Shikonin reverses HGF-induced resistance to gefitinib in lung cancer cells HCC827

AIM: To investigate the effect of shikonin on reversing hepatocyte growth factor(HGF)-induced resistance to gefitinib in lung cancer HCC827 cells, and to explore its possible mechanisms.METHODS: The gefitinib-resistant HCC827 cells induced by HGF were treated with shikonin and gefitinibthe alone or in combination. The inhibition rates of cell viability were determined by MTT assay. The invasive ability of HCC827 cells with HGF-induced resistance to gefitinib was determined by Transwell assay. The protein levels of epithelial-mesenchymal transition (EMT) and related signaling pathway in the HCC827 cells were detected by Western blot.RESULTS: The results of MTT assay showed that the cell activity of HCC827 cells was significantly inhibited by shikonin in a dose dependent manner. The IC₅₀ of shikonin in HCC827 cells was 3.06 μmol/L. And the IC₅₀ of gefitinib in HCC827 cells was 0.51 μmol/L. Under the condition of combined treatment with shikonin and gefitinib in the presence of HGF (20 μg/L), the IC₅₀ of gefitinib was 7.36 μmol/L, significantly lower than that treated with gefitinib alone (P<0.01), so did the result of the cell migration (P<0.01). HGF induced EMT, while shikonin reversed this effect. The protein expression level of p-AKT was significantly up-regulated by HGF, while markedly down-regulated treatment with shikonin and gefitinib compared with gefitinib alone (P<0.01).CONCLUSION: Shikonin reverses HGF-induced resistance to gefitinib in lung cancer HCC827 cells, and the mechanism may be likely related to the preventon of EMT and the inhibition of HGF-induced activation of p-AKT signaling pathway.     

应用SYBR Green I的Real-time PCR方法定量检测空肠弯曲杆菌

空肠弯曲杆菌(Campylobacterjejuni)被认为是人类主要食源性病原菌之一。由于空肠弯曲杆菌特殊的生长条件和容易进入不可培养但存活的状态(Viablebutnonculturable,VNC),所以传统的生化鉴定结果不一定可靠,并且是一项费时而繁琐的工作。核酸检测方法的出现为空肠弯曲杆菌的检测带来了方便。本文基于LightCycler为平台,利用SYBRGreenI染料能特异与双链DNA结合而发荧光的特性,建立一种Real-timePCR方法来定量检测空肠弯曲杆菌。用该方法检测时,发现所有空肠弯曲杆菌(11株)都呈阳性,所有其它弯曲菌(3株)和其它菌株(5株)都是阴性。整个检测过程60min内可以完成,检测限度为5CFU,标准曲线的相关系数为1。结果表明基于SYBRGreenI的定量PCR方法既为空肠弯曲杆菌提供了一种特异、敏感、快速和简洁的定量检测方法,又为研究空肠弯曲杆菌致病机理提供了一种重要方法。     

羊支原体性肺炎两标准株对抗菌药物敏感性的研究

本实验测定了环丙沙星、氧氟沙星、单诺沙星、红霉素、罗红霉素、泰勒菌素、泰妙菌素、四环素等8种药物对羊肺炎支原体两个标准株Y98和Y-goat的体外抑菌浓度以及红霉素与氧氟沙星、泰勒菌素对Y-goat和四环素与氧氟沙星、泰勒菌素对Y98的联合药敏作用。对羊肺炎支原体两个标准株对抗菌药物的敏感性进行了分析。     

兵团农机产业化的若干思考

猫鼻气管炎病毒、怀状病毒、泛白细胞减少症病毒的分离及其其形态学观察

根据病毒的理化特性，从猫三联活疫苗中分离出猫鼻气管炎、猫西洋太和猫泛白细胞减少症病毒；筛选出每种病毒敏感的细胞，并将该3种病毒分别分别在其中传代与扩增；通过电镜观察此3种病毒的形态学特征及其在宿主细胞内增殖部位、分布情况等，并以此对猫3种病毒进行了初步鉴定。

The Brief Introduction to the Sino-US Joint Centers for Soil and Water Conservation and Environmental Protection

Erosion and transport of soil has worldwide implications for agriculture, landscape stability, climate, natural hazards, and clean, renewable resources of water and air. Assured access to clean water and a healthy and safe environment requires an ethic of conservation and protection. The minimum scale in which these principles apply successfully is basin wide. These are the fundamental concerns of the Sino-US Centers for Soil and Water Conservation and Environmental Protection.     

Cybrid/hybrid plants regenerated from somatic fusions between male sterile Satsuma mandarin and seedy tangelos

Somatic hybridization provides an alternative for transferring mitochondria-encoded cytoplasmic male sterility (CMS). Herein, symmetric protoplast electrofusion was conducted between embryogenic callus protoplasts of Citrus unshiu Marc. cv. Guoqing No. 1 (G1), a CMS cultivar, and mesophyll-derived protoplasts of seedy ‘Page’ tangelo [C. reticulata Blanco × (C. reticulata Blanco × C. paradisi Macf.)] or ‘Nova’ tangelo [C. reticulata Blanco × (C. reticulata Blanco × C. paradisi Macf.)], to transfer CMS trait. Flow cytometry analysis showed that 14 plants recovered from G1 + ‘Page’ tangelo that displayed typical morphological character of ‘Page’ were diploid, and 6 plants regenerated from G1 + ‘Nova’ tangelo were tetraploid. Genetic compositions of regenerated plants from the two fusions were determined by SSR, CAPS and chloroplast-SSR analysis. Cybrid nature of diploids from G1 + ‘Page’ tangelo with nuclear DNA from ‘Page’, mitochondrial DNA (mtDNA) from the G1 and chloroplast DNA (cpDNA) derived from either parent was confirmed. Tetraploid plants from G1 + ‘Nova’ tangelo were identified as somatic hybrids with random cpDNA inheritance. The regenerated cybrid and hybrid plants hold great potential for Citrus seedless breeding at diploid or triploid levels.     

黄金树花器官发生及发育的形态观察

利用普通光学显微镜和扫描电镜技术对黄金树花器官的发生及发育过程进行了观察.结果显示:(1)黄金树花器官的形态发生及发育过程集中于3月下旬-5月上旬;(2)花原基分化形成花的整个过程符合一般的分化顺序:花萼原基-花冠原基-雄蕊原基-雌蕊原基,且各原基在分化顺序上存在交叉;(3)花药及胚珠的发育与花器官的形态发生之间有明显的连续性,当花蕾直径为3.0 mm左右时花粉母细胞及完整的花粉囊壁形成,直径达到3.5 mm左右时胚珠中出现孢原细胞的分化,它直接起大孢子母细胞的功能.
