Five diets were compared for their efficiency at maturing the gonads of the blue mussel, Mytilus edulis. The diets consisted of a 1:1:1 mixture of Isochrysis galbana (T-Iso), Pavlova lutheri, and Chaetoceros calcitrans given at concentrations of 2.4 × 1011 cells day?1 for the positive control treatment (PF) and 3.0 × 1010 cells day?1 (=1/8) for the negative control treatment (NF). The other three treatments, MB10+, MyStock+, and Frippak+, consisted of the NF diet supplemented with one of the micro-encapsulated diets MB10 (mixture of dried algae), MyStock (formulated diet), and FRiPPAK® Fresh #1 CAR (larval shrimp diet) at a level of 0.2% of the live weight (LW). Treatments PF, MB10+, and MyStock+ led to high percentages of spawning animals (80, 78, and 85%, respectively) and large numbers of eggs (on average 3.0 × 106 eggs female?1). Females given the NF and Frippak+ treatments produced only half the number of eggs per female, and only 17 and 6%, respectively, of the animals spawned. A high hatching rate was observed for all treatments, 71% for the pure algae diets PF and NF and more than 80% for the micro-encapsulated diets. The larvae resulting from the NF treatment were smaller, with 41% of D-larvae measuring less than 90 μm, whereas with the other treatments only 5–11% belonged to that size category. The four most important fatty acids found in mussel eggs were 16:0, 16:1(n?7), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA); these represented at least 50% of the fatty acids in all treatments. The high DHA content of MB10 and MyStock was not reflected in the fatty acid composition of the eggs whereas the high concentration of linoleic acid in MyStock was. To our knowledge, this is the first time that mussels are successfully conditioned with formulated feeds.  相似文献

The results of the study on the suitable algal feed for the mass rearing of holothurian larvae through hatchery system are presented. Auricularia larvae, after 48 h of fertilization, obtained from induced spawning of Holothuria spinifera, were fed with different algae Isochrysis galbana, Nanochloropsis salina, Pavlova lutheri, Tetraselmis chuii and Chaetoceros calcitrans as well as I. galbana+C. calcitrans to ascertain the effect of single and combined microalgal diet. The rate of feeding was 2 × 104 cells larvae–1 day–1 for a period of 9–12 days. The growth rate of 59 μm day−1 with 90% and 43 μm day−1 with 100% occurrence of late auricularia in the larvae fed with C. calcitrans alone and I. galbana+C. calcitrans, respectively, indicated that C.calcitrans itself or in combination with I. galbana is the effective feed for the larvae of H. spinifera.  相似文献

The aim of this study was to determine the effects of dietary probiotics on reproductive performance, growth, haematological and biochemical parameters of female rainbow trout broodstock. A total of 60 broodstocks with an average initial weight of 2,267.4 ± 54.9 g were divided into four groups and three replicates in raceway ponds. Fish were fed with diets containing 0 (control), 1 × 109 (P1), 2 × 109 (P2) and 4 × 109 (P3) CFU probiotic per kg diet twice a day for 8 weeks before spawning season. Results revealed that higher levels of dietary probiotics (P3) enhanced protein efficiency ratio and decreased feed conversion ratio (p < .05). There were no significant differences in haematological and other growth parameters among different treatments. According to the results, probiotic‐fed treatments had the lowest levels of cholesterol (237.7 ± 4.3 mg/dl) and the highest levels of total protein (11.3 ± 0.3 g/dl) and albumin (6.9 ± 0.3 g/dl), so that differences were found between P3 and control groups in all cases (p < .05). Based on the observations from reproductive parameters, the highest egg diameter, working and relative fecundities, fertilization and hatching rates, eyed eggs survival and alevins survival rates up to the absorption of the yolk sac were observed in P3 treatment (p < .05). Moreover, eyeing, hatching and yolk sac absorbing stages in P3 treatment were earlier than those in other treatments (p < .05). The results of present study demonstrated that probiotic supplementation improved reproductive performance and some growth and biochemical parameters in female rainbow trout broodstock and the best performance was observed in fish fed 4 × 109 CFU probiotic per kg diet. Due to an increase in egg production and egg size in probiotic‐fed treatments, this feed additive can be used as a suitable ingredient to increase high‐quality egg production in rainbow trout breeders.  相似文献

