The physiological and biochemical basis for quinclorac resistance in a false cleavers (Galium spurium L.) biotype was investigated. There was no difference between herbicide resistant (R) and susceptible (S) false cleavers biotypes in response to 2,4-D, clopyralid, glyphosate, glufosinate-ammonium, or bentazon. On the basis of GR(50) (growth reduction of 50%) or LD(50) (lethal dose to 50% of tested plants) values, the R biotype was highly resistant to the acetolactate synthase (ALS) inhibitor, thifensulfuron-methyl (GR(50) resistance ratio R/S = 57), and quinolinecarboxylic acids (quinclorac R/S = 46), resistant to MCPA (R/S = 12), and moderately resistant to the auxinic herbicides picloram (R/S = 3), dicamba (R/S = 3), fluroxypyr (R/S = 3), and triclopyr (R/S = 2). The mechanism of quinclorac resistance was not due to differences in [(14)C]quinclorac absorption, translocation, root exudation, or metabolism. Seventy-two hours after root application of quinclorac, ethylene increased ca. 3-fold in S but not R plants when compared to controls, while ABA increased ca. 14-fold in S as opposed to ca. 3-fold in R plants suggesting an alteration in the auxin signal transduction pathway, or altered target site causes resistance in false cleavers. The R false cleavers biotype may be an excellent model system to further examine the auxin signal transduction pathway and the mechanism of quinclorac and auxinic herbicide action.  相似文献

Parma hams at various processing stages were investigated by surface autofluorescence spectroscopy. Fluorescence "landscapes" of raw meat and salted (3 months), matured (11 and 12 months), and aged (15 and 18 months) Parma hams were obtained, and a three-dimensional data array (sample x emission x excitation) was used to develop a PARAFAC model including five components, which all exhibited characteristics of pure fluorophores regarding both excitation and emission spectra. The relative amount of each component related strongly to the processing stage, and sample age showed good correlation to fluorescence data (R = 0.98), with a relative error of prediction of approximately 1 month. Fluorescence measurements from samples of either semimembranosus or biceps femoris were used to predict chemical or sensory reference data, yielding good correlation for biceps femoris data, thereby enabling moisture content, sensory and instrumental color, and proteolysis value to be fairly well predicted. Overall, surface autofluorescence of Parma hams proved to hold relevant information, relating to major chemical/physical changes during processing. It is concluded that fluorescence spectroscopy has potential as an innovative method of quality control in dry-cured ham.  相似文献

Consumption of fruits and vegetables, which are rich in polyphenols, has been associated with a reduced risk of chronic diseases such as cancer. Dietary polyphenols have antioxidant and antiproliferative properties that might explain their beneficial effect on cancer prevention. The aim of this study was to investigate the effects of different pure polyphenols [quercetin, chlorogenic acid, and (-)-epicatechin] and natural fruit extracts (strawberry and plum) on viability or apoptosis of human hepatoma HepG2 cells. The treatment of cells for 18 h with quercetin and fruit extracts reduced cell viability in a dose-dependent manner; however, chlorogenic acid and (-)-epicatechin had no prominent effects on the cell death rate. Similarly, quercetin and strawberry and plum extracts, rather than chlorogenic acid and (-)-epicatechin, induced apoptosis in HepG2 cells. Moreover, quercetin and fruit extracts arrested the G1 phase in the cell cycle progression prior to apoptosis. Quercetin and strawberry and plum extracts may induce apoptosis and contribute to a reduced cell viability in HepG2 cells.  相似文献

A headspace solid-phase microextraction (HS-SPME) procedure at 30 degrees C with a 100 microm PDMS fiber of a saturated NaCl solution stirred at 1100 rpm combined to GC-ECD for the 2,4,6-trichloroanisol (TCA) determination in wines has been developed. Due to the matrix complexity and ethanol absorption into the fiber, the internal standard selection was crucial to obtain unbiased results. Thus, matrix effects were observed when analyzing different types of Spanish wines (white, early, and vintage red wines) spiked with TCA at low concentration levels (i.e., <40 ng L(-)(1)). In contrast, the use of 2,4,6-tribromoanisole (TBA) as internal standard overcame these matrix effects, whereas the use of 2,4,6-trichlorophenyl ethyl ether led to inconsistent results. The developed HS-SPME-GC-ECD methodology reaches a limit of quantitation for TCA in wine within 2.9-18 ng L(-)(1), with a relative standard deviation of 2.5-13.4%, depending on the TCA concentration level and wine characteristics. This analytical method is comparable to the existing methodologies based on HS-SPME followed by GC-MS in terms of accuracy, precision, length of determination, and length of quantification; however, analysis cost is reduced.  相似文献

