To improve the abiotic stress tolerance of maize (Zea mays L.), doubled haploid (DH) plants were produced by in vitro selection of microspores exposed to tert‐butyl hydroperoxide (t‐BuOOH) as a powerful prooxidant This study investigated the tolerance of the progenies of t‐BuOOH‐selected DH lines to oxidative stress, cold and drought in controlled environment pot experiments by analyses of photosynthetic electron transport and CO2 assimilation processes, chlorophyll bleaching and lipid peroxidation of leaves. Our results demonstrated that the t‐BuOOH‐selected DH plants exhibited enhanced tolerance not only to oxidative stress‐induced by t‐BuOOH but also to cold and drought stresses. In addition, they showed elevated activities of antioxidant enzymes such as superoxide dismutase, ascorbate peroxidase, catalase, glutathione reductase and glutathione S‐transferase when compared with the DH lines derived from microspores that were not exposed to t‐BuOOH and to the original hybrid plants. The results suggest that the simultaneous up‐regulation of several antioxidant enzymes may contribute to the oxidative and cold stress tolerance of the t‐BuOOH‐selected DH lines, and that the in vitro microspore selection represents a potential way to improve abiotic stress tolerance in maize. 相似文献

华北平原不同种植制度对粮食作物氮素利用和土壤氮库的影响 总被引:3,自引:2,他引:1
冬小麦-夏玉米一年两熟是华北平原粮食作物的主要种植制度,存在氮肥利用率低、土壤氮素过量累积问题。为探索华北平原氮素高效利用的适宜种植制度,采用15N示踪技术,基于3 a田间定位试验,对一年两熟冬小麦-夏玉米的常规水氮和优化水氮、两年三熟冬小麦-夏玉米-春玉米与冬小麦-夏大豆-春玉米及一年一熟春玉米3种种植制度的作物产量、15N利用率、氮素去向和土壤氮库表观平衡进行研究。结果表明,两年三熟的冬小麦-夏玉米-春玉米产量为32 248.52 kg/hm2,分别比一年两熟和一年一熟提高22.16%和52.88%;15N利用率为33.36%,比一年一熟提高26.12%。3种植制度的氮肥去向最高为土壤残留,其次为作物吸收和损失,两年三熟冬小麦-夏玉米-春玉米的作物吸氮量最高为151.82 kg/hm2,土壤氮库表观盈余量为21.22 kg/hm2,显著低于其他种植制度。综合分析,冬小麦-夏玉米-春玉米两年三熟在稳产高产和提高氮素利用率上具有可持续的潜力,是华北平原未来较为理想的种植制度。 相似文献

AIM: To explore the neuroprotective effect of novel Rho kinase inhibitor FSD-C10 on Alzheimer disease (AD) model of APP/PS1 double transgenic (Tg) mice. METHODS: Male APP/PS1 Tg mice (n=20) at 8 months of age were randomly divided into 2 groups:model group and FSD-C10 treatment group. The mice were treated with normal saline or FSD-C10 (25 mg·kg-1·d-1) by intraperitoneal injection, once daily for 2 months. Age-and sex-matched wild-type (WT) mice without treatment were used as the control. The Morris water maze (MWM) test and SMART 3.0 behavioral record system were applied to examine and analyze the spatial cognitive function of the mice. The protein levels and distribution of Aβ, p-tau and synapse-associated proteins such as synaptophysin, α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptor (AMPAR) and postsynaptic density protein 95 (PSD-95) were determined by immunofluorescence staining. The protein levels of phosphorylated amyloid precursor protein at Thr668[p-APP(Thr668)], beta-site APP-cleaving enzyme 1 (BACE1) and synapse-associated proteins in the brain were analyzed by Western blot. RESULTS: Compared with model group, FSD-C10 treatment significantly improved the cognitive function of the APP/PS1 Tg mice, accompanied by reduced Aβ deposition and p-tau level, increased protein level of p-APP (Thr668) in the central nervous system, decreased expression of BACE1, and increased expression of synapse-associated proteins in the brain of the mice (P<0.05). CONCLUSION: FSD-C10 has neuroprotective potential in the APP/PS1 Tg mice. The mechanism may be related to enhancing the non-amyloidogenic APP cleavage pathway, reducing the production of Aβ oligomers, the deposition of senile plaques and the amount of tau protein, up-regulating synapse-associated proteins, and increasing synaptic plasticity. 相似文献

