禽病原性大肠杆菌6种血清型分离株外膜蛋白型的研究   总被引:4,自引:0,他引:4  
从病鸡中分离的6 种血清型( O2、 O46、 O18、 O121、 O76、 O131)及5 种标准血清型( O1、 O15、 O35、 O36、 O78)大肠杆菌中提取外膜蛋白( Outer m em brane protein),经 S D S- P A G E 分析表明,分离株 O76、 O131及标准株 O1、 O35的外膜蛋白型由2 条带组成,为 O M P- 1 型;分离株 O2、 O46、 O18、 O121和标准株 O15、 O36、 O78的外膜蛋白型由3 条带组成,为 O M P- 2 型,结果表明,不同血清型菌株之间有相同的 O M P型。  相似文献

由扬州大学畜牧兽医学院高崧主持的“禽大肠杆菌病流行病学与免疫保护机理的研究”课题于1999年11月13日通过由江苏省科学技术委员会组织的专家鉴定。研究人员收集了我国18个省、市、自治区禽病原性大肠杆菌595株,在440个O血清型中确定了O18,O78,O2,O88,O11和O26为优势血清型,O18,O78,O2和O11具有多样性的OMP型,O88和O26分离株的OMP型具有高度的一致性。研究结果还表明:OMP抗体和LPS抗体均参与了免疫保护;用O18血清型分离株免疫,用同OMP型的O18分离株攻毒,能获得…  相似文献

从陕西省关中17个县鸡场分离出532株大肠杆菌,对其中136株进行血清分型,共分出11个血清型,主要血清型是:O2、O78、O26、O22、O53、O1。药敏试验利高霉素,丁胺卡娜霉素等10种药物对大肠杆菌敏感。选大肠杆菌分型分开菌株和重点菌株制备多价苗,用于易感鸡群,效果理想。  相似文献

微孔蛋白是禽源大肠埃希氏菌的致病因子和保护性抗原。本文用电洗脱法获得禽源大肠地面氏菌O78-g分离株的两种微孔蛋白(porinl和proin2)以皮内多点法免疫家兔,4周后强免疫一次,采血测得ELISA效价分别达1:12800和1:6400。在琼扩反应中porin2免疫兔血清与变性的外膜蛋白(OMPs)间出现了沉淀线,琼扩效价为1:2,该血清与未变性的OMPs间无反应;porinl免疫兔血清与任一  相似文献

通过在甘肃及西北部分地区进行鸡大肠杆菌病流行病学调查,采集病料120份,分出大肠杆菌105株,分离率为85%。对26株大肠杆菌进行了血清型鉴定出血清型10个,其中血清型O78、O1、O2、O114占受检菌株的76.92%。  相似文献

从江苏、广东、河南、新疆、四川、北京、黑龙江等18个省、市、自治区临 有典型大肠杆菌病病变的病禽1051羽采集病料,分离培养和鉴定出595株大肠杆菌。经O血清型测定,除144株未能定型、11株自凝外,共出440个分离株的O血清型。这些 50个血清型,但以O18、O78、O2、O88、O11、O26、O4、为、O1276、O131等10个血清型为主,这些分离株覆盖了60个血清型,但以血O18、O78  相似文献

对湖北省某鸡场110只蛋进行了鸡毒支原体(MG)、滑液支原体(MS)血清平板凝集试验(SPA)具有明显临床症状的鸡(眶下窦炎,呼吸困难,罗音等)均为MG-SPA阳性(54/54)同舍60只无症状鸡中有46只为MG-SPA阳性(76.6%)对4只病鸡及4只MG-SPA阴性健康鸡进行了支原体及细菌的分离鉴定。结果从3只病鸡中分离到鸡毒支原体,从2只病鸡分离到大肠杆菌O2血清型,证明为鸡毒支原体与大肠杆  相似文献

