A headspace solid-phase microextraction (HS-SPME) procedure at 30 degrees C with a 100 microm PDMS fiber of a saturated NaCl solution stirred at 1100 rpm combined to GC-ECD for the 2,4,6-trichloroanisol (TCA) determination in wines has been developed. Due to the matrix complexity and ethanol absorption into the fiber, the internal standard selection was crucial to obtain unbiased results. Thus, matrix effects were observed when analyzing different types of Spanish wines (white, early, and vintage red wines) spiked with TCA at low concentration levels (i.e., <40 ng L(-)(1)). In contrast, the use of 2,4,6-tribromoanisole (TBA) as internal standard overcame these matrix effects, whereas the use of 2,4,6-trichlorophenyl ethyl ether led to inconsistent results. The developed HS-SPME-GC-ECD methodology reaches a limit of quantitation for TCA in wine within 2.9-18 ng L(-)(1), with a relative standard deviation of 2.5-13.4%, depending on the TCA concentration level and wine characteristics. This analytical method is comparable to the existing methodologies based on HS-SPME followed by GC-MS in terms of accuracy, precision, length of determination, and length of quantification; however, analysis cost is reduced.  相似文献

Temporal variability is a key factor to understand the structure of belowground communities. Seasonal and annual variations are especially relevant in unpredictable desert ecosystems, where macroinvertebrates are poorly known, despite constituting an important group of soil organisms. In the present study, we analyse the composition and temporal (seasonal and annual) variations of soil macroinvertebrates in an arid area of southern Spain. During two years, macroinvertebrates were sampled in litter and belowground levels by means of soil cores. Results show that the assemblage was dominated by arthropods, especially Formicidae and Coleoptera. The assemblage differed between litter and belowground levels. In litter, detritivores dominated the community, while belowground fauna showed a similar proportion of detritivores and herbivores and a low percentage of predators. Litter and belowground assemblages showed seasonal variations in richness, abundance, biomass and composition, although variations were more marked in litter than belowground. Patterns of seasonal variation also differed between the two study years for both litter and belowground invertebrates. The seasonal and annual variability of the assemblage has potentially important implications for community dynamics in the study system, since the changes in species composition and trophic structure of soil invertebrate assemblages may affect species interactions and food web dynamics over time. Therefore, integrating temporal variability is likely to be crucial to understand soil community dynamics and food webs, especially in heterogeneous, variable systems as deserts.  相似文献

Soils were examined at 2505 m elevation in Haleakala's crater (Maui, Hawai?i) beneath 50 adult Hawaiian silversword plants (Argyroxiphium sandwicense DC.); mean canopy diameter was 42.0 cm. Exposed volcanic Inceptisols (Andic Humitropepts) seem significantly eroded beyond the dense rosette crowns, but remain unaffected below plants. Rosettes are perched on isolated basal soil mounds or pedestals 27–121 mm high (mean: 77.5 mm). Geomorphic field response of soils below rosettes and adjacent (∼ 100 cm apart) bare soils differs. Infiltration rates are higher under plants (mean: 158.7 mm/min) than in exposed control soils (60.0 mm/min). Soils below silverswords also show greater shear strength (146.1 g/cm2) and compressibility (2.795 kg/cm2) than unprotected soils (36.1 g/cm2, and 0.108 kg/cm2, respectively). Soil in the plant mounds contains more organic matter; this has influenced other pedological properties, which also differ substantially between sampling positions. Substrate under plants has a porosity ∼ 53% higher than exposed soil, while bulk density is 62% higher in soil outside the plant crown. The observed microtopographic differences are ascribed to greater soil erosion by rainsplash and runoff outside the silversword canopy. The dense rosette crown effectively intercepts raindrops; soils beneath plants also have a high surface cohesiveness provided by a dense network of fine plant rootlets and partially decomposed organic material. Higher runoff rates occur on the less permeable substrate beyond rosettes, which is affected by soil crusting.  相似文献

