out of 14 crosses attempted using B. rapa as the female parent, eight were successful. Embryo rescue was necessary to obtain interspecific plants. A total of 29 fertile interspecific plants were obtained after colchicine treatment. In the S2 generation, the expression of component characters in the majority of the resynthesized plants showed a negative trend. The resynthesized B. juncea lines are being maintained through repeated selfing and selection at each generation for desirable plant types. This process will continue till the progeny lines of the desirable plants achieve uniformity.  相似文献

In vitro meristem tissue cultures are used for production of virus-free rootstocks of hop (Humulus lupulus L.). Because use of plant tissue cultures is associated with occurrence of somaclonal variability, we assessed somaclonal variability in hop meristem in vitro cultures before and after thermotherapy by different molecular methods (RFLP, RAPD, STS, ISSR and AFLP) and compared it with existing clonal variability of Osvald's clones 31, 72 and 114. No molecular differences were observed between mother plants and in vitro mericlones by RFLP and STS analyses. Amplified molecular differences were found in RAPD and ISSR products of one from five in vitromericlones cvs. Eroica (E5) and Southern Brewer (SB2), respectively. Similarities with mother plants were 0.965 and 0.913 (JSC), respectively. Specific amplified polymorphic products were found for every mericlone and mother plant in AFLP reactions and variability of DNA sequence ranged from 0.824 to 0.993 (JSC). This variability was very similar to determined intra-clonal variability within Osvald's clones 31, 72 and 114 by AFLP analysis. Inter-clonal variability of DNA sequence was exactly higher than intra-clonal variability of DNA sequence in these clones. The molecular differences between Osvald's clone 72 normal and meristem derived were not verifiable. Thermotherapy increased frequency of molecular changes, since amplified differences were found in 14 from 20 in vitro mericlones of cv. Eroica, in 6 from 11 in vitro mericlones of cv. Yeoman and in 15 from 23 in vitro mericlones of cv. Southern Brewer by RAPD and ISSR analyses.  相似文献

Conditions for reliable induction of embryogenesis from isolated microspores were studied in ten genotypes of broccoli. Embryo yields were significantly increased in almost all of the broccoli genotypes by the incubation at 32.5 °C for 1 day, than when the standard incubation at 30 °C for 2 days was used. Treatments of 48 hours at 32.5 °C produced less than optimal results suggesting that broccoli microspores are more sensitive to high temperatures than those of B. napus. The use of the 1/2 NLN-13 medium yielded greater number of embryos than the standard NLN-13. The magnitude of the response to the redution of the concentration of major salts by half in the NLN medium varied with the different genotypes. High embryogenic broccoli cultivars, such as ‘Shogun’, ‘SDB9’, and ‘Green Valiant’, presented a better response to the reduction of the concentration of major salts by half in NLN-13. Reduction never produced a detrimental effect on embryo yield and seems not to have any effect in the subsequent development of embryos in plants. Women     
In New Zealand while women dominate leadership positions in constitutional, political and business areas, gender inequality remains entrenched. Despite significant gains in rights (employment, pay equity, property), as a group women earn less and perform more family care and unpaid work than men. In part these disparities result from the economic restructuring of the last 15 years which saw a loss in unskilled manufacturing jobs, less social support and greater rewards to business and public sector elites. Maori and Pacific Island women are the most disadvantaged groups. Labour/Alliance government initiatives to reduce poverty particularly among Maori and Pacific Island peoples by strengthening access to health and education services and supporting and replicating successful community initiatives are also likely to benefit other low income women.  相似文献

To overcome the cross-breeding barriers between the cultivated tomato Lycopersicon esculentum and the ‘peruvianum-complex’, hybrid production ability (HPA) and pistillate-parental differences were investigated. As a criterion of HPA, the number of germinated ovules per fruit (GPF) was used. GPF was expressed as GPF = OPF × GPO, where OPF is the number of ovules per fruit, and GPO is the proportion of germinated ovules to total ovules obtained. The interspecific crossing between nine varieties and three ‘peruvianum-complex’ accessions revealed that the cultivars ‘Sekaiichi’,‘Ponde Rooza’ and ‘Early Pink’ showed quite high and stable GPF over the years, but the cv.‘Best of All’ produced no hybrids. Variance analysis for GPF, OPF and GPO, and their correlation with seven sexual organ morphological traits and three fruit morphological traits were performed. These results indicated that choosing both the pistillate parents with wider reproductive organs for high OPF and appropriate environmental conditions for high GPO might be significant for enhancing GPF in interspecific crossing.  相似文献

