Mature black sea bass, Centropristis striata L. (200–800 g), were captured in coastal South Carolina during the spawning season and administered hormones for ovulation induction and strip spawning. During both study years, control groups of females were incorporated into the study design and administered sham injections containing physiological saline solution. In 2004, females received a single intramuscular injection of human chorionic gonadotropin (hCG) (330 IU kg−1) (n=8) or two injections of hCG at 24‐h intervals (n=8). In 2005, females received a single injection of hCG (n=10) or an analogue of luteinizing hormone releasing hormone (LHRHa) (n=10). In 2004, all fish administered a single dose of hCG ovulated at least once. Six fish ovulated on two consecutive days and one fish ovulated on 3 days consecutively. In contrast, six of eight fish receiving two doses of hCG ovulated once, five ovulated on 2 days successively and three fish ovulated 3 days in succession. Of the fish that spawned, no differences were found in any reproductive parameters. In 2005, all fish administered hCG or LHRHa ovulated at least once. Three fish administered hCG ovulated twice, four fish ovulated on three consecutive days and one fish 4 days successively. All fish administered LHRHa spawned at least twice, six fish ovulated thrice and three fish ovulated 4 days, successively. A significant difference in fertility was found between hCG (75.6±11.4%) and LHRHa (55.6±27.4%). The results of this study indicate that both hCG and LHRHa are effective for ovulation induction in prespawning black sea bass.  相似文献

Abstract The timing of ovulation and survival of eggs following exposure of 2-sea-winter Atlantic salmon, Salmo salar L., to different temperature regimes during the spawning season were investigated. Water temperature was either increased from 10 to 13–14°C (warm water), decreased abruptly from 10 to 5–7°C (cold water), or gradually decreased from 10 to 8°C (ambient control) from 1 November onwards. Median ovulation time was delayed by 5 weeks in the warmwater group compared with ambient controls, with 43% of the females remaining non-ovulated at the end of the study. Only minor effects were observed on timing of ovulation in the coldwater group compared with ambient controls. Survival of eggs to the eyed stage was significantly higher in the coldwater group (92·1%), compared with both the ambient control group (84·5%) and the warmwater group (76·6%). The results indicate that high water temperature during the spawning season inhibits ovulation and has a detrimental effect on gamete quality in Atlantic salmon.  相似文献

Abstract— Channel catfish Icralurus punctatus are commonly spawned for research purposes by pairing of a hormonally treated female with a male in flow-through aquaria. A technique that allows hormonal induction of ovulation in females without pairing would accelerate genetic improvement and production of hybrid catfish. Over a 3-yr period (1994, 1995, and 1996) we conducted a series of trials to demonstrate the potential for artificial spawning in recirculating systems, and in 1996 we included trials with grouped females in addition to male-female pairs. Females were induced to spawn with injection of synthetic leuteinizing hormone-releasing hormone, and those that ovulated were stripped and the eggs were artificially fertilized. During 1994 and 1995, all fish were spawned by pairing, and in 1996, half of the females were spawned by pairing and half were grouped in tanks without males. Spawning success (percent of females that produced eggs), latency (time between injection and ovulation), and percent fertilization were observed for the paired and grouped trials. Spawning success was 36% in 1994 (N= 36). 22% in 1995 (N= 54). 41% in 1996 (N= 27). and 58% for grouped females (N= 26). The latency period was 113 ± 69 h in 1994, 109 ± 57 h in 1995, 44 ± 8 h in 1996, and 50 ± 9 h for grouped females. Percent fertilization was 16 ± 26% for eggs stripped in 1994, 72 ± 25% in 1995, 43 ± 20% in 1996, and 16 ± 37% for grouped females. In 1995, water quality problems were associated with high mortality of females (24 of 44 females; 4 of 44 males). The metabolic demands of final oocyte maturation in combination with methemoglobinemia caused by high nitrite levels could account for the increased vulnerability of females. These trials indicate that with adequate biofiltration, artificial spawning is possible in recirculating systems and with females grouped rather than paired. Further research on hormone dosage and timing of egg stripping will increase the utility of grouped spawning of channel catfish.  相似文献

