The marine leech, Zeylanicobdella arugamensis, is a major threat to aquaculture in grouper-producing countries including Indonesia. This study aimed at investigating prevalence, intensity and histopathology of the ectoparasite in humpback and hybrid groupers cultured in different rearing systems. A total of 260 groupers (60 humpback groupers and 200 hybrid groupers) were used for samples. The marine leech was observed on skin, fins, gills and mouth, followed by histopathological assay on the skin tissue. The results showed that prevalence of the leech in both groupers was higher when they were cultured in the floating net cages compared with the hatchery, p < .05. Furthermore, humpback grouper had a higher prevalence than hybrid grouper when they were cultured in a similar system, p < .05. Meanwhile, there was no significant difference in intensity between the two groupers, p > .05. Within the hybrid groupers, the highest prevalence was obtained from hybrid groupers reared in the earthen ponds. Histopathological studies showed that the infected groupers exhibited inflammation, congestion and erosion of the epidermis layer. Hybrid grouper had more severe histopathological lesions in the skin tissues. These results suggested that species and type of aquaculture system had significantly determined the prevalence, intensity and severity of lesion in Z. arugamensis infestation.  相似文献

Early and accurate diagnosis is a critical first step in mitigating losses caused by plant diseases. An incorrect diagnosis can lead to improper management decisions, such as selection of the wrong chemical application that could potentially result in further reduced crop health and yield. In tomato, initial disease symptoms may be similar even if caused by different pathogens, for example early lesions of target spot (TS) caused by the fungus Corynespora cassicola and bacterial spot (BS) caused by Xanthomonas perforans. In this study, hyperspectral imaging (380–1020 nm) was utilized in laboratory and field (collected by an unmanned aerial vehicle; UAV) settings to detect both diseases. Tomato leaves were classified into four categories: healthy, asymptomatic, early and late disease development stages. Thirty-five spectral vegetation indices (VIs) were calculated to select an optimum set of indices for disease detection and identification. Two classification methods were utilized: (i) multilayer perceptron neural network (MLP), and (ii) stepwise discriminant analysis (STDA). Best wavebands selection was considered in blue (408–420 nm), red (630–650 nm) and red edge (730–750 nm). The most significant VIs that could distinguish between healthy leaves and diseased leaves were the photochemical reflectance index (PRI) for both diseases, the normalized difference vegetation index (NDVI850) for BS in all stages, and the triangular vegetation index (TVI), NDVI850 and chlorophyll index green (Chl green) for TS asymptomatic, TS early and TS late disease stage respectively. The MLP classification method had an accuracy of 99%, for both BS and TS, under field (UAV-based) and laboratory conditions.

Dual choice bioassays were used to evaluate the antifeedant property of essential oil and methanolic extract of Alpinia galanga (L.) (locally known as lengkuas) against two species of termites, Coptotermes gestroi (Wasmann) and Coptotermes curvignathus (Holmgren) (Isoptera: Rhinotermitidae). A 4-cm-diameter paper disc treated with A. galanga essential oil and another treated with either methanol or hexane as control were placed in a petri dish with 10 termites. Mean consumption of paper discs (miligram) treated with 2,000 ppm of essential oil by C. gestroi was 3.30 ± 0.24 mg and by C. curvignathus was 3.32 ± 0.24 mg. A. galanga essential oil showed significant difference in antifeedant effect, 2,000 ppm of A. galanga essential oil was considered to be the optimum concentration that gave maximum antifeedant effect. The essential oil composition was determined using gas chromatography-mass spectrometry. The major component of the essential oil was 1,8-cineol (61.9%). Antifeedant bioassay using 500 ppm of 1,8-cineol showed significant reduction in paper consumption by both termite species. Thus, the bioactive agent in A. galangal essential oil causing antifeeding activity was identified as 1,8-cineol. Repellent activity shows that 250 ppm of 1,8-cineol caused 50.00 ± 4.47% repellency for C. gestroi, whereas for C. curvignathus 750 ppm of 1,8-cineol was needed to cause similar repellent activity (56.67 ± 3.33%). C. curvignathus is more susceptible compare to C. gestroi in Contact Toxicity study, the lethal dose (LD50) of C. curvignathus was 945 mg/kg, whereas LD50 value for C. gestroi was 1,102 mg/kg. Hence 1,8-cineol may be developed as an alternative control against termite in sustainable agriculture practices.  相似文献

A series of treatments were designed to evaluate the efficacy of feeding commercial multispecies probiotics feeding with enzymes and fermentation process on the growth parameters and culture environment of Pacific white shrimp, Litopenaeus vannamei (Boone, 1931), in an intensive culture system. Commercial multispecies probiotics and enzymes (PEs) were continuously applied in three different doses, namely (i) 0.2, (ii) 0.4, and (iii) 0.6 mg L−1 and designated as 0.2, 0.4, and 0.6 PE during the first 30 days of intensive culture of Pacific white shrimp, L. vannamei (Boone, 1931). The probiotics were continually applied every alternate day, while the enzymes were added every sixth day throughout the trial period. The PE dose for all treated tanks was increased by 0.2 ppm after 30 days of culture and another 0.2 ppm after day 60. Meanwhile, fermentation technique, which has become the common method applied in Indonesia in the control treatment, was added on the same day with the PE group with increasing dosage, following the same trend with the 0.6 PE group. Results showed that the group receiving 0.6 PE showed higher final biomass, higher mean weight, and protein retention efficiency as well as a lower feed conversion ratio compared with the control treatment. Abundance of Vibrio spp. remained below 103 cfu mL−1 throughout the trial. Water quality indicators TAN, NO2 N and NO3 N peaked in weeks 3–5 and then declined after that until the end of the culture period in all tanks. This decline was significantly faster in PE-treated tanks. Multispecies PEs have potential applications in controlling Vibrio spp., maintaining proper water quality condition, and enhancing the growth of shrimp in intensive culture system.  相似文献

Rice sheath blight is an important disease caused by Rhizoctonia solani. The resistant and susceptible rice lines (32R and 29S, respectively) showed different responses to R. solani infection in metabolite levels. The aim of this study was to characterize the metabolite levels in rice lines during R. solani infection using capillary electrophoresis equipped with time of flight mass spectrophotometry (CE/TOF-MS) in positive ion mode. Hundred metabolites were identified and classified into six clusters by hierarchical cluster using Mass Profiler Professional software. Changes in metabolite level at inoculated 32R and 29S were mapped on branches of tricarboxylic acid and glycolysis pathway. Volcano plot successfully filtered the metabolites based on fold change and p-value. The volcano plot result showed that 10 metabolites were up and down regulated in inoculated 32R relative to 29S. One metabolite, chlorogenic acid, showed a positive response in 32R. Meanwhile, pipecolic acid showed as the highest magnitude of fold change and p-value significance level in 29S. In addition, eight amino acids; glutamate, γ-aminobutyric acid, glycine, histidine, phenylalanine, serine, tryptophan, and tyrosine showed increase in 29S after R. solani inoculation.  相似文献
