An integral component of breeding maize for resistance to Fusarium graminearum ear rot is the identification of resistant genotypes. Since natural infection is not consistent from year to year, maize researchers must use manual techniques to inoculate the plant material with fungal spores. Information is presented here on site resistance of commercial maize hybrids to F. graminearum over three years and at two locations. Additionally, results of an investigation on the two predominant techniques of inoculating maize, the silk channel and kernel inoculation methods, are reported. Of 61 commercial hybrids tested, only two were ranked as moderately resistant to the fungus by both inoculation methods. These two hybrids also had a stable response to the F. graminearum infection across seven environments when the silk channel inoculation method was used. The majority of the hybrids were ranked as either susceptible or highly susceptible and less than 10% of the hybrids had a stable response to fungal infection. In the investigation of methodology, it was concluded that silk browning would be the least laborious way to identify the ideal time to complete silk channel inoculations. It was found that kernel inoculations using the pin inoculation method should take place between 11 and 15 days after 50% silking to achieve proper hybrid discrimination. Stereotypies are invariant and repetitive behaviour patterns that seemingly have no function, which tend to develop in captive animals faced with insoluble problems and may be indicative of reduced welfare. A 4 year prospective study of the factors influencing the development of stereotypic and redirected behaviours (abnormal behaviour) in a population of 225 young Thoroughbred and part-Thoroughbred horses was conducted between 1995 and 1999. Abnormal behaviour affected 34.7% of the population. Multivariable analysis showed that foals of low- or middle-ranking mares were less likely to develop abnormal behaviour than foals of dominant mares (rate ratio (RR) 0.23, P<0.01; RR 0.48, P<0.01, respectively). Weaning by confinement in a stable or barn was associated with an increased rate of development of abnormal behaviour, compared with paddock-weaning (RR 2.19, P<0.05), and housing in barns, rather than at grass after weaning, was associated with a further increase (RR 2.54, P<0.01). Specific stereotypic and redirected behaviours were then considered as separate outcomes. Crib-biting was initiated by 10.5% of horses at median age 20 weeks, weaving by 4.6% of horses at median age 60 weeks, box-walking by 2.3% of horses at median age 64 weeks and wood-chewing by 30.3% of horses at median age 30 weeks. Wood-chewing developed at a lower rate in horses born to subordinate or mid-ranking mares than in horses born to dominant mares (RR 0.29, P<0.01; RR 0.41, P<0.01, respectively), and at a higher rate in horses kept in barns or stables rather than at grass after weaning (RR 4.49, P<0.001; RR 1A6, P<0.001, respectively). Feeding concentrates after weaning was associated with a 4-fold increase in the rate of development of crib-biting (RR 4.12, P = 0.02). The results of this study support the idea that simple changes in feeding, housing and weaning practices could substantially lower the incidence of abnormal behaviour in young horses. 相似文献
1. In commercial free-range systems for laying hens, popholes to the outdoor range are often installed on one side of the house only. In multi-tier systems, it is possible that some individuals fail to access the range due to internal barriers to movement.
2. Five commercial multi-tier flocks from different units were studied. For each flock, two different colour markers were used to distinguish 200 birds roosting near the popholes (NP-Roost) and 200 birds roosting far from the popholes (FP-Roost) at night. The following day, counts of marked birds on the range and inside the house were performed.
3. Significantly more NP-Roost birds were observed in all areas of the outdoor range than FP-Roost birds the next day. Distance of FP area from the popholes was very strongly positively correlated with effect size in the adjacent range area.
4. Additionally, in the indoor area far from the popholes (FP) more FP-Roost birds were observed the next day than NP-Roost birds. In the indoor area near to the popholes (NP) more NP-Roost birds were observed the next day than FP-Roost birds.
5. These results suggest that roosting location is associated with differential range use when popholes are only available on one side of the shed as birds that roosted far from the popholes used the range less. 相似文献
To determine sevoflurane’s minimum alveolar concentration (MACSEVO) and its cardiopulmonary effects in sheep.
Study design
Prospective experimental study.
