Summary Flowering plants of Rosa hybrida L. cv Meirutral have been obtained either from direct regeneration of adventitious shoots on leaf and root fragments, or through organogenesis and somatic embryogenesis on calli derived from anther, ovule, petal, sepal, receptacle, leaf, stem internode, root and zygotic embryo tissues. The calli derived from floral parts exhibited rhizogenesis. In this case direct induction of adventitious shoots from selected roots had to be performed in order to generate plants. A histological study of the morphogenetic calli was carried out. The plants regenerated directly and those regenerated from calli of leaf, stem internode, root and zygotic embryo tissues, together with reference plants propagated by cuttings, were compared on a phenotypic basis by taking into account petal number, form and colour, and plant growth habit. The morphological and genetic variations in somaclones of chili pepper (Capsicumannuum L.) derived from tissue culture were evaluated. Cotyledonary node explants of cultivars, Shishitou and Takanotsume, were cultured on Murashige and Skoog (MS) medium supplemented with benzyladenine (BA)5 mg/l for shoots regeneration and regenerated shoots were rooted on MS medium supplemented with naphthalene acetic acid(NAA) 0.1 mg/l and indol-3-butyric acid(IBA) 0.05 mg/l. The regenerated plants(R0) were selfed to obtain seeds for next generation (R1 lines). Qualitative characters were studied in R0generation and both qualitative and quantitative characters were studied in R1 generation. In R0 generation, variations were noticed in plant growth habit, stem color, flower color and color of unripe fruits, and expression of anthocyanin in unripe fruits. Comparison among the R1 lines and their parents were made for morphological and agronomic characters. Significant variation among R1 lines and differences between R1 lines and their parents were observed. Genetic variations among three somaclones were revealed by random amplified polymorphic DNA (RAPD) analysis. Variation, such as early flowering and increase of yield components, is an indication of the response to selection for any specific character. Occurrence of productive variants among somaclones of established cultivars, like Shishitou and Takanotsume, indicates the possibility of their improvement through somaclonal variation. Different wheat genotypes were treated with gamma-rays, sodium azide (SA) and EMS before tissue culture and immature embryos from M1 plants or plants shortly after exposure to gamma-rays were used to initiate callus culture. Thousands of plants were regenerated and used to investigate the effect of mutagenic treatments on the regenerated plants and somaclonal variation in the M3R2 and M2R2 generations. The results showed that mutagen-induced damage in terms of reduction in plant height, fertility and spike length were not outstanding in the regenerated plants as compared with the untreated control. In the M3R2 generation, only SA treatment had significantly higher frequencies of somaclonal variations than the control. Increases in the variation frequencies were observed when explant embryos were irradiated with 2.5 and 5 gy gamma-rays and the highest frequency appeared when embryos were exposed to 5 gy gamma-rays on the 5th day after anthesis. Increased variation spectra also resulted from mutagenic treatments and most of the variants recovered were unsuitable for plant improvement.  相似文献

The suitability of different protocols was compared for entire plant regeneration by somatic embryogenesis, of the forage plants Lolium multiflorum Lam. (Italian ryegrass) and Festuca arundinacea Schreb. (tall fescue). In the first protocol, miniature embryos were used as starting material, while mature seeds were retained in the other two. Whichever the considered protocol, undifferentiated calli were produced on Murashige and Skoog MS medium supplemented with 2,4-D. The calli were subcultured in the dark on solid MS agar medium, containing 5 mg/1 2,4-D (protocol 2) or on solid MS medium followed by transfer to a rotated liquid MS medium with 2 mg/1 2,4-D (protocol 1). In these conditions, induction of somatic embryogenesis occurred, and whole plants were regenerated during a limited lapse of time, upon transfer in the light, to MS medium supplemented with BAP but devoid of 2,4-D. The simultaneous elimination of 2,4-D and transfer to light appeared essential for full regeneration of the plants. Using this characteristic, an additional step was added to a new protocol (protocol 3) in which microcalli, cultured on liquid MS medium containing 5 mg/1 2,4-D, were transferred to the same medium with 2 mg/1 2,4-D, in the dark. In these conditions, the suspensions kept their embryogenic potential for months. In all cases, plantlets were successfully transferred into the soil. An evaluation of the somaclonal variation potential of the plants issued from each protocol is now underway.  相似文献

Regeneration and somaclonal variation in apomictic Paspalum dilatatum Poir   总被引:1,自引:0,他引:1  
Summary In an attempt to incorporate variation into a uniform obligate apomict, plants of apomictic common dallisgrass, Paspalum dilatatum Poir., were regenerated from callus derived from immature inflorescences. Plants developed through both organogenesis and embryogenesis. A total of 682 regenerants were produced and more than 400 were transplanted into a field nursery and screened for somaclonal variation. Eventually 20 regenerants were selected, increased, and planted into a replicated nursery along with normal common dallisgrass. The characteristics examined were maturity date, plant height, number of racemes per inflorescence, number of spikelets per raceme, pubescence, stigma and anther color, ergot resistance, seed germination, seed set, pollen stainability, method of reproduction, and chromosome number. There were differences among the regenerants and between them and common dallisgrass for all traits except chromosome number, stigma and anther color, and ergot resistance. One of the more important regenerants had significantly higher seed set than common dallisgrass. All regenerants reproduced by aposporous apomixis but some exhibited a high degree of abortion while others had more aposporous embryo sacs per ovule than common dallisgrass. These findings demonstrate that common dallisgrass can be regenerated through tissue culture and that somaclonal variation is expressed in some of the regenerants, even though some of the altered traits are deleterious.  相似文献

