OBJECTIVE: To determine synovial fluid gentamicin concentrations and evaluate adverse effects on the synovial membrane and articular cartilage of tarsocrural joints after implantation of a gentamicin-impregnated collagen sponge. ANIMALS: 6 healthy adult mares. PROCEDURES: A purified bovine type I collagen sponge impregnated with 130 mg of gentamicin was implanted in the plantarolateral pouch of 1 tarsocrural joint of each horse, with the contralateral joint used as a sham-operated control joint. Gentamicin concentrations in synovial fluid and serum were determined for 120 hours after implantation by use of a fluorescence polarization immunoassay. Synovial membrane and cartilage specimens were collected 120 hours after implantation and evaluated histologically. RESULTS: Median peak synovial fluid gentamicin concentration of 168.9 microg/mL (range, 115.6 to 332 microg/mL) was achieved 3 hours after implantation. Synovial fluid gentamicin concentrations were < 4 microg/mL by 48 hours. Major histologic differences were not observed in the synovial membrane between control joints and joints implanted with gentamicin-impregnated sponges. Safranin-O fast green stain was not reduced in cartilage specimens obtained from treated joints, compared with those from control joints. CONCLUSIONS AND CLINICAL RELEVANCE: Implantation of a gentamicin-impregnated collagen sponge in the tarsocrural joint of horses resulted in rapid release of gentamicin, with peak concentrations > 20 times the minimum inhibitory concentration reported for common pathogens that infect horses. A rapid decrease in synovial fluid gentamicin concentrations was detected. The purified bovine type I collagen sponges did not elicit substantial inflammation in the synovial membrane or cause mechanical trauma to the articular cartilage.  相似文献

Money has been said to change people's motivation (mainly for the better) and their behavior toward others (mainly for the worse). The results of nine experiments suggest that money brings about a self-sufficient orientation in which people prefer to be free of dependency and dependents. Reminders of money, relative to nonmoney reminders, led to reduced requests for help and reduced helpfulness toward others. Relative to participants primed with neutral concepts, participants primed with money preferred to play alone, work alone, and put more physical distance between themselves and a new acquaintance.  相似文献

Aortic aneurysm and dissection are manifestations of Marfan syndrome (MFS), a disorder caused by mutations in the gene that encodes fibrillin-1. Selected manifestations of MFS reflect excessive signaling by the transforming growth factor-beta (TGF-beta) family of cytokines. We show that aortic aneurysm in a mouse model of MFS is associated with increased TGF-beta signaling and can be prevented by TGF-beta antagonists such as TGF-beta-neutralizing antibody or the angiotensin II type 1 receptor (AT1) blocker, losartan. AT1 antagonism also partially reversed noncardiovascular manifestations of MFS, including impaired alveolar septation. These data suggest that losartan, a drug already in clinical use for hypertension, merits investigation as a therapeutic strategy for patients with MFS and has the potential to prevent the major life-threatening manifestation of this disorder.  相似文献

Successful clinical management of bacterial pneumonia in horses depends on the administration of an appropriate antimicrobial agent at an adequate dosage and frequency, given by the correct route of administration for an adequate duration. Empiric antimicrobial therapy should be based on the clinician's experience and current veterinary literature. Based on the frequency of Gram-positive bacterial pathogens isolated from horses with bacterial pneumonia, every effort should be made to provide antimicrobial coverage for this class of pathogens. Gram-negative pathogens may co-exist; therefore, broad spectrum antimicrobial coverage may be necessary. Antimicrobial agents posing potential drug related risks (e.g. nephrotoxicity in dehydrated patients, peracute colitis in racehorses) may not be appropriate for empiric use and enrofloxacin offers an enhanced spectrum of antimicrobial activity with reduced risk of complication. Our findings indicate that in cases of exclusive Gram-positive respiratory disease, solo therapy with enrofloxacin is not indicated.  相似文献

