In order to estimate the influence of the Extension (E) locus in cattle coat color, the melanocortin‐1 receptor (MC1R) gene in Japanese Black, Japanese Brown and Korean (Hanwoo) cattle were sequenced. The sequences of the coding region revealed three alleles (ED, E+ and e), which were previously reported. Polymerase chain reaction‐restriction fragment length polymorphism was performed to investigate the gene frequencies of the three breeds. Japanese Black was almost composed of ED and E+ individuals, ED = 0.481 and E+ = 0.514, and no homozygous e/e, therefore that is consistent with the hypothesis that ED and E+ induce black pigment synthesis. Allele frequencies between Japanese Brown and Hanwoo were obviously different; however, recessive red e allele frequency was 0.038 for Japanese Brown and 0.948 for Hanwoo, even though both breeds have quite similar coat colors (ranging from yellowish brown to dark brown including a red coat color). This result suggested that other genes are also associated with a coat color of red and brown in cattle.  相似文献

The differential variables for carcass grades were identified from morphological, behavioral and physiological measurements of young steers at a commercial farm. Thirty‐five Japanese Black × Holstein steers aged 6–10 months were randomly assigned to three pens. The steers had free access to ryegrass hay and a restricted amount of concentrate diet for the first 6 months of trial. They were then given oat straw ad libitum and an additional concentrate diet until slaughter. Behavioral observations (15 categories) were carried out once every 2 weeks for 2 h after feeding. Bodyweight was measured, blood sampled (for seven metabolic hormones and five metabolites), ultrasonic scanning conducted and physical measurements taken (10 parts for each animal) in months 1, 3 and 5 (early fattening stage, EFS) and in months 7, 9 and 11 (middle fattening stage, MFS) of the trial. Temperament scores during these procedures, entry order into restraint stalls, social rank and average daily gain were also assessed. A factor analysis and one‐way anova were used to identify clusters of variables which had different factor scores between carcass grades. As for the carcass yields, a cluster of chest girth and depth, bodyweight, withers and hip height, and rump length (P < 0.01), and a cluster of the frequency of scratching the body with facilities (P < 0.05) were identified as differential variables in the EFS. In the MFS, a cluster of concentrations of cortisol and epinephrine, and the frequency of eating straw were identified (P < 0.10). As for the carcass quality grade, a cluster of social rank, triglyceride concentration and the frequencies of stand‐chewing cud and eating hay, and a cluster of the frequency of investigating facilities (both P < 0.10) were identified in the EFS. In the MFS, a cluster of hip height, bodyweight, cannon circumference, chest depth, withers height and chest girth, and a cluster of the frequencies of eating a concentrate diet and stand‐chewing the cud, and social rank (both P < 0.10) were identified. In conclusion, bodyweight, chest girth and depth, withers and hip height were identified as good differential variables for future carcass grades of young steers. Facility enrichment that encourages steers to eat hay in the EFS, and to eat a concentrate diet in and after the MFS would be effective in upgrading carcass grades. High incidence of investigating and scratching the body with facilities and stand‐chewing the cud might lead to lower carcass grades.  相似文献

Commercial dairy cows milked in a parlor system are packed close together in the holding area before milking. The present study examined the relationships of ‘looking up’ behavior with some other behaviors and the productivity of 1116–1153 cows from five farms. The individual identities of the cows looking up in the holding area were recorded at 5 min intervals during six intermittent afternoon milking sessions. Entrance into the milking parlor and the numbers of steps and kicks by cows while the milking person was attaching the milking cups, were recorded in six milking sessions. Flight responses in the pasture after milking were recorded over four days intermittently. The frequency of ‘looking up’ behavior weakly, but significantly correlated with flight starting distance (r = 0.10, P < 0.05), while the correlation with the number of step‐kicks during milking was not significant. As for productivity, lactation number (r = ?0.18, P < 0.001), milk yield (r = ?0.15, P < 0.001) and fat content (r = ?0.15, P < 0.001) were negatively correlated with the frequency of ‘looking up’ behavior. Age of cows was correlated with the frequency of ‘looking up’ behavior as well as lactation number (r = ?0.21, P < 0.001). Entrance order was positively correlated with the frequency of ‘looking up’ behavior (r = 0.15, P < 0.001). The ‘looking up’ behavior was observed more frequently in cows in their third or less lactation compared with cows which were in their fourth or greater lactation (P < 0.05). The lactation number of cows was correlated with their milk yield (r = 0.36, P < 0.001) and flight starting distance (r = ?0.21, P < 0.001). In conclusion, ‘looking up’ behavior shown by cows in the holding area before milking might be an indicator of low motivation for milking, mainly because of fear of humans, and an aversion to milking caused by insufficient experience in being milked.  相似文献

The longitudinal changes in fecal steroid hormone concentrations and sexual behavior in 2 mated/pregnant and 3 non-mated female Hokkaido brown bears were investigated during the breeding season. Behavioral estrus (standing) lasted for 14 and 32 days in the mated females and for 25 to 36 days in the non-mated females. In non-mated females, sexual behavior, such as female-female mounting and masturbation, was observed for several days before and after the estrous period. In mated females, mean fecal estradiol-17beta concentrations were higher in the estrous period than in the post-estrous period, while fecal progesterone concentrations were higher in the post-estrous period than in the estrous period. The similar trends of steroid hormone changes were observed in the non-mated females.  相似文献