马铃薯与玉米复合种植对土壤化感物质及土壤细菌群落结构的影响 总被引:1,自引:0,他引:1
为探究马铃薯与玉米复合种植对化感物质积累与细菌群落结构的影响,分析轮作、间作缓解连作障碍的机制,本研究以马铃薯连作、玉米连作、马铃薯||玉米间作、马铃薯-玉米轮作第8年的土壤为对象,利用GC-MS测定土壤中化感物质含量,并采用Illumina Miseq高通量测序技术对土壤细菌16Sr DNA V4-V5区域进行测序,分析土壤中细菌多样性和群落结构的变化,并对化感物质和优势菌属进行相关性分析。结果表明:玉米连作和马铃薯连作会导致化感物质的积累,玉米连作土壤积累了更多的油酸、亚油酸、花生酸、木焦油酸等脂肪酸,马铃薯连作土壤积累了更多的硬脂醇、二十烷醇等脂肪醇类物质。轮作降低了大部分化感物质的积累,间作降低的化感物质种类相对轮作较少。不同种植方式下土壤细菌群落结构发生了显著变化,相对于连作,间作和轮作Ace指数和Chao指数显著升高。在门水平上,轮作土壤放线菌丰度显著高于马铃薯连作土壤,间作土壤拟杆菌门丰度显著低于玉米连作土壤,两种连作土壤中酸杆菌门丰度都较轮作显著升高。在属水平上,一些有益细菌如节杆菌属、溶杆菌属等在复合种植土壤中相对丰度更高。通过相关性分析发现土微菌属、小梨形菌属与脂肪醇类物质呈显著正相关,黄杆菌属、溶杆菌属、微杆菌属等与脂肪酸类物质呈显著负相关。马铃薯与玉米复合种植降低了化感物质在土壤中的积累,从而抑制了土壤细菌丰度的降低,提高了有益菌属丰度,消减了连作障碍。 相似文献

海南水稻生育期的时空变化特征及对气候变暖的响应 总被引:3,自引:0,他引:3
基于海南6个农业气象观测站点的水稻生育期资料和1961~2014年历史气象数据,分析了海南不同地区水稻生育期的时空分布特征,以及气候变暖对海南水稻生育期的影响。结果表明,近54 a海南水稻生长季内平均气温呈上升趋势,早稻气候倾向率为0.21~0.3℃/10 a,以苗期最为明显,晚稻为0.18~0.24℃/10 a,以成熟期最为明显。不同地区水稻生育期差异较大,东部地区早于西部地区,早稻播种时间最大相差55 d,早稻收获和晚稻播种最大间隔42 d,降水和高温可能是造成这一差异的主要原因。随着气候变暖,海南大部分地区早稻生育期提前,晚稻生育期延迟,苗期和成熟期持续时间缩短,与该阶段平均气温呈负相关,但全生育期持续时间变异较大,部分站点略有延长。在未来气候变暖背景下,海南早稻播期可适当提前,晚稻播期可适当延迟,并选用耐高温和抗干旱品种,以减轻高温的不利影响。 相似文献

We investigated the alleviative effects of mixed cropping using ice plant, which is one of the salt-accumulating halophytes, on the damage and growth inhibition of cowpea, which is not tolerant to high salinity. Three cropping patterns (mono cropping of cowpea and ice plant and their combination) were tested. The plants were treated with 0, 100, 200 and 300 mM NaCl for 14 days (consecutive NaCl). The plants were also treated with NaCl for 3 days, followed by 2 weeks (short-term recovery) and 1 month (long-term recovery) recovery. Salinity levels for short-term recovery were similar to those of the consecutive experiment, while the concentration of long-term recovery was 250 mM. The alleviative effects of mixed cropping in the consecutive NaCl experiment were observed at 200 and 300 mM NaCl. Mixed cropping significantly reduced the Na content in the cowpea leaves at 200 and 300 mM NaCl compared with mono cropping. In addition, the Na content in the soil of mix-cropped cowpea at 200 and 300 mM NaCl was statistically lower than that of the mono cropping. Mixed cropping was effective to recover from high concentration of NaCl in the experiments of short- and long-term recovery. These results indicate that mixed cropping with a halophyte could be effective in mitigating the damage and growth inhibition of a glycophyte not only under salinity but also under recovery periods. 相似文献

玉米单倍体育种技术加倍率低的问题是单倍体育种技术应用限制条件。本研究以雄穗低自然加倍材料DHL287为母本、雄穗高自然加倍材料吉Gjb335DH3为父本,构建单倍体雄穗高自然加倍群体。根据集群分离分析法(BSA,Bulked segregant analysis),结合SNP标记,使用滑动窗口分析3组BSA混池基因型数据,最终在1、3、4、5、7号染色体上筛选出23个与雄穗高自然加倍相关的QTL位点。其中,3组BSA混池结果都在1、3、4号染色体上定位到4个共有的显著QTL区段,说明4个QTL区段具有很强的重演性。同时,在5、7号染色体上分别定位到3个、2个与单倍体雄穗高自然加倍相关的QTL位点。 相似文献