This study addresses rapid methods to identify mature channel catfish, Ictalurus punctatus, females for induced spawning with luteinizing hormone releasing hormone analog (LHRHa) and common carp pituitary extract (CPE) and the effects of stage of maturity, hormone dose, season, and cannulation before hormone injection. Hormonal intervention is the most effective method for inducing maturation and spawning in channel catfish × blue catfish, Ictalurus furcatus, hybrids. A total of 80 mature channel catfish were staged for maturity based on secondary sexual characteristics and 20–30 oocytes cannulated to ascertain their maturation stage based on the position of gonadal vesicle. Twenty females were randomly assigned to one of the four hormone regimes (priming + resolving dose): CPE 2 + 8 mg/kg (CPEC); LHRHa 20 + 40 µg/kg (LHRHaL); LHRHa 20 + 60 µg/kg (LHRHaM); and LHRHa 20 + 80 µg/kg (LHRHaH) and a fifth treatment consisted of 20 females selected based on apparent maturity as evidenced by external appearance were injected with CPE, 2 + 8 mg/kg without cannulation (CPEO). Two trials were conducted in early part of the spawning season and two trials in peak season. External methods to identify maturity correlated (r = 0.63) with that of “germinal vesicle position” method to identify the stage of maturity in females. Mean percent of females that ovulated, hatched, and fry produced per kg body weight did not differ (P > 0.05) among the five hormone treatments. The mean percent females that ovulated was higher (P < 0.05) for CPE‐induced females that were identified as “excellent” females based on external examination. The performance of cannulated females did not differ (P > 0.05) with that of non‐cannulated catfish. The mean egg quality of hormone‐induced females and percent of females ovulated in response to hormone treatment established a weak but significant linear relationship (Y = 3.85 + 0.008 × ovulation, r2 = 0.39, P < 0.05).  相似文献

Hormone Induced Spawning of Summer Flounder Paralichthys dentatus   总被引:2,自引:0,他引:2  
During their first year in captivity, summer flounder Paralicthys denratus were induced to spawn with gonadotropin releasing hormone analogue (GnRHa) implants, injected carp pituitary extract (CPE) or human chorionic gonadotropin (hCG) injections. The percentage of fertile eggs was greatest (69%) in CPE-treated females. CPE, but not GnRHa or hCG, was capable of stimulating oocyte growth (increased follicle diameter during vitellogenesis) followed by ovulation. Fish with maximum ovarian follicle diameters between 180 and 435 μm at the initiation of CPE injections produced the greatest percentage of fertile eggs. For most females, fertilization rate was greatest for the first batch of eggs ovulated. The mean fertilization rate for the first spawn of CPE-treated fish was 42% compared with 14% for the second spawn from the same fish. Fish with maximum initial follicle diameters of 585 40 μm that were implanted with GnRHa ovulated the greatest number of eggs, but fertility was low and variable. Approximately 35% of females injected with hCG ovulated a limited number of eggs, but only one hCG-treated female produced fertile eggs. Only a limited number of spermiating males were available for spawning trials. Hormone treatments used on females were ineffective for inducing or maintaining spermiation in male summer flounder.  相似文献

Interest in commercial production of black sea bass has increased in recent years, but reliable spawning methods remain problematic. The objective of this study was to evaluate the effects of oocyte size and luteinizing hormone releasing hormone analogue (LHRHa) dosage and delivery systems on ovulatory success for in vitro fertilization. Vitellogenic females with maximum oocyte diameters of 400–625 μm were implanted with a 95% cholesterol–5% cellulose pellet containing 50 μg of LHRHa. Fish with maximum oocyte diameters < 450 μm failed to ovulate. In contrast, 90% of fish with 500 μm oocytes spawned within 36 h and 40% of this group ovulated a second time. All of the females containing oocytes > 550 μm ovulated. In a second experiment, females with uniformly vitellogenic oocytes (> 500 μm) and implanted with 50 μg LHRHa ovulated substantial numbers of eggs (45,000–192,000 eggs/kg body weight (BW), but fertility was consistently low (0–15%). In a third experiment, 19 of 39 females receiving implants containing 6.3–23.6 μg LHRHa/kg BW during the spawning season ovulated, but fecundity (17,000–339,000 eggs/kg) and fertilization (0–98%) were highly variable. When fish were grouped by developmental index, calculated as the number of oocytes with diameters > 400 μm/total number of oocytes measured, there were no statistical differences among groups with respect to the number of spawns, fecundity or fertilization success. In a fourth experiment, 11 of 13 females with a clutch of fully vitellogenic oocytes that were injected with 20 or 100 μg/kg BW LHRHa ovulated between 1 and 2 times on consecutive days. Five of seven given an implant containing 12.5 μg LHRHa ovulated one or more times. Fish implanted with shams or injected with vehicle alone did not ovulate in any of the experiments. No differences were found in the number of spawns, fecundity or fertilization success from fish receiving different doses of injected or implanted LHRHa. Incubation of pooled eggs produced 155,000 larvae (60% hatch) and 95,000 one-gram juveniles. These results demonstrate that injected or implanted LHRHa is effective for inducing ovulation in black sea bass with maximum oocyte diameters > 500 μm.  相似文献

