Fetal fluid, diaphragm muscle, heart, kidney, and liver from 14 four- to five-month-old calf fetuses were analyzed for Zn, Cu, Cd, Cr, Ni, Pb, and Fe. The same tissues and bone were examined from postparturient calves 30, 60, and 120 days of age. Weaned calves (8 to 9 months old) were necropsied and diaphragm muscle, heart, kidney, liver, brain, and bone were analyzed. Additional calves from cows exposed or not exposed to anaerobically digested sludge were placed in a feed yard at weaning for a feeding period of 150 days and then were killed and necropsied. Heavy metal content of the aforementioned tissues was compared with the same tissues obtained from peer calves weaned, killed, and necropsied 150 days earlier. Zinc, Cu, and Fe were up to 11 times higher in fetal livers than in control or exposed postparturient calves. These concentrations declined after birth and stabilized at approximately the concentrations observed in adult cattle at about 12 to 14 months of age. Cadmium accumulated to a greater extent in kidneys and livers of exposed calves than in control calves. However, the accumulation did not produce a detectable effect upon the functional or morphologic characteristics of the organs examined. Serum iron (SI)-related and hematologic changes were evaluated in a herd of weaned pigs inoculated with a strain of Salmonella cholerae-suis, causing 83% mortality within 22 days after inoculation was done. Serum iron concentrations decreased to 35% of base-line values 2 days after inoculation was done, but recovered to near base line subsequently. Total SI-binding capacity (TIBC) decreased gradually for 14 days after inoculation was done. Transferrin (TF) concentrations decreased to near half the base line throughout the postinoculation observation period. The calculated SI saturation coefficient decreased to half the base line, but recovered to or above the base-line value subsequently. Combined observations of SI, TIBC, TF, and SI saturation coefficient concentrations indicated that there was higher saturation of host iron-binding proteins and recruitment of additional iron-binding systems subsequent to 2 days after inoculation was done. Day 2 after inoculation seemed to be a critical period for host iron metabolism. Injection of supplemental iron dextran simultaneously with Salmonella infection resulted in lower mortality of iron-injected pigs (P less than 0.005). A highly significant negative correlation was observed between SI concentration and rectal temperatures after pigs were inoculated with Salmonella (r = -0.54; P less than 0.0001). Hemoglobin concentrations, mean corpuscular volume, and mean corpuscular hemoglobin were not significantly affected by Salmonella infection or iron injection concurrent with Salmonella infection.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献

Sodium selenite was mixed in a commercial pig food so as to increase the original selenium content (0.126 ppm) by 1.2 ppm. The supplemented food was given to 7 pigs during 78 days. A control group of as many pigs was given the non-supplemented commercial food. The pigs were killed, starting 4 hours after the last selenium-supplemented food had been given and then with intervals during the next 14 days. Small but significant increases of tissue selenium were noticed in liver and striated muscle as compared with the control animals. The kidneys showed no increase.  相似文献

Three litters of pigs were weaned at 21 days of age, and 3 others were left with the sow. Pigs were killed at 21, 23, 28, or 39 days of age. Weaned pigs exhibited anxiety, gastrointestinal dysfunction, and decreased rate of body weight gain. Plasma glucose or liver glycogen concentrations were not decreased by weaning. Adrenal gland weights and tyrosine hydroxylase (EC 1.14.3a), dopamine beta-hydroxylase (EC 1.14.2.1), phenethanolamine-N-methyl transferase (EC 2.1.1), and monoamine oxidase (EC 1.4.3.4) activities were increased after weaning. Adrenal catecholamine and cortisol levels and dopa decarboxylase (EC 4.1.1.26) and catechol-o-methyl transferase (EC 2.1.1.6) activities were not significantly altered, although some increases were indicated. Cranial cervical ganglionic choline acetyltransferase (EC 2.3.1.6) and tyrosine hydroxylase activities were increased after weaning. Weaning of swine at 21 days of age is a stressful experience, and many effects persist for at least 18 days; however, growth was no longer impaired 18 days after weaning.  相似文献

