Twelve Standardbred mares underwent blood sampling for 24 h to test the hypothesis that there is diurnal variation of humoral mediators of peripheral energy balance including active ghrelin, adiponectin, leptin, glucose, insulin, and cortisol. The experiment was conducted under acclimated conditions. Grass hay and pelleted grain were provided at 0730 and 1530. Plasma concentrations of active ghrelin and leptin concentrations both peaked (47.3 +/- 6.5 pg/ mL and 5.9 +/- 1.1 ng/mL, respectively; P < 0.05) at 1550, 20 min after feeding. Active ghrelin decreased (P < 0.05) to 28.9 +/- 4.5 pg/mL overnight. The nadir of leptin (4.6 +/- 0.9 ng/mL) occurred at 0650. Neither hormone showed variation (P > 0.05) after the morning feeding. Plasma glucose and insulin concentrations increased (P < 0.05) in response to feeding; however, the morning responses (glucose = 96.9 +/- 2.6 mg/dL; insulin = 40.6 +/- 7.3 uIU/mL) were greater (P < 0.05) than the afternoon responses (glucose = 89.9 +/- 1.8 mg/dL; insulin = 23.2 +/- 4.3 uIU/mL at 180 and 60 min after feeding, respectively). Cortisol concentrations increased (P < 0.05) during the morning hours, but did not respond to feeding, whereas adiponectin concentrations remained stable throughout the study. Hence, active ghrelin and leptin may be entrained to meal feeding in horses, whereas adiponectin seems unaffected. We concluded that there seems to be a diurnal variation in glucose and insulin response to a meal in horses. Furthermore, elevated glucose and insulin concentrations resulting from the morning feeding may be responsible for the increase in leptin concentration in the afternoon.  相似文献

This study examined the influence of sex steroid hormones on lipid metabolism in horses. The group of 34 clinically healthy Standardbred trotters aged 2 to 4 years was studied during an exercise test. The horses were divided into groups according to their sex. These groups were: 11 stallions, 16 mares, and seven geldings. Concentrations of testosterone, 17-β-estradiol, leptin, ghrelin, glycerol, free fatty acids (FFA), and triacylglycerols (TG) were measured in plasma obtained from blood samples taken at rest and after the end of the exercise. At rest, plasma ghrelin concentration was significantly higher in geldings than in stallions and mares (1,541 ± 206 vs 1,280 ± 288 and 1,310 ± 267 pg/mL, respectively; P = .012). Leptin was lower in geldings than in mares (2.65 ± 0.93 vs 4.70 ± 2.31 ng/mL; P = .036). The post-exercise rise in plasma ghrelin and TG concentrations was significantly higher in mares than in geldings (+220 ± 330 vs -25 ± 206 pg/mL; P = .049 and 0.31 ± 0.14 vs 0.13 ± 0.15 mmol/L; P = .016, respectively). The increase in plasma FFA level was higher in geldings than in stallions (535 ± 178 vs 334 ± 191 μmol/L, P = .046). In conclusion, lipolysis rate in geldings is higher than in noncastrated trotters.  相似文献

The epitheliochorial nature of the porcine placenta prevents the transfer of maternal immunity. Therefore, ingestion of the colostrum immediately after birth is crucial for neonatal piglets to acquire passive immunity from the sow. We performed a shotgun proteomic analysis of porcine milk to reveal in detail the protein composition of porcine milk. On the basis of the Swiss‐Prot database, 113 and 118 proteins were identified in the porcine colostrum and mature milk, respectively, and 50 of these proteins were common to both samples. Some immune‐related proteins, including interleukin‐18 (IL‐18), were unique to the colostrum. The IL‐18 concentration in the colostrum and mature milk of four sows was measured to validate the proteomic analysis, and IL‐18 was only detected in the colostrum (191.0 ± 53.9 pg/mL) and not in mature milk. In addition, some proteins involved in primary defense, such as azurocidin, which has never been detected in any other mammal's milk, were also identified in the colostrum.  相似文献

