In the present study, we examined the oral pharmacokinetics of the acidic drugs,
diclofenac (DF) and sulfamonomethoxine (SMM), which have different physicochemical
properties, in Shiba goats. DF and SMM were intravenously and orally administered to 5
male goats using a crossover design. The Tmax of DF and SMM were reached 1.5
and 5.6 hr after they have been orally administered, respectively, and this was followed
by their slow elimination. The elimination of both drugs was markedly faster after being
intravenously rather than orally administered, which indicated flip-flop phenomena after
the oral administration. The mean absorption times (MATs) of DF and SMM were 6 and 15 hr,
respectively. This slow absorption may have been due to slow gastric emptying in goats.
The large difference observed in MATs between DF and SMM may have been because DF, which
is more lipophilic than SMM, was partly absorbed from the forestomach. Therefore, these
results suggest that the absorption of highly lipophilic drugs from the forestomach may be
markedly high in Shiba goats. In case of drugs whose elimination is quite fast, their
efficacies may appear from the early stage after oral administration even in ruminants,
because elimination rate is the determinant factor of Tmax in flip-flop

The pharmacokinetics of acetaminophen was investigated following oral dosing to Shiba
goats in order to evaluate the properties of gastric emptying. Acetaminophen was
intravenously and orally administered at 30 mg/kg body weight to goats using a crossover
design with a 3-week washout period. The stability of acetaminophen in rumen juice was
also assessed. Acetaminophen concentrations were measured by HPLC. Since acetaminophen was
stable in rumen juice for 24 hr, the extremely low bioavailability (16%) was attributed to
its hepatic extensive first-pass effect. The mean absorption time and absorption half-life
were unexpectedly short (4.93 and 3.35 hr, respectively), indicating its marked absorption
from the forestomach, which may have been due to its smaller molecular weight. Therefore,
acetaminophen was considered to be unsuitable for evaluating gastric emptying in Shiba

Total spikelet number per panicle (TSN) is thought to be one of the most important agronomic traits associated with grain production in rice (Oryza sativa L.). We previously reported the development of 334 introgression lines (ILs) with variations in agronomic traits in the genetic background of indica rice variety IR64. Among these, an IL derived from high-yielding rice variety Hoshiaoba showed significantly higher TSN than IR64. We therefore have conducted a quantitative trait locus (QTL) analysis for TSN in an F2 population of IL × IR64 to ascertain the genetic basis for the high TSN of the IL. The analysis revealed the presence of a QTL for TSN on chromosome 7, where several QTLs for grain number have been previously reported. We developed a near-isogenic line (NIL) for this QTL by using DNA marker-assisted selection and characterized its effect. The NIL showed significantly higher TSN than IR64. These results suggest that the QTL and the NIL will be useful materials in breeding programs aimed at increasing TSN. 相似文献

In June 2006, a large number of small black spots containing a branching network of black hyphae developed on shipped Satsuma mandarin fruits grown in greenhouses, which markedly decreased their commercial value. The fungus that was frequently isolated from the damaged fruits was identified as Cladosporium cladosporioides. Inoculation of fruits with the fungus caused the same symptoms, and the fungus was reisolated from the diseased fruits. The disease caused by this fungus has not been previously documented in citrus fruits; we name this new disease “sooty spot” on the basis of its symptoms. 相似文献

Rhodococcus equi is a Gram-positive facultative intracellular bacterium that causes pyogranulomatous pneumonia in foals and immunocompromised people. In the present study, signature-tagged transposon mutagenesis was applied for the negative selection of R. equi mutants that cannot survive in vivo. Twenty-five distinguishable plasmid-transposon (plasposon) vectors by polymerase chain reaction (PCR), each containing a unique oligonucleotide tag, were constructed and used to select the transposon mutants that have in vivo fitness defects using a mouse systemic infection model. Of the 4,560 transposon mutants, 102 mutants were isolated via a real-time PCR-based screening as the mutants were unable to survive in the mouse model. Finally, 50 single transposon insertion sites were determined via the self-cloning strategy. The insertion of the transposon was seen on the virulence plasmid in 15 of the 50 mutants, whereas the remaining 35 mutants had the insertion of transposon on the chromosome. The chromosomal mutants contained transposon insertions in genes involved in cellular metabolism, DNA repair and recombination, gene regulation, non-ribosomal peptide synthesis, and unknown functions. Additionally, seven of the chromosomal mutants showed a reduced ability to multiply in the macrophages in vitro. Information about forest biomass distribution is important for sustainable forest management and monitoring fuelwood supply.
The objective of this study is to develop an accurate forest biomass map for Kampong Thom Province, Cambodia. We used a new
technique (object-based approach) and a conventional technique (pixel-based approach) for the estimation of forest biomass
using Landsat Enhanced Thematic Mapper Plus (ETM+). The object-based approach created segments of images, and calculated statistical
and textural attributes. Our results showed that estimation accuracy of the object-based approach, with the use of band 1
and an exponential fit, was the best (R
2 = 0.76), and this accuracy was comparable to that of the pixel-based approach (R
2 = 0.67). Although several textural variables were related to forest biomass, they did not contribute significantly to improvement
of estimation accuracy. However, the object-based method can be used for image segmentation so that the image objects are
spectrally more homogeneous within individual regions than with their neighbors. Hence, they can be regarded as management
units for policy-related spatial decisions. Therefore, it is possible to select either of the two methods depending upon what

In six normal beagles and 27 dogs with spontaneous focal or multifocal liver lesions, contrast-enhanced ultrasonography using Sonazoid® was performed. Sonazoid® is a newly developed second-generation contrast agent with the ability to be used for real-time contrast imaging along with parenchymal imaging. An appropriate protocol for the evaluation of all three phases (arterial, portal, and parenchymal) was established based on the results for normal beagles. By evaluation of the echogenicity of hepatic nodules during the arterial and parenchymal phases it was possible to differentiate malignant tumors from benign nodules with very high accuracy. In 15 of 16 dogs diagnosed as malignant tumors, nodules were clearly hypoechoic to the surrounding normal liver during the parenchymal phase. Additionally, malignant tumors had different echogenicity compared with the surrounding normal liver during the arterial phase in 14 of 15 dogs. In the portal phase, there were no characteristic findings. Contrast-enhanced ultrasonography with Sonazoid® appears to improve the characterization of canine focal and multifocal hepatic lesions. 相似文献