The phenotype and function of peritoneal cavity macrophage-derived dendritic cells (PEC-DC) was previously reported. In this study we have gone further in using our established culture system to generated discrete Peyer's patch dendritic cells (DPP-DC) from murine discrete Peyer's patch macrophages (DPP-M?), following stimulation with granulocyte macrophage colony stimulating factor (GM-CSF) plus interleukin 4 (IL-4) for 7 days. DPP-M? from murine small intestines were obtained by mechanical disruption of discrete Peyer's patches (DPP), followed by metrizamide density gradient centrifugation to remove Peyer's patch resident DC and debri, after which an overnight adherent step in tissue culture medium was carried out for macrophage enrichment. Characterization of the generated DPP-DC was carried out using well-established criteria of morphology, expression of membrane antigens and capacity for antigen presentation. Dendritic cells expressed DEC-205, F4/80 and CD34 at high levels, but exhibited very low CD11c levels. They were shown to present soluble protein antigen to CD3(+) spleen T cells. A comparison of the surface antigen expression in the progenitor DPP-M? population and the generated DPP-DC showed a significant decrease in MHC class II levels and a marked down regulation of the co-stimulatory molecule CD86 (B7-2). High expression of the haemopoietic progenitor marker CD34 indicates that the generated DC, possess a haemopoietic rather than myeloid origin. Taken together, these results may provide a better understanding of the complex network regulating mucosal immune responses.  相似文献

A suitable balance in the production of Th1/Th2-type cytokines has a crucial role in the control of microbial infections. We investigated cytokine production patterns and effects during Neospora caninum infection, based on two mouse models and an in vitro system. In the acute infection of N. caninum, BALB/c-background IFN-gamma-deficient mice that were sensitive to the N. caninum infection showed high levels of IL-10 production, whereas significant levels of interferon-gamma (IFN-gamma) and interleukin-4 (IL-4) production were observed in resistant wild type mice. BALB/c mice vaccinated with recombinant vaccinia virus expressing N. caninum surface protein NcSRS2 resisted parasite spread throughout the body, low levels of IFN-gamma production and high levels of IL-4 production were observed compared to unvaccinated animals. The treatment of N. caninum-infected cells with IFN-gamma or IL-10 decreased the host-cell viability in an in vitro system using mouse macrophage J774A.1 cells. On the other hand, IL-4, but not IL-10 administration, increased the viability of N. caninum-infected and IFN-gamma-treated cells. In the light of the balance of Th1/Th2-type cytokine production, an IFN-gamma/IL-4 balance may have a crucial role for the control of cellular responses against the parasite invasion.  相似文献

Using macrophage scavenger receptor-A knockout (SRKO) mice, we examined the role of macrophage class A scavenger receptors (MRS-A) on the immune response and acquisition of host resistance against repeated infestation with Haemaphysalis longicornis. Except for one batch of nymphs that infested one of the SRKO (SR-/-) mice and showed no appreciable reduction in body weight, all the other groups of nymphs manifested significant decrease in body weight. Both SR-/- and wild type (SR+/+) mice showed a sustained increase in anti-tick antibody titers, but SR+/+ mice showed significantly higher titers. The IFN-gamma assayed in SR-/- mouse immune sera was substantially less compared with that in SR+/+ mice. Immune sera from SR-/- and SR+/+ mice recognized the 51 and 44 kDa, and 44 kDa proteins, respectively, of the salivary gland antigen. The difference in the level of anti-tick resistance manifested by both groups of mice may be influenced by less efficient trapping and processing of tick antigens by macrophages in mice lacking for the macrophage scavenger receptors, and consequently affected the cascade of Th1 and Th2 responses. We have thus obtained valuable data that strongly infer the role of MSR-A in enhancing host defense against repeated infestation with H. longicornis.  相似文献

Transencapsidation of the Rice gall dwarf virus (RGDV) inner core by the Rice dwarf virus (RDV) outer capsid P8 protein was examined in vitro and in planta. When RGDV core particles were incubated with an extract from RDV P8-transgenic rice leaf tissue, RDV P8 encapsidated the RGDV core particles to form double-shelled virus-like particles in vitro. In contrast, when RDV P8-transgenic rice plants were inoculated with RGDV, progeny RGDV particles contained RGDV P8 but RDV P8 was not detectable in the virions. No significant differences were found in acquisition by the vector insects and subsequent transmission rates between RGDV infecting nontransgenic rice plants and those infecting RDV P8-transgenic rice plants. These results indicate that mechanisms of and/or requirements for interactions between P8 and the inner core particles of phytoreoviruses differ between in vitro and in planta.  相似文献

Resource recycling and the proper treatment of animal waste to reduce its environmental impact are currently important issues for the livestock industry. A microbial fuel cell (MFC), a new type of bioreactor, is expected to play roles in both waste‐water purification and energy recovery. However, the generation of electricity from cow waste has not yet been examined. In this study, using an MFC, we examined the possibility of generating electricity from dairy‐cow waste slurry, and analyzed the properties of the treated slurry as liquid manure for resource recycling. The MFC treatment of the slurry generated electricity in a dose‐dependent manner, and the maximum power output by the MFC from a 1 g of chemical oxygen demand/L slurry was 0.34 mW/m2. After the MFC treatment, 84% of the biological oxygen demand in the slurry was removed and three essential fertilizer elements (nitrogen, phosphorus, and potassium) were retained at 84, 70, and 91% levels, respectively. The amount of ammonia nitrogen in the slurry, as an element of fast‐release fertilizer, was increased by 1.9‐fold. Although the treated slurry displayed properties that made it preferable as liquid manure, further studies to improve the electrical power output by the MFC are required for practical use.  相似文献