A lipoxygenase from Terfezia claveryi Chatin ascocarp, a mycorrhizal hypogeous fungus, is described for the first time. The higher proportion of PUFA in T. claveryi ascocarps makes lipid rancidity the main factor limiting its storage life. Thus, the studies on LOX from T. claveryi are important because this enzyme, among other roles, may be involved in an alteration of lipids leading to consumer rejection. The enzyme has been purified to apparent homogeneity by phase partitioning in the presence of Triton X-114, followed by two steps of cation-exchange chromatography. The purified T. claveryi LOX preparation consisted of a single major band with an apparent molecular mass of 66 kDa after sodium dodecyl sulfate-polyacrylamide gel electrophoresis. The enzymic activity exhibited a strong specificity toward linoleic and linolenic acids as substrates, while only 32% activity was observed using gamma-linolenic acid. The pH optimum of this enzyme was pH 7.0. When the enzyme reacted with linoleic acid, it produced a single peak, which comigrated with standard 13-hydroperoxy-octadecadienoic acid; 13-hydroperoxy-octadecatrienoic acid was produced during the reaction with linolenic acid.  相似文献

In this work, a single flow injection multicommuted system using solid-surface fluorescence spectroscopy has been explored for the determination of diphenylamine in apples and pears. The native fluorescence signal of diphenylamine retained on the solid support (C18 silica gel) was used for its determination (lambda(exc)/lambda(em) 291/372 nm). The sample treatment consists of a liquid-liquid extraction with acetonitrile followed by a cleanup step using primary-secondary amines. The use of a continuous flow assembly implementing multicommutation, based on a set of three-way solenoid valves controlled by appropriate software, provides the automatic control of sample, carrier, and eluting solution with remarkable advantages in relation to conventional flow injection approaches. Using an optimized sampling time, the proposed method was linear in the range 0.25-5 mg kg(-1) with a detection limit of 0.06 mg kg(-1) and RSD (percent) values better than 3%. The proposed method was successfully applied to the analysis of diphenylamine in different apple and pear samples fortified at different concentrations, and recoveries between 78 and 104% were found. The results obtained illustrate the usefulness of the proposed method for the screening and evaluation of postharvest treatment of crops possibly containing diphenylamine.  相似文献

ABSTRACT

This study aimed to assess the physiological and biochemical responses of cotton plants to manganese (Mn2+) nutrition. Four cotton genotypes (G1 – TMG 47; G2 – FM 975 WS; G3 – TMG 11 WS and G4 – IMA 8405 GLT) were grown in nutrient solution under two Mn2+ concentrations (2 and 200 µmol L?1) for 10 days. No visible symptoms of Mn2+ toxicity were observed in the genotypes tested. All genotypes showed a marked increase in leaf chlorophylls, pheophytins, carotenoids, sucrose and total sugars concentration in response to high Mn2+ in a nutrient solution. However, the net photosynthetic rate, stomatal conductance, internal carbon dioxide concentration and transpiration decreased in genotypes G1 and G2 growing under 200 µmol L?1. Antioxidant enzymes such as superoxide dismutase (SOD), ascorbate peroxidase (APX) and glutathione reductase (GR) activities increased in genotypes G1, G3 and G4. Cotton genotypes showed an increased leaf antioxidant and sugar metabolism as a possible strategy to mitigate oxidative stress. The decrease in the net photosynthetic rate and stomatal conductance; the increased antioxidant enzymes activities (SOD, APX and GR); and the increase in leaf sucrose and total sugar concentration were the main physiological and biochemical responses in cotton plants to Mn2+ stress.  相似文献

