The objective of the study was to investigate different aspects on the efficacy of three anthelmintics on cyathostomin nematodes of Swedish horses. A faecal egg count reduction (FECR) test was performed on 26 farms. Horses were treated orally with recommended doses of ivermectin, pyrantel pamoate or fenbendazole. Faecal samples were collected on the day of deworming and 7, 14 and 21 days later. No resistance was shown against ivermectin; the FECR was constantly >99%. The effect of pyrantel was assessed as equivocal in 6 farms 14 days after treatment; the mean FECR was 99%. As many as 72% of the fenbendazole-treated groups met the criteria for resistance; the mean FECR was 86%, ranging from 56% to 100%. A re-investigation of two farms where pyrantel resistance had been suspected clearly revealed unsatisfactory efficacy of pyrantel on one of these farms; the FECR varied from 72% to 89%. Twenty-six of the horses previously dosed with pyrantel or fenbendazole, and which still excreted ≥150 eggs per gram of faeces 14 days after treatment, were dewormed with ivermectin and fenbendazole or pyrantel in order to eliminate the remaining cyathostomins. A total of 13 cyathostomin species were identified from horses that initially received fenbendazole and seven species were identified from pyrantel-treated individuals. The egg reappearance period (ERP) following treatment with ivermectin and pyrantel was investigated on two farms. The shortest ERP after ivermectin treatment was 8 weeks and after pyrantel was 5 weeks. We conclude that no substantial reversion to benzimidazole susceptibility had taken place, although these drugs have scarcely been used (<5%) in horses for the last 10 years. Pyrantel-resistant populations of cyathostomins are present on Swedish horse farms, but the overall efficacy of pyrantel is still acceptable.  相似文献

Two strains of Oesophagostomum spp., consisting of both O. quadrispinulatum and O. dentatum, were subjected to a controlled in vivo assay for resistance to levamisole and pyrantel by comparison with susceptible isolates. One strain (LEM) was recently isolated from a commercial herd, where sows showed high numbers of strongyle eggs in faeces within 2 weeks of farrowing and following treatment with levamisole at the manufacturer's recommended dose rate 1 week before farrowing. Levamisole had been used as the sole anthelmintic for treatment for at least 7 years on this farm. Treatment with pyrantel in this herd also indicated cross-resistance to this drug. A mixed population of O. quadrispinulatum and O. dentatum of this strain was subjected to controlled in vivo assays. Faecal egg count reduction (FECR) was found to be -573.3% (P greater than 0.05) and worm count reductions (WCR) of O. quadrispinulatum and O. dentatum were estimated as 44.5% (P greater than 0.05) and 96.4% (P less than 0.001), respectively. Treatment with pyrantel showed that cross-resistance existed to this drug, with FECR of 10.4% (P greater than 0.05) and WCR of 64.5% (P greater than 0.05) and 90.7% (P less than 0.05) for O. quadrispinulatum and O. dentatum, respectively. Another strain (VJ) was isolated from another commercial pig herd, which was dosed with pyrantel citrate four times a year for at least 8 years. This strain showed resistance to pyrantel, with FECR of 43.8% (P greater than 0.05) and WCR of 65.9% (P greater than 0.05) and 49.4% (P greater than 0.05) for O. quadrispinulatum and O. dentatum, respectively. However, both species were susceptible to levamisole. Our results suggested that selection with levamisole gave rise to levamisole resistance and automatically conferred resistance to pyrantel, whereas selection with pyrantel only resulted in resistance to this drug alone. These findings are discussed in relation to the location of the two species of Oesophagostomum in the large intestine of pigs and the mode of action of this class of anthelmintics.  相似文献

The prevalence of drug-resistant ovine parasites in the United States has not been widely reported. Thirteen flocks, typical of commercial sheep production units, were selected for survey. Four anthelmintics (fenbendazole, ivermectin, pyrantel pamoate, and levamisole) were tested for their ability to reduce herd mean pretreatment fecal egg count. If a properly dosed and administered drug failed to reduce herd mean pretreatment fecal egg count by 80%, it was considered ineffective in that flock, and the presence of parasites resistant to that drug was inferred. Fenbendazole administration changed pretreatment fecal egg counts by +9% to -100%. On the basis of the aforementioned definition, drug resistance existed in 6 of 13 flocks. Posttreatment larval culture indicated that Haemonchus contortus survived administration of fenbendazole. Levamisole, pyrantel pamoate, and ivermectin reduced pretreatment fecal egg count by -83% to -100%; resistance to these products was not evident in the flocks surveyed.  相似文献

