确定昆明种小白鼠超排的最佳激素剂量和分析其他影响超排的因素。将40只雌性昆明小白鼠随机分为四组。每组注射PMSG和HCG分别为5IU+5IU、5IU+10IU、10IU+5IU和10IU+10IU,注射间隔时间为48h,比较排出卵母细胞的数量。结果表明,试验3组小鼠排出卵母细胞的平均数量(57.1枚),显著(P<0.05)高于试验1组(42.7枚)和试验2组(41.7枚),极显著(P<0.01)高于试验4组(30.7枚)。其它三组间无显著差异。左右卵巢的排卵数也未表现出明显差异。初步说明昆明小白鼠超排的最佳激素剂量为10IU PMSG+5IU HCG,但要想得到很好的超排效果还要考虑其他因素。  相似文献

采用国产激素,以FSH LH、FSH HCG、PMSG LH、PMSG HCG4种不同的处理方法对32只皖江长毛兔进行了超排处理,旨在筛选确定皖江长毛兔最有效的超排方法及胚胎移植的最佳时期。结果表明:4种超排方法处理后的排卵数分别为42.5枚、46.25枚、30.63枚、33.38枚;回收胚胎数分别为27.63枚、31.25枚、22.37枚、20.75枚;有效胚胎数分别为20.50枚、24.13枚、17.63枚、15.25枚。这三项指标均以FSH HCG组最高。将收集到的237枚胚胎移植给24只受体,每只受体移植胚胎10枚左右。结果表明:2、4、8~16细胞期及桑椹胚移植后的妊娠率、产仔率及窝产活仔数分别为50%、16.6%和3.3只,66.7%、20.6%和3只,60%、24.5%和4.3只,71%、40.9%和5.4只。其中以桑椹胚组指标最高。  相似文献

采用杂交奶水牛及纯种奶水牛作为供体牛,将日本生产的FSH作为外源激素进行超数排卵,以确定胚胎移植供体牛超排的效果;采用CIDR+PMSG+PG+LHRH-A3法,确定受体牛胚胎移植和同期发情的效果。试验结果表明:5头奶水牛供体牛超排,共采卵7头次,采卵数21枚,其中10枚可用胚胎,平均1.4枚/头次,可用胚率为47.6%。受体牛的同期发情效果和可移植率分别为77.0%和62.0%。  相似文献

采用国产FSH和德国产PMSG对58只新西兰白兔进行超数排卵试验.结果表明:①两种激素均有较好的超数排卵效果,FSH组平均排卵点和平均回收胚胎数分别为28.5±18.0个、22.5±13.2枚;PMSG组分别为27.0±11.9个、23.6±9.8枚.经t检验,两者差异不显著(P>0.05),但从超排母兔回收胚胎的分布情况看,PMSG组的超排效果更稳定些.②右侧卵巢排卵数无论是FSH组(53.7%),还是PMSG组(50.5%)均高于左侧卵巢(分别为46.3%和49.5%),FSH组左、右侧卵巢排卵数差异极显著(P<0.01),PMSG组差异不显著(P>0.05).③两种激素在春季对母兔的超排效果都明显地优于秋季(P<0.01).  相似文献

采用PMSG +hCG(方案 1)和FSH +hCG(方案 2 )两种方案对青年和经产的日本大耳白兔进行超排处理。方案1处理的 2 7只兔中 ,16只经产兔共排卵 488枚 ,平均每只 ( 30 .5± 7.47)枚 ,卵子回收率为 71.1%;11只青年兔共排卵2 39枚 ,平均每只 ( 2 1.7± 7.1)枚 ,卵子回收率为 75 .7%。方案 2处理的 2 0只兔中 ,12只经产兔共排卵 5 47枚 ,平均每只( 45 6± 10 .2 )枚 ,卵子回收率为 6 3 .1%;8只青年兔共排卵 2 95枚 ,平均每只 ( 36 .9± 8.4)枚 ,卵子回收率为 6 7.46 %。结果表明 :方案 2超排效果优于方案 1(P <0 .0 1) ,经产兔比青年兔超排效果好 (P <0 .0 5 )。  相似文献