Studies considering broodstock conditioning of Ruditapes decussatus mostly used flagellates and diatoms for feeding, mainly Isochrysis galbana and Chaetoceros calcitrans, respectively. The present study evaluated the effect of different microalgal diets on gonadic maturation of R. decussatus including three microalgae species (C. calcitrans, Tetraselmis suecica, and Nannochloropsis oculata) which were used to prepare six diets: three monospecific and three bispecific mixed diets at a proportion of 1:1. Clams were daily fed at a ratio of 1% dry weight of algae/live weight of clam and unfed treatment was used as control. Results showed that clams fed C. calcitrans alone or mixed with T. suecica attained earlier full maturity with the highest response to spawning induction (after 34 conditioning days) and largest proportion in the partial spawning phase for both, followed by T. suecica alone after 48 days. In contrast, N. oculata alone or mixed diets demonstrated poor performance for gonadal maturation. Unfed treatment did not show any sign of maturation. The results indicated that T. suecica has high conditioning potential like C. calcitrans and their mixture gave the highest benefit. Therefore, T. suecica can be recommended for R. decussatus broodstock conditioning.  相似文献

The Portuguese oyster Crassostrea angulata shows great potential in oyster farming. The conservation of pure populations of this species is important for production diversification and biodiversity preservation. In this way, the zootechnological development for seed hatchery production is extremely important. Broodstock conditioning is a key step in the process of rearing bivalves in a hatchery. Many factors regulate the reproductive cycle, being food one of the most important ones. To evaluate the effect of different diets on C. angulata reproductive performance, broodstock were conditioned with different food regimes formulated fundamentally by flagellates (Diet 1 – Pavlova lutheri and Isochrysis galbana clone T‐ISO; Diet 2 – P. lutheri, T‐ISO and Skeletonema costatum) and constituted fundamentally by diatoms (Diet 3 – S. costatum and Chaetoceros calcitrans; Diet 4 – P. lutheri, S. costatum and C. calcitrans). During conditioning, samples of oysters were collected to evaluate condition index, gonadal development and biochemical composition. At the end of the conditioning period, oysters were induced to spawn to evaluate reproductive output (fecundity, fertilization rate and D‐larvae development). The diets had an impact on the gametogenesis process, energy storage and reproductive output performance, being the best results those obtained in broodstock fed with the diatoms‐predominant diets. However, those fed with diets majority flagellates had an unsuccessful performance. Holistic approaches incorporating all results in this study reveal and reinforce the idea that the diatom species used presented the nutritional requirements to C. angulata broodstock, being essential in the conditioning phase.  相似文献

This study aimed to examine the use of Streptococcus agalactiae polyvalent vaccine in tilapia broodstock and the effect of maternal immunity and resistance on their offspring against S. agalactiae strain. The broodstock was injected with polyvalent vaccine of S. agalactiae at a dose of 108 CFU per fish at 2nd gonadal maturity until spawning. Challenge test was carried out on the offspring at the 5, 10, 15 and 20 days after hatching using NK1, N17O, N14G, N3M, N4M strain respectively and combination of them. We observed immunological parameters in broodstock, eggs and larvae and relative per cent survival (RPS) of larvae after challenged with pathogenic S. agalactiae. The results showed that the leukocytes, phagocytic activity, respiratory burst, lysozyme activity and antibody levels of vaccinated broodstock had higher level compared with unvaccinated broodstock. The high level of the lysozyme activity, antibody levels and recombination activating gene 1 (RAG1) were also observed in eggs and larvae from vaccinated broodstock. Larvae produced from vaccinated broodstock when challenged with variety strain of pathogenic S. agalactiae had RPS value more than 50% until 20 days after hatching. In conclusion, polyvalent vaccine of S. agalactiae administrated in the broodstock could enhance immunity in the broodstock and protect their offspring from pathogenic S. agalactiae.  相似文献

