The effects of 60‐mg L?1 clove oil and 60‐mg L?1 tricaine methanesulphonate (MS‐222) on the blood chemistry of rainbow trout were compared after exposure to handling stress via caudal puncture blood sampling. Fish sampled by caudal puncture and subsequently exposed to anaesthetics showed a typical handling stress response over a 48‐h period. There were no significant differences between the responses of fish exposed to equal concentrations of clove oil and MS‐222, with the following exceptions: the blood glucose at full anaesthesia, and lactate at full recovery increased significantly in the clove oil‐exposed fish. In a subsequent experiment, the stress response observed in fish sampled by caudal puncture and exposed to clove oil and MS‐222 was compared with a non‐anaesthetized control group. The increases in plasma cortisol levels were significantly lower at recovery in fish treated with either anaesthetic compared with the control fish. Fish exposed to MS‐222 had significantly higher cortisol levels at 1 h. These findings show that few differences exist between the anaesthetic effects of clove oil and MS‐222 on the physiological response of fish to stress. However, clove oil is more effective at reducing the short‐term stress response induced by handling and blood sampling, and is recommended as an alternative fish anaesthetic.  相似文献

We examined the effects tricaine methanesulfonate (MS-222), clove oil and CO2 on feed intake and cortisol response in steelhead trout, Oncorhynchus mykiss. Even though a body of literature exists about the effects of different anaesthetics on fish, no comparative information seems to be available about their effects on feed intake after anaesthesia, which would be important to know especially in aquaculture research. We anaesthetised juvenile steelhead trout with these three anaesthetics, and then sampled them 4, 24 and 48 h later. Fish in all groups ate relatively well already 4 h after anaesthesia. However, feed intake in fish treated with clove oil or MS-222 was lower than in the controls. There were no differences in feed intake among anaesthetised groups. Plasma cortisol concentrations were elevated 48 h after anaesthetisation, but the treatment means were equal throughout the experiment. Our results support previous findings that clove oil is a reasonable alternative to MS-222.  相似文献

Clove oil has been demonstrated to be an effective, inexpensive anaesthetic and euthanizing agent for a number of fish species, including rainbow trout, used in aquaculture and fisheries research. However, the potential for clove oil to cause perturbations in important plasma hormone concentrations has not been investigated. The effect of anaesthesia and euthanasia in trout with eugenol (the active ingredient in clove oil) on plasma cortisol, glucose, growth hormone (GH) and two thyroid hormones [tri‐iodothyronine (T3) and thyroxine (T4)] was compared with tricaine methanesulfonate (MS‐222) anaesthesia, and stunning by cranial concussion in two experiments. Effects on blood chemistry were different when comparing the particular anaesthetic method being used. Stunning fish significantly increased plasma cortisol and glucose levels (both P<0.05), while euthanizing fish using either clove oil or MS‐222 had no effect on these hormone levels. In contrast, the levels of GH, T3 and T4 hormones were unaffected regardless of whether fish were euthanized by stunning, MS‐222 or clove oil. Variation in effects between hormones were observed using clove oil eugenol. In fish sampled 10 min after anaesthetizing with 150 mg L?1 of eugenol, cortisol levels were significantly decreased (P<0.03), while there were no differences in either glucose or GH levels. Tri‐iodothyronine and T4 also showed significantly elevated levels (P<0.05) after 10‐min exposure to eugenol. These results highlight the importance of investigating the potential effects of any new anaesthetic or euthanizing compounds on blood plasma parameters, prior to using them in a research setting, or when comparing results to other studies which have utilized alternative anaesthetic compounds.  相似文献

Two anaesthetics, clove oil and methane sulphonate (MS-222), were examined for their effects on the olfactory nerve response of masu salmon ( Oncorhynchus masou Brevoort) and rainbow trout ( O. mykiss Walbaum). Exposing both species to clove oil for 3 min at concentrations of 50 and 100 mg L−1, or for 10 min at 50 mg L−1, did not significantly reduce their olfactory response. Directly applying clove oil anaesthesia to the olfactory epithelium significantly reduced olfactory response though after 20 min, olfactory response recovered to 70% and 52% of pre-treatment levels in masu salmon and rainbow trout respectively. Compared with the post-anaesthetic recovery of responses after clove oil (50 mg L−1), buffered MS-222 (100 mg L−1) with NaHCO3 (100 mg L−1), and unbuffered MS-222 (100 mg L−1) treatment for 3 min, the response after MS-222 treatment declined gradually and significantly, but not after clove oil and MS-222+NaHCO3 treatments. Clove oil appears to be an effective and relatively safe anaesthetic for salmonids with little long-term impact on their olfactory response, which plays a crucial role in their life history.  相似文献

