AIM: To observe the effect of antisense locked nucleic acid (anti-LNA) blocking the translation initiation region of c-myc exon 2 on the viability and apoptosis of hepatocellular carcinoma cells.METHODS: The anti-LNA that was complementary to the translation initiation region of c-myc exon 2 was designed, synthesized, and introduced into the HepG2 cells by cationic liposome-mediated transfection. The mRNA and protein levels of c-Myc in the cells were determined by RT-PCR and Western blot. The change of cell apoptosis was analyzed by flow cytometry, and the toxicity of anti-LNA to the cells was detected by MTT assay.RESULTS: Five days after transfection, the mRNA level of c-Myc in anti-LNA group was 0.335±0.016, and the protein level was 0.448±0.037, significantly lower than those in control group (both P<0.05). The ratio of apoptotic cells in anti-LNA group was 32%±6%, which was higher than that in control group (P<0.05).CONCLUSION: Antisense locked nucleic acid targeting at the translation initiation region of c-myc exon 2 shows strong inhibitory effects on the apoptosis of hepatocellular carcinoma cells. 相似文献

桐油是一种重要用途的干性油脂,不同工艺会对油品质量产生重要影响。为了克服传统工艺在制油中的缺陷,本研究采用丁烷-亚临界法工艺进行桐油的制备,并基于Design-Expert软件进行响应面优化实验,获得最佳工艺参数:萃取温度40.64℃,萃取时间45.38 min,原料颗粒度39.71目,在此条件下桐油得油率达到66.12%。采用气质联用仪分析了桐油脂肪酸组成为α-桐酸74.93%,亚油酸8.62%,油酸7.12%,棕榈酸2.72%,硬脂酸3.85%,通过分析所得油样的5项指标,可知该工艺所得油品满足国标要求。 相似文献

AIM: To explore the neuroprotective effect of fasudil combined with bone marrow-derived neural stem cells (BM-NSCs) on the mice with experimental autoimmune encephalomyelitis (EAE). METHODS: Female C57BL/6 mice (8~10 weeks old, n=32) were immunized with myelin oligodendrocyte glycoprotein 35-55 (MOG35-55) to establish chronic EAE model. The mice were randomly divided into control (ddH2O) group, fasudil group, BM-NSCs group, and fasudil+BM-NSCs group. The clinical score and body weight were recorded every other day. The expression of neurotrophic factors was determined by immunofluorescence staining. RESULTS: In comparison with ddH2O group, fasudil combined with BM-NSCs delayed onset and ameliorated severity of EAE. The numbers of brain-derived neurotrophic factor, glial cell-derived neurotrophic factor, nerve growth factor, neurotrophin-3 and ciliary neurotrophic factor positive cells in fasudil group, BM-NSCs group and fasudil+BM-NSCs group were all increased in various extents. In particularly, the expression of these neurotrophic factors in fasudil+BM-NSCs group was significantly higher than that in the mice treated with fasudil or BM-NSCs alone (P<0.01). CONCLUSION: Fasudil combined with BM-NSCs promotes the expression of neurotrophic factors and improves microenvironment of central nervous system, thus playing a positive role in neural restoration and regeneration through a synergistic and superimposed effect. 相似文献

Quality control and screening of active substances in traditional Chinese medicines have been performed using fingerprint analysis. The spectrum-effect relationship between chromatography fingerprints and efficacy of herbal drugs is considered as a potentially useful method for determining active ingredients in complex mixtures. The study was designed to develop a method for determining the bioactive components of a compound Chinese medicine called Tiefeng based on spectrum-effect relationships between high-performance liquid chromatography (HPLC) fingerprints and anticoccidial activities. Four peaks of the established HPLC fingerprint indicate the main bioactive components of this medicine. In addition, pharrnacodynamic atlas was defined and used to assess the anticoccidial activity of Tiefeng from different sources for the first time. We found that the level of anticoccidial activity of Tiefeng was consistent with the degree of similarity between the pharmacodynamic atlas and chromatogram of any sample. 植物源蒸馏油是一种新型植物油,本文探讨了植物源蒸馏油在害虫控制中的应用潜力。以矿物油乳剂为参照,以柑橘木虱为试虫,分析比较植物油蒸馏油的触杀和拒避作用。植物油和矿物油对柑橘木虱的卵、若虫和成虫均有一定的触杀作用,其中对若虫尤其低龄若虫的触杀作用最强,可达90%以上。两种油剂对雌虫产卵均有一定的拒避作用,拒避雌虫产卵维持约2 d。两种油剂对成虫寄主选择没有明显的拒避作用。总体上,植物源蒸馏油对柑橘木虱的触杀作用以及对成虫的拒避作用与矿物油相当。如果进一步改进该植物油的乳化效果,可替代矿物油乳剂应用于害虫防治中。 相似文献

建立菠萝 DNA 甲基化水平的 HPLC 测定方法,分析菠萝愈伤组织 DNA 甲基化水平变化,为进一步研究菠萝 体细胞无性系变异机理奠定基础。通过对流动相和水解温度等条件的优化,建立菠萝 DNA 甲基化水平的检测方法。结 果表明,分离 C 和 5m-C 的最佳流动相为甲醇∶磷酸二氢钾∶三乙胺为 10∶90∶0.2(V/V),pH 3.0,DNA 的最佳水解 温度为 90 ℃。利用此体系分析菠萝愈伤组织和胚性愈伤组织的 DNA 甲基化变化,结果表明,菠萝愈伤组织在分化过 程中 DNA 总甲基化水平呈动态变化,变化范围为 5.14%~96.86%。此外,胚性愈伤组织甲基化水平低于非胚性愈伤组 织。推测 DNA 甲基化影响菠萝愈伤组织的分化及胚性愈伤组织的形成。 相似文献
