The activity of exotoxins produced by four strains of Corynebacterium pseudotuberculosis were compared by their ability to kill white mice, their haemolytic activity, staphylococcal haemolysin-inhibiting effect and activity in an enzyme-linked immunosorbent assay (ELISA). Exotoxins with a haemolytic titre of 1 in 256 or more killed all mice and had the most inhibitory effect (1 in 64 or more) on staphylococcal haemolysin. The haemolytic test and staphylococcal haemolysin inhibitory test of exotoxin activities were highly correlated (P less than 0.001) with the mouse toxicity of the four exotoxins examined. No significant correlation was evident between the ELISA and relative toxicity of the exotoxins. 相似文献

In arid shrublands, soil resources are patchily distributed around shrub canopies, forming well-studied “islands of fertility.” While soil nutrient patterns have previously been characterized quantitatively, we develop a predictive model that explicitly considers the distance from shrubs of varying canopy sizes. In 1-ha macroplots in both the Sonoran and Mojave Deserts, we used Plant Root Simulator? probes to measure nutrient availability along transects extending north and south from creosote bushes (Larrea tridentata). We modeled the decline of nutrients with distance from focal shrubs using hierarchical mixed models that included the effects of transect direction and shrub canopy size. Of the nutrients considered, nitrogen and potassium had the strongest response to distance from focal shrubs. In the Sonora, both depended on canopy size and had different patterns to the north versus the south. In the Mojave, potassium depended on size and direction, but nitrogen only on canopy size. We used the fitted model equations and the location and canopy size of all Larrea shrubs within the macroplots to estimate nutrient concentrations at a 20 cm resolution. This produced maps showing nutrient “hotspots” centered on Larrea. Our models predicted up to 60 % of the variation in nutrient availability the following growing season. Our models efficiently used a moderate number of sample locations to predict nutrient concentrations over a large area, given easily measured values of shrub size and location. Our method can be applied to many systems with patchily distributed resources focused around major structural landscape features. 相似文献

A 3 year old intact male pygmy goat developed progressive weakness and eventual recumbancy over the course of 1 week, while maintaining its ability to eat and drink. The animal died and at necropsy, the parietal pleural surfaces and the pericardial surface were noted to be covered with firm, white, variably sized nodules that often formed linear arrays or coalesced into larger clumped aggregates. The visceral pleural surfaces of the ventral lung lobes were also covered with similar nodules. Histopathological and immunohistochemical evaluation of the submitted tissues revealed a diagnosis of mesenchymal chondrosarcoma with extensive seeding throughout the thoracic cavity. 相似文献

Emissions of low molecular weight aldehydes (LMWAs) from deep-frying of extra virgin olive oil, olive oil, and canola oil (control) were investigated at two temperatures, 180 and 240 degrees C, for 15 and 7 h, respectively. The oil fumes were collected in Tedlar bags and then analyzed by gas chromatography-mass spectrometry. Seven alkanals (C2-C7 and C9), eight 2-alkenals (C3-C10), and 2,4-heptadienal were found in the fumes of all three cooking oils. The generation rates of these aldehydes were found to be dependent on heating temperature, showing significant increases with increases in temperature. The LMWA emissions from both kinds of olive oils were very similar and were lower than those observed from canola oil under similar conditions. These results suggest that frying in any type of olive oil, independent of its commercial category, will effectively decrease the generation of volatile aldehydes in the exhaust. This fact is important because less expensive refined olive oil is usually used for deep-frying operations, whereas extra virgin olive oil is usually used as salad dressing. 相似文献

An astronomical digital camera is equipped with a fish-eye lens and exchangeable filters to study the fine angular structure of radiation field in forests in the red and near infrared (NIR) spectral bands. A few examples of the angular distribution of downward radiances in conifer and broadleaf forests are given. The results of field measurements are compared with the spectral flux measurements by a field radiometer and with the calculations of radiance by a combined geometric-optical/radiative transfer (RT) model. The objective of this study was to determine whether the linear regression (LR) method could be used to validate genomic threshold models. Statistics for the LR method were computed from estimated breeding values (EBVs) using the whole and truncated data sets with variances from the reference and validation populations. The method was tested using simulated and real chicken data sets. The simulated data set included 10 generations of 4,500 birds each; genotypes were available for the last three generations. Each animal was assigned a continuous trait, which was converted to a binary score assuming an incidence of failure of 7%. The real data set included the survival status of 186,596 broilers (mortality rate equal to 7.2%) and genotypes of 18,047 birds. Both data sets were analysed using best linear unbiased predictor (BLUP) or single‐step GBLUP (ssGBLUP). The whole data set included all phenotypes available, whereas in the partial data set, phenotypes of the most recent generation were removed. In the simulated data set, the accuracies based on the LR formulas were 0.45 for BLUP and 0.76 for ssGBLUP, whereas the correlations between true breeding values and EBVs (i.e. true accuracies) were 0.37 and 0.65, respectively. The gain in accuracy by adding genomic information was overestimated by 0.09 when using the LR method compared to the true increase in accuracy. However, when the estimated ratio between the additive variance computed based on pedigree only and on pedigree and genomic information was considered, the difference between true and estimated gain was <0.02. Accuracies of BLUP and ssGBLUP with the real data set were 0.41 and 0.47, respectively. This small improvement in accuracy when using ssGBLUP with the real data set was due to population structure and lower heritability. The LR method is a useful tool for estimating improvements in accuracy of EBVs due to the inclusion of genomic information when traditional validation methods as k‐fold validation and predictive ability are not applicable. 相似文献

