Abstract

A priming method called sand priming was developed using sand as a priming solid matrix. The effect of sand priming on improving the field emergence performance of five super sweet corn cultivars was investigated. Sand priming significantly improved field emergence performance of all super sweet corn cultivars, and there was marked improvement by priming at 20ºC for 24 hr. After sand priming at 20ºC for 24 hr, field emergence percentage (FEP) of “Green Superman”, “Huatian 1”, “Yangtian 1”, “Mitian 8”, and “Chaotian 43” was increased by 52.1%, 37.5%, 38.0%, 40.9%, and 33.3%, respectively. Their field emergence speed (FES) was 2.3, 1.8, 2.0, 2.0, and 1.8 times of the control, respectively. To further elucidate the effect of sand priming on improving the field emergence performance of super sweet corn, we analyzed the membrane system integrity, α-amylase activity and protein content. Sand priming at 20ºC for 24 hr improved membrane system integrity and α-amylase activity in all super sweet corn cultivars. Furthermore, sand priming at 20ºC for 24 hr accelerated the degradation of embryo protein after 1 d germination in “Green Superman”.  相似文献

通过对酸性α-醋酸萘酯酶染色过程中的几个重要因素进行研究,确定了其最佳处理条件,并探讨了几个因素间的相互作用。结果表明,固定液用2.5%戊二醛的效果要优于甲醛-丙酮混合液;用pH值5.8~6.2的孵育液孵育1.5h,甲基绿复染1~10s效果最佳。变化各因素的处理条件对最终的观测效果和结果都有影响,其中以甲基绿复染时间过长影响最大。同时,该染色法应用于贵妃鸡的T淋巴细胞计数中,4次重复的结果表明,贵妃鸡酸性α-醋酸萘酯酶阳性细胞率为37.86±1.89%,各重复间差异不显著。  相似文献

AIM To investigate the effects of cytochrome P450 (CYP450) epoxygenase/epoxyeicosatrienoic acid (EET) pathway on insulin resistance in obese mice, and to explore the possible mechanisms. METHODS High-fat diet-induced obesity model was established in C57BL/6Cnc mice, and the obese mice were randomly divided into 3 groups, including obesity group (treated with saline; n=10), EET group (treated with 11,12-EET; n=10) and EET inhibitor 14,15-epoxyeicosa-5(Z)-enoic acid (EEZE) group (n=10). Normal C57BL/6Cnc mice (n=10) treated with saline served as control. Protein expression of CYP2J2 (one of CYP450 epoxygenases) and hypoxia-inducible factor-1α (HIF-1α) was measured by Western blot. Vessel-like structure was detected by immunofluorescence staining. The serum levels of insulin, tumor necrosis factor-α (TNF-α), interleukin (IL)-1β, IL-6 and monocyte chemoattractant protein-1 (MCP-1) were measured by ELISA. RESULTS In obese mice, homeostasis model assessment of insulin resistance (HOMA-IR) values were increased, the protein level of CYP2J2 was reduced, and the protein level of HIF-1α was increased in adipose tissues as compared with the controls (P<0.05). The serum levels of MCP-1, IL-1β, IL-6 and TNF-α were also significantly increased in obese mice (P<0.05). After treatment with 11, 12-EET, the HOMA-IR values were decreased compared with vehicle-treated obese mice, HIF-1α expression levels were decreased in the adipose tissue, and the serum levels of MCP-1, IL-1β, IL-6 and TNF-α were reduced (P<0.05). Immunohistochemical results of adipose tissue from vehicle-treated obese mice showed a marked decrease in vessel-like structures (CD31-positive) compared with normal control mice (P<0.05). EET treatment significantly increased the newly formed vessel-like structures in the visceral adipose tissues of obese mice as compared with vehicle-treated obese mice (P<0.05). CONCLUSION High-fat diet-induced obesity and insulin resistance are closely related to the CYP450 pathway. Exogenous EETs effectively decrease obesity-induced insulin resistance possibly through pro-angiogenesis and attenuation of hypoxia and inflammation.  相似文献

东北地区小麦赤霉病镰孢菌种群及其致病性测定   总被引:1,自引:0,他引:1  
为了明确东北地区小麦赤霉病镰孢菌的种群组成及分布,分别从辽宁省沈阳市、黑龙江省哈尔滨市和密山市、内蒙古自治区扎兰屯市、呼伦贝尔市和通辽市等地区采集小麦赤霉病病样,经单孢分离纯化共得到118株镰孢菌,传统形态学鉴定的基础上,采用基因组DNA的EF-1α序列分析技术进行了镰孢菌种类的辅助鉴定,确定属于7个种:禾谷镰孢菌为优势种,分离频率为64.41%,藤仓镰孢菌为次优势种,分离频率为18.64%;燕麦镰孢菌、尖孢镰孢菌、木贼镰孢菌、锐顶镰孢菌和轮枝镰孢菌的分离频率分别为5.08%,3.39%,3.39%,3.39%,1.70%。同时,在玉米成株期进行了致病性测定,结果表明,小麦赤霉病镰孢菌可以侵染玉米,较玉米茎腐镰孢菌的致病力低。  相似文献

Three new water-soluble alkaloids (1-3) together with twelve known compounds (4-15) have been isolated from the water extract of leaves of Suregada glomerulata. Their structures were determined by spectroscopic analysis and chemical method. Compounds 1-3 were evaluated for their in vitro inhibitory activity against α-glucosidase and HIV-1 replication. Evapotranspiration (ET) can be derived from satellite data using surface energy balance principles. METRIC (Mapping EvapoTranspiration at high Resolution with Internalized Calibration) is one of the most widely used models available in the literature to estimate ET from satellite imagery. The Simplified Surface Energy Balance (SSEB) model is much easier and less expensive to implement. The main purpose of this research was to present an enhanced version of the Simplified Surface Energy Balance (SSEB) model and to evaluate its performance using the established METRIC model. In this study, SSEB and METRIC ET fractions were compared using 7 Landsat images acquired for south central Idaho during the 2003 growing season. The enhanced SSEB model compared well with the METRIC model output exhibiting an r2 improvement from 0.83 to 0.90 in less complex topography (elevation less than 2000 m) and with an improvement of r2 from 0.27 to 0.38 in more complex (mountain) areas with elevation greater than 2000 m. Independent evaluation showed that both models exhibited higher variation in complex topographic regions, although more with SSEB than with METRIC. The higher ET fraction variation in the complex mountainous regions highlighted the difficulty of capturing the radiation and heat transfer physics on steep slopes having variable aspect with the simple index model, and the need to conduct more research. However, the temporal consistency of the results suggests that the SSEB model can be used on a wide range of elevation (more successfully up 2000 m) to detect anomalies in space and time for water resources management and monitoring such as for drought early warning systems in data scarce regions. SSEB has a potential for operational agro-hydrologic applications to estimate ET with inputs of surface temperature, NDVI, DEM and reference ET.  相似文献

对3份野生二粒小麦(Triticum dicoccoides)材料的抗虫24 kDaα-淀粉酶抑制因子编码基因进行分离克隆,得到72个24 kDaα-淀粉酶抑制因子基因,并进行序列分析。结果发现,α-淀粉酶抑制因子在野生二粒小麦中以多拷贝形式存在,经卡方检验初步推断野生二粒小麦中的α-淀粉酶抑制因子成熟蛋白编码基因序列变异类型在供试材料间无显著差异。序列分析表明,24 kDaα-淀粉酶抑制因子成熟蛋白编码基因序列无论在野生二粒小麦种内还是与小麦属的其他物种之间,都具有很高的一致性,说明该基因在进化过程中十分保守。  相似文献
