外源植物激素对东方山羊豆响应盐胁迫的调控效应初探   总被引:1,自引:0,他引:1

为确定东方山羊豆(Galega orientalis)种子萌发及苗期耐盐性,明确在盐胁迫条件下,植物生长激素对东方山羊豆种子萌发及苗期生理特性的影响,分析比较了不同NaCl浓度(0、25、50、100、150、200 mmol·L-1)梯度及不同植物生长激素处理下的东方山羊豆种子萌发率、幼苗脯氨酸、丙二醛和叶绿素含量等指标的差异。结果表明,东方山羊豆种子和幼苗对盐胁迫比较敏感,东方山羊豆能够适应50 mmol·L-1浓度以内NaCl胁迫,50 mmol·L-1浓度以内NaCl对种子萌发及幼苗生长影响小,随着NaCl浓度的增高,种子发芽率和幼苗丙二醛、叶绿素含量显著降低,脯氨酸含量显著增加;GA3可以提高150 mmol·L-1浓度以内NaCl胁迫条件下东方山羊豆的种子萌发率;NAA适合调控东方山羊豆幼苗在中、低盐浓度下的生长。  相似文献

2,3,7,8-Tetrachlorodibenzo-p-dioxin (TCDD) is one of the most toxic environmental pollutants that cause various biological effects on mammals. The purpose of our study was to identify the genes involved in hepatotoxicity and hepatocarcinogenesis caused by TCDD. C57BL/6 (AhR+/+, wild type) and B6.129-AhR/J (AhR-/-, knock out) mice were injected i.p. with a single treatment of TCDD at the dose of 100 microg/kg body weight. Relative liver weight was significantly increased at 72 hr after TCDD treatment without an apparent histopathological change in AhR+/+ mice (p<0.05). TCDD treatment also significantly increased activity of serum alanine aminotransferase in AhR-/- mice (p<0.05). The liver was analyzed for gene expression profiles 72 hr later. As compared with AhR-/- mice, the expression of 51 genes (>3-fold) was changed in AhR+/+ mice; 28 genes were induced, while 23 genes were repressed. Most of the genes were associated with chemotaxis, inflammation, carcinogenesis, acute-phase response, immune responses, cell metabolism, cell proliferation, signal transduction, and tumor suppression. This study suggests that the microarray analysis of genes in the liver of AhR+/+ and AhR-/- mice may help to clarify the mechanism of AhR-mediated hepatotoxicity and hepatocarcinogenesis by TCDD.  相似文献

Aluminum (Al) toxicity is an important factor in limiting crop production. The present study examined the Al alleviation effects on the growth of hybrid (breeding) and clonal (tissue culture) of D × P oil palm seedlings. The experiment was performed using calcium carbonate (CaCO3), and ground magnesium limestone (GML) and magnesium carbonate (MgCO3) as soil-amendments at different rates in Colombia and Malaysia, respectively. The effects of the treatments were evaluated monthly on vegetative variables and visual symptoms. Chlorophyll concentrations were recorded in Malaysia at the fifth month growing stage. The different amendments improved the soil fertility and it was reflected on better performance of shoot and root growth. The chlorophyll content in the frond number 3 for both materials enhanced significantly when Al saturation was low (0–30%). The results from the experiment revealed the importance of neutralization of Al in reducing its toxicity in oil palm.  相似文献

The change of crystalline structure of Nalita cellulose with tree age has been studied using X-ray diffraction and FT-IR spectroscopy. The proportion of crystallinity and crystal size were increased with tree age. FT-IR spectroscopy showed that the Nalita cellulose was a monoclinic unit cell structure (Iβ). The proportion of crystallinity and crystal size of the 30-month-old Nalita wood was higher as compared to aspen wood. The degree of polymerization (DP) of Nalita cellulose of different ages has also been studied. The DP of cellulose increased with tree age. The DP of Nalita cellulose was lower than that of aspen cellulose. The percentage of glucose in Nalita wood increased with tree age.  相似文献

Hypoxia is a consistent challenge for aquatic animals. It is a pressing environmental problem; hypoxia can cause cranial edema and ovarium dysfunction in fish. Although several studies have reported the effect of hypoxic insult to the visual system, the hypoxic effect on perinatal animals and in particular their offspring has yet to be elucidated. In this study, activated caspase-3 activity was investigated using immunohistochemistry in order to examine the perinatal hypoxic damage in offspring fish. Offspring were divided into groups based on different time points of sacrifice. This allowed assessment of ocular development for different age groups. The results indicated that perinatal hypoxia induced ocular developmental defects in the offspring. The defects took the form of trabecular cell death and fibre degeneration, corneal thinning and lens fibre derangement. A concomitant change in intraocular pressure was recorded by tonometer in the experimental animals compared with the controls. Further investigation should be initiated to develop strategies to prevent developmental disability due to perinatal hypoxia and to increase survivability of the offspring.  相似文献

