Forecasting the Genetic Impacts of Net Pen Failures on Gulf of Mexico Cobia Populations Using Individual‐based Model Simulations 下载免费PDF全文

Offshore net pen fish farming provides a cost‐efficient means for production of marine finfish, and there is great interest in development of net pen operations in domestic waters. However, there are concerns over the possible genetic and ecological impacts that escaped fish may have on wild populations. We used individual‐based simulations, with parameter values informed by life history and genetic data, to investigate the short‐term (50 yr) impacts of net pen failures on the genetic composition of cobia, Rachycentron canadum, stocks in the Gulf of Mexico. Higher net pen failure rates resulted in greater genetic impacts on the wild population. Additionally, the use of more genetically differentiated source populations led to larger influxes of non‐native alleles and greater temporal genetic change in the population as a result of net pen failure. Our results highlight the importance of considering the appropriate source population for broodstock collection in net pen aquaculture systems and help to provide a general set of best management practices for broodstock selection and maintenance in net pen aquaculture operations. A thorough understanding of the genetic diversity, stock structure, and population demography of target species is important to determine the impact escapees can have on wild populations. 相似文献

Surface runoff, erosion, compaction and the leaching of potential pollutants from land can degrade the soil resource and damage the water environment, reducing crop yields, causing loss of valuable nutrients and organic matter, together with increasing flood risk. Increasingly, it is recognized that scientific information must be translated into practical tools to change practices and protect the soil and water resource. Working alongside agencies in Scotland, we applied a suite of simple, transparent, rule-based models to identify areas most at risk of exporting sediment and pollutants that may degrade water quality based on field-scale (1:25,000) soil maps. The maps have been used by Scottish Environment Protection Agency, Scottish Water and Scottish Government to assess soil risks to waters from field to regional scale. The work brought scientists together with policy makers, agencies and the water industry to pool their knowledge to apply these practical tools for decision-making. It highlights the need to apply existing knowledge to answer salient questions. All three examples described show that providing the right type of information, which is based on fundamental soils data, can directly influence the implementation of policies, investment and monitoring decisions and provide evidence in support of government. However, this requires both researchers and agency scientists to develop skills as knowledge brokers and to normalize the use of soil data in everyday agency settings. 相似文献

The change in soil carbon (C) concentration, soil pH and major nutrients for approximately 1,000 topsoil sampled from on-farm experimental sites over a thirty-year period from 1950 to 1980 in north-east Scotland are summarized. This period coincided with increased agricultural intensification, which included regular liming and fertilizer additions. During 2017, 37 of these sites were resampled and reanlaysed. While pH and extractable phosphorus (P) and potassium (K) increased over this period, there was no detectable change in the percentage loss on ignition. Composite soil samples were taken by auger from a depth of 0–15 cm and compared with the corresponding archived samples collected at the initiation of each experiment. Analysis of these resampled soils indicated no significant change in soil carbon (C), although soil pH, extractable magnesium (Mg) and K and Nitrogen (N) concentrations were significantly greater (p < .001) but extractable soil P concentration was significantly less (p = .015) compared with the original samples. Even though measuring C concentration alone is a poor indicator of overall changes in soil C stocks, it does provide a relative quick “early warning” of C losses that would justify a more comprehensive measure of stocks. 相似文献

The aim of this study was to examine the effect of varying intracellular reactive oxygen species (ROS) levels during oocyte in vitro maturation with enzymatic ROS production systems (xanthine + xanthine oxidase or xanthine + xanthine oxidase + catalase), scavenger systems (catalase or superoxide dismutase + catalase) or cysteine on porcine oocyte maturation. Oocyte ROS levels showed an increase when H2O2 or O2?‐ production systems were added to the culture medium (p < 0.05). On the other hand, the presence of ROS scavengers in the maturation medium did not modify oocyte ROS levels compared with the control after 48 h of maturation, but the addition of cysteine induced a decrease in oocyte ROS levels (p < 0.05). The ROS production systems used in this work did not modified the percentage of oocyte nuclear maturation, but increased the decondensation of sperm head (p < 0.05) and decreased the pronuclear formation (p < 0.05). In turn, the addition of O2?‐ and H2O2 scavenging systems during in vitro maturation did not modify the percentage of oocytes reaching metaphase II nor the oocytes with decondensed sperm head or pronuclei after fertilization. However, both parameters increased in the presence of cysteine (p < 0.05). The exogenous generation of O2?‐ and H2O2 during oocyte in vitro maturation would not affect nuclear maturation or later sperm penetration, but most of the spermatozoa cannot progress to form the pronuclei after fusion with the oocyte. The decrease in endogenous ROS levels by the addition of cysteine would improve pronuclear formation after sperm penetration. 相似文献

Summary The receptivity of graft-polyacrolein on wood to hydroxylamine is discussed in terms of a geometric model for the voids and surfaces formed in wood by the swelling action of water. The receptivity of the carbonyl group of graft-polyacrolein varies inversely with the number of graft sites. This relationship is a consequence of decrease in the spacing between graft chains on the primary surface. The receptivity increases gradually with an increase in the amount of graft polymer, and this is a result of the swelling action of graft-polyacrolein which extends to the microscopically visible macropores of wood. 相似文献