The present study determined the effective concentrations of clove oil and MS‐222 in juvenile rohu Labeo rohita for quick induction and recovery. The immune‐biochemical responses due to 0, 1 and 24 hr exposure to those anaesthetics were also evaluated. Of four concentrations of the anaesthetics examined, the lowest effective concentration of clove oil and MS‐222 were 50 µl/L and 125 mg/L respectively. Clove oil and MS‐222 significantly increased the myeloperoxidase, total protein and alkaline phosphatase activity at some of the holding durations. However, superoxide anion production (after 0 and 1 hr) and antiprotease activity (after 24 hr) were significantly reduced in fish exposed to clove oil. Serum glucose content was significantly elevated in the MS‐222‐treated group. Furthermore, the clove oil‐treated group showed significantly higher levels of serum Na+ and K+, while the aspartate and alanine aminotransferase activities were significantly enhanced in the MS‐222 group. The use of both clove oil and MS‐222 is advised as an anaesthetic agent for rohu with a bias towards clove oil, considering its economic and operational feasibility.  相似文献

Clove oil has been tested for anaesthesia induction and recovery time as well as for haematology and stress indicators in the gilthead sea bream and rainbow trout. The former parameters were compared with those generated by 2‐phenoxyethanol. The results showed only slight differences between both anaesthetics in terms of anaesthetic efficiency and physiological effects. In addition, clove oil does not block the cortisol response to stress, as happens with other anaesthetics.  相似文献

Juvenile and adult black sea bass (Centropristis striata L.) were exposed to various concentrations of four anaesthetics to determine practical dosages for handling as well as for procedures such as bleeding, ovarian biopsy or tag implantation. In experiment 1, juveniles exposed to either 2.0 mg L?1 metomidate, 15 mg L?1 clove oil, 70 mg L?1 tricaine methanesulphonate (TMS) or 200 mg L?1 2‐phenoxyethanol (2‐PE) reached stage II of anaesthesia in 3–5 min and could be handled for weighing and measuring. All fish had completed recovery to stage III within 6 min. In experiment 2, the established concentrations of each anaesthetic were tested on juveniles to determine their ability to prevent a reflex to a subcutaneous needle puncture. All of the fish exposed to clove oil (20 mg L?1) and 40% of the TMS‐treated (70 mg L?1) fish reacted while none of the fish anaesthetized in metomidate (2.0 mg L?1) or 2‐PE (200 mg L?1) responded to the needle puncture. In experiment 3, metomidate (5.0 mg L?1), clove oil (30 mg L?1) TMS (125 mg L?1) or 2‐PE (300 mg L?1) were all effective for performing an ovarian biopsy or tag implantation on adults. In experiment 4, TMS (125 mg L?1) exacerbated the cortisol response to a short handling stressor during a 30 min exposure. Fish anaesthetized in 2‐PE (300 mg L?1), metomidate (5.0 mg L?1) or clove oil (40 mg L?1) had increased cortisol levels associated with the handling stressor but there were no further increases during the remainder of the experimental period. The results demonstrate that these anaesthetics are effective for sedation and anaesthesia of black sea bass and that the best choice is dependant upon the procedures to be performed.  相似文献

This study investigated the feasibility of using clove oil, 2-phenoxyethanol, or Propiscin as an alternative to tricaine methane sulphonate (MS 222) as a fish anaesthetic, particularly in regard to reducing fish stress. The biochemical blood profiles of perch Perca fluviatilis L. anaesthetized with either MS 222 (100 mg L−1), clove oil (33 mg L−1), 2-phenoxyethanol (0.40 mL L−1) or Propiscin (1.0 mL L−1), and a non-anaesthetized control group were compared. Biochemical profiles were determined from blood samples collected before treatment in controls. For each anaesthetic tested, fish were divided into two groups, one sampled immediately after 10-min anaesthesia and a second, sampled 24 h after 10-min anaesthesia. The values determined in the present study suggested that internal organs and tissues of perch were slightly altered by MS 222, clove oil and 2-phenoxyethanol anaesthesia, but not by Propiscin anaesthesia.  相似文献