The effects of dietary substitution of fishmeal with the combined dry Nannochloropsis oceanica (NO) biomass residue and casein on growth performance and carcass composition of juvenile abalone was determined. One thousand two hundred and sixty juvenile abalone were randomly distributed into the 18 L plastic rectangular containers. Five experimental diets in triplicate were prepared. The 28% fishmeal was included into the control (Con) diet. The 25%, 50%, 75% and 100% fishmeal were substituted with the combination of graded levels of dry NO biomass residue and casein, referred to as the NO25, NO50, NO75 and NO100 diets respectively. Finally, the salted sea tangle (ST), Laminaria japonica was prepared to compare the effect of the experimental diets on performance of abalone. The essential amino acids, such as isoleucine, leucine, phenylalanine, threonine and valine tended to increase with dietary substitution of fishmeal with NO biomass residue in the experimental diets. Arginine tended to decrease with dietary substitution of fishmeal with NO biomass residue. Survival of abalone fed the experimental diets was higher than that of abalone fed the ST diet for 16 weeks. Weight gain and specific growth rate (SGR) of abalone fed the NO100 diet were higher than those of abalone fed the all other diets. Weight gain and SGR of abalone linearly increased with dietary substation of fishmeal with NO. Shell length of abalone tended to increase with dietary substitution of fishmeal with the combined dry NO biomass residue and casein. The chemical composition of the soft body of abalone was different among treatments except for moisture content. In conclusion, fishmeal in the diets for abalone could be completely replaced with the combined dry NO biomass residue and casein when the 28% fishmeal was included.  相似文献

The antioxidant properties of the Pacific chub mackerel (Scomber japonicus) muscle protein hydrolysates prepared by enzymatic hydrolysis were investigated. After enzyme hydrolysis at 50°C for 60 min, more than 80% of the S. japonicus muscle protein was hydrolyzed. The highest 2,2-Diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging activity (71.69%) occurred in whole muscle protein hydrolysates treated at 50°C for 30 min with Protamex, and the highest 2,2?-azino-bis(3-ethylbenzothiazolin-6-sulfonic acid) (ABTS) radical-scavenging activity (95.39%) was observed in white muscle protein hydrolysates treated at 50°C for 30 min with Neutrase. The highest superoxide dismutase (SOD)-like activity (32.84%) was recorded in white muscle protein hydrolysates treated at 50°C for 120 min with Protamex. Changes in the molecular weight distribution of S. japonicus muscle proteins after enzymatic hydrolysis were observed by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE). A robust and a convenient enzyme hydrolysis technique for obtaining S. japonicus muscle protein hydrolysates with useful biological activities, within a short time (<2 h) is proposed.  相似文献

Effects of various concentrations of Scutellaria baicalensis (SB) extract in diets on growth, body composition, serum chemistry and disease challenge test of far eastern catfish (Silurus asotus) were determined and compared with a commercially available immune enhancer. Eight experimental diets were prepared in triplicate: control (Con) diet without supplementation of SB and SB‐0.25, SB‐0.5, SB‐1, SB‐2, SB‐3 and SB‐5 diets containing SB at concentrations of 0.25, 0.5, 1, 2, 3 and 5%, respectively. In addition, 0.1% of a commercial immune enhancer product (CP) was also tested. No significant difference in weight gain of fish was found. Feed consumption, feed efficiency ratio and protein retention of fish were not affected by the experimental diets. At the end of the 8‐week feeding trial, 10 externally normal fish from each tank were infected by Vibrio anguillarum or Strepotococcus iniae. Cumulative mortality of fish fed the Con diet was higher than that of fish fed the all other diets in 10 and 25 days after V. anguillarum or S. iniae infection. Results of this study indicated that dietary inclusion of SB extract was effective in improving survival of eastern catfish after V. anguillarum and S. iniae infection, but the various concentrations of SB did not affect fish performance.  相似文献

This study was performed to evaluate stabilization of extracellular matrix (ECM) proteins by polyphenolic fractions in hot water extract (HWE) from Pinus radiata bark against enzymatic degradation and radicals. To obtain polyphenolic fractions with different molecular weight (MW) distribution, HWE was fractionated into monomeric polyphenols (MPP), oligomeric proanthocyanidin (OPA), and polymeric proanthocyanidin (PPA) fractions by monitoring the UV–visible spectrum of the eluted fractions. For HWE, MPP, OPA, and PPA, the collagen (15 mg)-stabilizing capacities at 192 μg/mL were 86, 64, 76, and 88%, respectively, while the elastin-stabilizing capacities at 45.5 μg/mL were 37.6, 1.5, 10.8, and 41.8%, respectively. Among these fractions with different MW distribution, PPA played a significant role in the ECM (collagen and elastin) stabilization in a dose-dependent manner. It could be considered that adsorption of PPA onto the ECM is a significant mechanism of polyphenols for collagen protection. 1,1-Diphenyl-2-picrylhydrazyl (DPPH) free radical- and 2,2′-azinobis(3-ethylbenzothiazoline-6-sulfonate) (ABTS +)-scavenging activity of PPA adsorbed onto collagen showed a dose-dependent increase.  相似文献
