In simple epithelia, such as living epithelial pancreatic cancer cells (Panc-1), unusual amounts of keratin filaments can be found, which makes these cells an ideal model system to study the role of keratin for cell mechanical properties. In this work, the elastic moduli of Panc-1 cells and their extracted in-situ subcellular keratin intermediate filament network are determined and compared with each other. For this, the living adherent cells and their extracted keratin network were probed with local quasistatic indentation testing during large deformations using the Atomic Force Microscope (AFM). We determined the elastic modulus of the skeletonized but structurally intact keratin network to be in the order of 10 Pa, while the living cell elastic modulus ranged from 100 to 500 Pa. By removing microfilaments, microtubules, membranes and soluble cytoplasmic components during keratin network extraction, we excluded effects caused by crosslinking with other filamentous fibers and from the viscosity of the cytoplasm. Thus, the determined elastic modulus equals the actual elastic modulus inherent to such a keratin filamentous network. In our assessment of the effective mechanical contribution of the architecturally intact, skeletonized keratin network to living cell mechanics, we come to the conclusion that it plays only a very limited role. Evidently, the quantitative dominance of keratin in these cells does not reflect a strong influence on determining the cell's elastic modulus. Instead, keratin like other filamentous structures in the cell's scaffolding, e.g., F-actin and microtubuli, is one part of a greater whole.  相似文献

The deformation of suspended cells inside microchannel restrictions mimics passive cell transportation in the blood circulation system of the body. The cells traverse or get stuck in narrow vessels, as, e.g., during the metastasis of tumor cells. In this work, the mechanical responses of suspended pancreatic cancer cells as they move through and deform inside microchannel restrictions are assessed with a cantilever-based polydimethylsiloxane (PDMS) force sensor. Incorporated into a flow cell chip, the PDMS cantilever is integrated into the boundary wall of a narrow microrestriction. Upon being forced to enter the restriction by an applied flow, the cell exerts pressure on the cantilever, which then bends. By assuming a uniformly loaded cantilever, the total force and pressure on the cantilever can be calculated using elastic beam theory. This technique has the advantage of presenting an absolute and direct measure, which is independent of the applied flow and frictional processes at the channel-cell interface; in contrast to, e.g., measuring cell mechanics indirectly via cell sliding velocities. Furthermore, a high number of cells can be examined in a short time compared to other single cell mechanical testing devices.  相似文献

One of the great advantages of the fully sequenced rice genome is to serve as a reference for other cereal genomes in particular for identifying genes linked to unique traits. A trait of great interest is reduced lignocellulose in the stem of related species in favor of fermentable sugars as a source of biofuels. While sugarcane is one of the most efficient biofuel crops, little is known about the underlying gene repertoire involved in it. Here, we take advantage of the natural variation of sweet and grain sorghum to uncover genes that are conserved in rice, sorghum, and sugarcane but differently expressed in sweet versus grain sorghum by using a microarray platform and the syntenous alignment of rice and sorghum genomic regions containing these genes. Indeed, enzymes involved in carbohydrate accumulation and those that reduce lignocellulose can be identified.  相似文献

The scientific interest in the fate of antibiotic substances in the environment has increased during the past decades. Especially in animal husbandry, the assumed quantity of antibiotics excreted has caused major concerns as the environment can be contaminated by these compounds via different paths. In this study in Northern Germany, manure and leachate samples from dairy farms were analysed in order to verify if this kind of animal husbandry can be a source of antibiotic residues found in the environment. Neither in liquid manure nor in leachate were the analysed antibiotics detected. These results clearly show the minor importance of dairy cows as sources of antibiotic residues in the aquatic environment. To explain this fact, we propose the three following reasons: (i) the amount of antibiotics administered in dairy farming is low compared to pig or poultry production; (ii) manure storage is a useful process to minimize the antibiotic residues as the substances applied to dairy cows are sensitive to degradation processes during that period; (iii) excreted directly on grasslands, the substances degrade very quickly or adsorb to soil and are therefore not present in the leachate.  相似文献

Due to an increasing demand of forestry products, nitrogen (N) fertilization of coniferous forests may in the future become a standard silvicultural practice in parts of the boreal region that is not too remotely situated. Beside positive effects on productivity, forest fertilization may induce changes in species composition of the ground vegetation. More information on the magnitude of such side effects is needed to evaluate whether commercial forest fertilization is in accordance with principles of sustainable forestry and preservation of biodiversity.  相似文献

ABSTRACT: Antimicrobial peptides play a pivotal role as key effectors of the innate immune system in plants and animals and act as endogenous antibiotics. The molecules exhibit an antimicrobial activity against bacteria, viruses, and eukaryotic pathogens with different specificities and potencies depending on the structure and amino-acid composition of the peptides. Several antimicrobial peptides were comprehensively investigated in the last three decades and some molecules with remarkable antimicrobial properties have reached the third phase of clinical studies. Next to the peptides themselves, numerous organisms were examined and analyzed regarding their repertoire of antimicrobial peptides revealing a huge number of candidates with potencies and properties for future medical applications. One of these organisms is the horse, which possesses numerous peptides that are interesting candidates for therapeutical applications in veterinary medicine. Here we summarize investigations and knowledge on equine antimicrobial peptides, point to interesting candidates, and discuss prospects for therapeutical applications.  相似文献

Novel Decapod Iridescent virus (DIV1) infections emerged in mainland China around 2014 and have devastated shrimp aquaculture operations in Chinese coastal provinces. In 2020, DIV1 has spread to Taiwan with devastating results to shrimp and crayfish farms, in addition to being found in wild caught Penaeus monodon from the Indian Ocean. This trend is a major cause for concern and an urgent reminder to expand the tools needed to monitor the spread of DIV1 globally. Here, we describe a set of four different real-time polymerase chain reaction (PCR) assays positioned across the genome of DIV1 to detect the virus in shrimp tissues. All four assays show a wide dynamic range and high analytical sensitivity and specificity. In addition, the newly developed assays show excellent diagnostic sensitivity and specificity in clinical Litopenaeus vannamei samples of North Asian origin. The new molecular toolset will enhance global capabilities to monitor the spread of DIV1 and ultimately be used as an early warning system for farmers and authorities to engage in appropriate risk mitigation strategies.  相似文献

Novel types of anti-oomycetic compounds have been designed and prepared. The synthetic approach to these mandelamides is outlined. Biological data demonstrate their high efficacy against important plant diseases such as tomato and potato late blight (Phytophthora infestans De Bary) and grape downy mildew (Plasmopara viticola Berliner & de Toni). Two hundred eighty-eight crossbred feeder pigs were used in 2 trials to determine the effects of feed and/or water deprivation at an auction market, and the effects of restricting the intake of the receiving diet on their serum chemical profile. The study also was designed to assess the value of the serum chemical profile as a diagnostic data base for stress disorders in feeder pigs. Performance data indicated that feeder pigs provided water only at the auction facilities lost significantly more weight than did those provided feed and water. Feeder pigs deprived of both feed and water were not significantly different in body weight from either group. Several serum chemical values (creatinine, triglycerides, cholesterol, blood urea nitrogen, and lactate dehydrogenase) were significantly influenced by feed deprivation, but not by feed and water deprivation. However, only the serum creatinine values were significantly different after the 24-hour posttransport period. There were no significant differences in pig weight or serum chemical values 84 days after pigs had arrived at the finishing unit. The serum chemical profile, widely used in human medicine, appears not to provide a reliable marker for identification of short-term nutritional deprivation, nor for transport stress in feeder pigs.  相似文献
