The fracture properties of small, cylindrical samples of endosperm machined from single kernels of several varieties of wheat were measured using three methods: compression, wedge fracture and indentation. In addition, dynamic compression tests were also performed in impact loading. Density measurements using a variable density liquid gradient were carried out on machined endosperm samples that had previously been tested for fracture properties, enabling direct comparisons between their density and fracture properties. Within each variety, a distribution of density and hardness values was found. Soft wheat varieties (Riband, Apollo) showed a broad distribution of density with medians in the range 1340 to 1395 kg/m3, whilst hard varieties such as Mercia and a durum wheat exhibited much narrower distributions and higher mean densities, Mercia being skewed towards higher densities, with a median at around 1410 kg/m3. A considerable amount of overlap in density between the soft and hard variety occurred, with both containing a significant proportion of harder and higher density wheat grains, and the major difference appearing to be the presence in the soft wheat of a large proportion of lower density endosperm in the range 1280 to 1360 kg/m3. The differences in density between endosperm samples were attributed to variations in endosperm porosity, which were correlated with the fracture properties of the endosperm. It is proposed that increased levels of porosity weaken the endosperm structure and are responsible in part for soft endosperm texture. The relationship between endosperm density and fracture properties was non-linear, the failure stress and fracture toughness increasing rapidly as the density approached a limiting value corresponding to the density expected for pore-free endosperm. This indicates that endosperm is a notch sensitive material, where the pores concentrate applied stresses and can act as sites of crack initiation. It was shown that the notch sensitivity of endosperm is qualitatively consistent with models of brittle fracture based on fracture mechanics theory, i.e. stress concentration by voids. 相似文献

Nanoindentation is a powerful tool for hardness testing on a very small scale. Since the technique was first introduced for studying wood cell wall mechanics, it has been integrated as an important tool for measuring the modulus of elasticity and hardness of wood cell walls. In this study, hardness measured with nanoindentation (nanohardness) was compared with hardness measured by the standard Brinell test method (Brinell hardness) on jack pine (Pinus banksiana Lamb.) wood. Nanoindentation was performed on both the S2 layer of the secondary cell wall and the middle lamella (ML) of early- and latewood fibers. Four annual growth rings were studied. The influence of growth ring and initial spacing on both measurements was analyzed. The relationship between Brinell hardness, nanoindentation measurements, and average ring density was also studied. Results suggest that Brinell- and nanohardness are controlled by different mechanisms and factors. The location of nanohardness measurements (i.e., S2 layer or ML) also influenced hardness differently. It was concluded that nanomeasurements are not an exact representation of wood mechanical properties conducted at the macro level because of the hierarchical structure of wood. Sexual maturation and sperm release were induced in farmed European eels Anguilla anguilla kept exclusively in fresh water by using two dosages of human chorion gonadotropin (100 International Unit (IU)-Group one and 250 IU/individual per week-Group two). Sperm release took over 13 wk in both groups. The quality of sperm was investigated on the eighth, ninth, and tenth wk. The average cell densities were 0.94 ± 0.4 × 1010 (Group one) and 0.93 to. ± 0.6 × 1010 (Group two) spermatozoa/mL. The estimated motility of eel sperm was 33, 55, and 49% on the eighth, ninth, and tenth wk of treatment, respectively. The estimated average motility of samples selected for cryopreservation was 73 ± 10%, while the post-thaw motility of cryopreserved samples was 36 ± 11%. The extender originally developed for common carp sperm crypreservation together with methanol as cryoprotectant was found suitable for the cryopreservation of European eel sperm. 相似文献

Chemical signatures in the otoliths of teleost fishes represent natural tags that may reflect differences in the chemical and physical characteristics of an individuals' environment. Otolith chemistry of Atlantic bluefin tuna (Thunnus thynnus) was quantified to assess the feasibility of using these natural tags to discriminate juveniles (age 0 and age 1) from putative nurseries. A suite of six elements (Li, Mg, Ca, Mn, Sr and Ba) was measured in whole otoliths using solution‐based inductively coupled plasma mass spectrometry. Otolith chemistry of age‐1 T. thynnus collected from the two primary nurseries in the Mediterranean Sea and western Atlantic Ocean differed significantly, with a cross‐validated classification accuracy of 85%. Spatial and temporal variation in otolith chemistry was evaluated for age‐0 T. thynnus collected from three nurseries within the Mediterranean Sea: Alboran Sea (Spain), Ligurian Sea (northern Italy), and Tyrrhenian Sea (southern Italy). Distinct differences in otolith chemistry were detected among Mediterranean nurseries and classification accuracies ranged from 62 to 80%. Interannual trends in otolith chemistry were observed between year classes of age‐0 T. thynnus in the Alboran Sea; however, no differences were detected between year classes in the Tyrrhenian Sea. Age‐0 and age‐1 T. thynnus collected from the same region (Ligurian Sea) were also compared and distinct differences in otolith chemistry were observed, indicating ontogenetic shifts in habitat or elemental discrimination. Findings suggest that otolith chemistry of juvenile T. thynnus from different nurseries are distinct and chemical signatures show some degree of temporal persistence, indicating the technique has considerable potential for use in future assessments of population connectivity and stock structure of T. thynnus. 相似文献

