Red grouper (Epinephelus morio) were collected from the west coast of Florida, the central area of fishery harvest in U.S. waters, by fishery-dependent sources during 2000–2005. The west Florida shelf was divided into two regions: north (capture locations ≥28°N latitude) and south (capture locations <28°N latitude). Significant differences were found for age, length, and size-at-age by region and by gear; red grouper from the north were significantly younger and smaller on average than those from the south. Regional differences were also noted with respect to age progression; year class trends were only detected in the north. The 1996 year class dominated the landings in 2000–2001 (ages 4 and 5) and the 1999 year class dominated in 2004–2005 (ages 5 and 6). Regional data were fit to a size-modified von Bertalanffy growth model indicating smaller asymptotic length (L) and faster growth rate (k) in the north (north: L = 800 mm, k = 0.23 mm year−1, t0 = 1.12; south: L = 863 mm, k = 0.15 mm year−1, t0 = 0.05). Mortality estimates derived from catch curves resulted in higher total and fishing mortality in the north for both gears. Alternative explanations of regional differences likely depend on nursery delineation and correlation to periodic environmental events such as red tides and hurricanes; all possibly important factors based upon anecdotal information. Nevertheless, our finding of regional demographic differences in red grouper from the west coast of Florida suggests a more complex population spatial structure for red grouper.  相似文献

Two new cell lines have been established from the muscle and swim bladder tissues of barramundi, Lates calcarifer, and designated as BM (barramundi muscle) and BSB (barramundi swimbladder), respectively. The cells multiplied well at 28 °C in Leibovitz’s L‐15 medium supplemented with 10% foetal bovine serum, and have been continuously subcultured more than 100 times to date. Morphologically, BM cells were mostly fibroblastic, whereas BSB were mostly epithelial. Both cell lines were susceptible to grouper iridovirus (GIV) and displayed characteristics of apoptosis after viral infection. The induction of apoptosis was further assayed in GIV‐infected BM and BSB cells by various methods. The inhibition of cell growth by GIV was demonstrated by MTT [3‐(4,5‐dimethylthiazol‐2‐yl)‐2,5‐diphenyltetrazolium bromide] assay. Morphological observations revealed typical apoptotic features in the infected cells, including cell shrinkage and rounding, chromosome condensation and formation of apoptotic body‐like vesicles. Chromosome fragmentation was detected by DNA laddering and TUNEL assays. Finally, the appearance of phosphotidylserine on the outer leaflet of apoptotic cell membranes was confirmed by annexin V staining. This is the first report of apoptosis induced by GIV in fish cells.  相似文献

It was recently reported that Poly(I:C) immunization with live nervous necrosis virus (NNV) confers protection in sevenband grouper, Epinephelus septemfasciatus (Thunberg), from NNV infection. In the present study, we conducted field tests with sevenband grouper for the evaluation of Poly(I:C) immunization efficacy. In the first experiment, sevenband grouper were immunized with NNV followed by Poly(I:C) administration 7 weeks before natural occurrence of viral nervous necrosis (VNN). Survival rate of the naïve fish was 71.0%, whereas that of the immunized fish was 99.8%. In the second experiment, sevenband grouper were immunized 10 months before VNN occurrence and survival rate of the non‐treated and vaccinated fish was 79.5% and 97.5%, respectively. In the third experiment, we administered Poly(I:C) to sevenband grouper at 20 days after natural occurrence of VNN. The survival rate of the non‐treated fish was 9.8%, whereas that of fish administered Poly(I:C) was 93.7%. Based on these results, it was concluded that Poly(I:C) immunization conferred protection in fish against NNV infection in field tests and the protection lasted more than 10 months. Furthermore, even after occurrence of VNN, fish mortality could be reduced by Poly(I:C) administration and there was an unexpected curative effect on VNN‐affected fish.  相似文献

Six diets were formulated to contain corn starch, tapioca starch, or dextrin at 10% and 20% inclusion levels and fed to humpback grouper fingerlings to apparent satiation for 10 weeks. Growth and feed utilization efficiency of humpback grouper were not affected by dietary carbohydrate source and level. Only slightly higher growth was observed in fish fed 20% dietary carbohydrate compared to the fish fed 10% carbohydrate irrespective of carbohydrate source. Body indices and whole body proximate composition of fish in the present study were generally independent of diet, except that muscle lipid of fish fed diets with 7.6% lipid and 20% carbohydrate was significantly lower than the other groups. In view of the lower price and local availability of tapioca starch compared to corn starch and dextrin, tapioca starch is the preferred source of starch in practical diets for humpback grouper.  相似文献