The Prymnesiophycea Pavlova lutheriwas produced using a traditional technique (batch mode in carboys) and by an innovative technique (semi-continuous mode in alveolarphotobioreactor). Two experimental feedingtrials were carried out with Pacific oyster(Crassostrea gigas) larvae with freshand preserved (4 °C and 1 °C)P. lutheri concentrates used inmonospecific and bispecific diets, incombination with fresh Chaetoceroscalcitrans forma pumilum. Nodifferences in terms of gross composition andlarval performance were noticed between thefresh algae biomass produced by the twotechniques. The low temperatures allowedpreservation of the nutritionalcharacteristics of the P. lutheri freshculture for the time period considered (27days). The P. lutheri concentratespreserved at 4 °C or 1 °C didnot differ substantially in terms ofnutritional value. Larvae fed a monospecificdiet of P. lutheri, both fresh andpreserved, exhibited a modest growth rate (<2.1 µm day–1), in some cases notsignificantly different from that observed inthe unfed larvae (the control), and a lowsurvival rate. In contrast, when Chaetoceros calcitrans forma pumilumwas used as a monospecific diet, good growthperformance (>4.5 µm day–1) and ahigh survival rate (>86%) were observed inlarvae. A substitution of 50% (trial 1) or80% (trial 2) of fresh C. calcitransforma pumilum with the preserved P. lutheri concentrates did not adverselyaffect growth rate or survival of C.gigas larvae.  相似文献

A study was conducted to determine the effect of essential fatty acid in striped catfish (Pangasianodon hypophthalmus) broodstock diets on a level of oestradiol‐17β and vitellogenin, reproductive performance and larval quality in out‐of‐spawning season. Five female and five male broodstock were stocked into a net cage (3 × 5 × 1.5 m3) and fed with pelleted diets containing 1.5% fish oil and different amounts of corn oil (CO) in diet formulation, that is 0%, 1%, 2% and 3% respectively. Gonadal development, oestradiol‐17ß and vitellogenin concentrations were examined every 2 weeks. Oestradiol‐17ß and vitellogenin concentrations showed an increase during the maturation process. The significantly highest percentage of female mature, fecundity, hatching rate, larval production and larval survival rate were obtained from fish fed with 2% corn oil in the diet. This result revealed that the administration of 2% corn oil in the diet as a source of n‐6 (linoleic acid) and 1.5% fish oil as a source of n‐3 (linolenic acid) can improve the reproductive performance of striped catfish broodstock in out‐of‐spawning season.  相似文献

A 45‐day trial was performed to evaluate the effect of biofloc technology (BFT) with or without fresh food (FF) supplementation during pre‐maturation period on Farfantepenaeus duorarum spawning performance, biochemical composition and fatty acid profile of eggs as compared with conventional clear‐water system (CW+FF). Females raised in biofloc and that received FF supplementation (FLOC+FF) achieved better spawning performance in terms of number of eggs per spawn (49 × 103), number of eggs per spawn per g of spawner's body weight (2.1 × 103) and egg size (~275 μm) as compared with CW+FF (23 × 103, 1.1 × 103 and 263 μm respectively), but both treatments did not vary from FLOC (P > 0.05). High spawning activity was also observed in biofloc system as compared with clear‐water system as shown in number of spawns per ablated female (2.2–3.0 versus 0.6) and percentage of females that spawn at least once (80–82 versus 25%). Biochemical composition of eggs presented no significant differences among treatments. FA profile of eggs indicated that high spawning activity performed by females in FLOC+FF treatment was reflected in lower mean levels of EPA, DHA and sum of polyunsaturated fatty acids (n‐3) and (n‐6). The better reproductive performance demonstrated by females raised in biofloc justified the application of this technology in F. duorarum broodstock.  相似文献