This study was undertaken to determine whether anthelmintic-resistant Oesophagostomum spp. populations occur in Danish swine herds. A controlled field trial on selected sow herds suggested resistance to pyrantel citrate in a herd where faecal egg count depression in response to treatment was only 28.3%. This was confirmed by subsequent experimental infection of pigs where the suspected resistant Oesophagostomum isolate was compared with a susceptible worm isolate. After treatment with the recommended dose of the drug, worm burdens of the suspected isolate were only reduced by 42.6% (p greater than 0.05) in contrast to the susceptible isolate, which was reduced by 94.6% (p less than 0.01). Differential counts of the resistant isolate suggested that O. dentatum might be more resistant than O. quadrispinulatum. The resistant isolate originated from a herd, where the anthelmintic had been administered four times a year over a period of 7 years. The potential for development of anthelmintic resistance in Oesophagostomum spp. under management practices in our country is discussed in the light of the current views on predisposing factors.  相似文献

The effectiveness of anthelmintics was evaluated in four herds of captive ruminants, wapiti (Cervus elaphus), Armenian red sheep (Ovis orientalis), giraffe (Giraffa camelopardalis), and pronghorn (Antilocapra americana), by the use of fecal egg reduction tests (FERTs) and a commercial larval development assay (LDA) designed to evaluate susceptibility or resistance of nematodes to anthelmintics. Haemonchus sp. was the predominant nematode in the red sheep, giraffe, and pronghorn herds, whereas Ostertagia sp. and Trichostrongylus sp. were predominant in the wapiti. The LDA data indicated susceptibility by the worms to benzimidazoles except in the red sheep flock, which showed a high level of resistance. High levels of resistance to levamisole were seen in the worm populations from the wapiti and red sheep, moderate resistance in the pronghorn herd, and susceptibility in the giraffe herd. Worms were susceptible in all four herds to a combination of benzimidazole/levamisole. There was suspected avermectin resistance by Trichostrongylus sp. in the wapiti herd and by Haemonchus sp. in the giraffe. The FERTs agreed with the LDA in showing the Haemonchus in the giraffe was susceptible to fenbendazole and had suspected resistance to ivermectin, whereas Haemonchus in the red sheep and pronghorn were susceptible to ivermectin. There was correlation between the tests evaluating anthelmintics. The LDA is useful as a screening test in the selection of an anthelmintic for use in grazing ruminants, but the effectiveness of a drug in a host species may depend as much on the dose used, and the method of administration, as it does on the parasite's sensitivity to the anthelmintic.  相似文献

Since 2002, selected populations of Parascaris equorum in several countries have been reported to survive treatment with macrocyclic lactone (M/L) anthelmintics. Clinical treatment failures are characterized by negligible fecal egg count reduction, but M/L resistance has been confirmed in ascarids by controlled efficacy testing. Resistance was selected by current parasite control practices for foals, which often include exclusive and excessively frequent use of M/L dewormers, thereby minimizing refugia within the host and in the environment. Chemical control of M/L-resistant isolates can be accomplished with pyrimidine and/or benzimidazole anthelmintics, but a few M/L-resistant populations have recently exhibited resistance to pyrantel pamoate as well. Some specimens of Oxyuris equi regularly survive treatment with macrocyclic lactones, but it is uncertain whether this constitutes resistance or merely confirms the incomplete oxyuricidal efficacy of virtually all broad spectrum equine anthelmintics. Variations in other biological parameters of Oxyuris and Parascaris, specifically atypical infection of older hosts and shorter prepatent periods, have been reported anecdotally. These changes may represent genetic modifications that have evolved in parallel with resistance as a result of anthelmintic selection pressure.  相似文献

