Tests for free-living amoebae in water from 11 public fountains in Galicia were conducted at two seasons of the year, winter and summer. Only one fountain gave a negative result for the presence of amoebae in both samples. Twenty five strains were isolated, 14 belonging to the genus Acanthamoeba and 10 to the genus Naegleria. The Acanthamoeba strains were made up of the species A. polyphaga, A. quina, A. castellanii and A. paradivionensis. No relation was found between temperature, presence of free chlorine in the water and whether the water was drinkable and the presence or absence of amoebae, nor with the number of strains present in the water samples. The pathogenic capacity of the strains isolated from the genera Naegleria and Acanthamoeba was tested in vivo. It was found that of the 22 strains that would grow at 37 °C, 4 (3 strains of Acanthamoeba and 1 of Naegleria) caused the death of a statistically significant number of mice that had been inoculated intracerebrally, and the presence of amoebae was confirmed in the brains of all the animals inoculated. Intranasal inoculation caused less mice deaths than intracerebral inoculation, and less organs were found containing amoebae.  相似文献

Intermediate wheatgrass (IWG) (Thinopyrum intermedium) is a perennial grass with desirable agronomic traits and positive effects on the environment. It has high fiber and protein contents, which increase the interest in using IWG for human consumption. In this study, IWG flour was blended with refined wheat at four IWG‐to‐wheat ratios (0:100, 50:50, 75:25, and 100:0). Samples were analyzed for proximate composition, microstructure features, pasting properties (Micro Visco‐Amylo‐Graph device), protein solubility, and total and accessible thiols. Gluten aggregation properties (GlutoPeak tester) and mixing profile (Farinograph‐AT device) were also evaluated. IWG flour enrichment increased the pasting temperature and decreased the peak viscosity of blended flours. IWG proteins exhibited higher solubility than wheat, with a high amount of accessible and total thiols. The GlutoPeak tester highlighted the ability of IWG proteins to aggregate and generate torque. Higher IWG flour enrichment resulted in faster gluten aggregation with lower peak torque, suggesting weakening of wheat gluten strength. Finally, the addition of IWG to refined wheat flour resulted in a decrease in dough development time and an increase in consistency, likely because of the higher levels of fiber in IWG. The 50% IWG flour enrichment represents a good compromise between nutritional improvement and maintenance of the pasting properties, protein characteristics, and gluten aggregation kinetics.  相似文献

The use of a concentrate emulsion of Melia azedarach green fruits and a suspension of the fungus Beauveria bassiana was evaluated in the control of Rhipicephalus microplus on artificially infested cattle. The evaluation was conducted following the protocol established by the Brazilian Agriculture Ministry. Five groups of 4 or 5 animals were allocated to one of the following treatments: emulsion concentrate of M. azedarach at 0.25% (T AZED 0.25%), emulsion concentrate of M. azedarach at 0.5% (T AZED 0.5%), B. bassiana at 2.4 × 10(8) conidia (T BASS), association of the concentrate of M. azedarach at 0.25% with B. bassiana at 2.4 × 10(8) conidia (T AZED 0.25%+BASS), and control (untreated). The association of the two compounds provided better results than any one isolated treatment, indicating compatibility or perhaps a synergy between M. azedarach and B. bassiana. This treatment resulted in fewer engorged females (129 ± 70) than in the control group (233 ± 82), showing high performance against all developmental stages of the tick. Results revealed an apparent synergistic effect of M. azedarach and B. bassiana in the control of R. microplus that should be further investigated.  相似文献

Medicines and their metabolites have been found as water contaminants at very low concentrations; moreover, there is no extensive toxicological data to determine the risks associated with their occurrence in water resources. The ibuprofen genotoxicity potential to the Oreochromis niloticus fish (Tilapia), due to nanograms per liter exposure, was evaluated using the micronucleus test. Acute (48 h) and sub-chronic assays (10 days) were carried out at 300 ng/L ibuprofen aquatic concentration comparing with the negative control group (without treatment), with eight animals per group. The results were assessed from the average of triplicate analyses. The micronucleus frequency in peripheral blood of fish was determined using a sample size of 3,000 erythrocytes per animal. Significance was defined using t test (p????0.05). The bioassay results showed a statistically significant increase in the frequency of micronuclei for both exposure times in comparison to the negative control. The micronucleus frequency observed for the sub-chronic tests was higher than the one identified in the acute assays. The observed ibuprofen genotoxic effects demonstrated an aquatic environmental risk of this pharmaceutical, which occurs for the used fish experimental model in lower concentration than previously described for other aquatic organisms.  相似文献

The scanning tunneling microscope (STM) and the atomic force microscope (AFM) are scanning probe microscopes capable of resolving surface detail down to the atomic level. The potential of these microscopes for revealing subtle details of structure is illustrated by atomic resolution images including graphite, an organic conductor, an insulating layered compound, and individual adsorbed oxygen atoms on a semiconductor. Application of the STM for imaging biological materials directly has been hampered by the poor electron conductivity of most biological samples. The use of thin conductive metal coatings and replicas has made it possible to image some biological samples, as indicated by recently obtained images of a recA-DNA complex, a phospholipid bilayer, and an enzyme crystal. The potential of the AFM, which does not require a conductive sample, is shown with molecular resolution images of a nonconducting organic monolayer and an amino acid crystal that reveals individual methyl groups on the ends of the amino acids. Applications of these new microscopes to technology are demonstrated with images of an optical disk stamper, a diffraction grating, a thin-film magnetic recording head, and a diamond cutting tool. The STM has even been used to improve the quality of diffraction gratings and magnetic recording heads.  相似文献

