The Paleocene-Eocene Thermal Maximum (PETM) has been attributed to a rapid rise in greenhouse gas levels. If so, warming should have occurred at all latitudes, although amplified toward the poles. Existing records reveal an increase in high-latitude sea surface temperatures (SSTs) (8 degrees to 10 degrees C) and in bottom water temperatures (4 degrees to 5 degrees C). To date, however, the character of the tropical SST response during this event remains unconstrained. Here we address this deficiency by using paired oxygen isotope and minor element (magnesium/calcium) ratios of planktonic foraminifera from a tropical Pacific core to estimate changes in SST. Using mixed-layer foraminifera, we found that the combined proxies imply a 4 degrees to 5 degrees C rise in Pacific SST during the PETM. These results would necessitate a rise in atmospheric pCO2 to levels three to four times as high as those estimated for the late Paleocene. 相似文献
The signals that direct membrane proteins to the apical or basolateral plasma membrane domains of polarized epithelial cells are not known. Several of the class of proteins anchored in the membrane by glycosyl-phosphatidylinositol (GPI) are expressed on the apical surface of such cells. However, it is not known whether the mechanism of membrane anchorage or the polypeptide sequence provides the sorting information. The conversion of the normally basolateral vesicular stomatitis virus glycoprotein (VSV G) to a GPI-anchored protein led to its apical expression. Conversely, replacement of the GPI anchor of placental alkaline phosphatase with the transmembrane and cytoplasmic domains of VSV G shifted its expression from the apical to the basolateral surface. Thus, the mechanism of membrane anchorage can determine the sorting of proteins to the apical or basolateral surface, and the GPI anchor itself may provide an apical transport signal. 相似文献
Papua New Guinea has astonishing biological and cultural diversity which, coupled with a strong community reliance on the land and its biota for subsistence, add complexity to monitoring and conservation and in particular, the demonstration of declines in wildlife populations. Many species of concern are long-lived which provides additional challenges for conservation. We provide, for the first time, concrete evidence of a substantive decline in populations of the pig-nosed turtle (Carettochelys insculpta); an important source of protein for local communities. Our study combined matched village and market surveys separated by 30 years, trends in nesting female size, and assessment of levels and efficacy of harvest, each of which was an essential ingredient to making a definitive assessment of population trends. Opportunities for an effective response by local communities to these declines needs to consider both conservation and fisheries perspectives because local communities consider the turtle a food resource, whereas the broader global community views it as a high priority for conservation. Our study in the Kikori region is representative of harvest regimes in most rivers within the range of the species in Papua New Guinea, and provides lessons for conservation of many other wildlife species subject to harvest. 相似文献
This trial was conducted to evaluate if the effect of condensed tannin (CT) is associated with a true protein source on intake, ruminal and total digestibility, ruminal digestion rate, protein efficiency, and microbial efficiency in beef steers fed high concentrate diet (87% of DM). Four Bos indicus steers (407 ± 12 kg of BW) fitted with rumen cannula were assigned to a 4 × 4 Latin square design, arranged in a 2 × 2 factorial arrangement. Treatments consisted of either inclusion (0.4% of DM) or exclusion of condensed tannin (CT) from quebracho extract (76% of CT) with or without the use of soybean meal (SBM) as source of true protein. The level of inclusion was calculated to provide a daily intake of 1 g/10 kg of BW of CT. Intake of DM and nutrients was not affected (P > 0.10) by CT inclusion. However, there was an effect (P < 0.10) of CT inclusion on ether extract digestibility. An interaction (P < 0.10) was observed between CT and SBM on ruminal digestibility and digestion rate of crude protein (CP): when mixed with soybean meal, CT decreased the ruminal digestibility and, consequently, reduced the digestion rate of CP. Intake of CP increased (P < 0.10) with the inclusion of SBM. No differences in DM passage rate were observed (P > 0.10) among treatments. Effects of the interaction (P < 0.10) between CT and SBM were observed on flux of rumen undegradable protein (RUP), metabolizable protein (MP), and on the ratio MP:CP. In the presence of soybean