To evaluate the efficacy with which recommendations of means to avoid the spread of anthelmintic resistance in parasitic nematode populations are communicated to farmers on dairy goat farms in France, a questionnaire survey was undertaken on their use of antiparasitic drugs. Information was collected from 73 farms in two main areas of dairy goat production. The data referred to three years. Anthelmintics were used in 69 farms, the mean number of treatments per year being 2.74. Changing the drug from one year to another was not practised. Moreover, of the 58 farms using two or more treatments per year, only 37% used anthelmintics from different classes in the lactation and drying-off periods. Benzimidazoles and probenzimidazoles were given in all except two farms and these substances represented more than 80% of all the treatments. Levamisole/pyrantel or avermectins were used in 15% and 27% of the farms, respectively. Double the ovine dose, as recommended in goats to ensure efficacy of benzimidazoles, was applied in 55% of the farms. In addition, in all the flocks, the substances were given on the basis of a mean estimated live weight and not by reference to the heaviest animal. These results indicate that errors in the use of anthelmintics are still frequent in dairy goat farms in France, with probable consequences for the spread of anthelmintic resistance in the populations of parasites.  相似文献

In the present survey, 276 horses bred on 16 farms located in central and southern Italy were investigated for the presence of drug resistant cyathostomes by a Fecal Egg Count Reduction Test (FECRT). Sixteen to 20 animals were selected on each farm and randomly assigned to one of four equally sized treatment groups. Groups were treated with fenbendazole, pyrantel pamoate, ivermectin or moxidectin. Resistance to fenbendazole was declared on six farms (37.5%) and suspected in two farms (12.5%), with FECR values ranging from 41% to 88.3%. Resistance to pyrantel was found in two farms (12.5%) and was suspected in one case (6.2%), with FECR values ranging from 43% to 85.4%. Macrocyclic lactones remained effective on all farms. Only cyathostome third stage larvae (L3) were found in fecal cultures after treatment. This paper reports the first wide survey conducted in Italy for anthelmintic resistance in equine cyathostomes. The results indicate that multiple drug resistant equine cyathostomes are present in the central and southern regions of Italy. These data call for a geographically and numerically broader investigation of horse farms in all regions and for the development and implementation -among veterinarians, owners and managers of a plan to reduce the expansion of these anthelmintic resistant populations and control these important parasites.  相似文献

The aim of this study was to investigate the suitability of a larval development assay (LDA) for the determination of anthelmintic resistance in cyathostomin nematode populations of the horse. In addition, comparison of results between geographic regions, types of horse establishment, and the use of anthelmintics in Sweden, was established. Seventy horse herds from different parts of Sweden were sampled, and strongyle eggs from the faeces of 54 of those were investigated by an LDA (DrenchRite). The following anthelmintics were tested: thiabendazole (TBZ), levamisole (LEV), ivermectin monosaccharide (IVM-MS), ivermectin aglycone (IVM-AG) and pyrantel (PYR). The LC50 values for TBZ and LEV were generally lower than those previously reported in other LDA studies on horse nematodes. This could be related to the infrequent use of these compounds for the past 20 years in Sweden. In this study, there was a great variation within and between assay plates that could not be explained. Still the LC50 values differed significantly between the regions for all anthelmintics, except for pyrantel. The highest LC50s were observed in parasite populations from the south of Sweden. There were no significant differences between riding schools and studs. Limitations of this technique exist, namely the lack of established cut-off values for susceptible and resistant populations and interpretation problems related to multi-species infections. Although there are advantages with LDA such as the possibility of testing several compounds simultaneously without interference with the deworming programmes on the farms, we conclude that LDA currently is not a reliable alternative to the faecal egg count reduction test (FECRT).  相似文献

In order to assess the resistance situation against macrocyclic lactones in Parascaris equorum and against tetrahydropyrimidine derivatives in strongyles in Finnish trotter horses, 112 foals on 18 farms, mostly 1 year old, were examined for these parasites with a modified McMaster faecal flotation method. P. equorum positive foals (n=24) were given ivermectin orally at a dose of 200 μg/kg b.w., while strongyle positive but P. equorum negative foals (n=38) received pyrantel embonate orally at a dose of 19 mg/kg. Sixteen P. equorum infected foals, treated with ivermectin, also harboured strongyles. During the anthelmintic treatment visit to the farm, Faecal Egg Count Reduction Test (FECRT) reference (first) samples were collected. Fourteen days later, the second sampling (reduction samples) was done. The FECR was calculated for each foal/parasite combination. The reduction efficacies of ivermectin against P. equorum (mean 52%, calculated from the individual egg count reductions) and pyrantel against strongyles (43%) were strongly indicative of widespread resistance. Also indication of ivermectin resistance among strongyles was seen. The widespread use of anthelmintics for Finnish horses obviously has resulted in resistance, as has happened elsewhere, too.  相似文献

