Most veterinary drugs enter the environment via manure application. However, ecotoxic effects of antibiotics are varying as a function of their physicochemical characteristics and for most antibiotics it is still unclear how these substances interact with soil biota. It was the aim of the present study to investigate effects of manure containing different concentrations of the antibiotic amoxicillin (AMX) on microbial‐community function in two different soils over an incubation time of 18 d. Therefore, soil respiration, potential nitrification, and the products of N turnover were measured. We could show that CaCl2‐extractable amounts of AMX in soil are low, even shortly after the application of high doses together with manure. Thus, not surprisingly basal respiration in soil was not influenced by the addition of the antibiotic with manure. In contrast, mainly shortly after manure addition the kinetic of substrate‐induced respiration was clearly shifted by the treatments depending on the presence of AMX in the manure. Potential nitrification rates in the two different soils were not significantly affected when data were related to the overall incubation time by the application of AMX to the manure. However, shortly after the addition of the manure containing AMX, a tendency to lower turnover rates was visible compared to the application of pure manure. 相似文献

Difloxacin (DIF) belongs to the fluoroquinolones, a frequently detected group of antibiotics in the environment. It is excreted
in pig manure to a large extent and may consequently reach soils in potentially effective concentrations via manuring. The
aim of this study was to assess the effects of DIF-spiked manure on microbial communities and selected functions in soils
in a microcosm experiment up to 1 month after application. To test a dose dependency of the effects, three different concentrations
of DIF (1, 10 and 100 mg/kg of soil) were used. Microcosms with application of pure manure, as well as untreated microcosms
served as control. The addition of pure manure resulted in an increase of microbial biomass and soil respiration as well as
a reduced bacteria/fungi ratio. Due to the fast and strong immobilisation of DIF, effects of the antbiotic compound were only
visible up to 8 days after application (microbial biomass; respiration; potential denitrification; ratio of bacteria/fungi).
As expected these short-term effects resulted in reduced potential denitrification rates as well as a reduced bacteria/fungal
ratio in the treatments were DIF has been applied. Surprisingly, microbial biomass values as well as respiration rates were
increased by DIF application. Other parameters like nitrate and ammonium content in soil were not influenced by DIF application
at any time point. Long-term effects (32 days after application) were only visible for the potential nitrification rates.
For those parameters that were influenced by the DIF application a clear dose dependency could not be described. 相似文献

OBJECTIVE: This review evaluates the thermal and mechanical nociceptive threshold testing techniques that have been used in horses and discusses them with reference to their applications, limitations and the factors which can influence both the testing procedure itself and the animal's responses. Methods to optimise the reliability and repeatability of the testing procedures are suggested and the potential clinical applications discussed. DATABASES USED: Web of Science and Medline. Diseases of paranasal sinuses and nasal passages in horses can be a diagnostic challenge because of the complex anatomy of the head and limitations of many diagnostic modalities. Our hypothesis was that magnetic resonance (MR) imaging would provide excellent anatomical detail and soft tissue resolution, and would be accurate in the diagnosis of diseases of the paranasal sinuses and nasal passages in horses. Fourteen horses were imaged. Inclusion criteria were lesions located to the sinuses or nasal passages that underwent MR imaging and subsequent surgical intervention and/or histopathologic examination. A low field, 0.3 tesla open magnet was used. Sequences in the standard protocol were fast spin echo T2 sagittal and transverse, spin echo T1 transverse, short‐tau inversion recovery (STIR) dorsal, gradient echo 3D T1 MPR dorsal (plain and contrast enhanced), spin echo T1 fatsat (contrast enhanced). Mean scan time to complete the examination was 53 min (range 39–99 min). Lesions identified were primary or secondary sinusitis (six horses), paranasal sinus cyst (four horses), progressive ethmoid hematoma (two horses), and neoplasia (two horses). The most useful sequences were fast spin echo T2 transverse and sagittal, STIR dorsal and FE3D MPR (survey and contrast enhanced). Fluid accumulation, mucosal thickening, presence of encapsulated contents, bone deformation, and thickening were common findings observed in MR imaging. In selected horses, magnetic resonance imaging is a useful tool in diagnosing lesions of the paranasal sinuses and nasal passages. 相似文献