The effects of different stunning/killing procedures on flesh quality of European sea bass were investigated: (1) anaesthesia with clove oil, (2) anaesthesia with 2‐phenoxyethanol, (3) percussive stunning, (4) immersion in ice/water slurry, (5) chilling on ice and (6) anaesthesia with clove oil followed by immersion in ice/water slurry. Muscle pH values were significantly lower in sea bass anaesthetized or stunned by a blow to the head compared with fish immersed in ice/water slurry, chilled on ice or immersed in ice/water slurry after clove oil anaesthesia. Lightness was highest in sea bass anaesthetized by 2‐phenoxyethanol or percussively stunned and lowest in ice‐chilled fish. Redness and yellowness were highest in fish chilled on ice and lowest in fish anaesthetized with clove oil. Liquid loss, fat loss and shear values were not significantly different among the procedures. In general, lipid oxidation values during refrigerated or frozen storage did not significantly differ among treatment groups. Combination of clove oil anaesthesia followed by chilling on ice/water slurry appears to improve both flesh quality and welfare of sea bass, although the implementation of further studies is warranted to consolidate this finding.  相似文献

The efficacy of the anaesthetic agents benzocaine, metacaine (MS‐222), metomidate, 2‐phenoxyethanol, quinaldine and isoeugenol was studied in Atlantic halibut (Hippoglossus hippoglossus). Fish with an average body weight of 33 g were anaesthetized at 8 °C and fish with an average body weight of 1243 g were anaesthetized at 8 and 15 °C. Agents were tested individually and as combination anaesthesia comprising pre‐anaesthetic sedation, followed by anaesthesia. Induction and recovery times varied in relation to the body weight and water temperature. Large fish had longer induction times and shorter recovery times, and displayed reduced responsiveness to handling compared with small fish. A higher temperature resulted in shorter induction times, longer recovery times and increased responsiveness to handling. Lower dosages were used for all agents in combination anaesthesia. In small fish, this had no effect on the induction times but resulted in shorter recovery times and reduced responsiveness to handling. In large fish, combination anaesthesia resulted in shorter induction times whereas no uniform trend in recovery times and no differences in responsiveness to handling were observed. Neither individual agents nor combinations blocked all reflex reactions to external stimulation in all fish of any treatment group. MS‐222 and benzocaine, used separately or in combination anaesthesia, were the most effective agents in reducing reflex reactions.  相似文献

Anaesthetics are used in aquaculture and fisheries to facilitate routine procedures, such as capture, handling, transportation, tagging, grading and measurements that can often cause injury or induce physiological stress. Two experiments were performed to assess the efficacies of four anaesthetic agents, clove oil, benzocaine, 2‐phenoxyethanol and MS‐222 on juvenile marbled spinefoot rabbitfish (Siganus rivulatus). In the first experiment we tested the lowest effective doses that produced induction and recovery times in 3 min or less and 5 min or less respectively. Dosages were 70 mg L?1 for clove oil, 60–70 mg L?1 for benzocaine, 400 μL L?1 for 2‐phenoxyethanol and 100–125 mg L?1 for MS‐222. In the second experiment, we determined optimal concentrations of the four anaesthetics if they were to be used to transport rabbitfish fry. Anaesthetic concentrations suitable for handling and transport were: 10–15 mg L?1 of MS‐222, 5–10 mg L?1 of benzocaine, 5 mg L?1 of clove oil and 50–100 μL L?1 of 2‐phenoxyethanol. All anaesthetic agents are acceptable for use on S. rivulatus, however, 2‐phenoxyethanol, MS‐222 and clove oil appear to be more suitable than benzocaine. Further studies need to be conducted on effects of high and low doses of anaesthetic agents on physiology of marbled spinefoot.  相似文献

The present study evaluated the nutritional value of sesame oil cake (SOC) in rainbow trout fry (initial body weight of 1.42 g) in a growth trial performed for 45 feeding days at 15 ± 1 °C. A series of five isonitrogenous (380 g digestible protein kg?1 dry matter (DM)) and isoenergetic (18.1 MJ digestible energy kg?1 DM) diets were formulated in which the digestible SOC protein progressively replaced 0%, 13%, 26%, 39% and 52% of the digestible protein of a high quality fish meal (D0‐D52). Growth rate of fry significantly improved in fish fed SOC diets compared to the fishmeal control diet (D0) whatever the SOC inclusion level. This positive effect on growth was mainly related to a marked improvement of voluntary feed intake. The decrease of feed efficiency observed with increasing SOC was entirely explained by the reduction of DM and energy digestibilities with SOC incorporation. Nitrogen retention efficiency (nitrogen gain/nitrogen intake) was high (40–41%) and significantly reduced only in fish fed D52 (37%). Our results suggest that SOC can be a suitable protein source for a carnivorous fish and replace at least half of the fishmeal protein (without amino acid supplementation) without growth reduction in rainbow trout fry.  相似文献