The aim of this study was to investigate the effects of methylprednisolone treatment on serum leptin and ghrelin levels in healthy dogs (n=40). After 14 h of fasting, the dogs were injected intramuscularly with saline (control group) or methylprednisolone (1, 5 or 10mg/kg). Blood samples were collected prior to (baseline) and 2, 3, 4, 8, 12 and 24h subsequent to the treatments. Serum leptin and ghrelin were measured by radioimmunoassay. The mean baseline serum leptin and ghrelin were 2.5+/-0.1 ng/mL (n=40) and 35.0+/-2.1 pg/mL (n=40), respectively. In the control dogs, serum leptin, but not ghrelin levels showed a significant fluctuation during the 24h observation period. Serum leptin increased significantly (p<0.05-0.01) between 2 and 12h after 1mg/kg of methylprednisolone. Serum leptin levels showed biphasic response to 5mg/kg of methylprednisolone: its level decreased to 1.9+/-0.1 ng/mL (p<0.01) at 2h and increased at 12h (2.6+/-0.1 ng/mL) (p<0.01). In response to 10mg/kg of methylprednisolone, serum leptin levels decreased significantly (p<0.01) for 24h. Serum ghrelin levels decreased to 19+/-5 pg/mL at 2-3h (p<0.01) or increased to 87+/-18 pg/mL at 3-8h (p<0.05-0.01) after 1mg/kg of methylprednisolone or 10mg/kg of methylprednisolone, respectively. Serum ghrelin levels did not change at any time point during 24h observation period after 5mg/kg of methylprednisolone. There was a significant (p<0.001) inverse correlation (r=-0.635) between serum leptin and ghrelin levels. In conclusion, we found that methylprednisolone increases or decreases serum leptin and ghrelin levels depending upon its dose and there is a negative correlation between serum leptin and ghrelin levels after methylprednisolone administration.  相似文献

Ghrelin, the endogenous ligand of the growth hormone (GH) secretagogue receptor, is considered a pleiotropic regulator involved in a large array of functions, including control of energy balance, regulation of food intake and, more recently, modulation of the reproductive axis. The present study was aimed at determining the changes in plasma concentrations of acyl-ghrelin in pregnant and lactating sows, with special emphasis on the relationship with the levels of GH, leptin, non-esterified fatty acids (NEFA) and insulin-like growth factor (IGF-1). Blood samples were collected via jugular venipuncture from 22 multiparous sow 30, 60 and 90 days after artificial insemination, 7 and 21 days after farrowing and at first oestrus post-weaning. Plasma concentrations of acyl-ghrelin, leptin, GH and IGF-1 were quantified by validated radioimmunoassay; NEFA were determined using a colorimetric procedure. Plasma acyl ghrelin levels were highest at 30 days of pregnancy and decreased thereafter and during lactation. At the beginning of lactation, GH, IGF-1 and NEFA concentrations significantly increased, while a significant reduction occurred in leptin. In conclusion, ghrelin concentrations in sow maternal circulation does not seem to play an important role in maintaining circulating GH levels during lactation; moreover, ghrelin is not associated with leptin, NEFA and IGF-1 levels.  相似文献

The effect of interval vs. continuous exercise on plasma leptin and ghrelin concentration in young Standardbred horses was studied. The experiment was conducted on 27 trotters, in the age between 2 and 3 years. They were divided into two groups according to the type of exercise. Blood samples were collected through jugular venipuncture in the following experimental conditions: at rest, immediately after exercise and 30 minutes after the end of the effort. Plasma leptin and ghrelin concentrations were determined using RIA tests. The continuous exercise induced an increase in plasma leptin concentration whereas the interval type of exercise did not influence the level of this hormone (3.47 +/- 0.78 vs. 4.07 +/- 0.94 and 2.31 +/- 0.15 vs. 2.36 +/- 0.21 ng/mL, respectively). The plasma ghrelin concentration measured after the continuous exercise, significantly increased (720 +/- 27.4 vs. 814 +/- 13.8; p < or = 0.05) whereas concentration of this hormone assessed after the interval exercise, significantly dropped (982 +/- 56.5 vs. 842 +/- 35.6 pg/mL; p < or = 0.05). The changes in plasma ghrelin concentration measured after the end of the effort correlated inversely with blood lactic acid concentration. In conclusion, the obtained results showed that medium-intensive type of exercise, such as trot, interval or continuous, slightly affected plasma leptin level but significantly affected plasma ghrelin concentration in young Standardbred trotters.  相似文献