Animals
A group of 10 female nonpregnant Sardinian milk sheep.
Methods
Anesthesia was induced in each sheep twice with sevoflurane in oxygen. After a 30 minute equilibration at end-tidal sevoflurane concentration (Fe?Sevo) of 2.8%, an electrical stimulus (5 Hz/1 ms/50 mA) was applied to the right thoracic limb for 1 minute or until gross purposeful movement occurred. The Fe?Sevo was then changed using a 0.2% up-and-down protocol, dependent on whether or not the response was positive, and then noxious stimulation was repeated. The MACSEVO was defined as the mean Fe?Sevo between that allowing purposeful movement and that not. The group of 10 sheep were re-anesthetized and MACSEVO was re-determined. Thereafter, Fe?Sevo was maintained for 15 minutes each at concentrations corresponding to 1.0, 1.3, 1.6, 1.9 and 0.75 MACSEVO multiples, and cardiopulmonary, blood gas, acid-base variables and plasma electrolytes were determined. Also, time to induction of anesthesia, extubation and recovery were recorded.
Results
The mean ± standard deviation of the MACSEVO was 2.74 ± 0.38%. Median (interquartile range) time to intubation was 3.13 (2.98–3.33) minutes, time to extubation was 6.85 ± 2.65 minutes and time to recovery was 13.4 ± 5.2 minutes. With increasing Fe?Sevo, arterial blood pressures progressively decreased as did minute ventilation, which in turn caused end-tidal carbon dioxide, arterial partial pressure of carbon dioxide and bicarbonate values to steadily increase without significantly affecting arterial partial pressure of oxygen.
Conclusions and clinical relevance
The reported MACSEVO agrees with published data in this and other species. Administration of sevoflurane in sheep caused marked hemodynamic and respiratory depression, but soon after turning off the vaporizer, sheep could be extubated and recovered rapidly and event-free. 相似文献
Toumeyella parvicornis is an invasive soft scale insect native to North America that is rapidly spreading in Italy and France, provoking severe infestations on Pinus pinea L. To date, the control of this pest is entrusted to three endotherapic techniques whose short-term efficacy is partially known. No information on long-term efficacy is currently available, although fundamental. This work aims to report on the long-term effect that abamectin-based insecticides, injected with the three different techniques, have on adult female populations.
Results
The study was carried out in an infested P. pinea forest in the area of Rome, Italy. Results showed that the tested methods had a similar long-term effect, and only in one case there were differences with the untreated control. Multiresidue analysis reported a zero level of abamectin in plant tissues 14 months apart from injection, except for one treatment where pesticide concentration was just above the limit of quantification.
Mouse models have been widely used to test candidate vaccines against Chlamydophila abortus infection in mice. Although the induction of a systemic infection by endogenous or intraperitoneal inoculation is a useful tool for understanding the immune mechanism involved in the protection conferred by the vaccination, a different approach is necessary to understand other factors of the infection, such as mucosal immunity or the colonization of target organs. To test whether C. abortus intranasal model of infection in mice is a useful tool for testing vaccines in a first group of experiments mice, were infected intranasally with C. abortus to characterize the model of infection. When this model was used to test vaccines, two inactivated experimental vaccines, one of them adjuvated with QS-21 and another with aluminium hydroxide, and a live attenuated vaccine (strain 1B) were used. Non-vaccinated control mice died within the first 8 days, after displaying substantial loss of weight. Histologically, the mice showed lobar fibrinopurulent bronchointerstitial pneumonia. Prior immunization with QS-21 adjuvated vaccine or 1B vaccine presented mortality and the recipients showed a greater number of T cells in the lesions, especially CD8(+) T cells, than the control mice and mice immunized with vaccine adjuvated with aluminium hydroxide. The results confirm that the C. abortus intranasal model of infection in mice is a useful tool for testing vaccines. 相似文献
