The infection and colonization process of Colletotrichum acutatum on ripe blueberry fruit from two cultivars with different susceptibility to anthracnose were examined using light and confocal laser scanning microscopy. Ripe fruit from susceptible cv. Jersey and resistant cv. Elliott were drop-inoculated with a conidial suspension of C. acutatum, and epidermal peels were evaluated at selected times after inoculation and incubation. Results from pre-penetration studies demonstrated that there were significant differences in the rate of formation of melanized appressoria between the two cultivars, with the rate of formation being faster in the susceptible one. In both cultivars, penetration by the pathogen occurred via appressoria 48 h post-inoculation (hpi). However, in the susceptible cv. Jersey, C. acutatum then adopted an intracellular hemibiotrophic-like infection strategy, whereas in the resistant cv. Elliott subcuticular intramural-like infection occurred. In cv. Jersey by 108 hpi, intracellular growth of the pathogen led to the formation of numerous acervuli, with orange conidial masses. By 120 hpi, the conidial masses had coalesced covering the entire inoculated area. In cv. Elliott, acervuli were not seen until 144 hpi and contained few conidia. These results demonstrate for the first time the ability of C. acutatum to adopt a different infection and colonization strategy depending on the susceptibility of the host tissue being colonized.  相似文献

Host plant resistance is an important component for management of the melon fruit fly, Bactrocera cucurbitae (Coquillett), owing to difficulties associated with its chemical and biological control. Various biochemical traits including total sugar, reducing sugar, non-reducing sugar, tannins, phenols, alkaloids, flavinoid and pH contents of fruit were studied on 11varieties/ genotypes of muskmelon, Cucumis melo L., in relation to resistance against B. cucurbitae under field conditions. Significant differences were found in tested varieties/ genotypes for fruit infestation and larval density per fruit. AHMM/BR-1, RM-50 and AHMM/BR-8 were the most resistant; MHY-5, Durgapura Madhu and Pusa Sarabati were moderately resistant; AHMM/BR-13, Pusa Madhuras and Arka Jeet were susceptible; whereas Arka Rajhans and GMM-3 were the highly susceptible varieties/ genotypes to fruit fly in both seasons, 2011 and 2012. The larval density per fruit increased with an increase in percent fruit infestation and there was a significant positive correlation (r = 0.97) between percent fruit infestation and larval density per fruit. Total sugar, reducing sugar, non-reducing sugar and pH were lowest in resistant and highest in susceptible varieties/ genotypes, whereas tannins, phenols, alkaloids and flavinoid contents were highest in resistant and lowest in susceptible varieties/ genotypes. Total alkaloid and pH contents explained 97.96% of the total variation in fruit fly infestation and 92.83% of the total variation in larval density per fruit due to alkaloids and total sugar contents.  相似文献

Reactions of 60 water yam ( Dioscorea alata ) cultivars to three isolates of the yam anthracnose fungal pathogen ( Colletotrichum gloeosporioides ) were evaluated using tissue culture-derived whole-plant assay. Three disease parameters: single score on a scale of 0–6 at the seventh day after inoculation (SD7); area under the disease progress curve (AUDPC); and disease progress rate ( R d) were compared, and cultivars were classified into disease-response groups using a rank-sum method based on AUDPC scores for the two most virulent isolates. A wide range of variation in resistance of the D. alata cultivars, and significant effects of pathogen isolate and isolate–cultivar interactions, were observed for all disease parameters. The three disease parameters were positively correlated; however, four cultivars showed great dispersions from the regression lines for comparisons of SD7 with the multiple assessments based AUDPC and R d. The 60 cultivars were separated into resistant ( n  = 12), moderately resistant ( n  = 19), moderately susceptible ( n  = 18) and susceptible ( n  = 11) groups. The potential of the tissue culture-derived whole-plant assay to resistance breeding programmes and further understanding of the yam anthracnose pathosystem is discussed.  相似文献