Oxidative stress can induce abnormal tryptophan metabolism.The present study was mainly conducted to determine the effect of dietary tryptophan levels on oxidative stress in the liver of weaned pigs challenged by diquat.A total of 36 PIC piglets weaned at 21 days of age were randomly allotted to 1 of 3 diets containing dietary tryptophan levels of 0.18,0.30,and 0.45%for 14 d.On day 8,the piglets were injected intraperitoneally with sterile0.9%NaCI solution or diquat(10 mg/kg body weight).During the first 7 d of trial,increasing dietary tryptophan levels enhanced average daily gain(P=0.09) and average daily feed intake(P=0.08),and decreased the feed efficiency(P<0.05) of piglets.The growth performance was decreased by diquat injection(P<0.05).Diquat injection also decreased the activities of the superoxide dismutase(SOD) and glutathione peroxidase(GPx) in the plasma and liver(P<0.05),increased plasma malondialdehyde(MDA)(P<0.05) and urea nitrogen(P<0.05) concentrations,and enhanced MDA concentration(P =0.09) and tryptophan 2,3-dioxygenase(TDO) activity(P =0.07) in liver of piglets.Increasing dietary tryptophan levels could attenuate the effects of diquat injection on the MDA(P =0.06) concentration and the activities of SOD(P=0.09) and GPx(P=0.05) of the liver,and plasma urea nitrogen(P =0.06) concentration in the piglet.There was a synergistic role for increasing TDO activity in the liver between dietary tryptophan levels and diquat injection(P<0.05).These results suggest that increasing dietary tryptophan levels could attenuate the oxidative stress of the liver in weaned piglets intraperitoneally injected with diquat via enhancing the antioxidant capacity.  相似文献

To examine the effects of exogenous porcine (p) ST on measures of stress and immune function in weaned pigs with or without transport, pigs (20 +/- 1 d of age) received daily injections of pST (0.5 mg/kg; n = 16) or saline (n = 16) for 5 d. On d 5, a blood sample was collected immediately before injection. At 4 h postinjection, pigs were weighed, sampled for blood, injected with di-nitrophenyl-conjugated keyhole limpet hemocyanin, and weaned. One half of the pigs in each group were transported for 3 h before placement in the nursery. Pigs were weighed, and blood was collected on 1, 7, and 14 d postweaning. Statistical significance was set at P < 0.05. Serum IGF-I concentrations were increased by pST and decreased by weaning, but not affected by transport. The free cortisol index was elevated in all pigs 1 d postweaning, although less in transported versus nontransported pigs. By 7 d postweaning, the free cortisol index returned to prewean values. Serum concentrations of immunoglobulin (Ig) G increased in all pigs by 14 d postweaning, but were not affected by pST or transport. Serum IgM concentrations were elevated at 7 and 14 d postweaning. Before weaning and again 1 d postweaning, pigs treated with pST had greater concentrations of IgM than did control animals. Circulating neutrophils increased in pST-treated pigs 4 h after the final pST injection. Improved immune function in weaned pigs by pST may lead to greater health and growth in a commercial setting.  相似文献

The distribution in the mouse of Se75 after injection of Se75 sodium selenite was studied by autoradiography of sagittal whole-body sections. The animals were killed at various times, from 5 minutes to 8 days, after a single intravenous injection.The distribution was characterized by a high blood level of selenium during the whole observation period.The highest concentration of selenium soon after the injection was found in the liver, heart, kidney and adrenal cortex. Eight days after the injection the highest concentration was seen in the liver and the kidney. The selenium penetrated the placenta, but the uptake was much lower in the fetuses than in the maternal tissue.  相似文献

