Forty-two isolates ofColletotrichum gloeosporioidesfrom almond, apple, avocado, mango, pecan, and eight isolates ofC. acutatumfrom apple, peach and pecan were compared by molecular analyses and a pathogenicity assay in order to determine genetic variability and host specificity. Polymerase chain reaction (PCR) amplification of genomic DNA using four different primers andHaeIII digestion patterns of genomic DNA (A+T-rich DNA) grouped theC. acutatumisolates separately from theC. gloeosporioidesisolates. Based on arbitrarily primed PCR (ap-PCR), intraspecies similarity among the isolates ofC. acutatumandC. gloeosporioidesranged from 78 to 93% and from 0 to 38%, respectively. Similarity between the isolates ofC. acutatumandC. gloeosporioidesranged from 0 to 26.5%. A+T-rich DNA grouped theC. acutatumisolates separately from those ofC. gloeosporioides, corresponding to ap-PCR analyses. Artificial inoculations with nine representative isolates on almond, apple, avocado, mango and nectarine fruit showed a variation in levels of infection. TheC. gloeosporioidesisolates from almond grew more slowly, causing significantly smaller lesions on all inoculated fruit than the other isolates. TheC. acutatumisolates from apple and peach caused similar levels of infection on all fruit, but differed significantly from theC. gloeosporioidesisolate from apple. Variation in lesion size was also observed with isolates ofC. gloeosporioidesfrom apple, avocado and mango for most fruit inoculations.  相似文献

Almond anthracnose caused by Glomerella cingulata is a major disease of this crop in Israel. The pathogen infects young fruit resulting in fruit rot. Leaf wilting and shoot dieback accompany fruit rot, even though the pathogen cannot be isolated from leaves or twigs. Isolates of G. cingulata from diseased almond fruit were compared using vegetative compatibility grouping (VCG), molecular methods, fungicide sensitivity and pathogenicity assays in order to determine the genetic diversity and host specificity among different populations. Polymerase chain reaction amplification of genomic DNA, using four primers produced uniform banding patterns for all the almond isolates from different geographic locations in Israel. HaeIII digestion patterns of A + T-rich DNA, and Southern hybridization of the repetitive nuclear DNA element (GcpR1) to PstI-digested genomic DNA of almond isolates also revealed no polymorphism. Chlorate-resistant nitrate-nonutilizing (nit) mutants were generated and used in heterokaryon tests. Complementary heterokaryons formed between the mutants of different isolates indicated a single VCG. Isolates of G. cingulata from almond had optimal growth temperatures of 20–22°C as opposed to 26–28°C for avocado isolates. In addition, almond isolates of G. cingulata are insensitive to benzimidazole fungicides in contrast to sensitivity of isolates from avocado. In artificial inoculations, almond isolates infected almond, avocado, apple, mango and nectarine fruit at a slower rate than G. cingulata isolates from avocado, apple and mango. Only the anamorph Colletotrichum gloeosporioides has been detected on almond in Israel, whereas isolates of G. cingulata from other hosts produce ascocarps.  相似文献

The vegetative compatibility patterns among isolates ofElegans fusaria causing wilt disease of carnation were investigated. Nitrate non-utilizing mutants were generated from 16 isolates labelledF. redolens, nine of which came from carnation, and from 33 isolates labelledF. oxysporum, 19 of which came from carnation. Pairings of the mutants revealed five vegetative compatibility groups among the isolates from carnation, corresponding withF. oxysporum f.sp.dianthi race 1 (VCG1), race 2 (VCG2) and race 4 (VCG3),F. redolens f.sp.dianthi (VCG4) andF. redolens isolates from foot rot-diseased carnations (VCG5). Besides three isolates typical ofF. redolens, VCG4 comprised a now slightly deviating subculture of the type isolate ofF. redolens f.sp.dianthi of which the cultural characteristics correspond toF. oxysporum instead ofF. redolens. This observation may be taken to support previous conclusions that the distinction between both taxa is not justified. Otherwise, the compatibility patterns did not provide decisive evidence to accept or reject conspecificity of both taxa. Naast drie voorF. redolens karakteristieke isolaten omvatte VCG4 ook een afwijkende subculture van het type-isolaat vanF. redolens f.sp.dianthi, die in cultuureigenschappen overeen kwam metF. oxysporum in plaats vanF. redolens. Deze waarneming geeft enige steun aan eerdere conclusies dat het onderscheid tussen beide taxa niet gerechtvaardigd is. Daarbuiten gaven de compatibiliteitspatronen geen uitsluitsel over de mogelijke conspecificiteit van beide taxa. The relative virulence ofPhytophthora cactorum andP. syringae originating from almond trees, and ofP. citrophthora originating from citrus, to apple, pear, peach, cherry and plum rootstocks, was studiedin vivo andin vitro. Results of the different experiments were in good agreement. All testedPhytophthora isolates showed little virulence to pear rootstocks-causing only minor crown rot symptoms - and no virulence at all to apple rootstocks. In contrast, they were highly virulent to stone fruit rootstocks, causing crown rot disease. The non-pathogenicity of these isolates to pome rootstocks could be interpreted as strict host specificity.  相似文献