In this study, we subjected eels from Lake Balaton (Hungary) to a swimming period of 1 week and 2 or 6 weeks. Most eels were silver and were 13–21 years old. Time dependent changes in morphometrical parameters and developmental characteristics of the oocytes were determined. Already after 1 week of swimming, the gonadal mass increased and oocytes became larger, filled with large numbers of lipid droplets. After 2 and 6 weeks of swimming we found in addition a significant enlargement of the eyes, which is a sign of sexual maturation. In contrast to the resting eels, that had oocytes in the primary growth phase (stage 1–2); the swimming eels had oocytes in stage 3; the cortical alveolus or lipid droplet stage. The results indicate that lipid mobilisation induced by swimming is a requirement for the natural incorporation of lipid droplets in the oocytes, a crucial step in oocyte maturation. As the Balaton eels responded stronger to swimming than young farmed eels, it is suggested that older eels are more sensitive for maturation triggers. 相似文献

Foot-and-mouth disease (FMD) is endemic in Kenya and has been well studied in cattle, but not in pigs, yet the role of pigs is recognised in FMD-free areas. This study investigated the presence of antibodies against FMD virus (FMDV) in pigs sampled during a countrywide random survey for FMD in cattle coinciding with SAT 1 FMDV outbreaks in cattle. A total of 191 serum samples were collected from clinically healthy pigs in 17 districts. Forty-two of the 191 sera were from pigs vaccinated against serotypes O/A/SAT 2 FMDV. Antibodies against FMDV non-structural proteins were found in sera from 30 vaccinated and 71 non-vaccinated pigs, altogether 101/191 sera (53 %), and 91 % of these (92/101) also had antibodies measurable by serotype-specific ELISAs, predominantly directed against SAT 1 with titres of 10–320. However, only five high titres against SAT 1 in vaccinated pigs were confirmed by virus neutralisation test (VNT). Due to high degree of agreement between the two ELISAs, it was concluded that positive pigs had been infected with FMDV. Implications of these results for the role of pigs in the epidemiology of FMD in Kenya are discussed, and in-depth studies are recommended. 相似文献

Pigs and humans have shared influenza A viruses (IAV) since at least 1918, and many interspecies transmission events have been documented since that time. However, despite this interplay, relatively little is known regarding IAV circulating in swine around the world compared with the avian and human knowledge base. This gap in knowledge impedes our understanding of how viruses adapted to swine or man impacts the ecology and evolution of IAV as a whole and the true impact of swine IAV on human health. The pandemic H1N1 that emerged in 2009 underscored the need for greater surveillance and sharing of data on IAV in swine. In this paper, we review the current state of IAV in swine around the world, highlight the collaboration between international organizations and a network of laboratories engaged in human and animal IAV surveillance and research, and emphasize the need to increase information in high‐priority regions. The need for global integration and rapid sharing of data and resources to fight IAV in swine and other animal species is apparent, but this effort requires grassroots support from governments, practicing veterinarians and the swine industry and, ultimately, requires significant increases in funding and infrastructure. 相似文献

Articular cartilage defects are prevalent in metacarpo/metatarsophalangeal (MCP/MTP) joints of horses. The aim of this study was to determine and compare the sensitivity and specificity of 3-Tesla magnetic resonance imaging (3-T MRI) and computed tomography arthrography (CTA) to identify structural cartilage defects in the equine MCP/MTP joint. Forty distal cadaver limbs were imaged by CTA (after injection of contrast medium) and by 3-T MRI using specific sequences, namely, dual-echo in the steady-state (DESS), and sampling perfection with application-optimised contrast using different flip-angle evolutions (SPACE). Gross anatomy was used as the gold standard to evaluate sensitivity and specificity of both imaging techniques.CTA sensitivity and specificity were 0.82 and 0.96, respectively, and were significantly higher than those of MRI (0.41 and 0.93, respectively) in detecting overall cartilage defects (no defect vs. defect). The intra and inter-rater agreements were 0.96 and 0.92, respectively, and 0.82 and 0.88, respectively, for CT and MRI. The positive predictive value for MRI was low (0.57). CTA was considered a valuable tool for assessing cartilage defects in the MCP/MTP joint due to its short acquisition time, its specificity and sensitivity, and it was also more accurate than MRI. However, MRI permits assessment of soft tissues and subchondral bone and is a useful technique for joint evaluation, although clinicians should be aware of the limitations of this diagnostic technique, including reduced accuracy. 相似文献