The dynamics of selenium in the blood serum of 20 lambs of the Merino breed, traditionally reared, was followed from birth to weaning at the age of 110 days. Results of the observations revealed that the highest concentration of serum selenium was determined in lambs just after delivery (0.25 ppm) and the level was maintained throughout the period of colostral nutrition. In the period between the 15th and 60th day low selenium values (0.06-0.13 ppm) were recorded, which implies a critical period in relation to the origin of muscular dystrophy. At the end of the observations there appeared a significant selenium content increase in the blood serum of lambs.  相似文献

Twenty Angus cross heifers were fed a complete diet which contained 0.07 mg selenium/kg dry matter. Thirteen were injected subcutaneously with barium selenate at a dose rate of approximately 1 mg selenium/kg bodyweight and seven remained untreated. All the heifers were slaughtered during the following 121 days, the last of the treated group 119 days after injection. Glutathione peroxidase activity in blood increased within four weeks of administration and remained high thereafter. The selenium dependent glutathione peroxidase activity did not increase in liver kidney or muscle. The concentrations of selenium in the blood, liver and muscle were increased significantly from 30 days until 119 days. Between 76 and 99 per cent of the selenium injected remained at the site of injection.  相似文献

The investigation comprised 12 herds, totalling 660 sheep, in the counties of Stockholm and Uppsala. The selenium content of specimens of forage, whole blood, muscle, liver, and kidney was determined by a fluorimetric method. Data on the number of born and dead lambs were collected 4—5 months after lambing. Dead and sick lambs were subjected to necropsy and to clinico-chemical analyses, respectively.The selenium content of hay and corn was lower than 60 ng per g, which some authors regard as the minimum level of Se requirement for protection against white muscle disease in sheep. The analyses of forage mixtures containing oil concentrates and beet pulp showed selenium levels that were about 2—5 times as high as this minimum level.In herds in which the animals had not been treated with selenium preparations prior to sampling, there was a significant positive correlation between the selenium content of the forage and that of whole blood. A relationship was also noted between the selenium content of forage and of organs.In 9 out of the 12 herds the selenium concentrations in whole blood were on the level that is considered to be representative of white muscle disease.A high mortality rate was recorded for lambs whose mothers had low selenium levels in their blood.Prophylactic and curative treatment with selenium plus vitamin E orally and/or parenterally probably reduced lamb mortality rate in the herds fed a low-selenium diet.  相似文献

Attaching and effacing Escherichia coli (AEEC) adhere to mucosal epithelium in both small and large intestine and induce a distinctive lesion characterized by an irregular scalloped appearance of the epithelial layer. Infection with attaching and effacing E. coli was detected in 14 calves, 7 pigs, 2 lambs, and 3 dogs. Affected animals were from farms and kennels in South Dakota, Minnesota, Iowa, Nebraska, and Wisconsin. Ages of affected animals were calves, 2 days to 4 months; pigs, 1-6 weeks; lambs, 1 week; and dogs, 7-8 weeks. Clinical signs included diarrhea in all animals, but other nonenteric disease problems were present in some animals. Concurrent infection with other enteropathogens was detected in 9 calves and 5 pigs. Infection with AEEC appeared to be the sole cause of illness and death in some animals. There was evidence of intestinal hemorrhage in 5 of the calves and in all 3 dogs. Attaching and effacing lesions varied from small scattered foci to widespread involvement of large areas of intestinal mucosa. Verotoxin was produced by E. coli strains isolated from 9 calves, but not by strains from pigs, lambs, or dogs.  相似文献

The toxicity of fenbendazole, an anthelmintic for use in swine, was tested by feeding 8 growing pigs 2,000 mg of fenbendazole/kg of body weight daily for 14 days. A transient leukopenia developed on day 6, but values returned to base line on day 18, 4 days after discontinuation of dosing. Sorbitol dehydrogenase values were significantly (P less than 0.05) increased from day 4 and returned to base line on day 20. Marked gross or histopathologic lesions were not found.  相似文献