Induced spawning in bream, Abramis brama (L), was studied using acetone-dried common carp pituitary (CP) and bream pituitary (BP) with or without the addition of human chorionic gonadotrophin (hCG). The total dose administered to fish was of 5.0 mg kg?1 BP or 4.0 mg kg?1 CP with or without 2000-2200 IU hCG kg?1 for females and 2.5 mg kg?1 BP or 2.0 mg kg?1 CP with or without 1000–1100 IU HCG kg?1 for males. In all male treated groups 100% of spermiation was observed: in females the most effective method was a triple injection with hCG and carp pituitary, resulting in 79% of females ovulated (over 68% of eyed eggs). Biological quality of eggs, expressed as a percentage of eyed eggs, was negatively correlated with time elapsing between resolving (final) injection and ovulation. Spawning success, expressed as a value of Se (spawning effectiveness coefficient), was higher in fish treated with triple injection.  相似文献

Kutum Rutilus frisii kutum (Kamenskii, 1901), Cyprinidae is an endemic fish of the Caspian Sea. Iranian Fisheries Organization (Shilat) produce up to 200 million fry (1–2 g body weight (b.w.)) to restock the Caspian Sea population annually. Some of these fry are produced by spawning induction in broodfish by carp pituitary extract (CPE). The objective of this study was to assay the effectiveness of the gonadotropin releasing hormone analogue (d ‐Ala6, Pro9‐Net GnRH) alone or in combination with metoclopramide (MET), a dopamine antagonist, on the percentage of ovulated females, latency period, ovulation index and fertilization success. The following hormone treatments were tested: single injection of 2 mg kg?1 b.w. of CPE as a positive control, GnRHa alone 20 and 40 μg kg?1 b.w. and combination of GnRHa and MET as follows: 5 μg+2.5 mg, 10 μg+ 5 mg and 20 μg+10 mg kg?1 b.w. Negative control group was injected with 0.7% saline. The percentage of ovulated females, ovulation index and fertilization success were 90%, 71.3±1.24%, 68.4±2.3%, respectively, in the group treated with GnRHa+MET at a dose of 20 μg+10 mg kg?1 b.w. and were significantly higher than those in the positive control (60%, 64.5±0.23%, 69.1±4.5%) (P<0.05). However, the latency period in this group was longer than that in the positive control (P<0.05). Only 20% and 40% fish ovulated in groups that received 20 or 40 μg kg?1 b.w. GnRHa. We investigated the optimum term of 17α-methyltestosterone (MT) -treatment for behavioral test and changes of plasma sex steroids levels in MT-treated immature male masu salmon, Oncorhynchus masou parr. Behavioral responses of MT-treated fish to the urine of ovulated or immature females were observed in an experimental trough. After 5 min of behavioral tests, plasma sex steroids levels were measured by RIAs. MT-treated fish showed specific behavioral responses to the ovulated female urine. MT-treated fish without exposure to the urine showed no clear changes in plasma 17,20β-dihydroxy-4-pregnene-3-one (DHP). However, plasma DHP in MT-treated fish increased when they were exposed to the urine.  相似文献