本研究旨在优化肉用黑萨福克绵羊超排方案,筛选生产条件下最佳配种授精方式,提高黑萨福克体内胚胎生产效率,建立高效绵羊胚胎移植体系。供体超数排卵采用不同超排方案、不同FSH剂量、不同配种授精方式、不同侧卵巢超排效果、不同季节以及重复利用6组因素的比较试验。结果表明:采用CIDR+FSH+PMSG递减4d 8针注射法供体平均黄体数16.00个/只、可用胚数14.00枚/只和移植妊娠率71.43%,效果最好(P<0.05);FSH注射剂量范围在200260 IU,超排平均黄体数、平均可用胚胎数分别为14.87、13.28枚/只,高于160260 IU,超排平均黄体数、平均可用胚胎数分别为14.87、13.28枚/只,高于160200 IU组和260200 IU组和260320 IU组(P<0.05);开膣器输精+自然交配+开膣器输精方法供体冲出卵子的受精率97.32%、平均可用胚数11.25枚/只,高于其他2种配种方式(P<0.05);不同侧卵巢超排效果无差别;繁殖季节(秋季)供体平均黄体数13.00个/只、可用胚10.75枚/只,高于非繁殖季节(P<0.05);重复超排效果无明显差异。本研究获得了黑萨福克绵羊超数排卵的技术参数,优化了黑萨福克绵羊超排方法和配种授精方案,建立了高效的胚胎移植体系。  相似文献

对12只母兔分别用进口FSH+hCG(8只)和PMSG+hCG(4只)进行超数排卵处理.结果表明,FSH+hCG组(排卵点72.4±13.6个,获胚胎52.6±14.4枚)比PMSG+hCG组(排卵点32±6个,胚胎21.8±4.2枚)获得了更好的超排效果(P<0.01).将部分胚胎移植给4只同期化处理的母兔,怀孕2只,其中1只妊娠20 d流产,另1只顺利产下8只仔兔.  相似文献

对绒山羊分别进行FSH和PMSG超排效果试验:A组采用减量注射法肌肉注射FSH,每天2次,连续4天,总计量260IU;B组一次性肌注PMSG 1200IU。母羊发情后注射hCG 500IU和LH 200IU,随即配种,间隔8~10h复配一次。配种7天后,手术法从供试羊子宫角取胚。结果表明:使用FSH和PMSG进行绒山羊超数排卵均有较好的效果。两者相比,FSH处理组排卵整齐,且胚胎合格率高,效果优于PMSG。  相似文献

为了探讨孕马血清促性腺激素(PMSG)和人绒毛膜促性腺激素(HCG)对小鼠超数排卵的最佳组合剂量、最佳注射时间间隔,本试验以成熟实验小鼠为试验动物,分别采用不同剂量PMSG(5IU、7.5IU、10IU、12.5IU)与不同剂量HCG(5IU、7.5IU、10IU、12.5IU)组合注射小鼠进行排卵效果研究,并以44h、46h、48h、50h的间隔时间对小鼠进行超数排卵处理试验。结果表明,(10+10)IU剂量组合处理的见栓小鼠平均获卵数最高(10.3枚);在注射间隔为48h试验组中见栓小鼠平均获卵数最高(8.4枚)。提示本试验条件下,小鼠超排中PMSG与HCG组合剂量为10IU、注射时间间隔为48h时超排效果较好。  相似文献

选用6周龄雌小鼠48只,随机分为4组,分别用2.5IU、5.0IU、7.5IU、10.0IU的PMSG注射,48h后分别注射hCG2.5IU、5.0IU、7.5IU、10.0IU:次日早晨检栓,60h后冲胚。总见栓率为81.25%(39/48)。冲胚结果:2.5IU、5.0IU、7.5IU和10.0IU剂量组平均每只获胚分别为9.17(110/12)枚、32.67(392/12)枚、16.00(92/12)枚、11.67(140/12)枚,各组超排效果差异极显著(P〈0.01)。结果表明:采用5.0IU PMSG+5.0IU hCG对小鼠超排效果最好,输卵管分段冲胚法可行性良好。  相似文献