The LD50-values of sodium selenite after i/m injection to rabbits was determined by means of probit-regression straight line. Simultaneously a combination of vitamin E and sodium selenite was tested for toxicity in order to examine the effect of vitamin E on the selenium toxicity. The LD50/24h was 2.53 mg sodium selenite/kg body mass and in combination with vitamin E (30 IU vitamin E/5 mg NaSeO3) 2.73 mg/kg. Furthermore the selenium concentration was determined in blood, liver, kidney and muscles of animals which died or were killed after 24 h respectively. Not only chemical analysis but also relations of time-dose-efficiency and significantly increased LD50-values show a certain protective effect of vitamin E on selenium intoxication.  相似文献

The sequential tissue distribution of virus was investigated using virus isolation and immunofluorescence tests in 1-day-old piglets inoculated with porcine circovirus 2 (PCV2) and/or porcine parvovirus (PPV). Enlarged mesenteric lymph nodes were seen in the pig inoculated with PCV2 alone and killed at 26 days post-inoculation (PI). One of the pigs inoculated with PCV2 and PPV and killed at 21 days PI had an enlarged liver. The pig killed at 26 days PI in this group had enlarged liver, kidneys and heart. Histopathological changes were seen in lymphoid tissues of the pigs inoculated with PCV2 alone and killed at 14 and 26 days PI. Similar, but more severe, lesions were observed in the pigs infected with PCV2 and PPV and killed from 10 days PI onwards. Histological lesions of nephritis, pneumonia and hepatitis were also apparent in these animals. Mild nephritis was also seen in the pigs infected with PPV alone and killed at 14 and 26 days PI. Moderate amounts of PPV antigen were detected in tissues from the pigs inoculated with PPV alone and killed at 14 days PI. Low levels of PCV antigen were detected, mainly in lymphoid tissues, in the pigs inoculated with PCV alone and killed at 14 days PI. Low to moderate amounts of PCV antigen were detected in a wider range of tissues in the pig in this group killed at 26 days PI. In the pigs inoculated with both viruses, PPV antigen was detected in tissues of pigs killed from 3 to 26 days PI with maximal amounts detected between 6 and 14 days PI. PCV2 antigen was detected in low to moderate amounts in the tissues of pigs killed at 14 days PI. Large amounts of PCV2 antigen were detected in most of the tissues from pigs in this group killed between 17 and 26 days PI. Virus isolation results for PCV2 generally correlated well with the results for immunofluorescent staining. PPV was isolated from almost all tissues from pigs inoculated with PCV2 and PPV, a much higher incidence of positive tissues than observed for immunofluorescent staining.  相似文献

In May 1996, 150 grower pigs in 5 California counties were exposed to selenium-contaminated feed distributed by a single feed company. Feed samples from 20 herds had a mean selenium concentration of 121.7 ppm dry weight (range, 22.1-531 ppm). In San Luis Obispo County, 52 pigs in 24 herds were exposed to the feed, and 8 pigs died with signs of paralysis. Bilateral symmetrical poliomyelomalacia involving the ventral horns of the cervical and lumbar intumescence was evident on histologic examination of spinal cord from affected pigs. Of 44 surviving exposed pigs, 33 (75%) exhibited signs of selenosis, including anorexia, alopecia, and hoof lesions. Thirty-nine of 44 pigs (88.6%) had elevated (>1 ppm) blood selenium concentrations. Surviving exposed pigs were changed to a standard commercial ration containing approximately 0.5 ppm (dry weight) selenium. Blood selenium concentrations were determined weekly for 46 days following removal of the contaminated feed and were compared with values of 20 control pigs fed a standard commercial ration. Mean (+/-SD) blood selenium concentrations of exposed pigs were 3.2 +/- 2.6 ppm at the initial sampling and 0.4 +/- 0.1 ppm after 46 days. Mean blood selenium concentrations of < or = 0.3 ppm for control pigs at all samplings were significantly lower (P < 0.001) than concentrations for exposed pigs. Muscle and liver samples of 22 of the 44 exposed pigs were collected at slaughter approximately 72 days after withdrawal of the selenium-contaminated feed. Muscle samples had a mean selenium concentration of 0.36 ppm (wet weight). Liver samples had a mean selenium concentration of 1.26 ppm (wet weight). One liver sample had a selenium value in the toxic range for pigs (3.3 ppm wet weight; reference range, 0.4-1.2 ppm). A 1-compartment pharmacokinetic model of selenium elimination in exposed pigs was generated, and the geometric mean blood selenium elimination half-life was estimated to be 12 days. The 60-day withdrawal time recommended by the Food Animal Residue Avoidance Database was considered sufficient to allow safe human consumption of tissues from exposed pigs.  相似文献