The South Carolina Department of Natural Resources began a stock enhancement research program for red drum, Sciaenops ocellatus, as a possible management strategy for augmenting the availability of juveniles for recreational harvest in South Carolina estuaries. Between 1999 and 2011, approximately 6 million juveniles and 260 million larvae were stocked into the Charleston Harbor (CH) estuary, and proportions of stocked fish were found through standardized sampling as high as 49.6% within a subadult year‐class and up to 12% within the spatially separate spawning population. This study evaluated the genetic influences of stocking on the spawning population, using microsatellite loci, and examined any changes in genetic diversity metrics over the course of the stock enhancement program. Percent contribution of stocked fish among subadult and adult collections was compared against measures of genetic diversity. No statistically significant correlations were found among genetic diversity metrics, suggesting that responsible stocking of red drum as small juveniles has not altered the genetic diversity of adults spawning offshore the CH. Life‐history characteristics likely play a large role in mitigating changes in genetic diversity, despite high contributions of stocked fish in some years, because red drum exhibit a long adult life span and benefit from overlapping generations.  相似文献

This work investigated the action of neuropeptide Y (NPY) on thein vitro pituitary release of the maturing gonadotropic hormone (GtH) of the rainbow trout using a perifusion system employing trout balanced salt solution (pH 7.5) at 15°C and a 12.5 ml/h flow rate. In vitellogenic females a 20 minutes NPY application (10−7 M) induced a 20–30% decrease in GtH secretion. Removal of NPY was followed by a rebound in GTH secretion. On the contrary, in ovulated females, NPY (15 minutes, 10−7 M) directly stimulated GTH secretion. The greatest stimulation was obtained the day of ovulation where the stimulatory effect of NPY was similar to those induced by s.GnRH in the same conditions, reaching 400% of the basal GTH level. In vitellogenic females treated with 1-4-6 androstadien 3–7 dione, an inhibitor of aromatase activity, the pituitary response to NPY was similar to that obtained in ovulated females. Thus thein vitro action of NPY might depend on thein vivo steroidogenic environment.  相似文献

SUMMARY: Behavioral experiments concerning a releaser pheromone in the urine of female rainbow trout were performed using immature fish administered orally with 17α-methyltestosterone (MT) during the non-spawning season. The urine was collected by catheter. The frequency of entries of test fish was recorded in each channel scented by test and control solutions in a Y-maze trough. The behavior of both MT-treated and control fish demonstrated that they could not discriminate the differences between distilled and environmental water as control solutions. There was also no difference between MT-treated and control fish when distilled and environmental water were introduced. The MT-treated immature fish were attracted to the channel scented by ovulated female urine. Neither coelomic fluid nor the immature female urine had any effect on the behavioral responses of MT-treated fish, while immature control fish had no preference for the urine of ovulated females. These results suggest in rainbow trout that ovulated female urine contains a releaser pheromone to attract mature males, and that androgens are involved in the sensory mechanisms detecting the releaser pheromone in fish.  相似文献