The effects of mono‐species and bi‐species microalgal diets on body length, survival and metamorphosis of auricularia larvae of the California sea cucumber, Parastichopus californicus, were tested in two separate laboratory feeding experiments. The first examined eight single species of microalgae – Chaetoceros calcitrans (Cc), Chaetoceros muelleri (Cm), Dunaliella tertiolecta (Dt), Isochrysis sp. (Tahitian strain: TISO) (Ti), Pavlova lutheri (Pl), Phaeodactylum tricornutum (Pt), Tetraselmis suecica (Ts) and Thallassiosira pseudonana (Tp). The second experiment took the five most effective single species from the first experiment (i.e. Cc, Cm, Dt, Pl and Ti) and presented them in all possible binary combinations. In the first experiment, Cc was determined to be the best diet on the basis of generating a significantly higher metamorphic rate (65.5 ± 4.2%, mean ± SD) than any other species and leading to one of the best survival rates (72.3 ± 7.6%) and greatest body lengths (831.6 ± 5.6 μm at day 14). In the second experiment, diets containing Cc generally produced the highest survival and metamorphic rates and greatest body lengths (except Cc/Cm) of all mixed diets, although no bi‐species treatment was ever significantly superior to a control of Cc alone. Of the various mono‐species and bi‐species diets evaluated for rearing auricularia larvae of P. californicus, Cc (either alone or mixed with other species) is recommended.  相似文献

The present study was conducted to define a methodology to produce and store small‐scale microalgae paste to be used in a mollusk hatchery. Microalgae were cultured in 500 L fiberglass tanks, under temperature of 20 ± 2 C, Guillard f/2 culture medium, and continuous light intensity of 203–226 μmol photons/m2/sec. Cultures were centrifuged at 2000 g at the exponential growth phase. Microalgae cell quality after centrifugation and during storage was determined by analyses with Evan’s blue stain and by counting the number of total marine bacteria. Treatments with and without additive were applied to the microalgae paste produced, which was distributed into 100 mL plastic containers, capped, and stored under refrigeration at 4 ± 1 C. Results indicated that in the Chaetoceros muelleri paste, centrifugation did not damage the cells and the number of total marine bacteria reduced significantly from 2.9 × 106 to 8.3 × 105 colony‐forming units per milliliter. Chaetoceros muelleri and Chaetoceros calcitrans pastes stored with addition of 0.1% ascorbic acid had a shelf life shorter than 2 wk. For the treatment without additive, results with Evan’s blue stain showed that cells (99%) remained viable until the sixth week of storage for C. muelleri and seventh week of storage for Skeletonema sp. and C. calcitrans. The number of bacteria did not increase during storage for C. calcitrans and Skeletonema (P > 0.05). For C. muelleri, an increase in bacteria (P < 0.05) was observed after the sixth week of storage. This study demonstrated the feasibility to produce and store microalgae paste for a period of 2–8 wk, which allows it to be used as food source and also optimizes the use of microalgae cultured in laboratory.  相似文献

The present study was conducted to evaluate the effect of different stocking densities on the seed production of Nile tilapia, Oreochromis niloticus (L.), under intensive recycling hatchery system conditions. Males and females with mean body weights of 163.2 and 105.0 g, respectively, were stocked at three broodstock densities (4, 8 and 12 fish m–2) at a male:female ratio of 1:3 in 1 × 1 × 0.43 m (W × L × H) fibreglass tanks. The tanks were illuminated at 2500 lux for 18 h day–1 and the water temperature was maintained at 29 ± 1 °C. Effluent from spawning tanks was recycled through a biological filter with 10–15% replacement of new water per day. The experiment lasted for 126 days. The results showed that breeders stocked at 4 fish m–2 had significantly higher (P < 0.05) mean values for total seed production, seed kg–1 female day–1, seed female–1 day–1, seed m–2 day–1 and spawning synchrony than at 8 and 12 fish m–2 broodstock densities. The mean percentage of seeds in the yolk-sac and swim-up fry stages was highest at 4 fish m–2 broodstock density. However, the recovery rate was not affected by broodstock density. It is recommended that further research should be conducted to determine whether weight m–2 , number m–2 or age of broodstock should be the basis for stocking broodstock.  相似文献

