Blood metabolites in wild and seventh-generation cultivated shrimp were measured to determine how size-based selection could alter the nutritional and immunological conditions of Litopenaeus vannamei. Wild L. vannamei juveniles and a sample of seventh-generation cultured shrimp were acclimated under identical conditions. During 55 days, shrimp were fed a high (HCHO: 44%) or a low (LCHO: 3%) carbohydrate diet for 55 days. Wild shrimp showed a direct relation between dietary CHO and lactate, protein and hemocyte levels indicating that dietary CHO was used for protein synthesis via transamination pathways. In seventh-generation cultured shrimp these parameters were inversely proportional to dietary CHO level, indicating the capacity to synthesize protein from dietary CHO was repressed in cultured shrimp. Farmed shrimp showed a limited capacity to respond to LCHO diets demonstrating high protein dependence in their metabolism and immune response. These results demonstrate that during size-based breeding programs other metabolic process than CHO catabolism can be selected. The incapacity of shrimp to use dietary CHO could limit protein reduction of diets and limit the efforts of the shrimp industry to be ecologically and environmentally profitable.  相似文献

Amaranth is taking great attention as an important cereal crop that could fulfill food requirements for the growing population, especially in developing countries. However, the protein composition of these seeds is not well known yet. We have used the proteomics tools to characterize amaranth seed proteome. About 400 proteins spots were resolved on two-dimensional gel electrophoresis (2-DE) and protein spots were analyzed by LC-MS/MS (liquid chromatography tandem mass spectrometry). Identified proteins were related to stress and defense responses, metabolic, respiratory, and oxide-reduction processes. One abundant spot was identified as a Late Embryogenesis Abundant (LEA) protein and the gene was cloned and characterized. The AcLEA cDNA contains a 418 bp open reading frame (ORF) encoding a polypeptide of 139 amino acids. Bioinformatics analysis showed that AcLEA belongs to LEAs Group 5. Proteomics is a powerful technique that could be used even in non-sequenced organisms such as amaranth. The obtained information reveals that amaranth seed, beyond the classical seed storage proteins, contains proteins related to protection against stress. The identification of these proteins opens the door to the application of new strategies to improve the quality of amaranth production.  相似文献

We compared the abundance, species richness and diversity of saprobic filamentous microfungi in the forest and in coffee plantation systems (with different biophysical structures of the vegetation and agricultural management) and evaluated the degree of similarity in species composition among these sites. Soil washing was used to isolate the saprobic filamentous microfungi. Emerging colonies were quantified and transferred to tubes with culture medium and then mounted on semi-permanent slides for identification under the microscope. From 90 samples and 4,500 inoculated soils particles, 415 species were distinguished. The genera Trichoderma, Penicillium, Fusarium, Chaetomium and Humicola were the most frequent in all study sites. The transformation of the tropical montane cloud forest in coffee plantation had no significant effect on the abundance, species richness and diversity of the saprobic filamentous microfungi. Effects of the biophysical structures of the vegetation and the agricultural management of the sites were only detected at the level of dominant genera (Fusarium, Trichoderma and Penicillium) and on the evenness. The low degree of similarity among the six sites suggests the existence of a high exchange of species.  相似文献

Six Swedish and one Canadian single spore isolate of Drechslera teres f. teres were used to screen 109 barley lines for disease resistance and to select a differential set of barley lines for use in assessing pathogen virulence. A large variation for net blotch resistance was found among the 109 barley lines which were classified into four groups, those showing: 1) only resistant reactions; 2) differential reactions; 3) only intermediate reactions and 4) only susceptible reactions. The European commercial varieties included, showed susceptibility to all Swedish isolates, but a few were resistant to the Canadian isolate. The 18- member differential set separated 25 Swedish and two Canadian isolates of D. teres into 14 pathotypes, three of which made up 59% of the isolates. Only one barley differential (CI 9776) was resistant to all net form isolates. Host selection on the pathogen seems to be present as all six isolates obtained from cv. Golf belonged to the same pathotype and 4 of 5 isolates from cv. Karin shared the same virulence pattern. The net form of net blotch (D. teres f. teres) predominated in the sampled regions and only one of 26 Swedish isolates was of the spot form (D. teres f. maculata). Tetrahydro-beta-carbolines are biologically active alkaloids that occur and accumulate in mammalian tissues, fluids, and brain, but their ultimate origin or biological role is still uncertain. Four tetrahydro-beta-carboline alkaloids: 1,2,3,4-tetrahydro-beta-carboline-3-carboxylic acid, 1-methyl-1,2,3,4-tetrahydro-beta-carboline-3-carboxylic acid, 1-methyl-1,2,3,4-tetrahydro-beta-carboline, and 6-hydroxy-1-methyl-1,2,3,4-tetrahydro-beta-carboline, are found as naturally occurring substances in some fruit and fruit juices. These compounds occur in the microg/g level in those products, and a characteristic and distinct profile appears to exist depending on the type of fruit and juice involved. Thus, 1-methyl-1,2,3,4-tetrahydro-beta-carboline may appear in tomato, tomato juice, and kiwi; 6-hydroxy-1-methyl-1,2,3,4-tetrahydro-beta-carboline in bananas, pineapple, tomato, and their corresponding juices; and 1-methyl-1,2,3,4-tetrahydro-beta-carboline-3-carboxylic acid in oranges and grapefruits, although it also occurred in most juices. Fruit-occurring tetrahydro-beta-carboline alkaloids acted as antioxidants and free radical scavengers in the ABTS assay when compared with ascorbic acid and Trolox. This suggests that tetrahydro-beta-carboline alkaloids might act as antioxidants when absorbed and accumulated in the body, contributing to the antioxidant effect of fruit products containing these compounds.  相似文献