Canine oral melanoma (COM) is the most frequent tumour with oral localization in dogs. Copy number gains and amplifications of CCND1, a gene coding for Cyclin D1, are the most frequent chromosomal aberrations described in human non‐UV induced melanomas. Twenty‐eight cases of COM were retrieved from paraffin‐blocks archives. A total of 4 μm thick sections were immunostained with an antibody against human Cyclin D1 and Ki‐67. Cyclin D1 and Ki‐67 expressions were scored through two counting methods. DNA was extracted from 20 μm thick sections of formalin‐fixed paraffin‐embedded blocks. Pathological and surrounding healthy tissue was extracted independently. Cyclin D1 immunolabelling was detected in 69% (18/26) while Ki‐67 was present in 88.5% (23/26) of cases. Statistical analysis revealed correlation between two counting methods for Cyclin D1 (r = 0.54; P = .004) and Ki‐67 (r = 0.56; P = .003). The correlation found between Ki‐67 and Cyclin D1 indexes in 16/26 cases labelled by both antibodies (r = 0.7947; P = .0002) suggests a possible use of Cyclin D1 index as prognostic marker. Polymerase chain reaction analysis on CCND1 coding sequence revealed the presence of nine somatic mutations in seven samples producing synonymous, missense and stop codons. Since none of the single‐nucleotide polymorphisms was found to be recurrent, it is suggested that overexpression of Cyclin D1 may be the consequence of alterations of CCND1 upstream regions or other genetic aberrations not detectable with the methodology used in this study. Future studies are needed to verify the potential use of Cyclin D1 index as prognostic indicator and to highlight the molecular events responsible for Cyclin D1 overexpression in COMs. 相似文献

Beginning in April 2009, a novel H1N1 influenza virus caused acute respiratory disease in humans, first in Mexico and then around the world. The resulting pandemic influenza A H1N1 2009 (pH1N1) virus was isolated in swine in Canada in June 2009 and later in breeder turkeys in Chile, Canada, and the United States. The pH1N1 virus consists of gene segments of avian, human, and swine influenza origin and has the potential for infection in poultry following exposure to infected humans or swine. We examined the clinical events following the initial outbreak of pH1N1 in turkeys and determined the relatedness of the hemagglutinin (HA) gene segments from the pH1N1 to two H1N1 avian influenza (AI) isolates used in commercial turkey inactivated vaccines. Overall, infection of turkey breeder hens with pH1N1 resulted in -50% reduction of egg production over 3-4 weeks. Genetic analysis indicated one H1N1 AI vaccine isolate (Alturkey/North Carolina/17026/1988) contained approximately 92% nucleotide sequence similarity to the pH1N1 virus (A/Mexico/4109/2009); whereas, a more recent AI vaccine isolate (A/ swine/North Carolina/00573/2005) contained 75.9% similarity. Comparison of amino acids found at antigenic sites of the HA protein indicated conserved epitopes at the Sa site; however, major differences were found at the Ca2 site between pH1N1 and A/ turkey/North Carolina/127026/1988. Hemagglutinin-inhibition (HI) tests were conducted with sera produced in vaccinated turkeys in North Carolina to determine if protection would be conferred using U.S. AI vaccine isolates. HI results indicate positive reactivity (HI titer > or = 5 log2) against the vaccine viruses over the course of study. However, limited cross-reactivity to the 2009 pH1N1 virus was observed, with positive titers in a limited number of birds (6 out of 20) beginning only after a third vaccination. Taken together, these results demonstrate that turkeys treated with these vaccines would likely not be protected against pH1N1 and current vaccines used in breeder turkeys in the United States against circulating H1N1 viruses should be updated to ensure adequate protection against field exposure. 相似文献

Whiteflies are an insect group that comprises multiple species and biotypes, capable of affecting crops by phloem feeding,
virus transmission and promotion of fungal colonization. The distribution of these pests is worldwide. In Costa Rica, a country
located in the tropics, the most problematic whiteflies are Bemisia tabaci biotype B and Trialeurodes vaporariorum. In September 2009, two greenhouses in the Alfaro Ruiz region, northwest of the country’s capital, San Jose, were surveyed
as part of a larger effort to determine the occurrence of species and races of whiteflies in this agronomically important
region. In addition, the insect samples were analyzed to determine the presence of Tomato chlorosis virus (ToCV), a yield-affecting crinivirus transmitted by whiteflies. The results revealed the presence of the Q biotype of B. tabaci, and important invasive species, as well as the expected T. vaporariorum. Viral detection assays identified potentially viruliferous individuals for Tomato chlorosis virus. These results identified a new pest capable of harbouring plant viruses has been identified, as well as a viral agent (ToCV)