ABSTRACT

Acid-solubilized collagen (ASC) and pepsin-solubilized collagen (PSC) were extracted from the skin of giant groupers (Epinephelus lanceolatus) with yields of 39.51 and 19.12%, respectively. ASC and PSC consisted of two different α chains (α1 and α2) and were characterized to be type I collagen with no disulfide bond. The imino acid contents of the ASC and PSC from giant grouper skin were 189 and 181 per 1,000 residues, respectively. The maximum endothermic temperatures (Tmax) of ASC and PSC measured by differential scanning calorimetry (DSC) were 31.71 and 31.33°C, respectively. The denaturation temperatures of ASC and PSC measured by viscometry were 29.84 and 29.05°C, respectively. The maximum solubility in 0.5 M acetic acid was observed at pH 5 and pH 6 for ASC and PSC, respectively. A sharp decrease in solubility was observed for both ASC and PSC in the presence of NaCl above 3% (w/v).  相似文献

A major constraint in successful larviculture of groupers has been the small gape of the larvae and hence the requirement for small prey at first feeding. In this study, we examined how maintaining a phosphate concentration of 100 μg P L?1 and an inorganic nitrogen (N) level of 700 μg N L?1 via weekly fertilization with inorganic fertilizers affected phytoplankton, zooplankton and giant grouper larval survival in relation to a control group that was provided with rotifers immediately after larvae hatched. Unicellular algae, zooplankton within the size ranges of 10–50 μm and 50–100 μm and survival of giant grouper larvae were all significantly higher in the fertilized treatment compared with the control. Stomach analysis revealed that ciliates and flagellates were actively consumed by larval fish in the fertilized group, whereas few rotifers were consumed in the control. We conclude that the inorganic fertilization method provides high densities of suitable‐sized prey for larval groupers at the onset of exogenous feeding before they are able to consume larger, commercially available rotifers and copepods.  相似文献

Rock bream iridovirus (RBIV) causes huge losses, especially in rock bream Oplegnathus fasciatus. Rock bream injected with RBIV and held at 29, 26, 23 or 20 °C had 100% mortality. Conversely, all infected fish held at 17 °C survived even after the temperature was progressively increased to 26 °C at 100 dpi. Rock bream exposed to virus and held for 2, 4 and 7 days at 23/26 °C before the temperature was reduced to 17 °C had mortality rates of 26.6/73.2%, 66.6/100% and 93.4/100%, respectively, through 100 dpi. When surviving fish had the water temperature increased from 17 to 26 °C at 100 dpi, they did not exhibit signs of disease and had low virus copy numbers (below 103). To investigate the development of a protective immune, rock bream were infected with RBIV and held at 23 °C before shifting the water temperature to 17 °C at 4 dpi. All injected fish survived until 120 dpi. While 100% of the previously unexposed fish died, 80.2% of the previously infected fish survived. When the survivors were rechallenged again at 160 dpi, no further mortality occurred. The high survival rate of fish following rechallenge with RBIV indicates that protective immunity was established in the surviving rock bream.  相似文献

Trash fish/low valued fish is still the most commonly used feed in marine cage culture in Asia. Use of trash fish is controversial with regard to the sustainability of the farming practices, and related issues on environmental degradation. In this study, the results of farm based trials on Asian seabass (Lates calcarifer) and tiger grouper (Epinephelus fuscoguttatus) to evaluate the efficacies of the use of commercial pellet feed in comparison to trash fish/low valued fish in marine cages in Thailand are presented. In spite of the variations observed in each of the farm sites, the results indicate that the overall growth performances and fish survival rates between the two feed types were similar, for both species. However, better cost‐benefit and resource use were recorded for fish reared on trash fish/low valued fish. No significant differences were observed in water quality parameters, dissolved oxygen, transparency and ammonia, between sites where pellet or trash fish were used, inside and outside the cages and for both species. The results of the present study are discussed in the light of the current controversies on the use of trash fish/low valued fish in marine cage culture.  相似文献

Interspecies hybridization is widely used in aquaculture as a beneficial strategy. Diploid and triploid hybrids have been detected from the interspecies hybridization of Epinephelus coioides ♀ × Epinephelus lanceolatus ♂. This is the first report of triploidization through hybridization in grouper. Confirmation has been obtained through flow cytometry, karyotyping and erythrocyte nuclei measurement. The chromosome numbers of E. coioides, E. lanceolatus, diploid hybrid grouper are 48 and triploid hybrid grouper are 72. Measurements of erythrocyte nuclei indicate that triploid fish have a larger nuclear surface than the diploid groupers, and the average ratio of triploid to diploid surface area is 1.59. During the first 1.5 years, triploid hybrid groupers grow faster than diploid hybrid groupers or either parent species. The average growth rate of triploid hybrids is 1.61 times greater than that of diploid hybrids at 6 months of age and 1.43 times greater at 18 months of age. The triploid hybrid groupers are inferior in gonadal development, with no primary‐growth‐stage oocytes appearing in the gonads at 18 months of age. Morphological studies indicate that triploid hybrid groupers have distinctive differences in snout length, eye diameter, body trunk shape, and tail shape development compared with diploid hybrid groupers. Triploid hybrid groupers have an advantage in growth ability, and artificial breeding of triploid groupers might be of great potential use in the grouper aquaculture industry.  相似文献