The virulence ofPhytophthora citrophthora isolated from various host-plants on three peach rootstocks (GF677, PR204, KID I) was examined. There was no significant difference among the rootstocks with respect to their susceptibility to testedP. citrophthora isolates. The most virulent isolate originated from sycamore (Acer pseudoplatanus); isolates from pistachio trees (Pistacia vera) also showed high virulence but were significantly less virulent than the sycamore isolate. Isolates originating from plum (Prunus domestica), almond (Prunus amygdalus) and lemon (Citrus limon) trees were moderately virulent on peach rootstocks; those from cyclamen (Cyclamen persicum) showed the lowest virulence of those tested. There was thus great variation in virulence among the testedP. citrophthora isolates. Vegetative compatibility among 17 isolates ofVerticillium dahliae obtained from watermelon, originating from eight regions of Greece, was investigated using complementation tests between nitrate-nonutilizing(nit) mutants. Among 529 chlorate-resistant sectors obtained, only 107 werenit mutants. These mutants were paired with tester strains (from Greece and other countries) of previously described vegetative compatibility groups (VCGs), and also were paired in many combinations among themselves. All isolates were self-compatible. Sixteen isolates were found to belong to VCG2. Only isolate V75 could not be assigned to a VCG, because the threenit mutants obtained from it showed negative reactions with the tester strains of four VCGs and with complementary mutants from other isolates. Based on this sample, we conclude that the population ofV. dahliae from watermelon in Greece is homogeneous in respect to VCG.  相似文献

Since the 1980s a new disease has been affecting Australian lychee. Pepper spot appears as small, black superficial lesions on fruit, leaves, petioles and pedicels and is caused by Colletotrichum gloeosporioides, the same fungus that causes postharvest anthracnose of lychee fruit. The aim of this study was to determine if a new genotype of C. gloeosporioides is responsible for the pepper spot symptom. Morphological assessments, arbitrarily‐primed PCR (ap‐PCR) and DNA sequencing studies did not differentiate isolates of C. gloeosporioides from anthracnose and pepper spot lesions. The ap‐PCR identified 21 different genotypes of C. gloeosporioides, three of which were predominant. A specific genotype identified using ap‐PCR was associated with the production of the teleomorph in culture. Analysis of sequence data of ITS and β‐tubulin regions of representative isolates did not group the lychee isolates into a monophyletic clade; however, given the majority of the isolates were from one of three genotypes found using ap‐PCR, the possibility of a lychee specific group of C. gloeosporioides is discussed.  相似文献

为明确大豆胶孢炭疽菌Colletotrichum gloeosporioides对苯醚甲环唑的抗性风险及其抗性机制,采用药剂驯化法诱导获得其抗性突变体,并测定抗性突变体的适合度和交互抗性,同时分析抗性突变体和敏感菌株cyp51A和cyp51B基因的cDNA序列、启动区序列及相对表达量。结果表明:从大豆胶孢炭疽菌2株敏感菌株中筛选获得7株低抗苯醚甲环唑的突变体,突变频率为1.75×10-4,其抗药性可以稳定遗传。抗性突变体与亲本敏感菌株在对温度的敏感性、菌丝生长速率、产孢量和致病力方面均存在一定差异。苯醚甲环唑与丙环唑之间存在中等交互抗性(相关系数为0.80,P<0.05),苯醚甲环唑与咪鲜胺锰盐、多菌灵、氟啶胺和吡唑醚菌酯之间无交互抗性。抗性突变体cyp51A和cyp51B基因的cDNA序列未发生核苷酸突变,cyp51A和cyp51B基因的启动区无插入片段。经苯醚甲环唑处理后,4株抗性突变体lq27-7-1、lq27-7-2、lq30-2-1和lq30-2-2的cyp51A基因相对表达量被诱导上调倍数显著高于亲本敏感菌株;3株抗性突变体lq27-7-2、lq...  相似文献