Endogenous relationship to restraint and human handling were studied with growing–fattening steers. Thirty‐five crossbred (Japanese Black × Holstein) steers aged 6–10 months were randomly assigned to three pens. They had free access to an Italian ryegrass hay and a restricted amount of high‐concentrate diets (total digestible nutrients (TDN) 70.5%, digestible crude protein (DCP) 10.0%) for the first 6 months of trial. Then they had free access to an oat hay and another vitamin A‐restricted diet (TDN 72.0%, DCP 10.0%) until slaughter. The steers were individually driven into a restraint stall, and bodyweight was recorded. Blood samples were then collected under haltered conditions. These serial handling procedures started at 2 h after the morning feeding were conducted in months 1, 3, 5 (growing stage, GS) and in months 7, 9, 11 (fattening stage, FS) of the trial. Mean peripheral blood concentrations of epinephrine (A; GS, 117.4 ± 76.4 pg/mL; FS, 64.1 ± 34.2 pg/mL), norepinephrine (NA; GS, 257.7 ± 95.0 pg/mL; FS, 125.9 ± 44.1 pg/mL), cortisol (GS, 1.6 ± 0.8 µg/dL; FS, 1.2 ± 0.4 µg/dL), glucose (GS, 83.1 ± 7.5 mg/dL; FS, 71.9 ± 6.9 mg/dL), non‐esterified fatty acid (NEFA; GS, 0.13 ± 0.06 mEQ/L; FS, 0.10 ± 0.06 mEQ/L) and vitamin A (GS, 90.5 ± 24.6 IU/dL; FS, 37.2 ± 21.3 IU/dL) were higher (all P < 0.01) in the GS than in the FS, whereas those of insulin (GS, 1.06 ± 0.82 µU/mL; FS, 1.36 ± 0.61 µU/mL) and leptin (GS, 4.5 ± 1.8 ng/mL human equivalent (HE); FS, 6.8 ± 2.4 ng/mL HE) were lower (both P < 0.01). The metabolite that correlated with A and NA was glucose (A: r = 0.61, P < 0.001; NA: r = 0.53, P < 0.01) in the GS, and the metabolites correlating with A, NA and cortisol were NEFA (A: r = 0.31, P < 0.10; NA: r = 0.32, P < 0.10; cortisol: r = 0.41, P < 0.05) and triglyceride (A: r = ?0.37, P < 0.05; NA: r = ?0.39, P < 0.05) in the FS. Vitamin A was a mediator between A (r = ?0.38, P < 0.05) and NA (r = ?0.42, P < 0.05) and insulin (r = 0.31, P < 0.10) in the GS, and between NA (r = ?0.33, P < 0.10) and leptin (r = 0.38, P < 0.05) in the FS. In conclusion, when changing from the growing to the fattening stages, the stress of handling and restraint had caused the pathways to shift from carbohydrate metabolism to lipid metabolism. In addition, vitamin A seemed to be an important mediator in the endogenous pathways in both stages.  相似文献

The objective of this study was to investigate the relationship between marbling and serum leptin concentrations. First, we compared serum leptin concentrations between Japanese Black cattle and Holstein calves, and found that the mean serum concentration was significantly higher (P < 0.05) in Japanese Black cattle (2.67 ± 0.41 ng/mL, n = 13) than in Holstein calves (1.79 ± 0.43 ng/mL, n = 5). We then examined the effects of roughage intake or vitamin A on the serum leptin concentrations in Japanese Black cattle. Although the leptin concentrations for the high roughage group were significantly higher than those for the low roughage group at 4, 7 and 20 months old, there was no significant difference in the carcass characteristics of the two groups. In addition, although there was a significant difference in the marbling score between the high vitamin A group and low vitamin A group, there was no significant difference in serum leptin concentrations between the groups. These results suggest that although serum leptin concentrations were not significantly correlated to the marbling scores they may be related to the genetic advantage for marbling in Japanese Black cattle.  相似文献