The effect of feeding four semi‐purified diets A1, A2, A3, A4, containing different vitamin A acetate levels 0, 20, 40, 60 mg kg?1 diet, respectively, on fecundity, egg hatching rate, larval survival rate and vitamin A content in eggs of Chinese shrimp (Penaeus chinensis) broodstock was compared with a fresh clam diet (control) in a 60‐day feeding trial. The broodstock shrimp fed the diet with 60 mg kg?1 vitamin A acetate added exhibited significantly higher fecundity (P < 0.01). Hatching rate was highest with diet A4 (P < 0.05), whereas hatching rates were similar fed diets A1, A2, A3. Increasing levels of vitamin A in broodstock diet resulted in improvement in larval quality. The vitamin A levels in shrimp eggs from broodstock fed with diet A4 were higher compared with those from broodstock fed with diet A1, A2 (P < 0.01). The fecundity and hatching percentages were positively correlated with the vitamin A content in eggs in the present study. The results of this study showed that higher level of vitamin A in broodstock diet may have positive effects on fecundity and larval quality in P. chinensis.  相似文献

The effect of six different binding agents (agar, sodium alginate, cassava starch, gelatin, wheat gluten and kelp meal) in two concentrations (30 g kg−1 and 50 g kg−1) were evaluated with respect to physical quality of Litopenaeus vannamei broodstock pelleted feed, after 15, 30, 60, 90, 120, 150 and 180 min of water immersion. The best treatments in terms of water stability, water absorption and protein leaching were obtained with sodium alginate and wheat gluten at 50 g kg−1. In a second experiment, the feed ingestion and diet digestibility with these two binders and their combination (1 : 1) were compared against a control diet containing 50 g kg−1 wheat flour. There were no significant differences (> 0.05) in the daily feed ingestion rate 2.39–3.33% of the biomass. The most representative values of apparent digestibility of protein (ADP) and apparent dry matter digestibility (ADMD) were achieved with diets containing wheat gluten and alginate + gluten mixture as binder. Based on these results, combinations with 50 g kg−1 wheat gluten is recommended as binder in pelleted feed for broodstock L. vannamei.  相似文献

Filtration rate and ingestion rate of different stages of Paphia malabarica larvae (D‐shape, (80 μm), Umbo (120 μm) and veliger (180 μm)) were determined in relation to feeding on various micro algae. The micro algae tested were Nannochloropsis salina, Isochrysis aff. galbana, Dicrateria inornata, Tetraselmis gracilis and Chaetoceros calcitrans at 5 or 10 × 103 cells mL?1. Both filtration and ingestion rate of micro algae tested were increased with increasing larval size; however, at all larval stages, Ccalcitrans resulted in lower filtration and ingestion rate. Of the algal diets tested, Pmalabarica larvae showed greatest filtration rate and ingestion rate with Nsalina. Maximum filtration rate for Nsalina was 15.7, 26.3 and 33.9 μL h?1 and highest ingest rate was 15, 92 and 177 cells larva?1 h?1 in D shape larvae, umbo and pediveliger, respectively. Filtration rate and ingestion rate of Nsalina were always higher than those other algal species tested because of its small cell size (2 μm).  相似文献

The diatom Chaetoceros calcitrans is a microalgal species used as food for larva in aquaculture for many species worldwide. Chaetoceros calcitrans is an important source of omega 3 long chain (C ≥ 20) polyunsaturated fatty acids (n‐3 LC PUFA), chiefly eicosapentaenoic acid (EPA, 20:5n‐3). This article reports lipid content, lipid class composition and fatty acid profiles of each lipid class during the growth cycle of batch cultures of C. calcitrans. Total lipid content and the concentration of neutral lipid were highest in the late stationary growth phase (day 12). However, the amount of EPA was highest during the logarithmic growth phase (1.24 pg/cell on day 4). EPA was initially concentrated in the glycolipid fraction, but this fraction decreased during logarithmic growth, coinciding with the increase in neutral lipid. Docosahexaenoic acid (22:6n‐3, DHA) (0.91 pg/cell) was reported as a major fatty acid (>10 mg/100 g) in all lipid classes on day 1. DHA was depleted quickly from the neutral lipid and glycolipid classes, but at a slower rate from the polar lipid fraction. This work confirms that C. calcitrans is a good source of lipid, in particular EPA, for larval and adult filter feeders in aquaculture.  相似文献