Abstract. Observations of spawning by uninjured and injured Atlantic salmon, Salmo salar L., indicated that uninjured fish usually spawned in the outlets, or inlets, to pools and on coarse bottom sediments, whereas injured females spawned in different places. Frequently, the spawning sites lay in very shallow water with a slow current and on a bed of sand or fine gravel. Subsequent excavation of the spawning redds revealed that about 28% of the eggs laid by uninjured females were recovered, compared with only about 8% of those laid by injured females. Banded morwong (Cheilodactylus spectabilis) are of interest for marine finfish aquaculture in temperate southern Australia. To improve their ovulatory response, adult females were implanted during the autumn spawning season with slow‐release pellets containing 0–400 μg luteinizing‐hormone‐releasing hormone analogue (LHRHa)/kg body weight within 24 h of capture from the wild. Compared to the sham control group, animals treated with LHRHa produced significantly more eggs on each day after implantation for the following 7 d (91 ± 39 and 290 ± 38 mL) and a higher proportion ovulated (8/12 and 27/27). Of fish treated with LHRHa, 93% ovulated 2 d after implantation and 79% ovulated three times at 2‐d intervals, whereas control animals showed no cyclicity of ovulation and few ovulated more than once. Egg production was highest at the first ovulation after LHRHa treatment and declined at subsequent ovulations. In a second experiment investigating the range 100–400 μg LHRHa, there was no effect of dose rate on ovulation parameters, which additionally examined implantation either immediately after capture or after a 5‐d delay. Compared to immediate implantation, a delay resulted in a lower proportion of animals that could be stripped after implantation (100 and 50%, respectively) and the volume of eggs was lower (135 ± 15 and 107 ± 10 mL). The egg quality was poor following delayed implantation, resulting in no fertilization after artificial insemination compared with immediate implantation in which fertilization and hatch rates were higher for eggs collected on Day 2 after implantation (79 ± 8% and 58 ± 9%) than on Day 4 (23 ± 7% and 15 ± 6%). Thus, it is important to implant animals as soon as possible after capture to ensure optimum egg quality. Good‐quality eggs were buoyant and spherical and had a diameter of 1050 ± 25 μm with a single pigmented oil droplet of 190 ± 9 μm. When a separate large batch of eggs collected 2 d after implantation with 100 μg LHRHa was inseminated and cultured at 18 C, larvae hatched after 63 ± 2 h at a standard length of 2.6 ± 0.4 mm. Newly hatched larvae were buoyant and transparent with only a few melanophores, eyes were nonpigmented and jaws were nonfunctional. By the fourth day, jaws were functional and eyes were fully pigmented. Utilization of the endogenous yolk and oil was completed by Day 6, and swimming commenced with exogenous feeding. Larvae, initially fed lipid‐enriched rotifers followed by Artemia, reached 8.9 ± 0.7 mm length on Day 55, after which they metamorphosed to the postlarval paperfish stage of development, 22 ± 0.9 mm on Day 100, and 43 ± 1.0 mm at 6 mo of age. The results show that treatment of wild‐caught females with slow‐release pellets containing LHRHa is effective for the production of eggs for hatchery rearing.  相似文献

The effects of five dietary protein levels (25%, 30%, 35%, 40% and 45%) on the individual spawning frequency and the egg production of 135 tagged female Nile tilapia, Oreochromis niloticus (L.), were studied from 8 May to 19 November 1996 in outdoor concrete tanks. Virtually identical spawning patterns were found in all treatments, but there was a great deal of variation in the frequency of spawning and egg production. Overall, individual spawning frequency varied from one to 14 and individual egg production from 31 to 2828 eggs per spawning. The average number of spawnings and average number of eggs per spawning for fish receiving dietary protein levels of 25%, 30%, 35%, 40% and 45% were 8.0 ±1.6, 8.4 ± 2.2, 8.5 ± 2.7, 8.4 ± 2.5 and 9.4 ± 2.1 spawnings, and 1167 ± 305, 1082 ± 410, 1288 ± 324, 1145 ± 389 and 1328 ± 311 eggs per spawning, respectively. Fish receiving 45% dietary protein spawned more frequently than fish receiving 25% dietary protein. The total number of eggs produced per female was significantly higher for females fed 45% protein feed than females receiving 25% and 30% protein feeds. No definite trend was found in the number of eggs produced per spawning and the number of eggs produced per gram in females fed at different protein levels. Based on weekly checking, the time interval between spawnings varied from 7 to 77 days, and mean spawning intervals ranged from 15.8 to 17.1 days. Sixty per cent of females spawned after an interval 14 days, 15% after 21 days, 13.6% after 7 days, 7.2% after 28 days, 1.8% after 35 days and 1.0% after 42 days, and the time interval was 49-77 days for the rest of the females (1.5%). In all treatments, maximum spawning activity was recorded from May to August, and thereafter, it gradually decreased and no spawning females were found in November.  相似文献

Sarcoplasmic protein content decreased significantly and myosin heavy chain was degraded gradually during spawning migration of chum salmon (Oncorhynchus keta). Acid and neutral proteinase activities increased significantly during spawning migration. These proteinase activities were higher in females than in males. High levels of acid proteinase were not caused by the injury of lysosomal membranes. Considering the physiological states of chum salmon, neutral proteinase activity might be related to the degradation of muscle protein. As the changes in serum sex steroids were similar to those in the protease activities during spawning migration, it was considered that high levels of protease activity during spawning migration were closely related to the serum levels of androgens.  相似文献