Clove oil is an effective, local and natural anaesthetic. Many hatcheries and research studies use clove oil to immobilize fish for handling, sorting, tagging, artificial reproduction procedures and surgery and to suppress sensory systems during invasive procedures. Clove oil may be more appropriate for use in commercial aquaculture situations. Improper clove oil use can decrease fish viability, distort physiological data or result in mortalities. Because animals may be anaesthetized by unskilled labourers and released in natural water bodies, training in the proper use of clove oil may decrease variability in recovery and experimental results and increase fish survival. Here, we briefly describe many aspects of clove oil, including the legal uses of it, anaesthesia mechanism and what is currently known about the preparation and behavioural and pathologic effects of the anaesthetic. We outline methods and precautions for administration and changes in fish behaviour during progressively deeper anaesthesia and discuss the physiological effects of clove oil, its potential for compromising fish health and effectiveness of water quality parameters.  相似文献

Abstract. Juvenile rainbow trout (initial mean body weight 17 g) reared on practical trout diets containing different levels of available carbohydrate for 8–24 weeks were assayed for liver function based on the time required to recover from the anaesthetics tricaine methanesulphonate (MS222), 2-phenoxyethanol (2PE) and tertiary-amyl alcohol (TAA), as well as plasma clearance of 35S-sulphobromo-phthalein (BSP), Liver/body weight ratios (LBW) and percentage liver glycogen content (LG) increased in a linear manner with increased available carbohydrate. The mean recovery time (MRT) of trout anaesthetized with 2PE and TAA increased linearly with increased LBW and LG. No difference was observed in the MRT of trout anaesthetized with MS222 which is not metabolized in the trout liver. There was a significant linear regression of the biological half-life of plasma BSP on LBW (MR2= 95.7) and LG (MR2= 99.6). The results indicate that increased liver glycogen and liver weight due to higher levels of available carbohydrate affect the liver function of rainbow trout. These liver function tests may be applicable in determining acceptable levels of available carbohydrate in practical trout diets.  相似文献

The effects of protein source and nutrient density on growth efficiency, nutrient digestibility and plasma amino acid concentrations of rainbow trout were evaluated. A 3 by 2 factorial treatment design with three protein sources, fish meal–barley (F–B), plant concentrates (PC) and plant meals (PM), and two nutrient densities were used. A commercial reference diet was also fed. Triplicate tanks of 30 fish (initial wt. 28 g) were fed each diet, and the final weight averaged 240 g fish−1. Protein source and nutrient density affected feed intake, weight gain and feed conversion ratio. Weight gain of trout fed the PC and PM diets was approximately 10% less than fish fed the F–B diets. Protein retention was affected by protein source, but not nutrient density, and was the highest for the fish fed diets containing fish meal and the lowest for the fish fed PM diets. Apparent digestibility coefficients and apparent amino acid availabilities of the diets corresponded with differences in weight gain. This study provides further evidence that growth rates of trout fed fish meal‐free diets, using conventional and concentrated plant protein ingredients, are good but some limitation to growth exists in the fish meal‐free diets.  相似文献

Present study examined the effects of Ergosan on growth performance, digestive enzyme activities, hematological parameters and gastrointestinal structure of rainbow trout. Rainbow trout (mean weight 100–110 g) were fed basal diet (control) and diet treated with Aquavac Ergosan (5 g kg?1 of diet) for 50 days. Results of this study showed that Ergosan supplementation significantly increased weight gain (94.27 g vs. 65.04 g), specific growth rate (4.09 vs. 3.10) and feed intake (136.85 g vs. 111.22 g) and decreased feed conversion ratio (1.43 vs. 2.03) compared to control (P < 0.05). Lipase activity and leukocyte and erythrocyte count also increased in juvenile fish fed Ergosan-treated diet compared to control (P < 0.05). Light microscopy demonstrated that both groups of fish displayed normal morphology of proximal intestine and pyloric caeca. In Ergosan-treated group, higher percentage of goblet cell was shown in proximal intestine and pyloric caeca. Present study suggests that Ergosan effectively promotes growth performance, lipase activity and gastrointestinal structure in rainbow trout.  相似文献