Reproductive hormones in serum concentrations of progesterone, estradiol, and testosterone in female Indo-Pacific bottlenose dolphins (Tursiops aduncus, n = 12) housed in Ocean Park Hong Kong were investigated in the present study. Results showed that, onset of puberty of captive Indo-Pacific bottlenose dolphins was at 5 years while sexual maturity was at 6. Average serum progesterone concentrations in non-pregnant sexually mature individuals was 0.33 (0.25–0.97) ng/mL (interquartile), significantly higher than in immature ones 0.26 (0.25–0.38) ng/mL. This study found significant difference in serum estradiol concentrations between individuals at the onset of puberty (9.5 ± 1.7 pg/mL, ±SD) and not (below detection limit 9 pg/mL). A slightly seasonal breeding pattern, with progesterone values tend to be higher from February to October (0.38 [0.25–1.07] ng/mL) was inferred. During pregnancy, serum progesterone concentrations range from 10.54 ± 8.74 ng/mL (indexed month post-conception [IMPC] 0) to 25.49 ± 12.06 ng/mL (IMPC 2), and display a bimodal pattern with 2 peaks in early- (25.49 ± 12.06 ng/mL, IMPC 2) and late-pregnancy (21.71 ± 10.25 ng/mL, IMPC 12), respectively. Serum estradiol concentrations can seldom be detected in early-pregnancy and increase constantly in mid- (9.45 ± 1.83 pg/mL) and late-pregnancy (11.88 ± 3.81 pg/mL), with a spike (15.45 ± 6.78 pg/mL) 1 month prior to delivery. Serum testosterone concentrations elevate significantly in IMPC 7 (0.36 ± 0.10 ng/mL) compared to other months (0.16 ± 0.10 ng/mL) of the year. The present study provides normal concentration profiles for some reproductive hormones in female Indo-Pacific bottlenose dolphins and can contribute to the breeding monitoring of this species. Also, our study would shed further light on the reproductive physiology of small cetaceans.  相似文献

The plasma pharmacokinetics and mammary excretion of eprinomectin were determined in dairy yaks following topical administration at a dose of 0.5 mg/kg. The kinetics of plasma and milk concentrations were analyzed using a noncompartmental model. Plasma and milk concentrations of eprinomectin increased to reach maximal concentrations of 5.45 ± 2.84 and 2.29 ± 0.90 ng/mL at a Tmax of 1.79 ± 0.57 and 2.00 ± 0.82 days, respectively. The concentration of eprinomectin in plasma was remained >0.5 ng/mL for more than 30 days after administration. The mean residence times of eprinomectin in plasma and milk were 14.73 ± 6.22 and 9.37 ± 2.81 days, respectively. The AUC value in plasma (55.89 ± 18.16 ng day/mL) was threefold greater than that in milk (18.02 ± 6.48 ng day/mL). The AUC milk/plasma ratio was 0.33 ± 0.08. The systemic availability of eprinomectin in yaks was lower than that observed value in other domestic bovines. The low level of eprinomectin excretion in milk suggests that eprinomectin can be used in yaks with zero milk‐withdrawal time. The efficacy of eprinomectin against naturally acquired larvae of Hypoderma spp. was also determined in yaks. Topically administrated eprinomectin at a dose of 0.5 mg/kg was 100% efficacious against larvae of Hypoderma bovis, H. lineatum, and H. sinense.  相似文献

