Little is known about the transport of microorganisms through freeze-fractured clay soils. Normally consolidated clay (NCC) and compacted clay (CC) columns (representing a natural clay barrier and a compacted barrier, respectively) were exposed to six consecutive freeze–thaw cycles and permeated for 21 days with an Escherichia coli cell suspension (approximately 1?×?107 colony forming units (CFU)/mL) containing a 2.1-mM bromide tracer. An unfractured sand column was also examined for comparison with the clay columns. While no E. coli was detected in the effluent of both untreated NCC and CC control clay columns, a relatively low density of E. coli (between 228 and 270 CFU/mL compared to 1?×?107 CFU/mL in the influent) was first detected in the effluent of the freeze-fractured NCC and CC columns at 0.29 and 0.31 pore volumes (or at 5.4 and 4.1 h), respectively. It took 11 min for a full breakthrough of E. coli through the sand column, but only about 0.1% of the influent E. coli density was detected in the effluents of the freeze-fractured NCC and CC columns at day 21. These observations show that despite the high bacterial retention capacity of the freeze-fractured clay columns, the fractures were large enough for the E. coli to flow through. Based on batch sorption tests and the permeation data, it is estimated that 18%, 7%, and 84% of the freeze-fractured NCC, CC, and sand columns would be exposed to the influent, respectively, under a full E. coli breakthrough condition. Our data show that the high bacterial retention capacity of clay barriers can be compromised by freeze–thaw conditions.  相似文献

This study examines the technical efficiency (TE) of aquaculture farms in Hawaii. Policy makers and industry participants interested in assessing the economic outlook of aquaculture production not only have to assess measures of profitability and financial health, but also measures of production efficiency. We estimate the TE of the state's aquafarms with a data envelopment analysis (DEA) model. Using data from the US Census of Agriculture, we examine TE measures of the population of Hawaii's aquafarms across different types of farms and over time. The results show that only 12% of the farms in 2007 may be classified as efficient, with a steady decline in efficiency over time. We find TE was related to the size of farm, full‐time farms were significantly more efficient than part‐time farms and that the average excess real inputs for land and labor have slightly fallen over time. The sub‐sector analysis revealed a strong variation of TE across different type of farms. With few farms operating on the frontier, the findings of this study suggest that the majority of the farms could improve their TE.  相似文献

Complementary DNAs were isolated and used to deduce the primary structures of the alpha 1 and alpha 2 subunits of the dihydropyridine-sensitive, voltage-dependent calcium channel from rabbit skeletal muscle. The alpha 1 subunit, which contains putative binding sites for calcium antagonists, is a hydrophobic protein with a sequence that is consistent with multiple transmembrane domains and shows structural and sequence homology with other voltage-dependent ion channels. In contrast, the alpha 2 subunit is a hydrophilic protein without homology to other known protein sequences. Nucleic acid hybridization studies suggest that the alpha 1 and alpha 2 subunit mRNAs are expressed differentially in a tissue-specific manner and that there is a family of genes encoding additional calcium channel subtypes.  相似文献

A method using liquid chromatography electrospray ionization tandem mass spectrometry (LC/ESI-MS/MS) for the determination of trace levels of five macrolide antibiotics (spiramycin, tilmicosin, oleandomycin, erythromycin, and tylosin) in eggs is presented. Data acquisition under MS/MS was achieved by applying multiple reaction monitoring (MRM) of two or three fragment ion transitions to provide a high degree of sensitivity and specificity for both quantification and confirmation. Matrix-matched standard calibration curves were used to achieve the best accuracy of the method. A fully nested experimental design was used to study the measurement uncertainty arising from intermediate precision and trueness or proportional bias. The overall recoveries, that is, those determined by the nested experiments, of spiramycin, tilmicosin, oleandomycin, erythromycin, and tylosin at fortified levels of 60, 100, 200, and 300 microg/kg were 96.8, 98.2, 98.3, 98.8, and 95.4%, respectively. The LC/ESI-MS/MS method detection limits (S/N > or = 3:1) of five macrolides were <1.0 microg/kg.  相似文献

The present study was to examine effect of soy leaf powder (SLP) and soy leaf ethanol extract (SLEE) on serum lipoproteins in hamsters. The control group was fed a semisynthetic diet containing 0.1% cholesterol, while the tested groups were maintained on the same diet but supplemented with 3% SLP or the equivalent amount of SLEE derived from 3% SLP for 4 weeks. SLP supplementation led to a trend of lowering serum total cholesterol (TC) and nonhigh density lipoprotein cholesterol (non-HDL-C), with HDL-C being unaffected, whereas incorporation of SLEE into the diet led to an elevated level of HDL-C and a lower level of non-HDL-C with TC being unchanged. Both SLP and SLEE supplementation caused favorably a decrease in the ratio of non-HDL-C to HDL-C. The present results demonstrate that not only soybean seeds but also soy leaves are cardioprotective, by favorably modulating serum lipid profile.  相似文献

It is well documented that faecal contamination of drinking water has caused numerous disease outbreaks. Because the risks of disease outbreaks correlate with the incidence of faecal contamination, faecal bacteria are used as indicators of faecal contamination and hence, the possible presence of disease-causing organisms. However, different microbiological faecal indicators are used in different countries and jurisdictions. Therefore, it is important to understand the potentials and limitations of these indicator organisms before realistically implementing guidelines and regulations to safeguard our water resources. This review considers the history of indicator organisms, the evolution of the analytical methodologies (biochemical and molecular) and addresses the advantages and limitations of current faecal indicator microorganisms.  相似文献

Schisandrin B (Sch B) and schisandrin C (Sch C), but not schisandrin A and dimethyl diphenyl bicarboxylate, protected rat skin tissue against solar irradiation-induced oxidative injury, as evidenced by a reversal of solar irradiation-induced changes in cellular reduced glutathione and α-tocopherol levels, as well as antioxidant enzyme activities and malondialdehyde production. The cytochrome P-450-mediated metabolism of Sch B or Sch C caused ROS production in rat skin microsomes. Taken together, Sch B or Sch C, by virtue of its pro-oxidant action and the subsequent eliciting of a glutathione antioxidant response, may prevent photo-aging of skin.  相似文献

Recently, there has been a surge of interest in understanding the survival of Shiga toxin-producing Escherichia coli (STEC) in aquatic environments. Fifteen strains of STEC were monitored, individually, in untreated well water samples incubated at 10 and 22^C for 56 days. The strains were selected from three serogroups (O26, Ol11 and O157) and represented five distinct ERIC (enterobacterial repetitive intergenic concensus)-genotypes. The microcosms were prepared in triplicate and inoculated at an initial cell density of about 7.0 log CFU/ml well water. At 10^C, the cell density of five STEC strains fell below the detection limit of 0.8 log CFU/ml by day 56. Of the ten persisting strains, four showed superior survival with cell densities decreasing to an average of about 5 log CFU/ml while the remaining six strains showed moderate levels of survival, decreasing to an average cell density of about 3 log CFU/ml. At 22^C, strain H32 (genotype I) and H15 (genotype B) persisted at 1.1 log CFU/ml and 2.2 log CFU/ml in 56 days, respectively. The other 13 STEC strains dropped below the detection limit between weeks 3 to 8. The 15 strains demonstrated highly variable levels of survival with no correlation between ERIC-genotypes or serogroups and the strains' ability to persist in the well water samples. Although strain H32 (O157:H7) persisted significantly longer than strain H22 (O157:H7) in natural well water at both 10 and 22^C, both strains survived equally well in sterile well water, indicating that individual STEC strains vary in their ability to compete with background microbial populations.  相似文献