meal, the addition of CT increased (P < 0.10) the flux of RUP and MP, and improved the ratio MP:CP. The yield of microbial protein on the abomasum and the microbial efficiency did not differ among treatments (P > 0.10). There was no difference (P > 0.10) on the pH, VFA, and ruminal ammonia (N-NH3) with the addition of condensed tannin. The N-NH3 increased and the ruminal pH decreased with the inclusion of soybean meal (P < 0.10). The utilization of condensed tannin as an additive in beef cattle diets with high level of concentrate and soybean meal as a source of true protein implies positive effects on crude protein utilization, decreasing digestion rate and ruminal digestibility of crude protein and consequently increasing the levels of metabolizable protein, with no changes in the ruminal fermentation parameters. 相似文献
When continuous measurement of relevant above- and below-ground environmental factors is made, and the dynamics of crop growth and phenology is followed, it is shown that small-plot agronomic experimentation can provide information vital to the development and testing of simulation models. Such models for crops of maize (Zea mays) and the legume cowpea (Vigna unguiculata), grown as sole crops and in an intercrop system, were used to simulate yield at two separate sites in the main savanna farming zones in Ghana, West Africa. Simulation was carried out for these three cropping systems using 20 years of historical rainfall data for the two sites, investigating the effect on yield of management options which included time of planting, rate of nitrogen fertilizer application, and crop planting density.
The yield results of this series of simulations were then subject to two different types of analysis in order to assist the evaluation of agricultural options for the nation. The first analysis was an economic evaluation based on gross margins, but using the stochastic-dominance technique to rank the profitability of the alternative cropping and management systems.
The second, more general methodology employed was multi-criteria analysis (MCA). This analysis covered economic returns and their variability, the amount and quality of residue returned to the soil, and the aerial cover provided by the crop. Other factors which could affect the sustainability of production in the long term, but which could only be evaluated qualitatively, were also included in the analysis, as allowed in MCA methodology. The MCA was then carried out for three scenarios in which different weightings were given to economic outcomes and to factors which are believed to encourage ecological sustainability. These scenarios allow recognition of different priorities which might be given by traditional subsistence farmers as compared to commercial producers.
Outcomes of this broad analysis of development alternatives provided scientific support for the traditional Ghanaian practice of grain/legume intercropping which is also widespread in the tropics. The suite of methodologies illustrated by the case study appears to be suitable for the evaluation of alternative farming systems at the scale of a small nation such as Ghana. 相似文献
The primary structure of Brassica napus procruciferin 2/3a was engineered to elucidate structure-function relationships and to improve the functionality of cruciferin. The following mutants were constructed: (1) C287T, (2) DeltaII, variable region II was deleted; (3) C287T/DeltaII, mutation involving (1) and (2); (4) DeltaIV + A1aIV; and (5) DeltaIV + A3IV, variable region IV was replaced with variable region IV containing many charged residues from soybean glycinin A1aB1b and A3B4 subunits. Differential scanning calorimetry analysis revealed that the A1aIV region has a more favorable interaction with the procruciferin molecule than does A3IV as well as the original regions. On the basis of heat-induced precipitation analysis, it was concluded that replacement of the free cysteine residue with threonine (C287T) and insertion of charged regions (DeltaIV + A1aIV and DeltaIV + A3IV) could lead procruciferin to form soluble aggregates after heating. Low solubility was observed in mutants DeltaIV + A3IV, DeltaII, and C287T/DeltaII, especially between pH 4 and 6 at mu = 0.08, but not in DeltaIV + A1aIV, indicating that the number of acidic amino acid residues and the high number of glutamine residues are important factors for solubility at mu = 0.08. None of the mutants showed any improvements in emulsifying ability, indicating that destabilization and addition of the hydrophilic region are not effective for emulsification. The insertion of the A1aIV region in procruciferin made the molecule more susceptible to alpha-chymotrypsin. 相似文献