禽大肠杆菌病免疫保护机理的研究   总被引:12,自引:2,他引:10  
以禽病原性大肠杆菌O18、O78分离株制成超声波裂解铝佐剂灭活苗免疫14日龄鸡,以相同或不同外膜蛋白型(Outer membrane protein pattern,OMP型)的O18、O78分离株攻毒。结果表明:O78血清相同和不同OMP型分离株间能获得最大保护;O18血清型相同OMP型分离株间获得最大保护,而不同OMP型分离株间不能保护;上述两个血清型的分离株间不论OMP型是否相同,均缺乏保护。以间接ELISA试验、间接血凝试验分别测定了试验鸡临攻毒前针对大肠杆菌OMPs和脂多糖(Lipopolysaccharide,LPS)的抗体。结果表明:免疫组鸡血清上述两种抗体明显高于攻毒对照组;在免疫组,存活鸡临攻毒前血清中上述两种抗体滴度恒高于死亡鸡,但除3个组外,多数组差异不显著。攻毒对照组这一关系不稳定。结果说明:禽大肠杆菌疫苗的免疫保护,主要与O血清型有关,部分与OMP型有关,如O18分离株,免疫保护性抗原含OMPs,LPS等多个抗原表位。  相似文献

从陕西省部分优势血清型的禽源性大肠杆菌中提取外膜蛋白 (outer membrane protein,OMP) ,用 SDS- PAGE进行 OMP分型。 18株菌共产生了 3种 OMP型 ,其中 4株 O1菌株 ,4株 O2菌株 ,4株 O78菌株和 3株 O89菌株各属2个 OMP型 (OMP- 1,2型 ) ;3株 O75菌株属 3个 OMP型 (OMP- 1,2 ,3型 )。结果表明 ,陕西省分离的优势血清型禽源大肠杆菌具有多样性的 OMP型 ,且多种血清型间具有共同的 OMP型。  相似文献

Characters of Escherichia coli 078 isolated from septicaemic animals   总被引:2,自引:0,他引:2  
Twenty-one Escherichia coli isolates of serogroup 078 from animal septicaemia were obtained from laboratories in France, England and Canada. The bacteria were compared for outer membrane protein (OMP) patterns, lipopolysaccharide patterns, surface proteins of fimbrial types, biotypes, antibiotypes, colicin production, hydroxamate production and virulence in mice. Sixteen isolates from bovine, ovine, porcine and avian species in France and England had a similar OMP pattern. This characteristic associated with minor properties like surface proteins type, colicin V production and virulence in mice made these 16,078 E. coli isolates from 4 animal species, good candidates for the same clonal grouping. The five other bovine isolates with "Vir" or "31a" phenotypes were heterogeneous for most of the characteristics studied.  相似文献

某大型鸡场的28个分场分离到的菌株,经生化检验为大肠杆菌的有87株,经O血清测定定型65株,4株自凝,18株未能定型,其中O78占20株,O1占14株,O2占5株,这些分离株中O78、O1、O2有39株占定型菌株的60.0%,因此可以认定O78、O1和O2为该大型鸡场的优势血清型,以不同场分离的菌株,以每分离株10^7菌落形成单位(CFU)细菌气管接种1日龄易感鸡,根据接种后7天内发生死亡和病变鸡的比例,确定出高致病株、中等致病株和低致病株分别为87.36%、8.05%和4.59%。  相似文献

对2005年5月~2016年3月自河北、山东、河南、江苏、陕西、辽宁等省发病鸡场疑似大肠杆菌病的病死鸡病变部位分离的194株禽致病性大肠杆菌进行了生化特性测定、O抗原血清群鉴定及药敏试验。试验用5种大肠杆菌单因子(O1、O2、O57、O65、O78)阳性血清进行O血清群鉴定,确定了124株O血清群,占鉴定菌株数的63.9%(124/194),其中O1 3株、O2 26株、O57 15株、O65 47株和O78 33株,O65和O78为优势菌株,而O2和O57血清群菌株次之,O1血清群菌株最少。8种药物(氨苄西林、新霉素、多西环素、磺胺间甲氧嘧啶、替米考星、环丙沙星、头孢噻呋和氟苯尼考)的药敏试验结果表明,受试的5株大肠杆菌分离株均对环丙沙星和新霉素敏感,而对其他药物均有不同程度的耐受。本研究结果表明O65、O78、O2和O57血清群高度流行,为进一步研发多价菌苗奠定了基础。  相似文献