An attempt was made to increase free amino acids (FAA) concentrations in eggs of red snapper Lutjanus campechanus as part of an induced spawning protocol. Mature female red snapper were given intramuscular injections of human chorionic gonadotropin (HCG) at a 1100 IU kg?1 of female body weight. Immediately after the HCG injection, one set of females received an intramuscular injection of FAA in a buffered saline solution at a dose of 20 mg FAA cocktail kg?1 of body weight. The FAA cocktail was 25% isoleucine, 25% leucine, 25% lysine and 25% valine on a molecular weight basis. A second set of fish did not receive the FAA supplement. Both sets of fish were equally successful in spawning with ovulation occurring 21–32.5 h post injection. Injection of FAA into brooders as eggs began final maturation and hydration resulted in an increase in leucine (nmoles mg egg?1) and a slight increase in isoleucine (nmoles egg?1) in recently ovulated eggs. FAA supplementation via brood injection altered FAA utilization rates by embryos and during larval development, resulted in a greater yolk sac and oil globule reserve in larvae as they approached the time of first feeding.  相似文献

Abstract. In turbot, Scophthalmus maximus (L.), daily examination of broodstock females should be a hatchery management practice in terms of fry output. Eggs expelled shortly after ovulation by systematic stripping of females five times a week had high viability (72%) compared to eggs collected twice a week (46%), while handling frequency did not have any effect on female fecundity (total number of ovulated eggs per kg of female). Increase of stripping frequency raised hatching rates and larva production respectively from 11% to 35% and from 20000 to 65000 larvae/kg of spawning female. The enhancement of egg quality and of the resulting larva production are discussed.  相似文献

This study was conducted to evaluate the effects of stocking density and photoperiod in increasing the reproductive performance of Oreochromis niloticus. In experiment 1, a change in stocking density (from 47.7 to 6.8 kg m?3) was performed, with groups of 48 females moved to single compartments. In experiment 2, 36 females experienced a 6L:18D photoperiod for 21 or 28 days (stocking density: 31.3 kg m?3) before being placed in individual compartments (stocking density of 6.9 kg m?3, 12L:12D photoperiod). The spawning rates in experiment 1 (32.2%) and in experiment 2 (21 days: 65.2%, 28 days: 36.1%) were significantly higher than those in the control (17.7%). On the other hand, hatching and swim‐up fry rates were significantly lower in experiment 2 – 21 days (41.3%, 40.4%) but not in experiment 1 (64.0%, 56.3%) compared with the control (67.0%, 62.3%). The spawning rate in experiment 2 – 21‐day treatment group was the highest, while the number of eggs collected per female was significantly lower than that in the control. In experiments 1 (20.8%) and 2 (21 days: 44.4%, 28 days: 19.4%), the daily spawning rates were the highest 3 days after maintaining females in single compartments. The current experiment demonstrates how specific stocking density changes can be used to induce higher spawning rates in Nile tilapia.  相似文献

The effect of increasing levels of dietary phospholipids (PL) on the ovarian development and reproductive performance of Chinese mitten crab Eriocheir sinensis were investigated using four semipurified formulated diets supplemented with 0%, 1.2%, 2.4%, and 3.6% PL. Four groups of 40 females, with an average individual body weight of 95–120 g, were fed the experimental diets for a period of 7 months. Male crabs were introduced into the female rearing system in March, and mating, spawning, and egg hatching occurred in the following month. After 10 weeks of feeding, females fed the diet with 2.4% PL had a significantly higher gonadosomatic index (GSI) than females fed the diet with 0% PL, whereas females fed the diet with 1.2% PL had a significantly higher hepatic moisture content and lower hepatic lipid content than the other groups (< 0.05). After mating, the spawning rate was higher among females fed diets with 3.6% and 2.4% PL (95% and 92%, respectively) compared with females fed diets with 0% and 1.2% PL (both 81%). The results showed that egg production (total number of eggs/female) and fecundity increased with increasing dietary PL level, with females fed diets with 0% and 1.2% PL supplementation having significantly lower values than females fed the diet with 3.6% PL (< 0.05). In conclusion, our results suggest that dietary PL supplementation has a positive effect on ovarian development and reproductive performance of E. sinensis broodstock. Further study should aim to investigate the optimal PL level in the broodstock diet of E. sinensis in respect to offspring quality.  相似文献