An experiment was conducted with the objective of measuring the concentrations of total milk solids (TMS), CP, and 5'monophosphate nucleotides in sow colostrum and milk. Twelve multiparous sows (Landrace x Yorkshire x Duroc) were used. Litter size was standardized at 11 piglets for all sows at farrowing. Sows were fed an 18.45% CP corn-soybean meal-based diet throughout lactation. The experimental period was the initial 28 d of lactation, with colostrum collected within 12 h of farrowing and milk collected on d 3, 7, 14, 21, and 28. Colostrum and milk samples were analyzed for TMS, CP, adenosine 5'monophosphate (5'AMP), cytidine 5'monophosphate (5'CMP), guanosine 5'monophosphate (5'GMP), inosine 5'monophosphate (5'IMP), and uridine 5'monophosphate (5'UMP). Total milk solids decreased (P < 0.05) from 26.7% on d 0 to 23.1% on d 3. The TMS further decreased (P < 0.05) to 19.3% on d 7, but remained relatively constant thereafter at 18.2, 18.8, and 19.2% on d 14, 21, and 28, respectively. The concentration of CP decreased from 16.6% in colostrum to 7.7, 6.2, 5.5, 5.7, and 6.3% in milk collected on d 3, 7, 14, 21, and 28, respectively (linear and quadratic effect; P < 0.05). Concentrations of 5'AMP, 5'CMP, 5'GMP, and 5'IMP increased from d 0 to d 3 and d 7, and then decreased during the remaining lactation period (quadratic effect; P < 0.05). The concentration of 5'UMP decreased from d 0 to 28 of lactation (linear and quadratic effects; P < 0.05). In colostrum, 5'UMP represented 98% of all 5'monophosphate nucleotides, and in milk, 5'UMP accounted for 86 to 90% of all nucleotides, regardless of day of lactation. The results of this experiment suggest that the concentrations of TMS and CP in sow mammary secretions changed during the first week of lactation, but were constant thereafter. Likewise, the concentrations of 5'monophosphate nucleotides changed during the initial week postpartum, but during the last 2 wk of a 4-wk lactation period, the concentrations were constant.  相似文献

To determine the effects of standardized ileal digestible (SID) valine : lysine ratio on the performance, milk composition and plasma indices of lactating sows, 32 Large White × Landrace sows (219.78 ± 7.15 kg body weight; parity 1.82 ± 0.62) were allotted to one of four dietary treatments with eight sows per treatment based on parity, back fat thickness and body weight. The sows were fed corn‐soybean meal‐based diets containing 63, 83, 103 or 123% SID valine : lysine from day 107 of gestation until day 28 of lactation. The average daily feed intake of sows and daily weight gain of piglets increased linearly (P < 0.05) while back fat loss decreased linearly (P < 0.05) as the SID valine : lysine ratio increased. All of the analyzed amino acids in sow colostrum and valine concentrations of sow and piglet plasma increased linearly (P < 0.05) with the increasing SID valine : lysine ratio. In conclusion, 88 and 113% dietary SID valine : lysine ratios were optimal to achieve minimum back fat loss and maximum piglet growth rate using a linear‐break point model which exceeds the requirement of 85% that is estimated by the National Research Council (2012).  相似文献

Adequate Se transfer from ewes to lambs is important to prevent Se-deficiency diseases. To evaluate how different chemical forms of Se administered at comparative dosages to mature ewes affect Se status of their lambs, 240 ewes were divided into 8 treatment groups (n = 30 each) and drenched weekly (at an amount equal to their summed daily intake) with no-Se (controls); at recommended amounts (4.9 mg of Se/wk) with inorganic Na-selenite, inorganic Na-selenate, or organic Se-yeast; or at supranutritional amounts (14.7 and 24.5 mg of Se/wk) with Na-selenite or Se-yeast for 1 yr. Weekly drenching of Se was effective at increasing (P < 0.002) Se concentrations in ewe colostrum and milk at 30 d of lactation and in improving (P < 0.001) the Se status of lambs (whole-blood and serum-Se concentrations at birth, and skeletal-muscle Se concentrations at 14 d of age). Selenium concentrations in lacteal secretions were greater in ewes drenched with Se-yeast (colostrum: 374, 436, and 982 ng/mL at 4.9, 14.7, and 24.5 mg of Se/wk, respectively; milk: 26, 39, 64 ng/mL) compared with ewes drenched with Na-selenite (colostrum: 204, 334, 428 ng/mL; milk: 16, 21, 24 ng/mL), and were also greater (P < 0.001) in their lambs. Selenium concentrations continued to increase (P < 0.001) in lamb whole blood (558 and 695 ng/mL at 14.7 and 24.5 mg of Se/wk, respectively), serum (126, 183 ng/mL), and skeletal muscle (991, 1,696 ng/mL) with supranutritional concentrations of Se-yeast, whereas Se concentrations did not differ in whole blood (304, 332 ng/mL), serum (77, 85 ng/mL), or skeletal muscle (442, 482 ng/mg) of lambs from ewes drenched with 14.7 or 24.5 mg of Se/wk of Na-selenite. We conclude that weekly oral drenching of ewes during gestation and lactation with organic Se-yeast results in a more efficient transfer of Se (over a wide range of supplementation rates) from ewe to lamb than does inorganic Na-selenite.  相似文献