The effects of pneumonia on the pharmacokinetics of erythromycin administered IM and the tissue concentration changes with time were evaluated in 2-month-old calves. Pneumonia was induced by injection of Pasteurella haemolytica cultures through the thoracic wall into each lung. Six days prior to induction of pneumonia, erythromycin (15 mg/kg) was administered in a single IM dose. Erythromycin was administered again 48, 72, and 96 hours after injection of P haemolytica. On the third day of erythromycin administration (96 hours), the calves were serially euthanatized in groups of 4 calves each at 2, 5, 8, 12, 18, and 24 hours after the final dose was given. Tissue concentrations of erythromycin in kidney, liver, lung, muscle, CSF, and serum were determined. Neither the serum concentrations nor the overall pharmacokinetic values were significantly (P less than or equal to 0.05) changed by pneumonia. The concentrations of erythromycin were maximal at 5 hours for liver, muscle, and serum and at 8 hours for CSF, kidney, and lung. Serum and muscle concentrations were similar, whereas concentrations in CSF were lower than in serum and higher in kidney, liver, and lung. The lung/serum ratios were approximately 2.5 to 3 at 8 through 24 hours after IM administration. The peak concentration in lung was approximately 6 micrograms/g at 8 hours.  相似文献

Two trials were undertaken with lambs grazing selenium deficient pasture to determine if copper would enhance liveweight and fleece-weight responses to selenium supplementation. In the first trial, lambs given selenium or selenium plus copper gained significantly more weight and had higher fleece-weights after 260 days than did control lambs or lambs given copper alone. Copper given alone or together with selenium had no significant effect on liveweight or fleece-weight when compared with control lambs and lambs that were given selenium alone respectively. This finding was confirmed in a second trial when growth and fleece-weights of selenium and selenium plus copper treated lambs were compared and no significant differences found. In both trials copper significantly raised liver copper levels. In selenium supplemented but not in selenium deficient sheep, copper significantly increased blood selenium levels.  相似文献

Ghrelin action, which stimulates growth hormone (GH) secretion, may alter during the weaning period in calves. Our objective was to compare the effects of intravenous ghrelin injection on plasma GH, insulin and glucose concentrations in calves around the weaning period. Four Holstein bull calves were fed whole milk and allowed free access to solid feeds, and weaned at 7 weeks of age. Measurements were performed at weeks 1, 2, 4, 6, 7, 9, 11 and 13, when calves were intravenously injected with ghrelin (1.0 μg/kg body weight (BW)) through a catheter, and jugular blood samples were obtained temporally relative to the injection time. Estimated digestible energy intake per metabolic BW transiently decreased at week 7 because of low solid intake immediately after weaning, and thereafter gradually increased. Plasma insulin and glucose concentrations were not affected by ghrelin injection at all ages. In contrast, plasma GH concentrations increased with ghrelin injection at all ages. The incremental area of GH at week 7 was greatest and significantly higher compared with weeks 2, 4, 6 and 9. This result suggests that nutrient insufficiency immediately after weaning enhances GH responsiveness to ghrelin.  相似文献

