【目的】本研究基于试验测定的中国荷斯坦牛血液和乳汁中的孕酮浓度表型,探究了影响中国荷斯坦牛体内孕酮分泌的环境因素、孕酮浓度的变化趋势、孕酮浓度与乳成分的关联程度以及血乳中孕酮浓度的预测方法。【方法】试验于2021年8月在北京、山东两个牧场采集不同胎次、泌乳阶段和妊娠状态的中国荷斯坦泌乳牛的奶样、尾根血样,测定孕酮浓度,最终获得402条乳汁孕酮浓度和298条血液孕酮浓度表型用于数据分析。对孕酮浓度进行数据转化使其近似服从正态分布后,采用固定效应模型探究胎次、泌乳阶段、妊娠状态、牧场等固定效应对奶牛孕酮表型的影响,运用R语言cor函数计算孕酮与各乳成分间的关联,并利用偏最小二乘法和个体及乳成分信息对孕酮浓度进行预测,以建立孕酮浓度表型高通量获取手段。【结果】妊娠状态对转化乳汁孕酮浓度存在极显著影响(P<0.01),胎次、场效应对转化乳汁孕酮浓度均有显著影响(P<0.05),而转化血液孕酮浓度只受到妊娠状态影响(P<0.01);全乳固体、乳脂率、乳蛋白率、脂蛋比与转化乳血孕酮浓度均存在显著或极显著的正相关关系(r=0.14~0.37,P<0.05;P<0.01);基于本试验数据,乳成分与个体信息对转化乳血孕酮浓度的预测准确性不高(R2=0.030~0.17),但如果增加血液或乳汁的转化孕酮浓度对乳汁或血液的转化孕酮浓度进行预测,预测效果则有大幅提升(R2=0.40)。【结论】影响泌乳期中国荷斯坦牛转化孕酮浓度的因素除妊娠状态外,可能还包括饲养条件与胎次。此外转化孕酮浓度与乳脂率、乳蛋白率等乳成分呈极显著相关。基于乳成分信息与转化孕酮的关系,获得了对中国荷斯坦牛乳血转化孕酮浓度预测的可用策略,为今后牧场的繁殖辅助管理、奶牛育种新性状研发以及孕酮浓度的高通量获取等提供了新思路。 相似文献

Abstract A study to investigate the effect of lime on dry matter yield of maize (Zea mays) and beans (Phaseolus vulgaris) and nodulation of beans grown in three tropical acid soils (two humic Nitosols and one humic Andosol) was carried out in a greenhouse. The soils ranged from 4.2 to 5.0 in pH; 1.74 to 4.56 in %C; 21.0 to 32.0 meq/100g in CEC; 5.10 to 8.10 meq/100g in exchange acidity; 0.60 to 3.20 meq/100g in exchangeable (exch.) Al and 0.13 to 0.67 meq/ 100g in exch. Mn. Exchange acidity and exch. Al decreased with increasing levels of lime in the three soils. Exchangeable Al was reduced to virtually zero at pH 5.5 even in the soils which had appreciable initial amounts. Exchangeable Mn also decreased with increasing levels of lime in the two Nitolsos. Exceptional results, however, were obtained with the Andosol where exch. Mn increased ten‐fold with the first level of lime and then decreased with subsequent levels. In all the soils, mean dry matter yield of beans and maize, and mean nodule dry weight of beans generally increased significantly with increasing lime levels up to pH value of 6.0. The dry matter yield of beans and maize, and nodule weight of beans, however, decreased progressively with increasing lime levels beyond pH 6.0 value. pH range of 5.5 to 6.0 was considered optimum for the growth of maize and beans, and nodulation of beans in these soils. 相似文献

本试验旨在研究饲粮能量和蛋白质水平对滩羊小肠中小肽和氨基酸转运载体mRNA表达量的影响。选取112只健康、体重相近的滩羊,随机分成4组,每组4个重复,每个重复7只羊。标准水平的饲粮能量和蛋白质水平参考《肉羊饲养标准》(NY/T 816—2004),各组试验滩羊分别饲喂不同能量和蛋白质水平饲粮:0.84×标准水平(Ⅰ组)、0.96×标准水平(Ⅱ组)、1.08×标准水平(Ⅲ组)和1.20×标准水平(Ⅳ组)。试验根据羊体重分2个阶段:29~35 kg和36~40 kg。于每个阶段末,每个重复屠宰1只试验羊,取其小肠组织样,运用实时荧光定量PCR技术,研究小肽转运载体1(Pep T1)、y+型氨基酸转运载体1(CAT1)、兴奋性氨基酸转运载体3(EAAT3)mRNA表达量的变化。结果表明:1)在29~35 kg阶段末,小肠中Pep T1 mRNA的表达量随着饲粮能量和蛋白质水平的提高呈先下降再上升的趋势,Ⅱ组显著低于其他3组(P0.05);Ⅳ组小肠中CAT1 mRNA的表达量显著高于其他3组(P0.05);Ⅲ组小肠中EAAT3mRNA的表达量显著高于其他3组(P0.05)。2)在36~40 kg阶段末,Ⅱ组小肠中Pep T1mRNA的表达量显著高于其他3组(P0.05);Ⅱ组小肠中CAT1 mRNA的表达量显著高于Ⅲ组(P0.05);小肠中EAAT3 mRNA的表达量随着饲粮能量和蛋白质水平的提高呈上升趋势,Ⅲ组和Ⅳ组小肠中EAAT3 mRNA的表达量显著高于Ⅰ组和Ⅱ组(P0.05)。由此可见,饲粮能量和蛋白质水平会影响滩羊小肠中Pep T1、CAT1、EAAT3 mRNA的表达量,使机体对小肽和氨基酸的吸收利用率随之改变,以适应滩羊的生长发育。 相似文献