A method for the analysis of seven mineral contents—that is, potassium (K), calcium (Ca), phosphorus (P), iron (Fe), manganese (Mn), zinc (Zn), and copper (Cu)—was investigated in some oilseeds like flax, sesame, pumpkin, quinoa, and yellow mustard. X-ray Spectral Analysis method with Spectrometer EDX-900HS was used for the analytical concentrations of the macro and micro elements of the oilseeds. A relatively high level of Zn content was found in sesame and pumpkin seeds, while the highest level of Fe content was found in mustard seeds. Regarding the macro elements (K, Ca, and P) content, the highest levels of concentration were found in flax and mustard seeds. The highest values of Cu were obtained in sesame seeds while those of Mn were in pumpkin and brown flax seeds. The results showed that the method is a reliable and simple analytical procedure to characterize mineral elements in oilseeds with a very high analytical performance.  相似文献

Denaturation of proteins from striated and smooth muscles of scallop (Zygochlamys patagonica) was studied with differential scanning calorimetry (DSC) by monitoring maximum temperatures of transition and denaturation enthalpies. DSC thermograms of both striated and smooth whole muscles showed two transitions: Tmax 55.0, 79.2 degrees C; and Tmax 54.7, 78.7 degrees C, respectively. The DSC thermograms of myofibrils and actomyosin were similar to those corresponding to their respective whole muscles. As pH and ionic strength increased, the thermal stability of whole muscles decreased. The pH increase (5.0-8.0) significantly (p < 0.01) decreased the denaturation enthalpies (deltaH total, deltaH peakI, and deltaH peakII) of whole striated muscles. A significant decrease (p < 0.05) in the deltaH total and the deltaH peakI was also observed in DSC thermograms of smooth muscles at pH 8.0. Denaturation enthalpies (deltaH total and deltaH peakI) significantly decreased (p < 0.01) when the ionic strength increased from 0.05 to 0.5 in both types of muscles. Striated muscles were more affected than smooth muscles by changes in the chemical environment.  相似文献

Inter simple sequence repeat (ISSR) markers were used to evaluate levels of genetic similarity among Coffea arabica L. accessions from Tanzania and to estimate levels of genetic similarities in C. arabica and diploid coffee species. The six ISSR primers used generated a total of 82 fragments and the dissimilarity values ranged from 0.21 to 1. Mean dissimilarity values between provenances (0.56–0.85) were higher than within provenances (0.37–0.68). Cluster analysis based on Nei’s genetic distances showed C. arabica provenances grouping based on geographical origin. Two major clusters were formed that constituted of provenances from Kilimanjaro and Arusha in one sub-cluster; Tanga and Morogoro in the other; the second cluster had Mbeya provenances and diploid species, respectively. The implication is that Mbeya provenances are different from the rest of Tanzanian C. arabica. A principal coordinate analysis (PCA), whose first three coordinates explained 43% of the variation, showed similar groupings as in the cluster analysis. A separate cluster analysis of diploid species showed a distinct separation of the three species used. ISSR data gave results similar to previous findings from random amplified polymorphic DNA(RAPD) analysis. Microbial diversity might be a good indicator of ecosystem disturbance and functioning. We determined the effect of soil disturbances, such as N fertilization and tillage management, on soil microbial communities in a Typic Argiudoll of the El Salado river basin (Argentina). Microbial activity and substrate utilization provided a metabolic fingerprint of the soil microbial community. Univariate and multivariate analyses were used to differentiate responses to N fertilization and tillage at each of three growth stages of wheat (Triticum aestivum L.). Tillage had an adverse effect on microbial diversity, in which reduced and conventional tillage treated soils had different populations. However, N fertilization also altered microbial diversity depending on the crop developmental stage considered. Metabolism of carboxylic acids and carbohydrates were the main indicators of functional microbial activity and diversity. Although the substrate consumption profile of 32 C substrates did not provide insights into the fundamental ecological interactions that may induce changes in microbial population, it allowed us to demonstrate the alterations of microbial diversity as a result of tillage. We conclude that tillage and N fertilization altered microbial diversity.  相似文献