Changes in phenolic compounds, total glucosinolates, and vitamin C were monitored during the productive period along five inflorescence development stages of three broccoli commercial cultivars (Marathon, Monterrey, and Vencedor). In an attempt to identify differences due to agronomic factors, broccoli cultivars were grown under different sulfur fertilization with poor (15 kg/ha) and rich (150 kg/ha) rates. Phenolic compounds and vitamin C concentrations showed, in all broccoli cultivars, a rising trend from the first stage until the over-maturity stage, both for rich and poor sulfur fertilization. Significant differences were detected in the first two stages between rich and poor sulfur fertilization in total glucosinolates for all broccoli cultivars, where the highest concentration was always observed in the second development stage (used as minimally processed product) during poor fertilization. With regard to the last three stages, the glucosinolate concentration in the poor sulfur fertilization started to slope down until the over-maturity stage. Where rich sulfur fertilization is concerned, the highest level was reached during the third stage (used as minimally processed product also), and after that, glucosinolate concentration decreased until the fifth stage.  相似文献

Thirty-seven apricot varieties, including four new releases (Rojo Pasión, Murciana, Selene, and Dorada) obtained from different crosses between apricot varieties and three traditional Spanish cultivars (Currot, Mauricio, and Búlida), were separated according to flesh color into four groups: white, yellow, light orange, and orange (mean hue angles in flesh were 88.1, 85.0, 77.6, and 72.4, respectively). Four phenolic compound groups, procyanidins, hydroxycinnamic acid derivatives, flavonols, and anthocyanins, were identified by HPLC-MS/MS and individually quantified using HPLC-DAD. Chlorogenic and neochlorogenic acids, procyanidins B1, B2, and B4, and some procyanidin trimers, quercetin 3-rutinoside, kaempferol 3-rhamnosyl-hexoside and quercetin 3-acetyl-hexoside, cyanidin 3-rutinoside, and 3-glucoside, were detected and quantified in the skin and flesh of the different cultivars. The total phenolics content, quantified as the addition of the individual compounds quantified by HPLC, ranged between 32.6 and 160.0 mg 100 g(-1) of edible tissue. No correlation between the flesh color and the phenolic content of the different cultivars was observed.  相似文献

The oxidation of methyl linoleate (LMe) in the presence of bovine serum albumin (BSA) was studied to analyze both the processes involved when lipid oxidation occurs in the presence of proteins and the relative progression of the several reactions implicated. The disappearance of LMe, the formation of primary and secondary lipid oxidation products, the loss of essential amino acids, and the production of oxidized lipid/amino acid reaction products (OLAARPs) were studied as a function of incubation time. During the first steps of lipid oxidation, LMe was converted quantitatively to methyl linoleate hydroperoxides, which were very rapidly degraded to either secondary products of lipid oxidation or OLAARPs. No significant differences were identified in the major lipid oxidation products formed in incubations with or without proteins, indicating that mechanisms for formation of these compounds are similar in both cases. In addition, no significant differences were observed between the time-courses of formation of secondary oxidation products and OLAARPs, suggesting that hydroperoxide decomposition and OLAARP formation occur simultaneously when the lipid oxidation process takes place in the presence of proteins. Furthermore, OLAARP formation seems to be an unavoidable process that should be considered as a last step in the lipid peroxidation process.  相似文献

The Maillard reaction (MR) has a clear impact in food science, nutrition, and medical research. Free radical scavenging capacities of several MR mixtures made from single combinations of glucose or lactose and amino acids (gly, his, lys, trp, met, and cys) were evaluated by using the N,N-dimethyl-p-phenylenediamine radical cation assay. Medium-roasted coffee brew was used as reference of a thermally processed food. A novel approach has been applied in order to get more information about the kinetic behavior of the radical scavenging properties of MR mixtures in a watery environment. Antiradical efficiency (AE) concept has been applied, and it takes into consideration the reaction time, apart from the amount of antioxidant necessary to decrease by 50% the radical initial concentration (EC(50)). Cysteine and histidine reveal as powerful amino acids to exert a high AE in the MR mixtures. No relationship between AE parameter and browning was observed.  相似文献
