Chickpea (Cicer arietinum L.) yields are drastically reduced by water and cold stress that occur individually or simultaneously in northern region of India. The comparative effects of both the stresses were investigated at the metabolic level by examining the endogenous status of polyamines (PAS), active oxygen species and antioxidants. Chickpea plants (15‐day old) growing hydroponically under controlled conditions (light/ dark; 24/21 °C, 16/8 h; irradiance 250 μmol m?2 s?1) were subjected to water deficit stress (Ψs of ?0.2 to ?1.0 MPa) and cold stress (5–25 °C) for 4 days. LD50 in terms of root growth rate (RGR), electrolyte leakage and triphenyl tetrazolium chloride (TTC) reduction activity was observed at ?0.6 MPa and 10 °C for water and cold stress, respectively. In a subsequent experiment, 15‐day‐old plants were exposed to these stress levels under the above‐mentioned growth conditions for 7 days and analysed for various parameters. In cold‐stressed plants (CS), putrescine (PUT) was observed to be relatively higher while water‐stressed plants (WS) had more of spermidine (SPD). Spermine (SPM) levels increased more rapidly in WS and declined on the fourth day of stress while in CS, a gradual increase occurred that decreased on the seventh day. The accumulation of PAs was short‐lived under the combined presence of both the stresses. Hydrogen peroxide elevated abruptly in WS and remained higher than CS while the latter showed a marked increase in malondialdehyde content. Ascorbic acid increased sharply in WS that decreased on the fourth day while CS showed a relatively gradual increase that reached its maximum on the fourth day and declined subsequently. Glutathione was significantly higher in CS plants in comparison with WS and CS + WS plants. The activity levels of superoxide dismutase were higher up to 4 days and declined subsequently while those of WS stayed higher till the last day of stress. Ascorbate peroxidase levels were significantly higher in CS plants while catalase activity was comparatively more in WS. Exogenous application of PAs reduced the level of hydrogen peroxide, malondialdehyde content and raised the level of antioxidants. Put caused 44 and 32 % increase in RGR in CS and WS, respectively, while SPD resulted in 110 and 25 % enhancement in WS and CS, respectively. Under combination of stresses, RGR increased by 21, 53 and 10 % by Put, SPD and SPM, respectively. The effects of PAs could be reversed largely by their biosynthetic inhibitors. α‐difluoromethylornithine (a biosynthetic inhibitor of putrescine) caused more damage to CS while cyclohexylamine (inhibitor of SPD and SPM biosynthesis) was more inhibitory in WS.  相似文献

A PCR based method was developed for the identification of ceftiofur resistance genes (blaCMY-2, blaTEM-1, and ampC) in swine bacterial pathogens. Using this method, the ceftiofur resistant (n = 76) and susceptible (n = 45) strains of Bordetella bronchiseptica, Salmonella spp., Escherichia coli, and Pasteurella multocida were screened for the presence of these three genes. The resistant genes were detected in 70% (blaTEM-1), 68% (blaCMY-2) and 45% (ampC) of the resistant isolates and in 18% (blaTEM-1), 27% (blaCMY-2), and 36% (ampC) of the susceptible isolates. Results obtained in the present study showed widespread distribution of these three resistance genes in ceftiofur-resistant swine pathogens. It was also observed that more pathogens are acquiring these resistance genes.  相似文献

The effective utilization of crop diversity held in genebanks depends on our knowledge of useful traits and available markers associated with the target traits. Target region amplification polymorphism (TRAP) was used to evaluate the genetic diversity and underlying relationships among 263 accessions of chickpea landraces maintained by the USDA-ARS Western Regional Plant Introduction Station in Pullman, WA, USA. Two-hundred sixty-two TRAP markers were amplified by eight primer combinations. Altogether, 110 (42 %) markers were polymorphic, the other 152 (58 %) displayed no variation. These polymorphic markers revealed important differences among the accessions, with an estimated, mean pair-wise genetic distance of 25.82 %, ranging from 2.8 to 50.0 %. Genetic distance analysis divided the accessions into two major groups, with 113 and 150 accessions each, and substantial association between molecular diversity and geographic origin was evident. Bayesian analysis of population structure revealed two groups (K = 2) with evidence for six sub-groups. Additionally, the population structure of a subset of 110 lines was determined (K = 3) for testing marker-trait associations (MTAs). Phenotypic traits included the concentrations of protein and nine mineral elements in the seeds. Two MTAs were significant (p < 0.01) for concentrations of Ca and K, and three MTAs were significant for Cu and Ni concentrations. The results indicate that this population is useful for genome-wide association studies on other economic traits given the level of genetic diversity uncovered and the marker-trait associations in seed minerals discovered.  相似文献