Culture of Ruditapes decussatus is clearly limited by the availability of seed, as this production proceeds almost exclusively from natural recruitment. Artificial spawning and larval rearing programs could provide an alternative source of spat. This study was designed to evaluate the effect of different conditioning temperatures on the broodstock maturation, spawning success and larval viability of two geographically (north and south of the Iberian Peninsula) distinct populations of European clam (R. decussatus) collected at different periods of the year in order to create “optimal” artificial spawning and larval rearing programs. Two batches of clams from each population were collected in October and February, and conditioned at 18 ± 1°C, 20 ± 1°C and 22 ± 1°C. Of the three variables analysed the timing of broodstock collection was the most determining factor for gametogenic development, spawning and larval rearing. Geographic origin and conditioning temperature also greatly affected the spawning. The results also showed that the February conditioning was more effective than October and that the best conditioning temperatures were 20 ± 1°C and 22 ± 1°C for the northern and southern populations, respectively. These results suggest that the efficient conditioning temperature for each population of the same species is related to the seasonal temperature regime from their geographic origin. Larval viability and growth performance seemed to be independent of the broodstock conditioning.  相似文献

The composition of protein, carbohydrate, lipid and fatty acids of the gonad of wild female broodstock of black‐lip pearl oyster, Pinctada margaritifera, was compared with oysters fed on a ternary combination of microalgae in hatchery. Artificial feeding was found to be as good as natural feeding in terms of number and size of released eggs. Lipid, protein and carbohydrate reserves of unfed oysters were found to be insufficient to complete oogenesis. The proportions of saturated fatty acids (SFA), monounsaturated fatty acids (MUFA) and polyunsaturated fatty acids (PUFA) of the neutral and polar lipids extracted from female gonads were not influenced by variations in the fatty acid composition of the natural food and ternary combination of microalgae in hatchery. T‐Iso, Chaetoceros calcitrans and Chaetoceros muelleri were able to provide sufficient 22:6n‐3 (DHA) and 20:5n‐3 (EPA), two of the most important essential fatty acids required for gametogenesis. The n‐3/n‐6 and 22:5n‐3/20:4n‐3 ratios were consistently higher in the neutral lipids than in the polar lipids. Conversely, the ratio of 20:4n‐3/20:5n‐3, 22:6n‐3/20:5n‐3 and PUFA/SFA was higher in the polar lipids.  相似文献

Gonadic conditioning and maturation of queen clam Dosinia ponderosa fed three microalgae (Isochrysis galbana, Tetraselmis suecica and Chaetoceros calcitrans) at three temperatures (20, 25 and 30°C) were assessed. Histological analysis showed six gonadic stages, indicators of maturity. The evaluation included sex proportion, oocyte diameter and frequency of gonadic stages after 22 (middle of trial) and 44 days (end of trial). At day 22, around 60% of females conditioned at 20°C, reached stage III (advances grade of maturation) when fed T. suecica. At day 44, around 50% of females were on stage III, conditioned at 20 and 25°C and fed T. suecica and C. calcitrans. For males, the best combination was C. calcitrans at 20 and 25°C. The temperature of 30°C resulted inadequate for maturation, especially when fed the diatom Isochrysis galbana. High mortalities occurred at that temperature with the three microalgal diets, especially in females. The results permits conclude that T. suecica and C. calcitrans are good sources of feed for D. ponderosa and that the best range of temperature for gonadic development is 22–25°C. These results may be basic for a best management and culture of this clam.  相似文献