We report here validation of the Immulite 2000 Xpi cortisol immunoassay (Siemens; with kit lot numbers <550) for measurement of urine cortisol in dogs, with characterization of the precision (CV), accuracy (spiking-recovery [SR] bias), and observed total error (TEo = bias + 2CV) across the reportable range. Linearity assessed by simple linear regression was excellent. Imprecision, SR bias, and TEo increased markedly with decreasing urine cortisol concentration. Interlaboratory comparison studies determined range-based (RB) bias and average bias (AB). The 3 biases (SR, RB, and AB) and resulting TEo differed markedly. At 38.6 and 552 nmol/L (1.4 and 20 μg/dL), between-run CVs were 10% and 4.5%, respectively, and TEoRB were ~30% and 20%, respectively, similar to observations in serum in another validation study. These analytical performance parameters should be considered for urine cortisol:creatinine ratio (UCCR) result interpretation, given that, for any hypothetical errorless urine creatinine measurement, the error % on UCCR mirrors the error % on urine cortisol. Importantly, there is no commonly used interpretation threshold for UCCR, given that UCCR varies greatly depending on measurement methods and threshold computation. To date, there is no manufacturer-provided quality control material (QCM) with target values for urine cortisol with an Immulite; for Liquicheck QCM (Bio-Rad), between-run imprecision was ~5% for both QCM levels. Acceptable QC rules are heavily dependent on the desired total allowable error (TEa) for the QCM system, itself limited by the desired clinical TEa.  相似文献

In recent years, policy‐makers have sought the development of appropriate tools to prevent and manage introductions of invasive species. However, these tools are not well suited for introductions of non‐target species that are unknowingly released alongside intentionally‐introduced species. The most compelling example of such invasion is arguably the topmouth gudgeon Pseudorasbora parva, a small cyprinid species originating from East Asia. A combination of sociological, economical and biological factors has fuelled their rapid invasion since the 1960s; 32 countries (from Central Asia to North Africa) have been invaded in less than 50 years. Based on a combination of monitoring surveys (2535 populations sampled) and literature reviews, this paper aims to quantify and characterise important invasion parameters, such as pathways of introduction, time between introduction and detection, lag phase and plasticity of life history traits. Every decade, five new countries have reported P. parva introduction, mainly resulting from the movement of Chinese carps for fish farming. The mean detection period after first introduction was 4 years, a duration insufficient to prevent their pan‐continental invasion. High phenotypic plasticity in fitness related traits such as growth, early maturity, fecundity, reproductive behaviour and the ability to cope with novel pathogens has predisposed P. parva to being a strong invader. The Pseudorasbora parva invasion has provided quantitative data for the development of 1) early warning systems across different spatial scales; 2) rapid eradication programmes prior to natural spread in open systems and 3) sound risk assessments with emphasis on plasticity of life history traits.  相似文献

Livestock remains the world’s largest user of land and is strongly related to grassland and feed-crop production. Assessments of environmental impacts of livestock farming require detailed knowledge of the presence of livestock, farming practices, and environmental conditions. The present Europe-wide livestock distribution information is generally restricted to a spatial resolution of NUTS 2 (province level). This paper presents a modelling approach to determine the spatial distribution of livestock at the landscape level. Location factors for livestock occurrence were explored and applied to consistent and harmonized EU-wide regional statistics to produce a detailed spatial distribution of livestock numbers. Both an expert-based and an empirical approach were applied in order to disaggregate the data to grid level. The resulting livestock maps were validated. Results differ between the two downscaling approaches but also between livestock types and countries. While both the expert-based and empirical approach are equally suited to modelling herbivores, in general, the spatial distribution of monogastrics can be better modelled by applying the empirical approach.  相似文献

A multi-stage, continuous culture apparatus has been designed and tested for the production of algae for larval molluscs and crustacea. A single-line system produced a maximum of 2.4 × 1011 cells/day, or 5 g ash-free dry weight of Monochrysis lutheri. Multiple-line systems are recommended for hatcheries. The flow rate affected algal cell density, yield, biomass, protein level, and residual nitrate.Maximal cell yield occurred at 10 I flow per day, a dilution rate of 63% of the volume of the first growth carboy, or 30% of the volume of the total system. The system is also adaptable to growth of larger planktonic algae or mixed cultures of algae and protozoa and/or rotifers.  相似文献