One hundred and ninety four avian pathogenic Escherichia.coli (APEC) isolated from different pathotypes of chickens in Hebei province,Shandong province,Henan province,Jiangsu province,Shanxi province and Liaoning province in China between May 2005 and March 2016 were investigated by determination of biochemical characters,O serogroup identification and drug sensitivity test.Out of 194 isolates, 124 strains(63.9%) could be grouped to serogroups O1(3),O2(26),O57(15),O65(47) and O78(33).The drug sensitivity test used eight antibiotics (ampicillin, neomycin, doxycycline, sulfamonomethoxine,tilmicosin,ciprofloxacin ceftiofur and florfenicol) by one strain of O1,O2,O57,O65,O78 serogroup,respectively.The results showed that all strains were sensitive to ciprofloxacin and neomycin, and resistance to other drugs at varying degrees.This study demonstrated that the high prevalence of O65,O78,O2 and O57 isolates existed and provided the basis for the development of multivalent bacterin in future.  相似文献

Forty-four western Canadian isolates of Escherichia coli associated with colibacillosis of turkeys and chickens were examined for serotype, antibiotic resistance, and production of aerobactin. The isolates belonged to fourteen O serogroups, with 39% of the strains being non-typeable. A high frequency of resistance to tetracycline, kanamycin, neomycin, cephalothin, streptomycin and erythromycin was observed. Most isolates produced aerobactin. Ten E. coli belonging to serogroups O1, O2 and O78 were also examined for pili production, hemagglutination, serum sensitivity, production of iron-regulated outer membrane proteins (IROMPS), and virulence. All isolates examined produced pili, exhibited mannose-sensitive hemagglutination of avian red blood cells and produced IROMPS under iron-restricted growth conditions. The five isolates of serogroup O1 and O2 were resistant to killing by turkey serum and were highly virulent. Only two of the five isolates of serogroup O78 were serum resistant. No correlation between serum resistance and virulence was observed in serogroup O78.  相似文献

In total, 83 avian pathogenic Escherichia coli (APEC) isolates from avian colibacillosis during a period from 2001 to 2006 in Japan were investigated for serogroups, typical virulence factors, antimicrobial susceptibility, and genetic relatedness. The most common serogroup was O78 (30.1%); 80.7% of isolates harbored the iss gene and 55.4% of isolates harbored the tsh gene. Antimicrobial resistance of the isolates was found for ampicillin (77.1%), oxytetracycline (75.9%), kanamycin (36.1%), fradiomycin (33.7%), trimethoprim (25.3%), enrofloxacin (21.7%), and florfenicol (6.0%). Although multiple antimicrobial-resistant phenotypes (three or more antimicrobials) accounted for 54.2% of isolates, no isolate exhibited resistance to all agents tested. The fluoroquinolone-resistant isolates had point mutations in GyrA (Ser83 --> Leu, Asp87 --> Asn) and ParC (Ser80 --> Ile, Glu84 --> Gly). Of 18 enrofloxacin-resistant E. coli isolates, nine isolates belonged to serotype O78. In PFGE analysis, eight of the nine enrofloxacin-resistant O78 isolates were classified into an identical cluster. This suggests that a specific genotype of fluoroquinolone-resistant O78 APEC may be widely distributed in Japan.  相似文献

从山东省临沂、烟台、滨州、济南、聊城、德州等15地、市的解剖诊断门诊部收集临诊上有典型病变鸡(鸭)病的病料230余份中分离出大肠杆菌165株,115株大肠杆菌进行了O血清型鉴,共鉴定出84个分离株的O血清型,鉴定结果表明O78、O36、O6与O107是本次调查的优势血清型.挑选59株进行致病性试验.动物试验结果表明94.9%的菌株对7日龄内雏鸡具有很强的致病作用,多数菌株可使接种雏鸡100%死亡  相似文献

The objective of this study was to characterize virulence factors of Escherichia coli isolates from broilers with simultaneous occurrence of cellulitis and other colibacillosis lesions. Thirty flocks were sampled and 237 birds with cellulitis were examined. Eighty-two (34.6%) of 237 birds condemned for cellulitis had gross lesions in the heart, air sacs, joints, or liver. In 58 chickens, E. coli was isolated from both the cellulitis and other lesions of colibacillosis, and 18.9% of the E. coli isolates from the 2 types of lesions belonged to the same O group. Escherichia coli of serogroups O78, O1, and O2 predominated. Isolates of the same serogroup that were derived from different lesions in the same birds had similar patterns of biotype, aerobactin production, serum sensitivity profile, antibiotic sensitivity, and K1 capsule production. Escherichia coli derived from cellulitis lesions produced virulence factors similar to those found in E. coli isolated from other colibacillosis lesions in poultry.  相似文献