This experiment intended to assess the effect of sesame (SO), sunflower (SFO) and linseed (LO) oils on growth performance, fatty acid composition of fillet and liver or flesh quality traits of rainbow trout. Fish fed different four iso‐nitrogenous and iso‐lipidic experimental feeds. The control feed contained only fish oil as the primary lipid source. The fillet eicosapentaenoic acid and docosahexaenoic acid levels were the highest in fish fed control feed. In contrast, the liver eicosapentaenoic acid level was the highest in fish fed LO feed. Fish fed SFO feed had the highest level of total n?6 fatty acids in fillet and liver. Fish fed SO feed had the highest level of 18:1 n?9 fatty acid in fillet and liver. During the 12 days refrigerated storage period at 1°C, thiobarbituric acid (TBA) and pH values gradually increased in all dietary groups. However, trimethylamin nitrogen (TMA‐N) values increased in all dietary groups between days 0 and 9 during the storage period. Generally, pH value in fillets of control group was slightly higher than the other fish groups during 12 days refrigerated storage. Nevertheless, the chemical indicators of spoilage, TBA, TMA‐N and pH values were in the limit of acceptability for human consumption. Results of growth performance and chemical tests in the present study showed that sesame, linseed and sunflower oils could be used in feeds for rainbow trout. Moreover, we concluded that further researches should be carried out on the partial replacement of fish oil by sesame oil in rainbow trout nutrition.  相似文献

A feeding trial was conducted to investigate the effects of the replacement of dietary fish meal and fish oil with oilseed meals (soybean or canola) and canola oil on growth, nutrient utilization, body composition, diet digestibility and hematological parameters of rainbow trout Oncorhynchus mykiss. Seven diets were used; the control diet (designated FM) contained fish meal and fish oil as the main protein and lipid sources. For the experimental diets, 40% of fish meal protein was substituted with soybean meal, canola meal or a soybean/canola meal mixture, and these diets (designated SM, CM and SCM, respectively) contained fish oil as the lipid source. Three additional diets (SM?+?CO, CM?+?CO and SCM?+?CO) were formulated with the same vegetable protein meals but with fish oil replaced by canola oil. Fish were fed twice daily to apparent satiation for 11?weeks. The growth of fish fed the CM?+?CO diet was significantly lower than that of fish fed the FM, SCM, SM?+?CO and SCM?+?CO diets. The feed conversion ratio (FCR) was significantly better in fish fed the FM and SCM?+?CO diets than in fish fed the CM and CM?+?CO diets. Furthermore, feed intake was significantly lower for fish fed the CM?+?CO diet than in fish fed the SCM diet, and lipid digestibility of the CM?+?CO diet was significantly lower than that of all other diets. No significant differences of body composition were observed. Circulating leukocyte levels, leukocyte ratios and serum lysozyme activity remained unaffected by dietary treatment. However, it was observed that fish fed the CM?+?CO diet displayed hematocrit levels significantly lower (P?<?0.05) than that of fish fed the other diets. The results indicate that when diets contain either fish oil or canola oil, canola meal and soybean meal can be incorporated into rainbow trout feeds at a combined 32% inclusion level (replacing 40% of fish meal protein) without inducing significant negative effects on growth, nutrient utilization or health.  相似文献

This study investigated the feasibility of using clove oil as an alternative to tricaine methanesulphonate as a fish anaesthetic, particularly in fish stress research. The physiological stress responses of juvenile chinook salmon Oncorhynchus tshawytscha (Walbaum) anaesthetized with either tricaine (50 mg L?1) or clove oil (20 p.p.m.) were compared using unanaesthetized fish as controls. Haematocrit, serum cortisol and serum glucose concentrations, serum lysozyme activity and differential leucocyte counts were measured from blood samples collected before, during and upon recovery from anaesthesia and at specified intervals up to 72 h after recovery. Differences between the two anaesthetic groups were not significant for most of the physiological traits measured. Serum lysozyme activity of control fish, however, was significantly suppressed relative to the treated fish for 72 h after stress. Clove oil may be a safe and cost‐effective alternative to tricaine without significantly affecting study results. Furthermore, clove oil may be more practical for field‐based research, because a withdrawal period is unnecessary, and clove oil does not pose an environmental hazard.  相似文献

This study examined the effects of Hilyses?, fermented Saccharomyces cerevisiae (Scerevisiae), on rainbow trout growth performance, haematological parameters, digestive enzyme activities and gastrointestinal structure. Rainbow trout (mean weight 100–110 g) were fed dietary Hilyses? (5 g kg?1) and control diet without Hilyses? for 50 days. Results of this study demonstrated that yeast supplementation in treatment group resulted in increased feed intake, followed by improved feed conversion ratio (FCR) and growth performance. Significant increases were also observed in trypsin and amylase activities in juvenile fish fed treatment diet. Light microscopy demonstrated that both groups of fish displayed normal morphology of proximal intestine and pyloric caeca. In yeast‐treated group, higher density of the goblet cells per villus in the proximal intestine was shown. No effects on haematological parameters and carcass chemical composition were noted. It is therefore possible to use fermented Scerevisiae supplementation to significantly improve the gastrointestinal structure and growth performance in rainbow trout.  相似文献