The aim of this study was to investigate the effects of dietary supplementation of sows with alpha-tocopherol acetate (ATA) and vitamin C on deposition of alpha-tocopherol (AT) in piglet lymphoid organs, such as bone marrow, thymus, and spleen at birth and at weaning, as well as on indicators of immune response in piglets. Sows were given the following treatment diets: control, vitamin C 10 g/day, ATA 500 mg/kg feed, and combined vitamins (ATA 500+Vit-C 10). Supplementation with vitamins started at the beginning of pregnancy and lasted until weaning at 21+/-3 days of age. AT was determined in colostrum, milk, piglet plasma (cord blood) and tissues at birth and on day 21. Immunoglobulins were measured in piglet plasma, milk, and colostrum. Lymphocyte proliferation in response to PHA and ConA was determined in sow and piglet blood. ATA supplementation resulted in a significant increase (P<0.001) in the AT content of colostrum, milk, piglet plasma, liver, thymus, bone marrow, and spleen at weaning. The AT content of colostrum and milk significantly (P<0.001) influenced the AT content of piglet plasma and tissues at weaning (day 21). Total Ig and IgG concentrations in piglet plasma were significantly increased in piglets given the combined vitamin treatment. No effect of AT supplementation was observed on IgG and IgA in colostrum and milk. In sows, vitamin C given alone significantly increased lymphocyte response to ConA and PHA; whereas, in piglets, there was no significant effect of treatments on lymphocyte response to PHA and ConA.  相似文献

In sows, a strong relationship exists between body condition and reproductive efficiency and milk yield. Leptin may act as a metabolic gate which permits the activation of reproductive axis: in the sow, serum concentration of leptin was positively correlated with adiposity at farrowing. An interesting aspect useful to clarify the biology of leptin, was the discovery that the placenta expresses the ob gene, the ob receptor gene and it is a site of leptin production, suggesting a possible role of the hormone in fetal growth; after birth, the placenta functions were taken over from milk, especially to the delivery of maternal hormones and growth factors to the neonate. The exact role of maternal leptin in the physiology of neonatal piglets remains to be determined. Our aim was to evaluate if maternal leptin levels at the beginning of lactation and at weaning could predict the resumption of cycle activity and/or the piglet gain. Thirty-eight Large White × Landrace pregnant sows (16 nulliparous and 22 pluriparous) were used. Blood samples were taken from sows and piglets at d 5 and d 21 after farrowing; in the same days, milk samples were taken after oxytocin injection by means of complete manual milking of all mammary glands of one side. On the basis of the blood leptin at d 5, sows were divided into 3 groups (Low: < 2.3 ng/ml; Medium: 2.3 to 2.6 ng/ml; High: > 2.6 ng/ml). Our results show a correlation at d 5 between backfat thickness and blood leptin (r = 0.342; P < 0.05). The resumption of the cyclic activity was faster in sows with a leptin level at d 5 greater than 2.3 ng/ml (P < 0.01). Milk composition at d 5 and 21 was not affected by parity and leptin. Piglet ADG was significantly (P < 0.05) influenced by sow leptin groups (0.180 kg day− 1 for piglets from Low group and 0.224 for High group). Piglets weaned by High group sows have shown a greater blood leptin content at weaning (P < 0.01) than other groups. In conclusion we have found a significant correlation between leptin and productive and reproductive performances of pigs. This paper underlines the pleiotropic actions exerted by leptin in the productive sow.  相似文献