Orchard and vineyard producers conduct preplant site evaluations to help prevent planting permanent tree and vine crops on
lands where the crop will not perform to its highest potential or attain its full life expectancy. Physical soil characteristics
within specific soil profiles and spatially throughout an orchard influence decisions on land preparation, irrigation system
selection, horticultural choices, and nutrient management. Producers depend on soil surveys to help them understand the soil
characteristics of the land and may be interested in technology that provides additional information. Electromagnetic induction
(EM38) and four-probe soil resistance sensors (VERIS) are being used in combination with global positioning systems to map
spatial variability of soils using apparent soil electrical conductivity (ECa). The hypothesis evaluated in this study is
whether rapid, in situ, and relatively low-cost methods of measuring ECa (EM38 and VERIS) can effectively identify and map
physical soil variability in non-saline soils. The supposition is that in non-saline soils, ECa levels will relate well to
soil texture and water-holding capacity and can be used to map physical soil variability. In turn, the information can be
used to guide decisions on preplant tillage, irrigation system design, water and nutritional management, and other horticultural
considerations. Two sites in the Sacramento Valley were mapped each with EM38 and VERIS methods. Site-specific management
zones were identified by each provider on ECa maps for each site, and then soil samples were collected by University of California
researchers to verify these zones. Results showed that on non-saline soils, ECa measured with both EM38 and VERIS correlate
with physical soil properties such as gravel, sand, silt, and clay content but the relationship between conductivity and these
physical soil properties varied from moderately strong to weak. The strength of the correlation may be affected by several
factors including how dominant soil texture is on conductivity relative to other soil properties and on methods of equipment
operation, data analysis and interpretation. Overall, the commercial providers of ECa surveys in this study delivered reasonable
levels of accuracy that were consistent with results reported in previous studies. At one site, an ECa map developed with
VERIS provided more detail on physical soil variability to supplement published soil surveys and aided in the planning and
development of a walnut orchard. At a second site, almond yield appeared to correlate well with distinctly different soil

Microsatellites or Simple Sequence Repeats(SSRs) are informative molecular genetic markers in many crop species. SSRs are
PCR-based, highly polymorphic, abundant, widely distributed throughout the genome and inherited in a co-dominant manner in
most cases. Here we describe the presence of SSRs in cDNAs of cotton. Thirty one SSR primer pairs of 220 (∼14%) tested led
to PCR amplification of discrete fragments using cotton leaf cDNA as template. Sequence analysis showed 25% of 24randomly
selected cDNA clones amplified with different SSR primer pairs contained repeat motifs. We further showed that sequences from
the SSR-containing cDNAs were conserved across G. barbadense and G. hirsutum, revealing the importance of the SSR markers for comparative mapping of transcribed genes. Data mining for plant SSR-ESTs
from the publicly available databases identified SSRs motifs in many plant species,including cotton, in a range of 1.1 to4.8%
of the submitted ESTs for a given species.

The objective of this study was to establish a withdrawal period for flunixin in milk by quantifying 5-hydroxyflunixin, the marker residue, in bovine milk as a function of time, following intravenous treatment of lactating dairy cows with flunixin-N-methyl glucamine (Banamine or Finadyne). Lactating dairy cows were dosed on three consecutive days at 2.2 mg of flunixin free acid/kg of body weight/day. Milk was collected twice daily and assayed using a liquid chromatography-mass spectrometry/mass spectrometry (LC-MS/MS) procedure. The method was validated at concentrations in the range 0.5-250 ppb. The concentrations for 5-hydroxyflunixin measured 12 h after the last administration of drug ranged from 1.56 to 40.6 ppb for all cows. Milk concentrations for 5-hydroxyflunixin were used to establish withdrawal periods of 36 h using guidelines established by the U.S. Food and Drug Administration/Center for Veterinary Medicine and 24 h using guidelines established by the European Medicinal Evaluation Agency/Committee on Veterinary Medicinal Products. 相似文献

Yessotoxins from a large-scale culture (226 L) of Protoceratium reticulatum strain CAWD129 were harvested by filtration followed by solid-phase extraction. The extract was purified by column chromatography over basic alumina and reverse-phase flash chromatography to afford pure yessotoxin (193 mg). Isolation of yessotoxin was greatly facilitated by selection of a strain which did not produce analogues that interfered with yessotoxin isolation. In addition to yessotoxin, numerous minor yessotoxins were detected by LC-MS in other fractions. From one of these, an early eluting minor analogue with the same molecular weight as yessotoxin and a similar mass spectrometric fragmentation pattern was isolated. This analogue was identified by NMR and mass spectrometry as a novel yessotoxin analogue containing a furan ring in the side chain. This finding reveals biosynthetic flexibility of the yessotoxin pathway in P. reticulatum and confirms earlier findings of production of many minor yessotoxin analogues by this alga. Production of these analogues appeared to be a constitutive trait of P. reticulatum CAWD129. 相似文献