Botryodiplodia theobromae, Colletotrichum gloeosporioides andGliocephalotrichum microchlamydosporum are the causal fungi of three rambutan postharvest diseases, stemend rot, anthracnose and brown spot, respectively. Two different treatments of rambutan fruits to control the diseases were investigated: application of potassium metabisulphite (250 ppm) or culture filtrate ofTrichoderma harzianum (TrH 40) followed by controlled atmosphere storage (CA) at 13.5°C and 95% r.h. Potassium metabisulphite at 250 ppm under CA effectively controlled the incidence and severity of the three postharvest diseases and maintained the eating quality and color of the fruit for 21 days. The greatest effect of this treatment was on brown spot disease, caused byG. microchlamydosporum. Forty-three isolates ofVerticillium lecanii from insects, phytopathogenic fungi and other substrates were tested for vegetative compatibility by observing heterokaryon formation among complementary nitrate-nonutilizing (nit) mutants.nit mutants were isolated from 42/43 strains examined. Twenty-one isolates were self-incompatible, and the remaining 21 isolates were divided into 14 vegetative compatibility groups (VCGs): ten containing only a single strain each, and the remaining four containing two to four isolates each. Members of isolates in each of these VCGs all shared the same IGS haplotype. Further, the isolates within a VCG were correlated with one another in part by fragment patterns of mt-LrDNA, -SrDNA, Bt-2 and H4 region, by PCR-RFLP and -SSCP, but not by dsRNA. Two isolates belonging to VL-J2 have high virulence to aphids, whereas strains from VL-J1 lack this character. These findings indicate that two VCGs (VL-J1 and -J2) may originate from two distinct clonal lineages. Alternatively, high VCG diversity and HSI frequency ofV. lecanii might be associated with an array of distinct lineages. Eleven pathotype groups (A-K), including five not previously reported, ofDidymella rabiei (anamorphAscochyta rabiei), representing isolates of the pathogen from Ascochyta blight-affected chickpeas mainly from India, Pakistan, Spain and the USA, were characterized using 44 single-spore isolates tested against seven differential chickpea lines. Of 48 isolates tested for mating type, 58% belonged to MAT 1-1 and 42% to MAT 1-2. Thirty-nineD. rabiei isolates, as well as two isolates ofAscochyta pisi and six isolates of unrelated fungi, were analyzed using Randomly Amplified Polymorphic DNAs (RAPDs) employing five primers (P2 at 40°C, and OPA3, OPC1, OPC11 and OPC20 at 35°C). Computer cluster analysis (UPGMA / NTSYS-PC) detected a relatively low level of polymorphism among all theD. rabiei isolates, although atca 7% dissimilarity,ca 10 RAPD groups [I-X] were demarcated, as well as subclustering within the larger groups. By the same criteria, the maximum dissimilarity for the whole population ofD. rabiei isolates wasca 13%. No correlation was found between different RAPD groups, pathotype, or mating type ofD. rabiei, although some evidence of clustering based on geographic origin was detected. The use of RAPDs enabled us to identify specific DNA fragments that may have a potential use as genetic markers in sexual crosses, but none which could be used as virulence markers.  相似文献

为有效防控由胶孢炭疽菌Colletorichum gloeosporioides引起的辣椒炭疽病,自辣椒上分离得到内生细菌,通过平板拮抗和辣椒离体生防试验筛选对胶孢炭疽菌有抑制作用的拮抗菌株,通过形态学特征、生理生化特征以及分子生物学技术对其进行鉴定,并于室内测定其对胶孢炭疽菌菌丝生长的影响、对辣椒炭疽病的防效及接种后辣椒内抗病活性物质含量以及防御酶活性。结果显示,从辣椒上共分离纯化获得46株细菌,其中菌株SQ-6对胶孢炭疽菌有明显的抑制作用,抑制率为61.11%,显著高于其他45株。结合菌株SQ-6的形态学特征、生理生化特征以及分子生物学特征,将该菌株鉴定为解淀粉芽胞杆菌Bacillus amyloliquefaciens。SQ-6菌株的50%无细胞滤液可引起胶孢炭疽菌菌丝畸形、断裂等,对其抑制率为57.87%。SQ-6菌株的10%、50%发酵液和10%、50%无细胞滤液均能显著降低由胶孢炭疽菌引起的辣椒炭疽病的发病率和病情指数,其中50%无细胞滤液的防效最好。SQ-6菌株能够提高辣椒内Vc、酚类和黄酮类物质含量,诱导辣椒内过氧化物酶(peroxidase,POD)、丙氨酸解氨酶(p...  相似文献