Abstract

Serum concentrations of progesterone (P) and estradiol (E2) were measured over a 7-month period in two captive juvenile female manatees Trichechus manatus. The animals, aged 5.17 and 6.58 years, had mean (±95% confidence interval) serum P concentrations of 0.45 ± 0.27 ng/mL and 0.67 ± 0.24 ng/mL, and mean serum E2 concentrations of 91.1 ± 71.9 pg/ mL and 121.6 ± 50.6 pg/mL, respectively. A mature male was sampled twice: serum P was 0.92 and 0.79 ng/ mL, and serum E2 was 117.6 and 207.8 pg/mL. A gravid female was sampled once during her first trimester: serum P was 3.42 ng/mL, and E2 was 27.7 pg/mL.  相似文献

The concentrations of vitamin E, glutathione peroxidase (GSH-Px) and retinol were analysed in blood plasma of six primiparous sows during gestation and lactation and in their suckling piglets, and the concentrations of vitamin E, selenium and retinol were analysed in colostrum and in sows' milk. During late gestation the concentrations of vitamin E and retinol in blood plasma increased slowly while no changes were found for GSH-Px. This was followed by a large decline on the day of farrowing, when the lowest mean values of 1.07 mg, 0.15 mg and 2879 U per litre (l) of blood plasma, respectively, were recorded. During lactation the concentrations of vitamin E, GSH-Px and retinol in blood plasma of the sows increased to levels observed during late gestation. The highest mean concentrations of vitamin E (17.9 mg kg- 1 ), selenium (200 w g l- 1 ) and retinol (2.45 mg kg- 1 ) were found in colostrum, but with a large variation between sows. One week postpartum the concentrations of vitamin E, selenium and retinol in sow milk had decreased, on average by 86, 68 and 71%, respectively, after which only small changes occurred. Mean concentrations of vitamin E (1.77 mg l- 1 ), GSH-Px (682 U l- 1 ) and retinol (0.07 mg l- 1 ) were low in the blood plasma of the piglets at birth, but a large increase was observed on the next sampling occasion at the age of 3 days (vitamin E: 5.31 mg l- 1 ; GSH-Px: 996 U l- 1 ; retinol: 0.19 mg l- 1 ). The mean concentration of vitamin E in blood plasma of the piglets declined thereafter until the age of 6 weeks (1.04 mg l- 1 ), when there was a small increase, while the mean concentrations of GSH-Px and retinol continued to increase during the suckling period. A high correlation was found between the concentration of vitamin E in blood plasma and sidefat thickness of the sows (r = 0.94), and between the concentration of vitamin E in colostrum and vitamin E in blood plasma of the sows on the day of farrowing (r =0.77). High correlations were also found between the concentration of vitamin E in the blood plasma of the piglets at 3 days and 5 weeks of age and the concentration of vitamin E in colostrum (r = 0.78) and (r =0.89), respectively. The results indicate that vitamin E storage in the adipose tissues of the sow has a great influence on the supply of vitamin E via colostrum and milk to the piglet.  相似文献

Therap. 33 , 480–484. The purpose of this study was to evaluate the pharmacokinetics of ketamine in mature Holstein cows following administration of a single intravenous (i.v.) dose. Plasma and milk concentrations were determined using a high‐performance liquid chromatography assay. Pharmacokinetic parameters were estimated using a noncompartmental method. Following i.v. administration, plasma Tmax was 0.083 h and plasma Cmax was 18 135 ± 22 720 ng/mL. Plasma AUC was 4484 ± 1,398 ng·h/mL. Plasma t½β was 1.80 ± 0.50 h and mean residence time was 0.794 ± 0.318 h with total body clearance of 1.29 ± 0.70 L/h/kg. The mean plasma steady‐state volume of distribution was calculated as 0.990 ± 0.530 L/kg and volume of distribution based on area was calculated as 3.23 ± 1.51 L/kg. The last measurable time for ketamine detection in plasma was 8.0 h with a mean concentration of 24.9 ± 11.8 ng/mL. Milk Tmax was detected at 0.67 ± 0.26 h with Cmax of 2495 ± 904 ng/mL. Milk AUC till the last time was 6593 ± 2617 ng·h/mL with mean AUC milk to AUC plasma ratio of 1.99 ± 2.15. The last measurable time that ketamine was detected in milk was 44 ± 10.0 h with a mean concentration of 16.0 ± 9.0 ng/mL.  相似文献

