【目的】近年来由白背飞虱传播的南方水稻黑条矮缩病给水稻生产造成了巨大损失,开展该病的抗性遗传分析和基因精细定位,将为抗性育种提供材料和理论依据。【方法】分析了抗性材料D4对南方水稻黑条矮缩病的抗性特征,并通过广恢998/D4F2群体分析该病抗性的遗传规律,利用QTL-seq技术联合遗传连锁分析定位主效抗性QTL。【结果】D4对南方水稻黑条矮缩病的抗性表现为抗病毒性而非抗虫性,且受主效基因和微效基因共同控制。QTL-seq和连锁分析将南方水稻黑条矮缩病主效抗性QTL定位于第9染色体上,命名为qSRBSDV9。利用代换作图法进一步将qSRBSDV9定位在102.3kb的区间内,该区间包含21个预测基因,其中9个基因与赤霉素信号传导相关。【结论】揭示了D4对南方水稻黑条矮缩病的抗性特征及遗传规律,精细定位了南方水稻黑条矮缩病主效抗性QTL qSRBSDV9。这为该QTL的图位克隆及育种利用奠定了基础。 相似文献

土壤盐渍化严重制约水稻生产发展,提高耐盐性已成为水稻育种的重要目标之一。挖掘水稻耐盐新基因,解析其分子作用机制可以为水稻耐盐性遗传改良奠定基础。本文从定位群体、耐盐性鉴定时期和鉴定方法、耐盐性评价指标、鉴定到的耐盐QTL、耐盐QTL的精细定位和图位克隆等方面,总结了近年来水稻耐盐QTL定位研究中所取得的进展;介绍了水稻耐盐/盐敏感突变体筛选和基因克隆以及耐盐性关联分析的研究近况;并对水稻耐盐性分子标记辅助选择改良的现状作了概述。 相似文献

应用候选基因定位水稻抗稻瘟病QTL 总被引:3,自引:2,他引:3
应用经克隆了的已知功能或有潜在功能的DNA序列,即候选基因,作为分子标记,在中156/谷梅2号F8重组自交系群体中进行水稻抗稻瘟病QTL的分析。大部分候选基因在水稻染色体上成簇分布,并且位于已知抗病基因簇区域。应用复合区间法检测到1个调控病斑大小和1个调控病斑数量的QTL,前者位于第1染色体CG36a~RM212区间,贡献率为4.17%,抗性等位基因来自父本谷梅2号;后者定位于第2染色体CG18a~RM263区间,贡献率为6.25%,抗性等位基因来自母本中156。同时检测到2对控制病叶面积和1对控制病斑大小的基因互作。这些QTL和互作基因涉及抗性基因同源序列、离子通道调控子以及编码致病相关蛋白和几丁质酶的基因,表明候选基因的应用有助于揭示QTL的功能。玉米锈病抗性基因Rp1与稻瘟病抗性有关,提示了利用水稻这个模式作物来克隆较大基因组中有利基因的可能性。 相似文献

【目的】发掘水稻抗细菌性条斑病新基因,丰富抗病基因资源,为水稻抗细菌性条斑病基因克隆及分子育种提供依据。【方法】以抗病材料广西普通野生稻WP1和感病品种9311及其衍生的F8:9代RIL群体为研究材料,利用QTL-Seq初定位与细菌性条斑病抗性相关的区间,之后利用QTLIciMapping4.1进行复合区间作图以验证结果并精细定位。【结果】分别在第4、8、10染色体上鉴定了一个与细菌性条斑病抗性相关的位点,复合区间作图验证了第4染色体抗细菌性条斑病QTL位点qBLS4.1,表型贡献率和LOD值分别为10.65%和5.03,并进一步将qBLS4.1精细定位在521kb的范围内。抗感亲本序列比对分析发现,在41个基因的编码区共有252个非同义突变,可能与细菌性条斑病抗性相关。【结论】通过QTL-seq分析结合复合区间作图法可以更快速高效地对水稻QTL进行定位,鉴定了一个抗细菌性条斑病性QTL新位点qBLS4.1,为水稻抗细菌性条斑病新基因鉴定与克隆奠定基础。 相似文献

小麦白粉病是世界范围内广泛流行的小麦叶部真菌病害,显著影响小麦的产量和品质。由于现有品种的白粉病抗性不断丧失,使得进一步挖掘新的白粉病抗源及抗病基因十分重要。本研究以硬粒小麦品种UC1113和Kofa构建的包含93个株系的F8代RIL群体为材料,利用235个多态性标记构建遗传图谱,并结合白粉病成株抗性表型数据,对4个环境下的白粉病成株抗性进行QTL定位。结果表明,在RIL群体中共定位到4个与白粉病成株抗性相关的QTL,分别位于1AL、4AL(2)和5AL染色体上。其中,来自UC1113的3个QTL( QPm.hnau-1AL 、 QPm.hnau-4AL.1 和 QPm.hnau-4AL.2 )为新的白粉病成株抗性QTL。本研究为小麦白粉病抗性基因挖掘和抗病育种提供了重要信息。 相似文献

