Abstract

For most markets for wood, it is used without any treatments or modifications. When wood is used in adverse environments, it may be treated with chemicals to help prevent decay, improve water resistance, reduce the effects of ultraviolet radiation or increase fire retardancy. Many of these treatments involve the use of toxic or corrosive chemicals that can harm the environment. Chemical modification of wood provides an alternative by providing protection against water, decay, UV and thermal degradations by bonding chemicals to the cell wall polymers that do not leach out. Dimensional stability and decay resistance are two major properties that can be greatly improved by simple reactions with acetic anhydride.  相似文献

为了解决现有孢子检测系统体积大、成本高等问题,提出了一种基于衍射重构技术的作物真菌病害孢子检测方法。根据惠更斯-菲涅尔原理、角谱理论,利用衍射成像复合重构计算设计了一种包含富集、进样机构的真菌病害孢子检测系统。该系统可以定时完成富集、进样、拍摄、重构和检测等操作,并通过重构算法实现对真菌病害孢子原像的重建,根据重构后的图像提取面积(Area)、细度(Thinness ratio)两个重要形态学特征,对稻瘟病孢子进行检测识别。实验结果表明,所设计装置对稻瘟病孢子的检测结果与人工显微镜识别结果高度线性相关,决定系数为0. 99,平均检测误差为5. 91%,具有较好的准确性。  相似文献

Amino sugars, as a kind of microbial residue, are strongly associated with cycling of microbial-derived soil organic matter. However, responses of amino sugars to agricultural practices on the Loess Plateau in North-western China are poorly known. The objective was to evaluate effects of film mulching (no film mulching + NPK fertilizers, CK; film mulching + NPK fertilizers, PF; film mulching + NPK fertilizers + cow manure, FM) on accumulations of amino sugars in this region. FM significantly increased total amino sugar by 190.46 mg kg?1 in 0–10 cm layer and 214.66 mg kg?1 in 10–20 cm layer relative to CK, but PF significantly decreased it by 139.28 mg kg?1 in 0–10 cm layer. Ratios of glucosamine to muramic acid were markedly decreased by 2.50 in 0–10 cm layer and 2.28 in 10–20 cm layer in FM than CK, suggesting a tendency of microbial residues pool shift towards bacterial residues in this agroecosystem. These results indicated film mulching alone was not benefitial to accumulation of amino sugar while organic manure contributed to the build-up of amino sugar partly due to manure contained microbial residues. The different patterns of amino sugars suggested significant changes in the quality of microbial-derived organic matter.  相似文献

不同林型红松林土壤真菌群落组成和多样性   总被引:1,自引:1,他引:0  
采用基于ITS(Internal Transcribed Spacer)序列的Illumina MiSeq高通量测序技术,分析凉水国家级自然保护区的云冷杉红松林、椴树红松林、红松人工林和红松天然次生林四种林型的土壤真菌群落组成和多样性。共获得1 316个OTUs,四种林型的土壤含有大量未知真菌,在已知真菌类群中,子囊菌门(Ascomycota)和担子菌门(Basidiomycota)为优势菌门,其中后者在四种林型土壤中的分布差异显著(P0.05)。α多样性分析结果表明:原始红松林(云冷杉红松林和椴树红松林)真菌群落丰富度指数(Ace和Chao1指数)显著高于红松天然次生林(P0.05)。β多样性分析显示:四种林型土壤真菌群落组成差异显著(R=0.537,P=0.001),但两种原始红松林之间、人工林和天然次生林之间组成无显著差异。土壤全氮、总有机碳、pH、水分含量和总孔隙度是显著影响四种林型土壤真菌群落组成的环境因子(P0.05)。研究结果为原始红松林的保护、人工植被恢复和真菌资源的开发利用提供依据。  相似文献