Thiabendazole was the first of several benzimidazole anthelmintics and was superior to previously available drugs in both-efficiency and safety. Of the “second generation” of benzimidazoles, mebendazole and cambendazole are widely used inhorses and provide wider efficacy spectra and lowerdose rates compared with thiabendazole. Thiabendazole has proven activity against larval strongyles, but requires high dose rates. Two organophosphates, dichlorvos and haloxon have been formulated as horse anthelmintics. Pyrantel and its analogue morantel, are reported as efficient equine anthelmintics, but only in-feed and pasteformulations are recommended by the manufacturer as suitable for horses. Strains of small strongyles tolerant to benzimidazoles have been reported in U.S.A. and U.K. The addition of a piperazine salt to approximately half recommended dose rate, or the use of pyrantel, removed the tolerant nematodes.  相似文献

REASONS FOR PERFORMING STUDY: Possible anthelmintic resistance on a breeding farm where a rapid rotation anthelmintic programme had been implemented for 9 years was investigated. Cyathostomins resistant to fenbendazole and pyrantel were documented by faecal worm egg count reduction test (FWECRT). OBJECTIVES: To 1) manage small strongyle transmission in a herd of horses in which resistance to both pyrantel pamoate and fenbendazole was identified and thereby reduce the risk of clinical disease in the individual animal, 2) monitor the change in resistance patterns over time and 3) monitor the efficacy of ivermectin over the study period. METHODS: Targeted ivermectin treatment of horses on the farm was instituted for mature horses with faecal worm egg counts (FWEC) > 200 eggs/g (epg) and for horses < age 2 years with FWEC > 100 epg. RESULTS: Over a 30 month period, targeted ivermectin treatment achieved acceptable control in mares, as judged by FWEC, and improved control of patent cyathostome infection in consecutive foal crops. Egg reappearance time (ERT) after treatment with ivermectin was < 8 weeks in mares and foals more frequently in the second year of the study than in the first year. Numbers of anthelmintic treatments were reduced by 77.6 and 533% in the mare and foal group, respectively. CONCLUSIONS: Targeted ivermectin treatment may be an economically viable method of managing multiple drug resistant cyathostominosis. POTENTIAL RELEVANCE: Use of ivermectin should be monitored closely for development of resistance.  相似文献

SUMMARY Following the apparent failure of levamisole to control infections of Haemonchus contortus in sheep at Lawes in south eastern Queensland, a strain of this parasite was isolated at the Animal Research Institute, Yeerongpilly. This strain was used to infect sheep at Yeerongpilly and the Merrindale Research Station, Victoria where four experiments to classify the resistance pattern of the parasite were carried out. Resistance to thiabendazole was first suspected in 1969, and these experiments confirmed that resistance to this drug was still present. They also showed that a strong degree of resistance had been developed to both levamisole and morantel tartrate. Other benzimidazole anthelmintics and also the organophosphorus compound naphthalophos were only moderately effective against the original isolate but rafoxanide, nitroxynil and phenothiazine were almost 100% effective. Other highly effective chemicals were disophenol and closantel. After passaging the strain for four generations with both levamisole and albendazole, resistance to both naphthalophos and the newer benzimidazole anthelmintics increased dramatically. This is the first report of a field strain of H. contortus exhibiting resistance to benzimidazole, non-benzimidazole and organophosphorus anthelmintics.  相似文献