为探讨昆明鼠超数排卵的最佳条件,提高超排效果,比较促性腺激素使用剂量、孕马血清促性腺激素(PMSG)与人绒毛膜促性腺激素(hCG)注射间隔时间、超排小鼠所处发情周期阶段及小鼠周龄等因素对昆明鼠超数排卵结果的影响。PMSG 8IU+hCG 8IU、间隔46h~48h注射组超排效果最好;3周龄、8周龄~10周龄雌鼠较4周龄~7周龄雌鼠超排效果好,小鼠发情间期或发情前期开始超排的效果显著高于发情周期的其他时期。  相似文献

本文以近交系C57BL/6小鼠为实验对象.研究注射不同剂量的PMSG和hCG对小鼠超排效果的影响。取C57BL/6小鼠各30只,按照注射剂量不同分为A、B、C三组,每组10只,A组注射PMSG2.5IU,HCG2.5IU,B组注射PMSG5.0IU,HCG5.0IU,C组注射PMSG7.5IU,HCG7.5IU。每只小鼠腹腔注射PMSG,间隔48h后分别注射HCG进行超数排卵,再与性成熟同系公鼠合笼,次日早上检查阴道栓.有栓雌鼠用颈椎脱臼法处死。在实体显微镜下由输卵管膨大部冲卵.收集卵母细胞置于盛有M2培养液的表面皿中检查计数.分析超排效果。结果表明。C57BU6小鼠B组的平均取卵数极显著高于A组的平均取卵数(P〈0.05);B组的平均取卵数显著高于C组的平均取卵数(P〈0.05);C组与A组的平均取卵数差异不显著(P〉0.05)。  相似文献

Embryo transfer in pigs normally involves surgery. In connection with the development of nonsurgical or endoscopic transfer techniques, it is important to know whether the uterine site to which embryos are transferred has an effect on the success rate. In the present investigation, prepubertal donor gilts were treated with 1,500 IU of PMSG and, 72 h later, with 500 IU of hCG. Gilts were artificially inseminated 24 and 36 h after hCG injection. Embryos at the expanded blastocyst stage were collected from donor gilts. Recipient gilts were treated synchronous with the donors, using 1,000 IU of PMSG followed, 72 h later, with 500 IU of hCG. After a maximum of 3 h in vitro, embryos (n = 15 to 20, mean = 17.3) were transferred surgically to the middle of the uterine horn, to the caudal quarter of the uterine horn, or to the uterine body. Recipients were slaughtered between 28 and 34 d after transfer. The pregnancy rate of the recipients was low when the embryos were deposited in the uterine body (12%), compared with the middle (88%) or the caudal quarter of the uterine horn (81%) (P < .01). The corresponding average number of viable fetuses per pregnant recipient was 8.2 in the uterine body, 5.6 in the middle, and 4.5 in the caudal quarter. Average survival rate of embryos after transfer to the middle of the uterine horn was 41% vs 29 and 3% after transfer to the caudal quarter or the uterine body, respectively (P < .01). Hence, the uterine body seems to be an unsuitable site for embryo transfer in pigs. These results may explain the unsatisfactory results achieved with nonsurgical embryo transfer in the past.  相似文献

The objectives of this study were (a) to assess the ovulatory response and embryo production of Hungarian Merino ewes after superovulation, (b) to investigate the factors influencing the efficiency of embryo transfer (ET) in Hungarian Merino ewes, (c) to compare the results of two ovarian stimulation protocols (PMSG and PMSG + FSH treatment) in Hungarian Merino ewes, and (d) to study how superovulation, laparoscopic insemination and surgical embryo retrieval (ER) affect the subsequent reproduction of Hungarian Merino donor females after an ET programme. There was no significant difference between the ovarian stimulation protocols in the ratio of donor ewes responding to superovulation nor in the average number of corpora lutea. However, the number of transferable embryos recovered per donor ewe was higher in the PMSG + FSH group. The proportion of transferable embryos, unfertilized oocytes and degenerated embryos did not differ between the treatment protocols. The total pregnancy rate was 53.4% (179/335). Neither the developmental stage of the embryo nor the number of transferred embryos affect the implantation of embryos. However, the increased number of transferred embryos positively influenced the pregnancy rate. No difference was found in the pregnancy rate between synchronised and non-synchronised groups of recipients. Thirty-six out of 45 donor ewes (80%) became pregnant within one year after the ET programme, indicating that ovarian stimulation and surgical ER did not affect adversely their reproduction.  相似文献