High carbohydrate diets can affect the health and behaviour of foals, but the mechanisms are not always fully understood. The objective of this study was to compare the effects of feeding a starch and sugar (SS), or a fat (oil) and fibre (FF) rich diet to two groups of eight foals. Diets were fed from 4 to 42 weeks of age, alongside ad libitum forage. Faecal pH levels did not differ significantly between groups and endoscopic examination showed that the gastric mucosa was healthy in both groups at 25 and 42 weeks of age. At 40 weeks of age, SS foals had significantly higher total blood glucose and lower total blood gastrin than FF foals during the 6h period following ingestion of their respective diets, but insulin levels did not differ significantly. The ratio between serum tryptophan and other large neutral amino acids showed a trend towards an interaction between diet and sampling time. The results provide preliminary information about the effects of diet on the physiology of young horses. 相似文献

Much evidence exists detailing how animals respond to pathogen challenge, yet information explaining how the various behavioural, immunological, and physiological systems in chickens interplay during such challenges remains limited. 2. To gain an understanding of this interplay while controlling for genetic variation, the current study collected a variety of behavioural, physiological and immunological measures from three inbred lines (P, O and N) of laying hens before and after a sub-clinical infection with Salmonella enterica Typhimurium at 56 d of age. For comparison, an equal number of control birds were inoculated with a Salmonella-free broth. To identify an underlying profile, which might result in reduced susceptibility to infection, data were also collected in the pre-infection period. Post-infection blood and faeces were collected at 1-d post infection (dpi) and faeces again at 8 dpi. Animals were killed 15 d after infection and faeces, caecal contents, and spleen were examined for the presence of Salmonella. 3. Statistical analysis was performed to identify pre- and post-infection differences between genetic lines, changes in bird behavioural patterns between the two periods, and associations between a positive test for Salmonella and the various response measures. 4. Tissues from Line P birds were more often negative for Salmonella than those from birds of other lines, though this was inconsistent and tissue-dependent. The P line was also characterised by relatively greater serum concentrations of immunoglobulins at 1 dpi and α(1)-acid glycoprotein at 15 dpi. In addition, P line birds were more timid and their growth was reduced during the pre-infection period suggesting the possibility of a profile with reduced susceptibility to the bacterial challenge. 5. The current work has identified correlations between attributes of chicken strains and improved clearance. Future work using hypothesis-based testing will be required to determine whether the identified correlations are causally related. 相似文献
Olive (Olea europaea L.) is one of the most important tree crops of the Mediterranean regions. In spite of the increasing appreciation of typical extra virgin olive oils at world level, based on the use of local traditional varieties, very few studies have focused on the genetic characterisation of olive cultivars of regional interest, such as those grown in Veneto, a North-Eastern Italy region. A deep knowledge of the varieties cultivated in this territory is a key step to address the product quality, to increase market demand and to certify the origin of local olive oils. Here we have analyzed olive cultivars and cultivar groups within the olive cultivation area in Veneto, from the Garda Lake to the Euganean and Trevisan hills, by using discriminant SSR markers, in order to obtain a systematic genetic survey of the Veneto regional olive germplasm patrimony. A total of 203 previously uncharacterized olive samples were collected from ancient trees still grown by local farmers. The analyzed samples included also 36 olive reference cultivars from Veneto and neighbour Regions. We found 57 unique molecular profiles out of this set of olive accessions that were split into 15 cultivar groups corresponding to genetically distinct STRUCTURE clusters. Based on a common SSR database, our 239 Venetian accessions were compared with 280 olive reference genotypes representative of the Mediterranean cultivation area. From the genetic structure analysis, it has been observed that 80% of Venetian cultivars clustered in the central Mediterranean group, about 9% and 2% with the eastern and western varieties, respectively, and all the others resulted intermixed among two or three populations. We found that regionally the most common variety was “Casaliva”, corresponding to the widely diffused cultivar “Frantoio”, while others showed identity with known varieties grown in close regions, such as “Leccino”, “Miniol”, “Capolga” and “Bianchera”. Besides these genotypes, others were not matching any known reference and therefore they could be classified as true local varieties of indigenous origin, possibly deriving from the hybridization and selection made by farmers and from their adaptation to the local soil and climate conditions. 相似文献