Clinical trials using fecal egg count reduction tests and coproculture were conducted with yearlings and mares on a farm in 1997. Fecal samples were taken from each horse to estimate the number of strongyle eggs/g feces with Cornell-Wisconsin centrifugal flotation and Cornell-McMaster dilution techniques. Eleven of 15 yearlings, which had been on a daily feeding of grain with pyrantel tartrate for 66 d were found with strongyle eggs in feces. This was the first time the in-feed medication had been used on the farm. Nine yearlings were randomised into three groups; continuation of daily pyrantel tartrate or one treatment with pyrantel pamoate or moxidectin. Two of three yearlings given pyrantel tartrate or pamoate had no reduction in the eggs/g feces. These six yearlings were then given moxidectin and in all yearlings the eggs/g feces was reduced to zero. The 66 d of pyrantel tartrate use was an inadequate time for development of resistant cyathostomes and a hypothesis was the resistance was due to extensive use on the farm over many years of pyrantel pamoate at twice the label dose for control of tapeworms. That hypothesis was tested with 12 mares with strongyle eggs in the feces randomised into two treatment groups: pyrantel pamoate at label dose or moxidectin. Five of six mares given pyrantel had <80% reduction in egg/g feces. These mares were then given moxidectin and in all mares the eggs/g feces was reduced to zero. Only cyathostomes were found on culture and apparently there was side resistance among the pyrantel salts.  相似文献

The increase of anthelmintic resistance in the last years in the nematode population of veterinary importance has become a major concern. The objective of the present study was to evaluate the efficacy of the main anthelmintic drugs available in the market against small strongyles of horses in Brazil. A total of 498 horses from 11 horse farms, located in the states of Paraná, São Paulo, Rio de Janeiro and Minas Gerais, in Brazil, were treated with ivermectin, moxidectin, pyrantel and fenbendazole, orally at their recommended doses. The fecal egg count reduction test (FECRT) was used to determine the product's efficacy and fecal culture was used to determine the parasite genus. Reduction on anthelmintic efficacy was found for fenbendazole in all horse farms (11/11), pyrantel in five yards (5/11) and ivermectin had low efficacy in one of the yards studied (1/11). Multidrug resistance of up to 3 drugs classes was found in one of the tested farms (1/11). Cyathostomin were the most prevalent parasite. The results showed that resistance to fenbendazole is widespread; the efficacy of pyrantel is in a critical situation. Although the macrocyclic lactones compounds still showed high efficacy on most farms, suspected resistance to macrocyclic lactones is of great concern.  相似文献

Trials were conducted in ponies to evaluate the efficacy of pyrantel pamoate (Strongid-T®) and two newer anthelmintics not yet commercially available, nitramisole and avermectin B1a, against migrating Strongylus vulgaris larvae. Ponies were removed from their mares within 24-48 hr after birth and reared in isolation, worm free. Between six and 14 weeks of age they were infected with 2000 or 2500 infective S. vulgaris larvae. Subsequently, they were monitored daily for clinical signs until the experiment terminated at 28 days postinfection. All ponies showed increased body temperature and reduced appetite within the first week of infection. All anthelmintics were administered on day 7 and in addition pyrantel pamoate was given on day 8 postinfection. The anthelmintics were in liquid formulation. Nitramisole and pyrantel pamoate were given by stomach tube and avermectin B1a by subcutaneous injection.

Following administration of these compounds toxic reactions were not observed. All anthelmintics caused a reduction in body temperature and increased appetite and effected a clinical cure. In ponies which were not treated with an anthelmintic, temperatures remained elevated and appetites never returned completely to normal. These ponies also showed variable degrees of lethargy, depression, recumbency and colic and the majority died between two and three weeks postinfection. At necropsy, these control ponies showed variable degrees of adhesions involving the abdominal organs, necrosis of the ileum and cecum and severe arteritis and thrombosis of the major abdominal arteries and their branches.

Although pyrantel pamoate, used at eight times the therapeutic dose for intestinal nematodes in the horse, effected a clinical cure it did not produce a radical cure. At necropsy, ponies treated with pyrantel pamoate had arteritis and thrombosis of the cranial mesenteric artery and its major branches. Nitramisole and avermectin B1a were able to effect both a clinical and radical cure.