Two experiments were conducted to evaluate the effects of weaning swine at 2 or 5 wk of age on postweaning performance and fat metabolism. In the first experiment, 52 pigs were weaned at 2 or 5 wk of age with body weights determined from birth to 8 wk. The early weaned group was fed a 20% protein corn-soybean meal-oat diet containing 25% dried whey from 2 to 5 wk while both groups were fed a 20% protein cereal grain-based diet from 5 to 8 wk of age. In a second experiment, a total of 90 pigs weaned at similar ages and fed the same diet sequences were killed at weekly intervals from 2 to 8 wk of age to evaluate body fat content and lipogenesis in liver and adipose tissue. Lipogenic capacity was measured by incorporation of acetate-1 14C into the total lipid fraction in liver slices and adipose tissue minces or by monitoring liver ATP citrate lyase activity. The results demonstrate that pigs weaned at 2 wk experience a slower postweaning growth rate with lower empty body weights than those either concurrently nursing the dam or weaned at 5 wk of age. Both groups had similar body weights from 6 to 8 wk of age. The body fat content of nursing pigs increased from 2 to 5 wk of age. Pigs weaned at 2 wk lost approximately 25% of their body fat the first week postweaning while later-weaned pigs did not lose body fat postweaning. Body fat composition of both groups was similar by 8 wk of age. Lipogenic activity was higher in liver than in adipose tissue from 2 to 5 wk of age and remained relatively constant throughout the trial. Adipose tissue lipogenic activity was lower in the nursing pig but increased dramatically at 5 wk in the early-weaned group and 7 wk of age in the late weaned group. These results suggest that weaning age can affect postweaning body fat composition and that adipose contributes a greater lipogenic capacity than liver tissue as the pig matures.  相似文献

In a series of five 17-d replicate trials, a total of 54 cannulated and 12 noncannulated pigs were used to determine the effects of weaning age (17 d or 24 d) on pH, dry matter percentage, aerobic and anaerobic microflora, lactate, and volatile fatty acid (VFA) concentrations in the jejunum, ileum, and cecum of weanling pigs. At -14 d of age, cannulated pigs were surgically fitted with T-cannulas in the jejunum (n = 20), ileum (n = 18), or cecum (n = 16). Upon weaning, cannulated pigs were individually caged in an environmentally controlled room with ad libitum access to a phase starter diet and water. Noncannulated pigs were killed at weaning and samples were collected from the jejunum, ileum, and cecum. Digesta and fecal swabs from cannulated pigs were collected twice weekly. The pH of cecal contents was lower (P < 0.05) and dry matter percentage was greater (P < 0.05) than those ofjejunal or ileal contents. Pigs weaned at 24 d of age had increased (P < 0.05) E. coli populations 3 d postweaning compared to preweaning populations, regardless of site of collection, whereas this increase was not observed in pigs weaned at 17 d of age. Unweaned pigs maintained higher (P < 0.05) lactobacilli populations compared to weaned pigs; however, populations declined (P < 0.05) in both groups by 3 d postweaning, with pigs weaned at 24 d of age having lactobacilli populations greater than pigs weaned at 17 d of age. Fecal populations of E. coli and lactobacilli declined (P < 0.05), whereas fecal bifidobacteria populations increased (P < 0.05) postweaning, regardless of weaning age. Concentrations of total fecal anaerobes declined (P < 0.05) in pigs weaned at 17 d of age but were maintained in pigs weaned at 24 d of age. Volatile fatty acid concentrations were greater (P < 0.05) in the cecum than in the jejunum or ileum, and acetic acid concentrations decreased (P < 0.05) postweaning regardless of weaning age. A tendency for L+ lactate concentrations to be greater (P < 0.07) in the ileum and jejunum vs the cecum was observed. Results indicate that weaning and weaning age have significant effects on microbial populations and VFA concentrations.  相似文献