Rumple is a serious peel collapse of Primofiori lemons in the southeast of Spain with an unresolved aetiology. Symptoms typically occur on fruits at ripening under wet conditions as dark sunken lesions producing premature fruit drop and damaged fruits unacceptable for fresh commercialization. A total of 16 Colletotrichum spp. isolates established from rumple-affected lemons collected during the autumn of 2007 from two different orchards were characterized by molecular and phenotypic assays and compared with reference isolates. Species-specific PCR reactions using β-tubulin 2 nucleotide sequences showed Colletotrichum gloeosporioides to predominate (81.5%) with limited occurrence of C. acutatum (18.75%). Among the C. gloeosporioides isolates, five (38.5%) showed benomyl resistance and eight (61.5%) were highly sensitive to the fungicide. The limited occurrence of C. acutatum could be related to factors such as the presence of both species on the same fruit, unfavourable meteorological conditions and low disease incidence. This work reveals an association of C. gloeosporioides and C. acutatum isolates with rumple disease of lemons and expands the range of C. acutatum on citrus.  相似文献

Differential interactions ofColletotrichum gloeosporioides isolate KG 13 with green and red pepper fruits (Capsicum annuum were found when it was inoculated on unwounded and wounded fruits. The isolate produced the typically necrotic, sunken anthracnose symptom on unwounded and wounded green fruits, and wounded red ones, but not on unwounded red ones. Appressorial formation of the fungus on the surfaces of compatible green fruits was higher than on incompatible red ones up to 12 h after inoculation. More and longer infection pegs from appressoria were produced on green than on red fruits. When cuticular wax layers of green and red fruits were removed by dipping in chloroform, red ones only produced larger lesions and more conidia than water-dipped controls did. However, differences in lesion diameter and conidial production were not observed between green and red fruits wounded by pin-pricking. In addition, concentrations of wax extracted from the surface of green and red fruits affected conidial germination and appressorial formation of the fungus. These findings suggest that the isolate KG 13 ofC. gloeosporioides may react differentially to green and red pepper fruits, probably due to the physical and chemical differences in cuticular layers of the fruits.  相似文献

The aim of the present study was to analyse the genetic and pathogenic variability of Colletotrichum spp. isolates from various organs and cultivars of mango with anthracnose symptoms, collected from different municipalities of São Paulo State, Brazil. Colletotrichum gloeosporioides isolates from symptomless citrus leaves and C. acutatum isolates from citrus flowers with post‐bloom fruit drop symptoms were included as controls. Sequencing of the ITS region allowed the identification of 183 C. gloeosporioides isolates from mango; only one isolate was identified as C. acutatum. amova analysis of ITS sequences showed larger genetic variability among isolates from the same municipality than among those from different populations. fAFLP markers indicated high levels of genetic variability among the C. gloeosporioides isolates from mango and no correlation between genetic variability and isolate source. Only one C. gloeosporioides mango isolate had the same genotype as the C. gloeosporioides isolates from citrus leaves, as determined by ITS sequencing and fAFLP analysis. Pathogenicity tests revealed that C. gloeosporioides and C. acutatum isolates from either mango or citrus can cause anthracnose symptoms on leaves of mango cvs Palmer and Tommy Atkins and blossom blight symptoms in citrus flowers. These outcomes indicate a lack of host specificity of the Colletotrichum species and suggest the possibility of host migration.  相似文献

由胶孢炭疽菌Colletotrichum gloeosporioides引起的炭疽病是建兰的重要病害.为建立快速检测该病原菌的方法,以ITSl/ITS4为引物,对15个建兰胶孢炭疽菌的ITS进行PCR扩增及测序,将测定的序列与炭疽菌属其它种的ITS序列进行比对分析,设计特异性引物CFl/CR1,并通过常规和巢式PCR对建兰胶孢炭疽菌进行检测.结果显示,15个菌株中有13个菌株ITS序列与该菌的模式种序列相似性高达99%以上,而另外2个菌株相似性则为86%;供试菌株在系统发育树上聚为2个不同的分支;引物CFl/CR1通过常规PCR可从1 ng的建兰胶孢炭疽菌基因组DNA中扩增到目的条带,而利用引物ITSl/ITS4和CF1/CR1通过巢式PCR可从1 pg的基因组DNA中扩增到目的条带,即巢式PCR反应检测灵敏度较常规PCR至少高1 000倍.表明建立的巢式PCR法可从自然感病的建兰叶片组织中检测到胶孢炭疽菌.  相似文献