This study vividly presents results from a seasonal particulate matter measurement campaign conducted at world’s largest ship-breaking yard i.e., Alang-Sosiya (Gujarat, India) at six locations and a reference station at Gopnath which is 30 km south of this ship-breaking yard. The collected suspended particulate matter (SPM) 24-h samples were critically analyzed for heavy metals (Pb, Cd, Co, Ni, Cr, Mn, Fe, Cu, Zn). The average concentration of SPM within the ship-breaking yard during the investigation was 287.5 ± 20.4 μg m?3 and at reference station it was 111.13 ± 5.81 μg m?3. These values are found to be in excess of the permitted national standards. The levels of heavy metals at Alang-Sosiya are very high as compared to US EPA and WHO guidelines. The mean concentrations of all metals are in the order: Fe >>Zn >Cu > Mn > Cd >Pb > Co >Ni >Cr. The results on enrichment factors (EF) suggest that most of the metals in the ship-breaking yard exhibit EF values of near or above 100 which must have been comprehensively affected by ship-breaking activities. Metal data was used to evaluate the role of spatial factors on their distribution characteristics. Thereafter, factor analysis was carried out to identify the main components liable for the variance of the data set.  相似文献

High concentrations of heavy metals are known to be toxic to many soil organisms. The effects of long-term exposure to lower levels of metals on the soil microbial community are, however, less well understood. The southern Pennines of the U.K. are characterised by expanses of ombrotrophic peat soils that have experienced deposition of high levels of heavy metals since the mid to late 1800s. Concentrations of metals in the peat remain high but the effect of the contamination on the in-situ microbial communities is unknown. Geochemical and molecular polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE) and sequencing techniques were used to derive new information on the metal chemistry and microbial populations in peat soils from six locations in the southern Pennines. All sites were highly acidic (pH 3.00–3.14) with high concentrations of potentially toxic heavy metals, particularly porewater Zn and particulate-associated Pb. The results also reveal a split in site characteristics between the most polluted sites with the highest levels of bioavailable metals (Bleaklow, FeatherBed Moss and White Hill) and those with much lower bioavailable metals (Cowms Moor, Holme Moss and Round Hill). There was no difference in the number of dominant bacterial species between the sites but there were significant differences in the species composition. At the three sites with the highest levels of bioavailable metals, bacterial species with a high similarity to acidophilic sulphur- and iron-oxidizing bacteria and those from high metal environments were detected. The transformations carried out by these metal mobilising and acid producing bacteria may make heavy metals more bioavailable and therefore more toxic to higher organisms. Bacteria with similarity to those typically found in forest and grassland soils were documented at the three sites with the lowest levels of bioavailable metals. The data highlight the need for further studies to elucidate the species diversity and functionality of bacteria in heavy metal contaminated peats in order to assess implications for moorland restoration.  相似文献

Results from examining shellfish collected from ‘safe’ (2–34Escherichia coli/100 mL) and ‘unsafe’ (> 34E. coli/100 mL) waters for the presence ofYersinia enterocolitica and related bacteria are reported. The recovery of these microorganisms was low in bivalve mollusks destined for food consumption, high in those derived from prohibited areas. No human pathogenic bio-serogroup f Yersinia was found during the entire survey.  相似文献

Archaeologists often encounter problems in interpreting the genesis of layers and the context in which specific materials occur in excavations. Field visits of geologists, soil scientists or biologists can solve a number of questions, but often, problems remain that only can be studied by microscopic research as, for example, soil micromorphology and palynology. In the past, the employment of these two disciplines, one in earth science and one in biology, generally occurred independently of each other, also in interdisciplinary research and contract research. Consequently, results could not always be correlated, permitting only a restricted integration of data. It was realised that soils, as archives, contain much more information than generally utilised. A skillful employment and balancing of both specialties create a real integrated approach that offers a substantially improved interpretation potential without hardly additional efforts and costs. The results of 7 years of experience were very stimulating and presented here.In this article, the essence of soil micromorphology and palynology applied to archaeology is briefly outlined and illustrated by selected examples of the different research themes covered, showing the possibilities and results of integrated research. Both specialties generate essential information that, when combined and integrated properly, lead to improved understanding of the landscape genesis and human activities in the past. To be able to employ the full interpretive potential of this integrated approach, the research procedures followed by both specialties had to be compared. The key requirements for optimal integration of each other's databases comprised a combined effort to compare the pros and cons during the first four steps of the research procedure: defining research questions, field survey, selection of sampling locations and sampling. This approach enabled optimal correlation of the individual results, made cross-checking and references possible and led to higher levels of interpretation that would not have been possible otherwise. The integrated research elucidated lacunae in the other's discipline, resulting in a distinctly more complete reconstruction of paleoenvironments and past land uses, important research themes of archaeologists for the specialties concerned.  相似文献