Production of haploids, followed by chromosome doubling to produce doubled haploids (DH) represents the most rapid means of achieving complete inbreeding. In order to improve the androgenetic responses and maximize the production of green regenerants from selected plants of twenty-five triticale (× Triticosecale, Wittmack) populations (BC1F1, TC1F1, and F2) we used a uniform and optimal growth environment for anther donor plants within a greenhouse hydroponic system. Non-orthogonal analysis of deviance showed highly significant differences (p < 0.001) among populations for both induction and regeneration. The overall induction response of the populations was very high with a mean of 50.4 embryoids per 100 anthers plated. Among all tested-populations, M86-6068/TW179//EP80 (TC1F1) was the most responsive for both induction and regeneration which could be associated to its Triticum timopheevii cytoplasm. On the other hand, although populations 80465/II83-194 both as BC1F1 and F2 had a high level of induction response, only a few green plants were regenerated. These populations probably possess a partial Secale montanum genome, which could be contributing to the low regeneration ability. In conclusion, the need for optimization of donor plant growth conditions to effectively assess the androgenetic ability of individual populations/lines would be emphasized. 对5个小麦不同组合基因型的F1,F2,F3花药进行培养,结果表明:不同组合基因型在同一杂种世代的花药愈伤组织诱导率、愈伤组织鲜重和绿苗分化率等方面存在较大差异,而同一组合基因型在不同杂种世代的花药愈伤组织诱导率、愈伤组织鲜重和绿苗分化率等方面差异较小。花药愈伤组织诱导率与绿苗分化率具有显著的相关性(r2=0.903)。以石4185为亲本配制的两个组合基因型,在F1,F2,F3都表现出较高的花药愈伤组织诱导率和绿苗分化率,说明石4185是理想的花培育种桥梁亲本。  相似文献

Microspore populations of eight Fhybrids of winter wheat (Triticum aestivum L.) whose parents had different levels of resistance to Fusarium were screened in vitro, using phytotoxins of Fusarium as biochemical probe. Two selection methods were compared for the in vitro selection: either embryoids and calli were first initiated from anthers in toxin-free medium and then grown on medium with 0.3—0.9 %Fusarium toxin; or anthers were immediately cultured in modified liquid potato-2 medium in the presence of 1.5, 2.0 and 2.5ml Fusarium toxin per liter culture medium, a concentration which reduced the number of calli and embryoids to about 10 % compared to the toxin-free controls. Microspores from donor hybrids which were produced from very susceptible cultivars were killed by lower toxin concentrations than micro-spores from hybrids of less susceptible parents. From surviving calli and embryoids, originally initiated from 242,000 anthers in both procedures a total of 375 green lines could be regenerated. The results indicate that it is possible to enrich the fraction of regenerating microspores by those which contain the gene complex responsible for reduced susceptibility to Fusarium by the use of a pathotoxin.  相似文献

Summary A combination of compatible second pollinations and embryo rescue was applied for systematic production of true tetraploid hybrids from crosses between disomic tetraploid Solanum acaule and tetrasomic tetraploid potato, S. tuberosum. Several genotypes of tetraploid potatoes were pollinated with S. acaule, and the compatible second pollinations were made on the following day, with a genotype of S. phureja, IvP 35 to promote fruit development. Embryo rescue was carried out in 21 families, 14 to 27 days after the first pollination. A total of eight plants were obtained from the embryo rescue and their chromosome numbers were counted in the root tips. Three of the eight plants were identified as tetraploid, and five others as diploid. Morphology, isozyme banding patterns, and pollen stainability, as well as potato spindle tuber viroid (PSTVd) resistance, indicated the hybrid nature of the three plants. This is the first report of successful tetraploid hybrid production between disomic tetraploid S. acaule (4x) and tetrasomic tetraploid potatoes. Seed set from the crosses between one of hybrids and diploid potatoes indicated workable levels of both male and female fertility for introgression of valuable genes from S. acaule into the cultivated potato gene pool. The methodology used may be applied to other disomic tetraploid tuber-bearing Solanum species and with some modifications also to distantly related solanaceous species and genera.  相似文献
