ABSTRACT The basidiomycete Oncobasidium theobromae was identified as the cause of a devastating disease of cacao named vascular-streak dieback (VSD) in Papua New Guinea in the 1960s. VSD now causes losses among cacao seedlings and kills branches in mature cacao trees throughout Southeast Asia and parts of Melanesia. The characteristic symptoms include a green-spotted chlorosis and fall of leaves beginning on the second or third flush behind the stem apex, raised lenticels, and darkening of vascular traces at the leaf scars and infected xylem. Eventually complete defoliation occurs and, if the fungus spreads to the trunk, the tree will die. O. theobromae is a highly specialized, near-obligate parasite of cocoa. It is a windborne, leaf-penetrating, vascular pathogen, and may have evolved as an endophyte on an as yet unidentified indigenous host. The rate of disease spread on cocoa is limited because basidiocarps develop only on fresh leaf scars during wet weather, and basidiospores remain viable for a few hours on the night they are shed. Consequently, very few new infections occur beyond 80-m from diseased trees. Transmission of the disease through seed or infected cuttings has not been demonstrated. Strict quarantine measures applied to the movement of intact plants are crucial in reducing spread of the disease. Integrated management, including the planting of less susceptible genotypes, nursery construction and management, canopy pruning and shade management, provides effective control.  相似文献

The spotted-wing drosophila, Drosophila suzukii Matsumura, is an invasive pest causing significant damage to soft skinned fruits. Control of D. suzukii is critical since there is no tolerance for infested fruit in the market. While most insecticides control one or more D. suzukii life-stages (e.g., egg, larvae, and adult), the impact of insecticides that are toxic to immature stages  is unclear on the subsequent generation of a field population. Insecticides were applied at field recommended rates on cherries and blueberries in the laboratory to determine immature D. suzukii mortality. Spinetoram, cyantraniliprole, malathion, methomyl, spinosad, and phosmet resulted in relatively high mortality of all immature life stages. Zeta-cypermethrin, cyclaniliprole, and fenpropathrin resulted in lower mortality of egg and all larval instars. Malathion was also applied to lowbush blueberries with different fruit sizes (small, medium, and large) in the laboratory and there was no statistical difference in mortality rates depending on fruit sizes. Mortality data from the laboratory experiments were used to parameterize a refined D. suzukii population model. The model revealed that the timing and order of different insecticide classes are important to control D. suzukii population. Model runs that included early applications of more effective insecticides resulted in high immature mortality and greater reduction of D. suzukii populations compared to treatments applied later.

Studies were conducted to determine the potential for large scale culture of the mysid shrimp Mysidopsis almyra. Reproduction was consistent, as newly hatched mysids were always present in the culture trays. At the end of 45 day preliminary trials, the populations in the culture trays had increased 323.3% and 256.6%. A larger pilot-scale system connected to a biological filtration tank was constructed and operated for 17 weeks. Two rectangular trays (125 cm × 50 cm × 8 cm deep) were placed one above the other; water volume in each tray was 20 l. The room was kept dark. Temperatures were maintained at 26(2) °C and salinities at 20(2). A total of 1,000 adult mysids were placed in the culture tray and the hatchlings were collected and moved into a hatchling tray. Water circulation was static except for twice-daily water exchanges; newly hatched Artemia nauplii (24 h incubation) were fed to the mysids immediately after each water exchange. Feeding presented no technical problem to the pilot-scale culture of mysids in static water systems but the cost of Artemia did represent the largest expense. Mean (SD) mysid production throughout the 17 weeks of the trial was 133(69) hatchlings d–1 with highest production [244(30) hatchlings d–1] occurring between weeks 11 and 13. Mean survival in the hatchling tray after the 14 day growth periods was 41.4%. Reproduction occurred at ammonia-nitrogen and nitrite-nitrogen concentrations as high as 1.5 mg l–1 and 0.250 mg l–1 respectively, and at pH values as low as 7.6. Landscape and site-scale data analyses aid the interpretation of biological data and thereby help us develop more cost-effective natural resource management strategies. Our study focused on environmental influences on stream assemblages and we evaluated how three classes of environmental variables (geophysical landscape, land use and cover, and site habitat), influence fish and macroinvertebrate assemblage richness in the Brazilian Cerrado biome. We analyzed our data through use of multiple linear regression (MLR) models using the three classes of predictor variables alone and in combination. The four MLR models explained dissimilar amounts of benthic macroinvertebrate taxa richness (geophysical landscape R 2 ≈ 35 %, land use and cover R 2 ≈ 28 %, site habitat R 2 ≈ 36 %, and combined R 2 ≈ 51 %). For fish assemblages, geophysical landscape, land use and cover, site habitat, and combined models explained R 2 ≈ 28 %, R 2 ≈ 10 %, R 2 ≈ 31 %, and R 2 ≈ 47 % of the variability in fish species richness, respectively. We conclude that (1) environmental variables differed in the degree to which they explain assemblage richness, (2) the amounts of variance in assemblage richness explained by geophysical landscape and site habitat were similar, (3) the variables explained more variability in macroinvertebrate taxa richness than in fish species richness, and (4) all three classes of environmental variables studied were useful for explaining assemblage richness in Cerrado headwater streams. These results help us to understand the drivers of assemblage patterns at regional scales in tropical areas.  相似文献