Equine welfare charities have identified the education of (primarily novice) horse owners as the principal route to tackling the majority of equine welfare problems in the United Kingdom. To reach and educate owners effectively, we must understand where they seek advice about issues that arise when owning or caring for a horse. Questions regarding information-seeking behavior were included in an Internet survey completed by a convenience sample of 1,326 UK leisure horse owners, recruited online and through electronic and postal mail shots to equestrian clubs and establishments. The questions asked where the owners sought advice regarding 4 different subject areas: horse behavior, health, stable care, and training. Respondents were asked to select their main sources of information from a list of 7—books/magazines, vet/farrier, instructor, Internet/forums, friends/family, tack shops, and other. Although books/magazines were the most frequently used information source across all 4 subject areas overall, there were differences between the 4 subject areas in the most commonly selected sources (χ2 = 992, P < 0.001) and the number of sources consulted (χ2 = 254, P < 0.001). Vets/farriers were the source consulted most frequently for health advice (Z = ?13.32, P < 0.001), instructors for training advice (Z = ?3.93, P < 0.001), and books/magazines for advice regarding both behavior (Z = ?9.49, P < 0.001) and stable care (Z = ?8.26, P < 0.001). Despite the survey mode and that many participants were recruited online, Internet/forums were only the third to the fifth most used of the 7 information sources, depending on the subject area. Logistic regression analyses identified significant associations between owner demographics (age, self-rated skill level, and self-reported rider type) and the likelihood of selecting a particular source for advice regarding a specific subject area. The findings have important implications for those trying to reach the horse-owning population to disseminate research findings and ultimately improve the welfare of their horses. 相似文献

Combination effects of heavy metals and fluoranthene on soil bacteria 总被引:16,自引:0,他引:16
The effects of (1) Cd, Cu, Zn, and fluoranthene (FLA), separately applied, and (2) combinations of one of these heavy metals
with FLA on the growth of bacteria were studied in agar plate experiments. The bacteria were extracted from A horizons of
a Eutric Regosol and a Calcic Chernozem. Significant reductions of bacterial counts were observed for both soils at concentrations
> 1.0 mg Cd l–1, 0.5 mg Cu l–1, and 0.5 mg Zn l–1, respectively. Additions of FLA up to 100 mg l–1 did not result in increasing reductions of bacterial growth in the Regosol. Only 0.5, 2, and 100 mg FLA l–1 caused significant reductions of 22–27%. Bacterial counts were not affected by 0.2 mg FLA l–1. Low concentrations of heavy metals which were not affective when added separately were found to reduce bacterial growth
when applied in combination with 0.2 mg FLA l–1. At higher levels of heavy metals up to 2.5 mg l–1, addition of FLA also increased the toxicity of the metals. It is assumed that the enhancement of toxicity by FLA is due
to an alteration of the permeability of bacterial cell membranes.

How ion channels are gated to regulate ion flux in and out of cells is the subject of intense interest. The Escherichia coli mechanosensitive channel, MscS, opens to allow rapid ion efflux, relieving the turgor pressure that would otherwise destroy the cell. We present a 3.45 angstrom-resolution structure for the MscS channel in an open conformation. This structure has a pore diameter of approximately 13 angstroms created by substantial rotational rearrangement of the three transmembrane helices. The structure suggests a molecular mechanism that underlies MscS gating and its decay of conductivity during prolonged activation. Support for this mechanism is provided by single-channel analysis of mutants with altered gating characteristics. 相似文献

Cooling of blood platelets clusters the von Willebrand factor receptor complex. Macrophage alphaMbeta2 integrins bind to the GPIbalpha subunit of the clustered complex, resulting in rapid clearance of transfused, cooled platelets. This precludes refrigeration of platelets for transfusion, but the current practice of room temperature storage has major drawbacks. We document that alphaMbeta2 is a lectin that recognizes exposed beta-N-acetylglucosamine residues of N-linked glycans on GPIbalpha. Enzymatic galactosylation of chilled platelets blocks alphaMbeta2 recognition, prolonging the circulation of functional cooled platelets. Platelet-associated galactosyltransferase produces efficient galactosylation when uridine diphosphate-galactose is added, affording a potentially simple method for storing platelets in the cold. 相似文献