Anthracnose fruit rot, caused by Colletotrichum acutatum, is among the most important diseases of blueberries. Most cultivars are susceptible but ‘Elliott’ is resistant. Our objective was to identify possible antifungal compounds that play a role in the resistance response. Chemical fractions from freeze-dried, ripe fruit of ‘Elliott’ and a susceptible cultivar (Jersey) were extracted with methanol and ethyl acetate. Extracts were screened on solid media for suppression of microconidiation of C. acutatum. The methanolic extract was fractionated and the soluble methanolic fraction from ‘Elliott’ was the most biologically active. This fraction was dried, dissolved in water, and screened in vivo by pre-treating ripe ‘Jersey’ fruit with 0.5, 1, 2, and 4% solutions (w/v) and subsequently inoculating the fruit with C. acutatum. An 88% reduction in infection incidence was observed after 12 days with the 4% solution. Anthocyanins and other flavonoids were then quantified in fruit of the two cultivars using HPLC-MS. ‘Elliott’ fruit contained more anthocyanins (4.87 mg/g of freeze-dried tissue) than ‘Jersey’ (3.27 mg/g of freeze-dried tissue); however, the same compounds were found in both cultivars. ‘Elliott’ fruit also contained more non-anthocyanin flavonoids (0.18 mg/g of freeze-dried tissue) than ‘Jersey’ (0.12 mg/g of freeze-dried tissue), including two distinctive compounds in ‘Elliott’. The non-anthocyanin flavonoid fractions of both ‘Elliott’ and ‘Jersey’ significantly decreased the growth of C. acutatum in a liquid bioassay but the effect was more pronounced in the ‘Elliott’ fraction. The two distinctive compounds in ‘Elliott’ were further characterized by MS/MS and were identified as quercetin 3-O-rhamnoside and, putatively, syringetin rhamnoside. Additionally, we evaluated ‘Elliott’ and ‘Jersey’ methanolic extracts for their ability to inhibit lipid peroxidation, and the extract obtained from ‘Elliott’ was almost twice as active at inhibiting peroxidation. These results provide new insights into the role of antifungal compounds in the resistance response of ripe ‘Elliott’ blueberries to infection by C. acutatum.  相似文献

The grey mould disease caused by Botrytis cinerea leads to substantial economic losses in strawberry production all over the world. Control of the disease requires an extensive amount of fungicide that is applied in varying complexes because the pathogen easily develops resistance against the active compounds. Planting of resistant cultivars seems to be a promising alternative for fruit growers, but there are currently no cultivars available combining resistance to B. cinerea with attractive horticultural traits. Breeding of new cultivars requires the effective identification of resistant strawberry genotypes; therefore the current study was aimed at the evaluation of strawberry genetic resources under controlled conditions by establishing an artificial inoculation assay. The method presented in this study is an artificial inoculation of ripe fruits with a defined spore suspension under laboratory conditions. The results show that this assay is fast and simple and leads to reproducible results that correlate with field observations. Over 3 years a total of 107 strawberry genotypes of the German National Fruit Genebank at the JKI in Dresden‐Pillnitz were evaluated. Five partly resistant genotypes, cultivars Diana, Joerica and Kimberly, and Fragaria virginiana ‘Wildmare Creek’ and F. vesca subsp. bracteata, were identified with mean disease levels of <20% at 6 days post‐inoculation. The obtained results are discussed with regard to future breeding activities.  相似文献

1991 ̄1995年采用自然发病和土壤接种方法鉴定了1550份玉米品种对茎腐病的抗病性,表现高抗的397份,中抗304份,中感364份,高感485份,经重复鉴定,筛选出76份抗性稳定的自交系和杂交种。不同抗性品种用肿囊腐霉菌和串珠镰刀菌接种测定,抗性反应基本一致,人工接种比自然发病鉴定效果好,通过病圃自然发病初筛,人工土壤接处复选,可准确鉴定大量材料的抗病性。  相似文献