The epidemic outbreak in northern Europe of Neonectria neomacrospora, the causal agent of dieback in Abies spp., led the European and Mediterranean Plant Protection Organization (EPPO) to include the pathogen on its alert list in 2017. Effective monitoring of this pathogen calls for a rapid and sensitive method of identification and quantification. A probe‐based real‐time PCR (qPCR) assay based on the β‐tubulin gene was developed for the detection and quantification of N. neomacrospora in infected wood samples, and directly for ascospores. This study presents the first published species–specific molecular detection assay for N. neomacrospora. The analytical specificity was validated on taxonomically closely related fungal species as well as on 18 fungal species associated with the host (Abies sp.). The analytical sensitivity was tested on naturally infected wood, on purified pathogen DNA in a matrix of host DNA and on N. neomacrospora ascospores for detection of airborne inoculum. The latter was tested both with a DNA extraction step prior to qPCR and without DNA extraction by direct qPCR on collected ascospores. The assay was specific to N. neomacrospora, with a sensitivity of 130 fg purified DNA, or 10 ascospores by direct qPCR. Omitting DNA extraction and amplifying directly on unpurified ascospores improved assay sensitivity significantly.  相似文献

The use of wrapped forage bales with high dry‐matter (DM) content implies risk of fungal growth inside the wrapping, and impaired feed quality. Since fungi may be unevenly distributed in bales, the method of sampling can influence the outcome of the analysis. Three common sampling methods for detection of fungi in wrapped forage were compared: direct plating of visible mouldy patches on bale surfaces (Method I), direct plating of pieces of foliage from drilled core samples (Method II) and quantitative analysis dilution series of drilled core samples (Method III). All samples were cultured on two media at two temperatures. Samples were collected from 124 farms in Sweden and Norway. Using Methods I, II and III, fungi were detected on 52%, 77% and 56% of the farms respectively. Fifty‐two fungal species were identified using a combination of culturing and molecular methods. The most predominant species were Arthrinium spp., found on 55% of the farms, followed by Penicillium roqueforti at 48% of the farms. Incubation at 25°C resulted in higher numbers of all genera, except Aspergillus, compared to incubation at 37°C. The different methods of sampling and culturing did not identify the same fungal species within the same bale. Analysing one bale per farm resulted in detection of less than half the number of species compared to analysing three bales. Of the sampling and culturing methods compared, direct plating of drilled core samples cultivated at malt extract agar at 25°C performed best in terms of qualitative analysis of fungi species.  相似文献

[目的]研究黄土高原矿区重构土壤植被恢复对土壤理化性质、酶活性以及真菌群落的影响,准确评价植被恢复对该地区的土壤生态恢复效果。[方法]在内蒙古自治区准格尔旗选择3种典型植被恢复类型(灌木、乔木、草地)和未复垦地采集0—20 cm土壤样品,采用扩增子测序技术分析了土壤真菌多样性和群落组成,同时结合土壤理化性质探讨了不同植被恢复类型影响真菌群落多样性的重要因素。[结果]①与未复垦地相比,3种典型植被恢复类型不仅对土壤理化性质和酶活性均有明显提升,还提高了土壤真菌可操作分类单元(OUTs)和多样性指数,且差异显著,以灌木的效果最好。②真菌主要菌门相对丰度在各样地变化趋势为子囊菌门(Ascomycota)担子菌门(Basidiomycota)球囊菌门(Glomeromycota),相对丰度之和已超过50%,土壤真菌群落结构在4个样地之间保持了一定的稳定性。③土壤含水量为影响真菌群落结构的最主要因素。[结论]植被恢复影响土壤理化性质、酶活性及真菌群落多样性,黄土高原矿区生态恢复是一个漫长的过程,在生态恢复过程中,宜种植灌木来改善重构土壤的生态环境。  相似文献

选取黔中地区的贵阳市乌当区百宜乡(WD)、花溪区孟关(MG)和贵州省黔南龙里县龙架山森林公园(LL)3个地区的马尾松-杜鹃群落为研究对象,采集杜鹃根围土壤进行高通量测序,分析物种多样性,并通过数量生态学原理分析植物群落物种α多样性及土壤养分对真菌群落的影响.结果表明:从杜鹃的根围土壤中获得有效序列505 444条,隶属于9门23纲84目169科296属真菌(3 435 OTU),其中LL有真菌9门19纲53目90科127属(495 OTU)、 WD有8门21纲78目150科246属(1 076 OTU)、 MG有6门17纲55目99科129属(522 OTU).菌群α多样性分析显示, Chao1,ACE,Shannon指数均表现为WD大于MG和LL.菌群群落结构受生境植物多样性、土壤养分的综合作用,土壤有机质、 pH值以及速效磷、速效氮、全氮含量较大程度解释了真菌对土壤养分变化的响应.  相似文献
