Ammonium (NH ) nutrition causes retardation of growth in many plant species. In Arabidopsis grown with NH as the sole N source, growth retardation occurs already at early stages before photosynthesis has come to its full power. In order to describe the peculiarities of these retarded plants, they were compared with nitrate (NO )‐grown plants of the same age of 15 d. Photosynthetic activity as measured by CO2 uptake per unit chlorophyll is half as high in NH ‐grown seedlings as in NO ‐grown ones. This finding is confirmed by the analysis of chlorophyll fluorescence. Chloroplasts of NO ‐grown, but not of NH ‐grown, seedlings show starch deposits after 5 h of illumination with 40 μmol m–2 s–1. Gene‐expression analysis based on cDNA microarray and on Northern blots provide a clue about the biochemical background. After the first 2 weeks of growth, it seems that NO ‐grown seedlings subsist mainly on normal photosynthesis, whereas NH ‐grown seedlings still use lipids from the seeds stored in oleosomes. Corresponding to this observation, the mRNAs for enzymes of β‐oxidation are more strongly expressed in NH ‐grown seedlings. Different carbohydrate sources for sucrose synthesis are indicated by different gene expression. Higher gene expression of fructose bisphosphate aldolase (cytosolic isoform) in NO ‐grown seedlings indicates the dependence on photosynthesis, whereas a higher gene expression of PEP carboxykinase in NH ‐grown seedlings points to a prominent role of β‐oxidation of storage lipids still present.  相似文献

Exogenous pigments produced by ascomycetous filamentous fungi belonging to the genera Penicillium, Epicoccum, and Monascus, preselected based on chemotaxonomic knowledge, have been extracted and characterized by quantitative colorimetry. The color characteristics of the fungal extracts were compared to water soluble natural colorants derived from sources currently in use. The tested fungal extracts also included some commercially available Monascus colorants. The a values for the fungal extracts were found to be both positive and negative, the b values were found to be positive, while the hue angles of the fungal color extracts ranged from 40 to 110 indicating the color distribution of fungal extracts over the red-orange-yellow region of the CIELAB color space. The fungal extracts exhibited additional color hues in the red spectrum and similar hues in the yellow spectrum as compared to the reference natural colorants. They were also found to be similar or brighter in terms of chroma to some of the reference natural colorants. Principal component analysis was performed to group and distinguish different colors based on the a and b values. The fungal color extracts could be grouped in accordance with the similarity or difference in the color to those of the existing natural colorants. The diversity of colors was not only found among different fungal genera and/or species but also within the same species on changing the media. There was a marked change in the color composition of the extracts resulting in relatively different hues. Our results, thus, indicate that there exists pigment-producing genera of ascomycetous fungi other than Monascus that produce color shades in the red and the yellow spectra in addition or similar to reference colorants. These color shades could add to the color palette of the natural colorants currently in use. In addition, the multivariate approach in distinguishing and classifying the colorants was shown to be a very useful tool in colorimetric comparison of colorants.  相似文献

Since the mid-1990s, the cultivation of white lupin (Lupinus albus L.) has rapidly decreased in Germany and other countries in Central Europe because of the occurrence of Colletotrichum lupini, the causal agent of the fungal disease anthracnose, and the lack of varieties with a sufficient resistance towards this disease. To reestablish the relevance of cultivating white lupins, the development of new varieties with improved resistance is vital. In the study presented here, new breeding lines were evaluated from 2012 to 2014 on a total of five experimental sites in Germany. In each year, at least on one site a high disease pressure with good differentiation built up from natural seed infestations, so that evaluation and further selection for resistance was possible in all 3 years. The breeding lines showed improved performance of resistance towards C. lupini (anthracnose disease score 3.8 on average of all breeding lines and environments) compared to the reference varieties (anthracnose disease score 5.8). Improved resistance had a favorable effect on grain yield, particularly in environments with high disease pressure, and yield stability. Average grain yields were 2.6 t ha?1 for the breeding lines and 1.5 t ha?1 for the reference cultivars. Among the tested range of white lupins, the breeding material showed consistently higher grain yields on average of all environments compared to the rather low and variable yielding reference cultivars. The alkaloid content varied among environments and breeding lines.  相似文献