Several factors influencing the incidence of internal fruit rot of cucumber caused byDidymella bryoniae were studied.Internal infection of fruits is achieved via the flower. However, in most cases the majority of the fruits escaped infection after flower inoculation. It took more than two days for the fungus to reach the fruit after infection of the style. A mechanical barrier was not detected in the fruit tip within three days after inoculation of the open flower.Inoculation of wilted flowers resulted in 60% less infection than inoculation of fresh flowers. Blossom excision reduced fruit infection with ca 75%. Growing plants under drought stress markedly increased the incidence of internal fruit rot.Neither the method of inoculation, nor the composition of the inoculum, nor the relative humidity influenced the incidence of internal fruit rot. Fruit thinning, duration of fruit growth, flowering period and the removal of parts of the flower had no effect either on fruit infection.Cultivars resistant to powdery mildew were also resistant to internal fruit infection. The resistance was associated with a long style and a short flowering period.Growing cultivars in which the flowers quickly fall away from the fruitlets or in which the flowers have no style may solve the problem of internal fruit rot in cucumber.Samenvatting Factoren die van invloed kunnen zijn op het ontstaan van inwendig vruchtrot van komkommer, veroorzaakt doorDidymella bryoniae, werden onderzocht.Inwendige vruchtinfectie vindt plaats via het bloempje. Vruchtdunning, duur van de vruchtgroei, bloeiduur en het verwijderen van delen van de bloem hadden ook geen effect op de aantasting.Cultivars die resistent waren tegen echte meeldauw vertoonden ook resistentie tegen inwendige vruchtaantasting. Bitter rot, caused by the fungus Greeneria uvicola, is one of the most important fruit rot diseases that threaten the burgeoning winegrape (Vitis vinifera) industry in the southeastern United States. Epidemiological studies were conducted to examine the period of fruit susceptibility of V. vinifera to G. uvicola, influence of temperature and duration of wetness on infection, and relative susceptibility of cultivars to bitter rot. In field studies, susceptibility of Merlot, Chardonnay, and Cabernet Franc fruit increased from bloom until véraison in 2003 and from bloom until 2 weeks before véraison in 2004. When detached V. vinifera fruit were inoculated and incubated at 14, 22, 26, and 30 degrees C for 6, 12, 18 or 24 h of wetness, 22.4 to 24.6 degrees C and 6 or 12 h of wetness were the optimal conditions for infection of fruit by G. uvicola. The relative susceptibility of 38 cultivars and selections, including 23 V. vinifera cultivars and five French-American hybrids, was determined in a detached fruit inoculation assay. A wide range in susceptibility was observed among the cultivars and selections. Fruit of cultivars of V. vinifera were significantly more susceptible than French-American hybrids. Isolates of G. uvicola differed in aggressiveness when tested on cv. Chardonnay.  相似文献

Head rot is a major disease of broccoli caused by the soft rot pathogens Pseudomonas fluorescens and Erwinia carotovora. Two in vitro pathogenicity tests were evaluated as a methods to identify broccoli cultivars susceptible or resistant to bacterial head rot. One test used mature heads excised from the plant and inoculated with squares of cotton lint which had been soaked in a bacterial suspension. The other test involved stab-inoculating axenically grown seedlings. With the excised head test, susceptible cultivars showed a black soft rot, whilst less susceptible or moderately resistant cultivars showed only watersoaking, or browning and slight softening of the tissue. No cultivar was completely resistant. Ten cultivars were tested, and their susceptibility ratings corresponded with previously recorded field data, with one exception. This laboratory test could be used to screen for susceptibility to head rot in broccoli breeding programmes. The seedling test distinguished differences in aggressiveness among bacterial isolates but not cultivar susceptibility. Increasing head size correlated negatively with disease resistance. Head shape, i.e. cultivars which showed a domed shape rather than a flat shape, was positively correlated with disease resistance. Thus small domed heads are more resistant to head rot than large flat heads. Other morphological characteristics, viz. floret prominence and number, and sepal stomatal number were not correlated with host resistance.  相似文献

Early anthracnose caused by Colletotrichum acutatum has become an increasingly serious disease on green, unripe bell pepper fruit in Florida. This contrasts with earlier reports of anthracnose occurring on bell pepper primarily as a ripe-rot disease of mature, colored pepper fruit caused by Colletotrichum gloeosporioides. Management of anthracnose on green bell pepper fruit using fungicides and a commercial inducer of systemic acquired resistance, acibenzolar-S-methyl (ASM), was evaluated during three seasons. In two of the three trials, all the fungicides tested including azoxystrobin, fludioxonil + cyprodinil, mancozeb, famoxadone + cymoxanil, copper hydroxide, and ASM significantly increased the number of marketable fruit compared with control plants. These trials identified fungicides that could contribute to a successful pest management program on pepper for controlling anthracnose caused by C. acutatum. The cross-infectivity potential of C. acutatum was investigated on tomato and strawberry by in vitro and field inoculation. Anthracnose lesions formed readily on wound-inoculated detached fruits of all hosts in in vitro assays. Under field conditions, after inoculation, anthracnose lesions occurred on pepper fruit but no lesions of anthracnose were found on either ripe or unripe tomato or strawberry fruit in adjacent plots.  相似文献