The hatching envelope elevated around the spawned eggs of penaeoidean eggs presents a formidable barrier for manipulation of the embryo. By spawning eggs into seawater containing 3-amino-1,2,4-triazole and subsequently passing the eggs through Nitex screening approximately 45 min after spawning, the hatching envelope can be effectively removed with little or no damage to the embryos. The time and extent to which the envelope is removed can be varied. Embryos treated in this manner continue to develop and the method provides a new technique with which investigators can gain access to the surface of the egg or embryo.  相似文献

This study evaluated wood chips and wheat straw as inexpensive and readily available alternatives to more expensive plastic media for denitrification processes in treating aquaculture wastewaters or other high nitrate waters. Nine 3.8-L laboratory scale reactors (40 cm packed height × 10 cm diameter) were used to compare the performance of wood chips, wheat straw, and Kaldnes plastic media in the removal of nitrate from synthetic aquaculture wastewater. These upflow bioreactors were loaded at a constant flow rate and three influent NO3–N concentrations of 50, 120, and 200 mg/L each for at least 4 weeks, in sequence. These experiments showed that both wood chips and wheat straw produced comparable denitrification rates to the Kaldnes plastic media. As much as 99% of nitrate was removed from the wastewater of 200 mg NO3–N/L influent concentration. Pseudo-steady state denitrification rates for 200 mg NO3–N/L influent concentrations averaged (1360 ± 40) g N/(m3 d) for wood chips, (1360 ± 80) g N/(m3 d) for wheat straw, and (1330 ± 70) g N/(m3 d) for Kaldnes media. These values were not the maximum potential of the reactors as nitrate profiles up through the reactors indicated that nitrate reductions in the lower half of the reactors were more than double the averages for the whole reactor. COD consumption per unit of NO3–N removed was highest with the Kaldnes media (3.41–3.95) compared to wood chips (3.34–3.64) and wheat straw (3.26–3.46). Effluent ammonia concentrations were near zero while nitrites were around 2.0 mg NO2–N/L for all reactor types and loading rates. During the denitrification process, alkalinity and pH increased while the oxidation–reduction potential decreased with nitrate removal.

Wood chips and wheat straw lost 16.2% and 37.7% of their masses, respectively, during the 140-day experiment. There were signs of physical degradation that included discoloration and structural transformation. The carbon to nitrogen ratio of the media also decreased. Both wood chips and wheat straw can be used as filter media for biological denitrification, but time limitations for the life of both materials must be considered.  相似文献

Soil pH is one of the most influential variables in soil, and is a powerful factor in influencing the size, activity and community structure of the soil microbial community. It was previously shown in a century old artificial pH gradient in an arable soil (pH 4.0-8.3) that bacterial growth is positively related to pH, while fungal growth increases with decreasing pH. In an attempt to elucidate some of the mechanisms for this, plant material that especially promotes fungal growth (straw) or bacterial growth (alfalfa) was added to soil samples of the pH gradient in 5-day laboratory incubation experiments. Also, bacterial growth was specifically inhibited by applying a selective bacterial growth inhibitor (bronopol) along the entire pH gradient to investigate if competitive interaction caused the shift in the decomposer community along the gradient. Straw benefited fungal growth relatively more than bacterial, and vice versa for alfalfa. The general pattern of a shift in fungal:bacterial growth with pH was, however, unaffected by substrate additions, indicating that lack of a suitable substrate was not the cause of the pH effect on the microbial community. In response to the bacterial growth inhibition by bronopol, there was stimulation of fungal growth up to pH 7, but not beyond, both for alfalfa and straw addition. However, the accumulation of ergosterol (an indicator of fungal biomass) during the incubation period after adding alfalfa increased at all pHs, indicating that fungal growth had been high at some time during the 5-day incubation following joint addition of alfalfa and bronopol. This was corroborated in a time-series experiment. In conclusion, the low fungal growth at high pH in an arable soil was caused to a large extent by bacterial competition, and not substrate limitation.  相似文献