This paper reports a survey conducted in France during 2011 to evaluate the efficacy of commonly used anthelmintics against horse cyathostomins. A total of 40 farms and 1089 horses were screened for the presence of cyathostomins. All farms but one were positive, with an overall animal infection rate of 53.7%, ranging from 9% to 83% on individual farms. On 445 horses from 30 of these farms, a faecal egg count reduction test (FECRT) was performed to evaluate the efficacy of oral formulations of fenbendazole (FBZ), pyrantel embonate (PYR), ivermectin (IVM) and moxidectin (MOX). Calculation of the mean FECR and 95% confidence intervals (95% CI) around the mean was performed using bootstrap analysis. Resistance to FBZ was found on 17 of 18 farms investigated, with a mean reduction of 57% (95% CI: 38.5-71.2%). Suspected resistance for PYR was found on 6 of 30 farms, and confirmed on another 3 of 30 farms, with a mean reduction for PYR of 94.7% (95% CI: 88.9-98.5%). Reduced efficacy simultaneously of FBZ and PYR was found in 7 farms. Reduced efficacy of IVM was found in one animal on one farm and of MOX in one animal on another farm, and was combined with resistance against FBZ and/or PYR. These results indicate that single and multiple drug resistance and reduced efficacy in equine cyathostomins is present in France. Macrocylic lactones proved to be highly effective compounds against cyathostomins, with reduced efficacy for IVM and MOX in two farms only. These results extend present knowledge on the occurrence of drug resistant cyathostomins in Europe, and illustrate the necessity to use anthelmintics in appropriate worm control programmes.  相似文献

In order to study the prevalence of benzimidazole-resistance in equine cyathostomes, 30 stables in south east England were selected according to strict criteria but with minimum bias to provide three matched groups of 100 horses. One group was treated with fenbendazole, one with pyrantel embonate and the third was left untreated. The overall efficacies of fenbendazole and pyrantel embonate, as judged by the faecal egg-count reduction test, were 56.3 and 95.8 per cent, respectively. The numbers of horses at individual stables were too small to draw conclusions at each stable, but whereas efficacies greater than 85 per cent were recorded for pyrantel at 26 of 27 stables, the corresponding figure for fenbendazole was five of 27. Cyathostomes accounted for more than 90 per cent of the larvae cultured from faeces before and after treatment. Trichostrongylus axei was detected on 20 sites, but Strongylus species were present on only five. In the area of the survey, benzimidazole-resistance was very widespread in stables which used benzimidazole anthelmintics regularly and had more than nine horses per stable, but the benzimidazole-resistant cyathostome strains were adequately controlled with the non-benzimidazole anthelmintic pyrantel embonate.  相似文献

The prevalence of anthelmintic resistance on 27 sheep farms in Slovakia was investigated in 2003 and 2004 using the faecal egg count reduction test (FECRT) according to the WAAVP guidelines. Resistance to albendazole was detected on one farm (3.7%) and suspected on two farms (7.4%) out of 27 sheep flocks. Resistance to ivermectin was tested on 26 farms. On six (23.1%) farms, results indicated the presence of ivermectin resistance. Resistance to ivermectin was suspected on eight farms (30.8%). However, it is also possible that generic ivermectin anthelmintics used in survey have a lower efficacy against sheep nematodes.  相似文献

The present study included 19 stud farms, including 243 horses, that were investigated for the occurrence of anthelmintic resistant cyathostomes. The number of horses on the farms varied from nine to more than 100, and horses of all ages were included. A minimum of seven horses were used for faecal egg count reduction (FECR) tests. The anthelmintics included were: fenbendazole (paste formulation), ivermectin (paste formulation) and pyrantel (powder). Resistance to benzimidazoles was detected on 14 farms, with FECR values ranging from 65.1 to 86.3%. Larval cultures after fenbendazole treatment revealed exclusively cyathostome larvae. Ivermectin was tested on eight farms and proved to be effective on all. Pyrantel was tested on two farms and FECR test indicated high efficacy (92-97%). Egg hatch assay (EHA) results showed that mean concentrations of thiabendazole that inhibited hatching in 50% of the eggs (ED(50)) in resistant populations were over 0.1 microg ml(-1). The results of our study suggest widespread resistance to fenbendazole in equine cyathostomes in Slovakia, and possible strategies to delay anthelmintic resistance are discussed briefly.  相似文献