研究了玉米灰斑病菌侵染四个抗病和感病的玉米品种时,叶片内部活性氧代谢酶及细胞过氧化产物含量的动力学变化。结果表明,抗、感病品种的SOD、CAT及POD酶活性在病茵侵染后都变化显著,抗病品种各酶活性变化幅度比感病品种大。叶片内过氧化产物MDA含量则相反,抗病品种沈试29在接种第13天时叶片MDA含量只为18.46nmol/g,而感病品种铁单9为23.14nmol/g,抗病品种比感病品种增加幅度小。说明活性氧代谢在植物抗病机制中起着重要作用,抗病品种对活性氧代谢的酶调节能力强,病菌侵染后细胞过氧化程度低,在痛菌侵染时活性氧清除酶活性最大增加值与发病程度呈显著正相关。  相似文献

葡萄霜霉病是葡萄生产上重要的病害之一,通过对辽宁省不同葡萄品种进行室内离体叶片接种和田间自然发病情况调查,以期为葡萄抗性品种的选育和葡萄霜霉病的防治提供科学依据。结果表明,在供试品种中,没有对霜霉病完全免疫的品种,室内离体叶片接种和田间调查结果基本一致,不同品种间霜霉病的抗性存在差异。供试的65个品种中,室内离体叶片接种评价高抗品种3个,抗病品种23个,感病品种24个,高感品种15个;田间自然发病调查评价高抗品种1个,抗病品种19个,感病品种35个,高感品种10个。欧美杂交品种(系)相对欧亚杂交品种(系)较抗病。  相似文献

Neonectria galligena can cause European canker of apple as well as fruit rot. Healthy unwounded fruits on potted trees of cvs Cox, Bramley and Gala were inoculated with conidia of N. galligena to investigate the effects of wetness duration and fruit maturity on rot development. Overall, the incidence of fruit rot was influenced more by fruit maturity at the time of inoculation than by duration of wetness (6–48 h). Young fruit were most susceptible to infection, with 50% of fruit infected when inoculated up to 4 weeks after full bloom. The susceptibility decreased initially until c. 2 months after full bloom and then increased gradually until harvest. Almost all preharvest symptoms (eye rot) developed only on the fruit inoculated up to 4 weeks after full bloom. All other rots were observed after six‐month postharvest storage under controlled atmospheric conditions. However, the relative proportion of preharvest eye rots and postharvest storage rots varied greatly among three years. The effect of wetness duration was only significant for fruit inoculated in their early stages of development but not for those inoculated near harvest. Regression models were developed to describe the observed effects of fruit maturity and wetness on the incidence of total nectria rots.  相似文献

Six inoculation techniques were compared for their ability to evaluate resistance toColletotrichum acutatum of five strawberry cultivars. Inoculation by dipping the whole cold stored plants in a suspension of conidia adjusted to 2.106 conidia ml–1 made it possible to screen cultivars resistant to crown rot at 28 days after inoculation. Using the dipping technique, 44 strawberry cultivars were evaluated for their resistance to one strain ofC. acutatum, 1267b. Twelve of them did not show wilt symptoms and could be classified as resistant. When another strain ofC. acutatum, 494a, was inoculated to seven cultivars, all of them including Dover, resistant to 1267b, showed wilt symptoms. This result showed the importance of investigations on genotype × isolate interactions to conduct an efficient breeding programme for screening resistance toC. acutatum.  相似文献

Black rot, caused by Xanthomonas campestris pv. campestris, (Xcc), is one of the most serious diseases of crucifers world-wide. Forty-nine genotypes were evaluated for resistance under field conditions in Tanzania after artificial inoculation with Xcc race 1. Open pollinated white cabbage cultivars were generally susceptible, while Portuguese and pointed cabbages exhibited partial resistance. Some F1 white cabbage cultivars were highly susceptible, whereas others exhibited a high level of partial resistance. The most promising of the hybrid cultivars were T-689 F1, Gianty F1, No. 9690 F1, N 66 F1, and SWR-02 F1. Breeding line Badger I-16 exhibited the highest level of resistance of all genotypes. The genotypes accounted for 72.9–75.5% of the variation of the disease severity when assessed on the leaves, and 71.4% of the variation when assessed as internal black rot in heads at harvest. High correlations (equal to or above 0.7) were found between disease severities assessed on leaves three times during the growing season and also with the amount of internal black rot in heads. Leaf loss also was correlated with disease severity. The high genetic determination of the trait and the high correlations between disease assessments indicate that selection for resistance to black rot will be efficient when field screenings are carried out. Evaluation of genotypes for disease severity on leaves during the growing season combined with evaluations of head resistance in the most promising genotypes may be a simple method to select resistant cultivars.  相似文献