Widespread multinutrient deficiencies in the semi-arid tropics (SAT) are among major factors for large gaps between farmers’ current crop yields and potential yields. In this study, we adopted a stratified soil sampling method to assess soil fertility-related constraints in farmers’ fields in eight districts of Andhra Pradesh in the semi-arid tropics of India. Most of the fields across all eight districts were critical in sulfur (61%–98% deficient fields); and up to six districts each in boron (83%–98% deficient fields), zinc (50–85% deficient fields), and soil organic carbon (55–97% deficient fields). Low soil organic carbon specifically indicates nitrogen deficiency. Phosphorus deficiency was critical in three districts (60–84%) while potassium in general was adequate. Soil test-based nutrient balancing through the application of sulfur, boron, and zinc in addition to farmers’ practice of adding only nitrogen, phosphorus, and potassium increased crop productivity by 8%–102%. Benefit–cost ratio (1.60–28.5) proved favourable to scale-up balanced nutrition. Better post-harvest soil health and residual benefits of sulfur, boron, and zinc up to four succeeding seasons indicated sustainability of the practice. Results showed that balanced nutrition is a way forward for sustainably improving farm productivity and livelihoods.  相似文献

The relationships between the concentrations of zinc (Zn), lead (Pb), copper (Cu), nickel (Ni) and chromium (Cr) as measured by X-ray fluorescence analysis (XRF), aqua regia, and HNO3 pressure digestion were studied in soil samples covering a wide range of heavy metal concentrations. The soils were contaminated by sewage sludge, exhaust depositions, river sediments of mining residues, and dump material. The question was addressed whether the source of heavy metals or other soil properties affect the relationship between these three methods. The aqua regia-digestible fraction of the five heavy metals reached on average 64% of the XRF-detectable content. The pressure accelerated HNO3-digestible fraction of the five heavy metals was on average 71% of the XRF-detectable content; the respective phosphorus (P) fraction reached a median of 75%. This suggests that HNO3 pressure digestion can also be used for characterizing the total P content of soils. Aqua regia extraction and HNO3 pressure digestion gave similar values for Zn, Pb, and Cu, which dominate the heavy metal load of most contamination sources. Significantly higher Cr values were obtained by HNO3 pressure digestion than by aqua regia extraction. Additionally, the Cr contents were affected by the source, e.g. sewage sludge had relatively high contents of aqua regia and HNO3 pressure extractable contents in comparison to the XRF values. The element-specific relationships between the three methods were all highly significant. However, the respective multiple linear regression models were in most cases affected by soil organic carbon (C), in some cases by clay or soil pH.  相似文献

The oleo-gum resin of Commiphora wightii (Arn.) Bhandari, a pharmacologically important balsamiferous woody shrub, has been used in treating various ailments and disorders since ancient times (2000 B.C.) due to the presence of steroidal compound guggulsterone. Two bioactive isomers of guggulsterone, E and Z, are responsible for lipid- and cholesterol-lowering and anti-cancerous activities. Further, guggul has been approved as food supplement by US-FDA as well as Council of Europe. Indiscriminate harvest of C. wightii from wild with negligible conservation efforts has lead to its inclusion in IUCN assemblage of endangered plant species. For identification of high guggulsterone yielding ecotypes of C. wightii, using high-performance thin-layer chromatographic (HPTLC) analysis, stem samples were collected from 50 plants from eleven locations in arid tracts of Haryana, Gujarat and Rajasthan. Dried, powdered material was subjected to extraction with petroleum ether using soxhlet apparatus. Samples were spotted on precoated activated silica plates (60F-254) and were developed using toluene–acetone (9:1 v/v) as mobile phase. The analysis was carried out in the absorbance mode at 250 nm using HPTLC scanner. The regression analysis data for the calibration plots for E and Z guggulsterone showed good linear relationship with R2 = 1 and 0.9897, respectively. Highest concentration of guggulsterone E (284 μg/g dry wt) was found in the accession collected from Palana, Bikaner whereas highest guggulsterone Z concentration (89.5 μg/g dry wt) was found in the accession collected from CAZRI, Jodhpur.  相似文献