AIM: To investigate the relationship between the expression of estrogen receptor α36 (ER-α36) and the invasion of gastric cancer cells. METHODS: SGC7901 cells were treated with 17β-estradiol at high or low concentration. The invasion of gastric cancer cells and the expression of ER-α36 were detected. The SGC7901 cells with the characteristics of stable low expression and high expression of ER-α36 were constructed. The invasion ability and microRNA sequences were determined in above-mentioned recombinant cells. RESULTS: The decreased invasion ability and ER-α36 expression were detected in SGC7901 cells treated with low concentration of 17β-estradiol. The situation was the opposite in the cells treated with high concentration of 17β-estradiol. The expression of miR-143 was significantly decreased in the SGC7901 cells with stable high expression of ER-α36 and was increased in the SGC7901 cells with stable low expression of ER-α36. CONCLUSION: The expression of ER-α36 is positively related to the invasion of gastric cancer cells. It is possible that miR-143 plays an important role in the regulation of gastric cancer invasion. 相似文献

Astaxanthin, a powerful antioxidant, is a good candidate for the prevention of intracellular oxidative stress. The aim of the study was to compare the antioxidant activity of astaxanthin present in two natural extracts from Haematococcus pluvialis, a microalgae strain, with that of synthetic astaxanthin. Natural extracts were obtained either by solvent or supercritical extraction methods. UV, HPLC-DAD and (HPLC-(atmospheric pressure chemical ionization (APCI)+)/ion trap-MS) characterizations of both natural extracts showed similar compositions of carotenoids, but different percentages in free astaxanthin and its ester derivatives. The Trolox equivalent antioxidant capacity (TEAC) assay showed that natural extracts containing esters displayed stronger antioxidant activities than free astaxanthin. Their antioxidant capacities to inhibit intracellular oxidative stress were then evaluated on HUVEC cells. The intracellular antioxidant activity in natural extracts was approximately 90-times higher than synthetic astaxanthin (5 µM). No modification, neither in the morphology nor in the viability, of vascular human cells was observed by in vitro biocompatibility study up to 10 µM astaxanthin concentrations. Therefore, these results revealed the therapeutic potential of the natural extracts in vascular human cell protection against oxidative stress without toxicity, which could be exploited in prevention and/or treatment of cardiovascular diseases. 相似文献

AIM: To explore the role of nucleotide-binding oligomerization domain-like receptor protein 1 (NLRP1) inflammasome in atorvastatin-induced reduction of interleukin-1β (IL-1β) and interleukin-18 (IL-18) releases from the THP-1 macrophages. METHODS: Lipopolysaccharide (LPS, 10 μg/L) was used to trigger the secretion of IL-1β and IL-18 in the THP-1 macrophages. The cells were incubated with different concentrations of atorvastatin (1, 10 and 20 μmol/L) for 24 h, or treated with 10 μmol/L atorvastatin for different time (12 h, 24 h and 48 h). NLRP1 siRNA was transfected into the THP-1 cells. The mRNA expression of NLRP1 inflammasome was detected by RT-PCR. The protein expression of NLRP1 inflammasome was determined by Western blot. The secretion of proinflammatory cytokines IL-1β and IL-18 was quantified by ELISA. RESULTS: Atorvastatin inhibited the mRNA and protein expression of NLRP1 inflammasome in the THP-1 macrophages in a dose- and time-dependent manner. Transfection of NLRP1 siRNA significantly decreased the protein expression of NLRP1 and promoted the suppressive effect of atorvastatin on IL-1β and IL-18 secretion in the THP-1 macrophages. CONCLUSION: Atorvastatin inhibits the production of IL-1β and IL-18 in the macrophages through decreasing NLRP1 inflammasome expression, possibly contributing to the anti-inflammatory effect of atorvastatin on atherosclerosis. 相似文献