In this study, we analyzed insulin-like growth factor I (IGF-I), growth hormone receptor (GHR) and uncoupling protein (UCP) mRNA expression in the muscle and liver of high- (0.23 g/g) and low- (0.17 g/g) feed-efficiency (FE) Japanese quail at three different air temperatures: comfortable (25 °C), heat stress (38 °C) for 12 h or cold stress (10 °C) for 12 h. Total RNA was extracted from the liver and breast muscle of each quail, and cDNA was amplified using specific primers for the target genes. Expression was analyzed using quantitative real-time PCR (qRT-PCR). IGF-I mRNA expression was higher in the livers of high-FE quail than in the livers of low-FE quail under both heat and cold stress conditions. High-FE birds also showed higher GHR mRNA expression independent of temperature. UCP mRNA expression in the liver was lower in high-FE birds and higher under heat stress compared with the other conditions. IGF-I mRNA expression was higher in the muscle of high-FE quail under the three conditions tested, and UCP mRNA expression was higher under cold stress. Our results suggest that air temperature affects the expression of genes related to growth and mitochondrial energy production, and quail with different feed efficiencies respond differently to environmental stimuli.  相似文献

Background – In humans, thymic stromal lymphopoietin (TSLP) plays a central role in the development of allergic inflammation, such as atopic dermatitis (AD), but it is unknown whether it is involved in the pathogenesis of canine AD (CAD). Hypothesis/Objectives – Our aim was to characterize canine TSLP and to assess its expression in CAD. Methods – Canine TSLP was identified based on sequence homology with human TSLP and the complementary DNA (cDNA) cloned by RT‐PCR. Real‐time quantitative RT‐PCR was established to assess the expression of canine TSLP in cultured canine keratinocytes and in skin biopsy specimens from lesional and nonlesional skin of 12 dogs with CAD and eight healthy control dogs. Results – Partial canine TSLP cDNA was cloned and characterized. It contained four exons that shared 70 and 73% nucleotide identity with human and equine TSLP, respectively, encoding the signal peptide and full‐length secreted protein. We found significantly increased TSLP expression in lesional and nonlesional skin of dogs with CAD compared with healthy control dogs (P < 0.05), whereas no difference was measured between lesional and nonlesional samples. In cultured primary canine keratinocytes, we found increased TSLP expression after stimulation with house dust mite allergen extract or Toll‐like receptor ligands lipopolysaccharide and poly I:C. Conclusions and clinical importance – Increased TSLP expression in the skin of dogs with CAD supports an involvement of TSLP in the pathogenesis of CAD similar to that in humans. Further studies should elucidate the function and therapeutic potential of TSLP in CAD.  相似文献

The effectiveness of seeding introduced species, with or without straw mulching, was tested as a measure of post-fire erosion control in gypsiferous (Xeric Haplogypsid) and calcareous soils (Xeric Torriorthent) in Central Ebro Valley (NE-Spain). Paired control, seeding, and combined seeding and mulching plots were established in four replicated plots for each soil tested. Seeding rate was 30 g m -2 and straw mulch was applied at 100 g m -2 in each plot. Plant projective cover (total and specific), plant biomass, bare soil cover and sediment yield were determined over a 2-year period. During the first year of sampling, species introduced by seeding increased plant cover (about 30%) without significant differences observed between soils. Plant cover was similar in seeding-only and seeding-mulching treatments, although the latter treatment significantly enhanced plant weight. During the second year these differences disappeared because species introduced by seeding practically did not survive. So, the introduced herbs did not interfere with native plants. Bare soil cover remained significantly lower in treated plots than on the control plots for both soils during both years of sampling. In the second year, the reduction of bare soil was attributed to both the straw mulch and litter from the seeding species. Soil protection was significantly higher in calcareous soils than in gypsiferous soils which is related to their physical and chemical properties. Cumulative sediment yield decreased significantly over time with both treatments on both soils. Soil losses from control plots were three times higher than seeding plots and 3.3 times higher than from seeding and mulching plots in gypsiferous soils. Soil losses from control plots were two times higher than from seeding plots and 2.7 times higher than the combined seeding and mulching plots in the calcareous soils. Cumulative soil loss was higher from the gypsiferous soils than from the calcareous soils due to the lower plant cover.  相似文献

The microbiological quality of 18 commercially available in Spain ready-to-eat fish products containing Engraulidae was evaluated through application of the corresponding ISO procedures for total mesophilic aerobic microbial counts, detection and enumeration of enterobacteria, and detection of Staphylococcus spp. All isolates were identified to the species level using two different biochemical methods: the API® test and the Biolog® identification system. The most commonly occurring contaminants found were Enterobacteriaceae—such as Citrobacter freundii and other Citrobacter species, Enterobacter cloacae, Cronobacter sakazakii, Hafnia alvei, Pantoea, Proteus ssp., and Escherichia coli. The presence of such opportunistic pathogens and contaminant microflora was confirmed in 61% of the foods sampled.  相似文献
