Saponin-containing feed additives have shown positive effects on pig performance. Quinoa hull has high saponin content and may be of interest as a feed additive. This study aimed to evaluate quinoa hull meal (QHM) as a feed additive in a pig diet. The effects of QHM were assessed for three dosages of South American (SA) origin (100, 300 and 500 mg/kg) and one dosage of Danish (DK) quinoa (300 mg/kg). In addition, the effect of dietary SA-QHM and SA-QHM-extract on jejunal epithelial physiology was studied ex vivo in Ussing chambers. The experiment included 400 piglets weaned at 28 ± 2 days of age and the experimental period was 4 weeks. Piglets were weighed initially and finally and feed intake registered. The ex vivo studies were performed with epithelium from 40 pigs receiving control or SA-QHM. Epithelium from each pig was placed into eight Ussing chambers, where four concentrations of SA-QHM-extract were added. Epithelial permeability, Na(+)-dependent glucose transport and serotonin (5-HT) and theophylline-induced secretion were measured. The results showed that QHM had no influence on piglet's growth (p = 0.41) or feed intake (p = 0.17). In spite of a large difference in saponin content between SA-QHM and DK-QHM (28.7% and 2.0% w/w respectively) the source did not affect pig performance. The ex vivo studies revealed no effect (p > 0.05) of adding QHM-extract into the medium. The permeability and glucose induced absorption were highest (p = 0.003 and p = 0.04 respectively) in epithelium from pigs that consumed 100 or 300 mg/kg SA-QHM. The secretory response to 5-HT was not affected (p = 0.59) by dietary treatments, but the theophylline-induced secretion decreased (p = 0.02) with increasing dietary SA-QHM. The changes in epithelial physiology measured ex vivo did not affect animal performance in vivo in this study.  相似文献

Arabinoxylans (AX) are the main dietary fiber (DF) polysaccharides in rye where they represent ≈55% of the total polysaccharides. Rye AX consist of a backbone of (1→4)‐β‐d ‐xylopyranosyl residues (X) mainly substituted with α‐l ‐arabinofuranosyl residues (A) to varying degrees at the O‐2 position, the O‐3 position, or both. The A/X ratio of total AX is 0.49–0.82 and extractable AX ratio is 0.34–0.85 in different studies. AX also contain small amounts of ferulate residues bound to arabinose as esters at its O‐5 position. The weight average molecular weight varies from 40,000 to 900,000 with an average of ≈200,000. AX influence physiology in different segments of the gastrointestinal tract. The complex molecular structure of rye AX makes them resistant against microbial modification in the small intestine; consequently, rye AX have a much higher influence on the viscosity in the small intestinal digesta than does β‐glucan from oats and barley. In spite of that, it has not been possible in studies with AX‐rich foods such as bread to demonstrate a significant effect on the postprandial glucose response, however, a significantly reduced insulin response has been seen. Nevertheless, addition of 6 g and 12 g of AX‐rich wheat fiber to a breakfast meal has significantly lowered postprandial glucose and insulin response. Studies with hypercholesterolemic pigs fed rye buns rich in AX have resulted in dramatic reductions in plasma total and LDL cholesterol, whereas a gender difference was seen in studies on the effect of AX on plasma lipids in humans. Only certain species of bacteria from the human gut produce the enzymes needed for the degradation of AX. Nevertheless, wheat AX stimulate prebiotic bacteria presumably brought about by cross feeding of lactobacilli and bifidobacteria with degradation products from versatile carbohydrate‐degrading bacteria. Soluble AX are readily fermented in the large intestine, the majority is broken down between the ileum and the cecum. AX, characterized by a low degree of substitution and virtually no doubly substituted xylose, are slowly degraded at more distal locations. The remaining AX, characterized by a high degree of substitution, are not degraded at all. Although the fermentation pattern of AX may vary in different experimental models, in vitro fermentation studies and in vivo intervention studies with animals and humans point to AX as substrates that enhance the formation of butyrate in the large intestine.  相似文献