Influence of estrus synchronization of prepubertal gilts on embryo quality   总被引:4,自引:0,他引:4  
Synchronization and superovulation are commonly used to obtain large numbers of embryos for experimental and practical purposes. This study compared the number, quality, and in vitro development of embryos recovered from gilts following single or double estrus synchronization and superovulation. Prepubertal gilts from the single synchronization group were injected with 1500 I.U. PMSG and 1000 I.U. hCG 72 h later. The double synchronized group of gilts was treated with 750 I.U. PMSG and 500 I.U. hCG 72 h later. After 17 days, 1500 I.U. PMSG followed by 1000 I.U. hCG was administered. Five days after insemination embryos were recovered and cultured for 6 days. The effects of pregnant mare serum gonadotrophin (PMSG) dose and PMSG antiserum on superovulation in crossbred beef cows were studied. In experiment I, three groups were treated with 1200, 2400 or 3600 IU of PMSG and 48 h later with prostaglandin (PGF). The mean numbers of corpora lutea (CL), unovulated follicles, and total ova/embryos collected increased as the PMSG dose increased. The percent of fertilized ova and transferable embryos was lowest in the highest dose group (p < 0.05). In experiment II, all cows received 2500 IU of PMSG; groups 1 and 2 were treated with sheep anti-PMSG serum at 48 h or 60 h after PGF; group 3 cows were PMSG-only controls. The number of CL was lowest and the number of unovulated follicles highest in the PMSG-only group (p < 0.05). The number of CL was higher in group 2 (anti-PMSG at 60 h) than in the control group, with the anti-PMSG at 48 h not different from the other groups. Numbers of total ova/embryos, fertilized ova, and transferable embryos were higher (p < 0.05) in both antiserum-treated groups relative to the PMSG-only group. We conclude that superovulation of beef cows with PMSG and treatment with PMSG antiserum will induce a higher superovulatory response and will result in higher CL numbers and fewer unovulated follicles. Further, the variability in the superovulatory response to PMSG treatment was still evident when PMSG antiserum was administered.  相似文献

[目的] 评价引进品种肉羊的连续重复超数排卵(超排)、冲胚、胚胎移植等繁育技术集成模式的应用效果。[方法] 于2016—2018年,对选取的1 575只澳洲白(n=230)、白头杜泊(n=228)、黑头杜泊(n=541)、白头萨福克(n=200)、黑头萨福克(n=376)5个品种母羊,进行间隔30 d以上的连续同期发情(孕酮海绵栓)与超排(330~450 IU FSH+200~300 IU PMSG+0.1 mg PG)处理;经过腹腔内窥镜输精后进行子宫角冲胚,回收胚胎后计数并评估质量,挑取部分胚胎进行移植;对比分析品种、季节、重复超排次数等对胚胎生产指标的影响。[结果] 共获得18 496枚胚胎,有14 971枚可用胚;胚胎移植后的受胎率为69.8%,产羔率为93.2%;只均总胚数和只均可用胚数2项指标显示,胚胎生产特性以白头萨福克、黑头萨福克、黑头杜泊较好,澳洲白次之,白头杜泊最差;不同品种母羊连续重复超排冲胚后回收的只均总胚数和只均可用胚数持续增长,直到第4或第5次;澳洲白的超排冲胚效果在春季较好,而其他品种在冬季较好。[结论] 连续重复超数排卵、冲胚技术对于扩繁引进肉羊品种群体是可行的,应根据不同品种及其表现出的季节特性科学、适时应用。  相似文献