Verticillium dahliae Kleb. with a complicated genetic diversity is a widely distributed major pathogen resulting in cotton wilt, which causes high economic losses in cotton lint production in the cotton belt of Turkey. A collection of 70 TurkishV. dahliae isolates (68 from wilted cotton plants in 28 districts and two from watermelon plants in two districts) were tested for vegetative compatibility by observing heterokaryon formation among complementary nitrate-nonutilizing (nit) mutants. The mutants were tested against international reference tester isolates and also were paired with one another. Thirty-nine isolates were assigned to vegetative compatibility group (VCG) 2B, 19 to VCG2A and three to VCG4B. One isolate was self-incompatible and eight others could not be assigned to any of the identified VCGs because theirnit mutants showed negative reactions with the tester isolates of four VCGs or theirnit mutants reverted back to the wild type. This is the first report of VCGs inV. dahliae from cotton in Turkey.  相似文献

Microbial isolates from living petals, petal residues and leaf residues of rose, and from laboratory collections, were evaluated for control ofBotrytis cinerea in rose. In leaf residues artificially infested withB. cinerea, isolates of the filamentous fungiGliocladium roseum, FR136 (unidentified) andTrichoderma inhamatum reduced sporulation of the pathogen by >90%, other filamentous fungi were 25–90% effective, and those of yeasts and bacteria were <50% effective. In artificially inoculated petal residues, no microbe reduced sporulation ofB. cinerea by >75%, but isolates ofCladosporium oxysporum and four yeasts were 51–75% effective, and three filamentous fungi, eight yeasts andBacillus subtilis isolates were 26–50% effective. Isolates ofT. inhamatum, C. oxysporum andG. roseum performed best againstB. cinerea among isolates evaluated in leaf residues naturally infested with the pathogen and indigenous microorganisms. Totals of ten isolates of filamentous fungi (includingC. oxysporum andC. cladosporioides), two of yeasts and five ofBacillus subtilis completely prevented lesion production byB. cinerea in detached petals, and a further six isolates of filamentous fungi (includingG. roseum) and six yeasts were 90–99% effective. Isolates ofC. oxysporum, C. cladosporioides andB. subtilis, the most effective microorganisms againstB. cinerea in flower buds, reduced number of lesions in the range of 42–65% compared with 59–89% for à standard fungicide (vinclozolin). It is suggested that application of leading antagonists Jo living rose leaves and flowers should optimize control of inoculum production byB. cinerea when the tissues die. Optimal biocontrol of lesion production in flower buds requires a better understanding of the microenvironment of petals.  相似文献

为明确引起福建省大豆炭疽病的病原菌种类,2018—2019年从福建省福州、三明、莆田、泉州和南平市采集具有大豆炭疽病症状的大豆豆荚,采用组织分离法分离病原菌,结合形态学特征和多基因系统发育分析对病原菌进行鉴定。结果表明,从采集的豆荚样本中共分离获得152株炭疽菌菌株,经形态学初步鉴定分属于3种类型,从3种类型的病原菌中分别选取3株代表菌株进行多基因系统发育分析,这9株代表菌株分别与Colletotrichum plurivorum、平头炭疽菌C.truncatum和胶孢炭疽菌C.gloeosporioides聚在一起。形态学和多基因系统发育分析结果表明,引起福建省大豆炭疽病的病原菌有3种,分别为C.plurivorum、平头炭疽菌和胶孢炭疽菌,其占比分别为10.53%、50.00%、39.47%。致病性测定结果表明,C.plurivorum、平头炭疽菌和胶孢炭疽菌对大豆均有致病作用,但不同的菌株致病力存在差异。表明平头炭疽菌是引起福建省大豆炭疽病的主要病原菌,而C.plurivorum为引起我国大豆炭疽病的新纪录种。  相似文献

Pepper cultivars from diverse geographic origins were evaluated for resistance to different isolates ofPhytophthora capsici under controlled environmental conditions. All accessions tested were susceptible at the four-leaf stage to the six isolates ofP. capsici. Inoculation at the eight-leaf stage resulted in significantly different interactions among the accessions andP. capsici isolates. The Korean and U.S. cultivars tested were highly susceptible to the isolates ofP. capsici at this stage. In contrast, PI 201234 and PI 201238 had a differential interaction with someP. capsici isolates. At the twelve-leaf stage, Phytophthora blight developed slowly in the Korean and U.S. cultivars that were highly susceptible at the eight-leaf stage. Furthermore, the accessions from the Asian Vegetable Research and Development Center (AVRDC) became highly resistant toP. capsici at this stage.  相似文献