Abstract

Ninetysix pigs, half females and half castrated males from 12 litters, were housed in 24 groups of 4 litter mates (2 females, 2 castrates). The pigs were weaned at 4 weeks of age and from the age of 115 days half of the groups were subjected to a schedule of unpredictable, inescapable electroshocks for 33 days, and half served as control. The adrenocortical reactivity to ACTH stimulation and to an emotional stressor (the procedure associated with initiation of blood collection) was measured in females, whereas the behavioural and pituitary-adrenocortical reactivity to an open field test was measured in castrates, as was the behavioural reactivity in a social test. Intermittent stress increased adrenocortical reactivity to ACTH stimulation significantly. This effect was present within the first week of intermittent stress (p < 0.06), but no difference was found after 4–5 weeks of stress. In contrast, the adrenocortical reactivity to additional emotional stressors was unaffected by stress after one week of intermittent stress, but increased after 4–5 weeks. In the open field test, centre location increased after one week of intermittent stress, whereas exploration decreased and walking and ambulation increased after 4–5 weeks of stress. In addition, 4–5 weeks of intermittent stress decreased aggressiveness in a social test. In conclusion, the change in reactivity to an additional acute stressor during intermittent stress appeared in different variables and contexts at different times during the course of intermittent stress. Thus, assessment of stress based on changed reactivity has to include several distinct measures of behavioural and hormonal reactivity.  相似文献

The epidemic outbreak in northern Europe of Neonectria neomacrospora, the causal agent of dieback in Abies spp., led the European and Mediterranean Plant Protection Organization (EPPO) to include the pathogen on its alert list in 2017. Effective monitoring of this pathogen calls for a rapid and sensitive method of identification and quantification. A probe‐based real‐time PCR (qPCR) assay based on the β‐tubulin gene was developed for the detection and quantification of N. neomacrospora in infected wood samples, and directly for ascospores. This study presents the first published species–specific molecular detection assay for N. neomacrospora. The analytical specificity was validated on taxonomically closely related fungal species as well as on 18 fungal species associated with the host (Abies sp.). The analytical sensitivity was tested on naturally infected wood, on purified pathogen DNA in a matrix of host DNA and on N. neomacrospora ascospores for detection of airborne inoculum. The latter was tested both with a DNA extraction step prior to qPCR and without DNA extraction by direct qPCR on collected ascospores. The assay was specific to N. neomacrospora, with a sensitivity of 130 fg purified DNA, or 10 ascospores by direct qPCR. Omitting DNA extraction and amplifying directly on unpurified ascospores improved assay sensitivity significantly.  相似文献

The turnover of I131-albumin has been studied in two sows with hypertrophic gastritis (Hyostrongylus rubidus infestation) and in three pigs with terminal ileitis. The sows showed few characteristic digestive symptoms and their albumin turnover data were essentially normal (Table 1). Pigs with terminal ileitis showed more active symptoms of gastrointestinal disease (diarrhea). Albumin turnover rates were greatly accelerated, with daily degradations of nearly 50 % of the vascular albumin pool (Table 2).It is concluded that the pattern of albumin metabolism in pigs with gastrointestinal disease is related to the actual clinical phase of the disease rather than to pathoanatomical lesions (hypertrophy etc.). The pathophysiological features determining the clinical picture are, accordingly, of great importance.  相似文献

Yield and water productivity of potatoes grown in 4.32 m2 lysimeters were measured in coarse sand, loamy sand, and sandy loam and imposed to full (FI), deficit (DI), and partial root-zone drying (PRD) irrigation strategies. PRD and DI as water-saving irrigation treatments received 65% of FI after tuber bulking and lasted for 6 weeks until final harvest. Analysis across the soil textures showed that fresh yields were not significant between the irrigation treatments. However, the same analysis across the irrigation treatments revealed that the effect of soil texture was significant on the fresh yield and loamy sand produced significantly higher fresh yield than the other two soils, probably because of higher leaf area index, higher photosynthesis rates, and “stay-green” effect late in the growing season. More analysis showed that there was a significant interaction between the irrigation treatments and soil textures that the highest fresh yield was obtained under FI in loamy sand. Furthermore, analysis across the soil textures showed that water productivities, WP (kg ha−1 fresh tuber yield mm−1 ET) were not significantly different between the irrigation treatments. However, across the irrigation treatments, the soil textures were significantly different. This showed that the interaction between irrigation treatments and soil textures was significant that the highest significant WP was obtained under DI in sandy loam. While PRD and DI treatments increased WP by, respectively, 11 and 5% in coarse sand and 28 and 36% in sandy loam relative to FI, they decreased WP in loamy sand by 15 and 13%. The reduced WP in loamy sand was due to nearly 28% fresh tuber yield loss in PRD and DI relative to FI even though ET was reduced by 9 and 11% in these irrigation treatments. This study showed that different soils will affect water-saving irrigation strategies that are worth knowing for suitable agricultural water management. So, under non-limited water resources conditions, loamy sand produces the highest yield under full irrigation but water-saving irrigations (PRD and DI) are not recommended due to considerable loss (28%) in yield. However, under restricted water resources, it is recommended to apply water-saving irrigations in sandy loam and coarse sand to achieve the highest water productivity.  相似文献