The objective of this experiment was to investigate the effect of fat supplementation during the transition period on pre and postpartum body weight (BW), body condition score (BCS), non‐esterified fatty acids (NEFA), glucose and leptin concentrations in Holstein cows. Holstein cows (n = 15) received a low fat diet (LF; 1.61 Mcal net energy for lactation (NEL)/kg of dry matter [DM]), moderate fat diet (MF; 1.68 Mcal NEL/kg DM) or a high fat diet (HF; 1.74 Mcal NEL/kg DM) for 4 weeks prior to calving. All cows were fed similar lactation diets ad libitum (1.74 Mcal NEL/kg DM) for 30 days after calving. Increasing diet energy density during transition period had no effect on prepartum DMI, BCS, BW, glucose and NEFA concentrations (P > 0.05); but leptin concentrations and energy balance (EB) were affected by treatments (P < 0.05). Animals fed HF had less plasma leptin prepartum. After parturition, BW, milk production, milk fat, protein, urea nitrogen and plasma glucose concentrations were affected by prepartum diets (P < 0.05). Fat supplementation prepartum did not affect postpartum NEFA. In conclusion, prepartum fat supplementation decreased leptin concentration prepartum.  相似文献

The sow at parturition is challenged with respect to energy status due to increases in energetic expenses associated with 1) nest building 2) uterine contractions, and 3) colostrum production. A previous study indicated that sows were depleted of glucogenic energy around farrowing. The aim was to investigate whether intravenous infusion of glucose from observed nest-building behavior to 24 h postpartum affected the farrowing kinetics and colostrum production in sows. Ten multiparous sows (DanBred landrace × DanBred Yorkshire) were fitted with a jugular vein catheter on each side (one for infusion and the other one for blood sampling). Sows were infused with either 0.9% saline (CON; n = 5) or 10% glucose (GLU; n = 5) solution at a constant rate of 125 mL/h. From day 108 of gestation, sows were fed once daily with 3.33 kg of a standard lactation diet. During farrowing, sows were monitored to register the onset of farrowing, time of birth, birth status (live or dead), sex, stillbirth rate (SR), and weight of newborn piglets. Farrowing assistance (FA) was provided when the birth interval exceeded 60 min. In late gestation, 1 mL of blood was collected every third hour for blood gas analysis and every sixth hour for harvesting plasma. During farrowing, 1 mL (for blood gas) and 9 mL of blood were collected at 0, 3, 6, 9, 12, 15, 18, 21, and 24 h in milk (HIM). Colostrum and milk samples were collected at 0, 6, 12, 18, 24, and 36 HIM and also at 3, 10, 17, and 24 d in milk. Compared with CON sows, GLU infusion decreased the SR (16.1% vs. 7.4%; P = 0.03), FA (21% vs. 9.0%; P = 0.01), and surprisingly also blood glucose at the onset of farrowing (5.53 vs. 5.09 mmol/L; P = 0.03), respectively. A tendency to higher plasma lactate at the onset of farrowing (P = 0.05) but decreased piglet survival from 0 to 24 h (P = 0.06) was also found for GLU sows. No effects of treatment on farrowing duration or mean birth intervals were found. Lactate in whole blood (P = 0.003) and plasma (P = 0.002) was increased for GLU sows as compared with CON sows during the colostrum period. No effect of GLU infusion was seen on colostrum and milk composition and yield. The increase in lactate was most likely due to a shift toward a greater proportion of glucose oxidation and insufficient O2 supply to fuel uterine contractions. In conclusion, infusion of glucose reduced the frequency of SR and FA, and improved energy status of the sow which seems to be crucial to enhance total piglet survival.  相似文献