AbstractA total of 32 rice genotypes carrying different dwarf or semi-dwarf genes were inoculated with the fungus Fusarium moniliforme Sheldon or treated with 50 mg l-1 GA3 in order to select resources resistant to rice bakanae disease from the dwarf materials. The length of the elongated seedlings was measured, and the percentage of death of the seedlings after transplanting to field was also counted. A significant correlation was found between the length of the seedling treated with GA3 and disease injury by bakanae fungus. Rice materials carrying dwarf gene such as sd1 were not only sensitivity to GA3 but also susceptive to rice bakanae disease. Materials carrying dwarf gene d1 were insensitive to GA3 but susceptive to bakanae. On the other hand, all materials carrying d29, sd6 or sdq(t) genes showed resistance to bakanae. The present study indicated that dwarf and semi-dwarf rice materials might be useful resources for improvement of bakanae resistance in rice breeding programs. 相似文献

【目的】本研究旨在了解江苏省水稻主栽品种种子携带水稻恶苗病原菌的情况。【方法】采用环介导等温扩增技术(loop-mediated isothermal amplification,LAMP)对江苏65个水稻主栽品种种子携带藤仓镰孢、层出镰孢、拟轮枝镰孢和Fusarium andiyazi等4种水稻恶苗病菌进行检测。【结果】65个品种中有54个品种的种子样本携带恶苗病菌,表明江苏省稻种携带水稻恶苗病菌是一种普遍现象。其中,藤仓镰孢为优势种,检出率高达83.07%,其次是层出镰孢和Fusarium andiyazi,检出率分别为12.30%、6.15%,但供试的65个品种种子均未检测到携带有拟轮枝镰孢。【结论】该技术可用于水稻种子携带恶苗病菌的快速检测。 相似文献

The baseline sensitivity of Fusarium verticillioides to the novel cyanoacrylate fungicide 2-cyano-3-amino-3-phenylancryic acetate (experimental code JS399-19) was established, and its impact on the germination of rice seeds and efficacy in controlling rice bakanae disease by seed treatment were evaluated. The EC50 values for JS399-19 inhibiting mycelial growth of wild-type F. verticillioides isolates ranged from 0.162 to 0.559 μg/ml (a 3.5-fold difference between the lowest and highest value), and the average (±SE) EC50 value was 0.316 ± 0.072 μg/ml. The sensitivity curve was unimodal over the sensitive range and the sensitivity data can be used as a baseline for monitoring any future changes in sensitivity to JS399-19 by F. verticillioides populations. Germination of healthy seeds was greater than 77% for all the selected cultivars, and was unaffected by treatment with JS399-19. Treatment of infected seeds (harvested from diseased plants) with all concentrations of JS399-19 in this study significantly decreased the abnormal elongation and damping-off of seedlings in plastic bottles in the greenhouse, indicating that JS399-19 has excellent efficacy in controlling rice bakanae disease, and was safe to rice seeds. It could be used as a disinfectant of rice seeds infected by F. verticillioides. The results also suggested that the F. verticillioides isolates exhibited little sensitivity shift to this fungicide after a single application. If properly applied, JS399-19 might be used as a major fungicide or a good companion fungicide for the control of rice bakanae disease in China. 相似文献

Rice false smut disease, which is caused by the fungus Ustilaginoidea virens, is currently one of the most devastating rice fungal diseases in the world. Rice false smut disease not only causes severe yield loss and grain quality reduction, but also threatens food safety due to its production of mycotoxins. In this review, the most recent progresses regarding the life cycle, infection processes, genome and genetic diversity, pathogenic gene and disease resistance in rice were summarized in order to provide theoretical basis for the control of U. virens. We also proposed some future directions and key questions that need to be addressed for a better understanding of the molecular mechanism that leads to rice false smut disease and the prospects for sustainable control of rice false smut. 相似文献

20世纪80年代,中国杂交水稻抗白叶枯病育种进展迅速。在大面积种植的籼型三系和两系杂交稻中主要应用白叶枯病抗性基因Xa4,白叶枯病在生产中几乎“销声匿迹”了20年。但近年来,此病害却在长江流域一些种植新组合的稻田里又爆发了。什么原因引起它再度流行?品种的抗性是否还有效?抗病育种是否还是水稻改良的主要目标之一?通过哪些途径才能更有效地利用抗病基因培育出更为持久的抗性品种?为回答这些问题,就我国杂交水稻对白叶枯病抗性的改良,寄主与病菌群体的互动演变效应进行了分析,讨论了拓宽抗性遗传基础、白叶枯病菌毒性群体结构、基因轮换、合理利用基因等问题。 在采用了包括传统方法、分子标记辅助选择和转基因技术的综合策略后,杂交水稻抗白叶枯病育种研究将出现新的局面。 相似文献