Background

Infections with bovine respiratory syncytial virus (BRSV) and bovine coronavirus (BoCV) are endemic to the cattle populations in most countries, causing respiratory and/or enteric disease. It has been demonstrated that herds can remain free from these infections for several years also in high prevalence areas. Organically managed (OM) dairy herds have been shown to have lower seroprevalence of both viruses compared to conventionally managed (CM) herds. The objective of this study was to challenge the hypothesis of a lower occurrence of BRSV and BoCV in OM compared to CM dairy herds.In November 2011, May 2012 and May 2013 milk samples from four homebred primiparous cows were collected in 75 to 65 OM and 69 to 62 CM herds. The antibody status regarding BRSV and BoCV was analysed with commercial indirect ELISAs. Herds were classified as positive if at least one individual sample was positive.

Results

The prevalence of positive herds ranged from 73.4% to 82.3% for BRSV and from 76.8% to 85.3% for BoCV among OM and CM herds, over the three sampling occasions. There was no statistically significant difference between OM and CM herds at any sampling occasion. The incidence risk of newly infected herds did not differ statistically between OM and CM herds at any sampling occasion, neither for BRSV nor for BoCV. The incidence of herds turning sero-negative between samplings corresponded to the incidence of newly infected. Bulk tank milk (BTM) samples were also sampled in the herds and analysed. Several herds were negative on individual samples but positive in BTM. Herd-level data on production, health and reproduction were retrieved from VÄXA Sweden and the study herds were representative of the source population.

Conclusion

There was no difference in prevalence of or incidence risk for BRSV or BoCV between Swedish OM and CM herds. Because the incidence of herds becoming seropositive was balanced by herds becoming seronegative it should be possible to lower the prevalence of these two infections among Swedish dairy cattle herds if biosecurity is improved.

Current techniques for three-dimensional (3D) optical microscopy (deconvolution, confocal microscopy, and optical coherence tomography) generate 3D data by "optically sectioning" the specimen. This places severe constraints on the maximum thickness of a specimen that can be imaged. We have developed a microscopy technique that uses optical projection tomography (OPT) to produce high-resolution 3D images of both fluorescent and nonfluorescent biological specimens with a thickness of up to 15 millimeters. OPT microscopy allows the rapid mapping of the tissue distribution of RNA and protein expression in intact embryos or organ systems and can therefore be instrumental in studies of developmental biology or gene function.  相似文献

Group A streptococci, a common human pathogen, secrete streptokinase, which activates the host's blood clot-dissolving protein, plasminogen. Streptokinase is highly specific for human plasminogen, exhibiting little or no activity against other mammalian species, including mouse. Here, a transgene expressing human plasminogen markedly increased mortality in mice infected with streptococci, and this susceptibility was dependent on bacterial streptokinase expression. Thus, streptokinase is a key pathogenicity factor and the primary determinant of host species specificity for group A streptococcal infection. In addition, local fibrin clot formation may be implicated in host defense against microbial pathogens.  相似文献

Climate variations influenced the agricultural productivity, health risk, and conflict level of preindustrial societies. Discrimination between environmental and anthropogenic impacts on past civilizations, however, remains difficult because of the paucity of high-resolution paleoclimatic evidence. We present tree ring-based reconstructions of central European summer precipitation and temperature variability over the past 2500 years. Recent warming is unprecedented, but modern hydroclimatic variations may have at times been exceeded in magnitude and duration. Wet and warm summers occurred during periods of Roman and medieval prosperity. Increased climate variability from ~250 to 600 C.E. coincided with the demise of the western Roman Empire and the turmoil of the Migration Period. Such historical data may provide a basis for counteracting the recent political and fiscal reluctance to mitigate projected climate change.  相似文献