Summary

Seven pesticide application strategies were investigated to control apple scab (Venturia inaequalis) and powdery mildew (Podosphaera leucotricha) and, at the same time, fulfil the new quality standards implemented by some German retailers. These demand that pesticide residues should be below 80% of European Maximum Residue Levels (EU MRL) and that the number of residues present at levels above 0.01 mg kg?1 should be limited to a maximum of four. The strategies fulfilled the requirement to use combinations of different active substances in order to prevent the emergence of resistance to pesticides. The trials were conducted at two sites in Switzerland, in 2007, and all strategies and applications were in accordance with actual practice. Four replicates of apple samples from each strategy were then analysed for pesticide residues. The incidence of infection with apple scab and powdery mildew were monitored during the season in order to evaluate the efficacy of the different strategies. The efficacies of the different strategies against apple scab and powdery mildew were between 84% and 100% successful. In general, the level of pesticide residues found correlated with application rate and time, and no measured residue level exceeded the EU–MRL. The numbers of residues present at > 0.01 mg kg?1 were between two and five.  相似文献

Gas dispersion in a set of three different porous materials with similar particle size, as a function of material tortuosity and anisotropy ratio, was investigated. The materials were packed with different spatial orientations of the individual particles so as to create media with different tortuosity and anisotropy ratios. Three different media (slate chips, wood chips, and pebbles) and four particle orientations have been used to generate a total of nine different porous media mimicking single porosity, dual porosity isotropic, anisotropic, aggregated, or granular materials. Resulting values of tortuosity and anisotropy ratio for each medium were determined via measurements of gas permeability and molecular gas diffusion coefficient. These values were then compared to measured values of gas dispersivity for each medium. The results showed that dispersivity is inversely proportional to tortuosity but directly proportional to anisotropy ratio and that the relations were approximately linear within the range of tortuosities and anisotropy ratios investigated. Wood chips (dual porosity material) yielded higher values of gas dispersivity compared to slate chips (single porosity material). A likely reason is in part the difference in pore structure between the materials and in part a difference in particle surface roughness (which was highest for wood chips) both of which affects dispersion.  相似文献

In this study, we assessed a broad range of barley breeding lines and commercial varieties by three hardness methods (two particle size methods and one crush resistance method (SKCS—Single-Kernel Characterization System), grown at multiple sites to see if there was variation in barley hardness and if that variation was genetic or environmentally controlled. We also developed near-infrared reflectance (NIR) calibrations for these three hardness methods to ascertain if NIR technology was suitable for rapid screening of breeding lines or specific populations. In addition, we used this data to identify genetic regions that may be associated with hardness. There were significant (p<0.05) genetic effects for the three hardness methods. There were also environmental effects, possibly linked to the effect of protein on hardness, i.e. increasing protein resulted in harder grain. Heritability values were calculated at >85% for all methods. The NIR calibrations, with R2 values of >90%, had Standard Error of Prediction values of 0.90, 72 and 4.0, respectively, for the three hardness methods. These equations were used to predict hardness values of a mapping population which resulted in genetic markers being identified on all chromosomes but chromosomes 2H, 3H, 5H, 6H and 7H had markers with significant LOD scores. The two regions on 5H were on the distal end of both the long and short arms. The region that showed significant LOD score was on the long arm. However, the region on the short arm associated with the hardness (hordoindoline) genes did not have significant LOD scores. The results indicate that barley hardness is influenced by both genotype and environment and that the trait is heritable, which would allow breeders to develop very hard or soft varieties if required. In addition, NIR was shown to be a reliable tool for screening for hardness. While the data set used in this study has a relatively low variation in hardness, the tools developed could be applied to breeding populations that have large variation in barley grain hardness.  相似文献