Sixteen inbred lines and one hybrid of manse were tested for their capability of somatic embryogenesis, and fully developed plants could be regenerated, from ten inbred, lines. The highest frequency of plant regeneration was expressed in the inbred line CHI 31, and when this line was crossed with a recalcitrant, non-regenerating line, the F1 and BC hybrids were regenerable. The results of reciprocal crosses demonstrated that dominant nuclear genes and cytoplasmic factors are primarily responsible for the heritable determination of embryogenic callus proliferation and in vitro regeneration of maize plants. Somaclonal and radiation-induced variability was studied in maize to assess their nature and potential contribution to plant breeding., The inbred line CHI 31 possessing a high in vitro capacity of somatic embryo formation was used as experiments.] material. CHI 31 plants were selfed and twelve-day old zygotic embryos irradiated with 60Co gamma radiation in situ. Mature caryopses were harvested and assigned as M1 material. In another series, immature zygotic embryos (size 1.2—1.5 mm) were cultured in vitro on N-6 medium supplemented with 2,4-D (2.5 μM), and somatic embryos regenerated into plants; these were transplanted into soil and self-pollinated. Regenerants from non-irradiated cultures were grown as R1 generation, while regenerants from irradiated explants were considered as M1R1 generation. The genetic variability was evaluated in the M2, R2 and M2R2 generations, respectively, and compared with a non-treated seed control. Irradiation induced a variety of chlorophyll and morphological variants segregating in the M; generation; however, the frequency of deviant types obtained in the R: generation (somaclonal variation) was significantly exceeding the one derived from the M2 populations. The combination of expert irradiation and in vitro regeneration was most effective for the manifestation of chlorophyll and morphological o if types in the M2R2 generation, and increased drastically the frequency of early flowering variants. Differences in the segregation patterns of mutant phenotypes amonsister somaclones in the R3 and M3R3 generations indicate a different genetic basis, of plants originating from the same explant. This phenomenon suggests a mutational sectoring of the callus during culture. Radiation induced and somaclonal variation exerted a similar spectrum of chlorophyll and morphological deviants.  相似文献

A protocol for in vitro induction of tetraploids via colchicine-treated somatic embryos from immature zygotic embryos of diploid grapevine (Vitis vinifera L.) is reported. Embryogenic callus was initiated from immature zygotic embryos cultured on Nitsch and Nitsch (NN) medium supplemented with 1.0 mg/l 2,4-dichlorophenoxyacetic acid (2,4-D). The callus was transferred to NN medium containing 1.0 mg/l α-naphthalene acetic acid (NAA) and 0.5 mg/l benzyladenine (BA) to establish somatic embryogenesis. The vigorously growing globular embryos were selected and treated by 0, 10 or 20 mg/l colchicine for 1, 2 or 3 days, and then immediately transferred to NN medium supplemented with 0.03 mg/l NAA and 0.5 mg/l BA, for somatic embryo conversion and plant regeneration. The number of surviving embryos and regenerated plantlets following colchicine treatment decreased with increasing colchicine concentration and treatment time. Among 29 randomly investigated plantlets regenerated from colchicine-treated somatic embryos, five solid tetraploids (2n = 4× = 76) were identified by chromosome counting analysis; all others were diploid (2n = 2× = 38). Ploidy level of plant regenerated was also determined from leaves using flow cytometry. No chimeras with both 2C and 4C nuclei was produced from colchicine-treated somatic embryos. Significant differences in leaf stomata parameters were observed between diploid and induced tetraploid plantlets.  相似文献

为建立橡胶树不同品系的体胚发生和植株再生体系,为不同品系橡胶树良种生产奠定基础。以橡胶树优良品系‘热研106’、‘热试59’、‘热试62’的花药为材料,采用正交设计,对不同品系、不同植物生长调节剂配比培养基的花药愈伤和体胚诱导效果进行了研究。[结果]结果表明:3个品系的愈伤诱导率和体胚诱导效率差异明显,同一品系不同培养基的愈伤诱导率和体胚诱导效率差异明显;愈伤诱导培养基中的植物生长调节剂对下一阶段的体胚诱导有明显影响。根据本研究,‘热研106’的花药愈伤诱导最佳浓度组合为2,4-D 1 mg/L+NAA 2 mg/L+KT 1 mg/L,‘热试59’的花药愈伤组织诱导最佳浓度组合为2,4-D 0.5mg/L+NAA 1 mg/L+KT 1 mg/L,‘热试62’的花药愈伤组织诱导最佳浓度组合为2,4-D 1 mg/L+NAA 1 mg/L+KT 1 mg/L。‘热试62’的体胚发生最佳浓度组合为KT 3 mg/L+NAA 0.05 mg/L+GA3 0.05 mg/L+6-BA 0.5 mg/L。[结论]本研究获得了不同品系的愈伤诱导和体胚发生最佳培养基组合,为多品系橡胶树花药植株再生体系的建立奠定了基础。  相似文献

A protocol was developed for plant regeneration from leaf explants (laminaand petiole) of Aster cordifolius cultivar `White Elegans', which ischimeric for chromosome number and the ligulate flower colour. Explantswere cultured on MS or Gamborg's B5 medium in presence of IAA, 2,4-Dand BAP, alone or in combination. The highest frequency of shootregeneration was obtained on B5 medium supplemented with 2,4-D(0.09 mg l-1) and BAP (1.8 mg l-1). After the invitro rooting stage and acclimatization, 90% of plantlets survived in thegreenhouse. The regeneration process occurred by indirect organogenesisas shown by cyto-histology. The regenerants maintained the originalchimerism for chromosome number. Pink flowers instead of theexpected white ones were obtained in all regenerants. Ahypothesis for the uniform colour change is discussed. Variations weredetected in the regenerant generation (R0) for quantitative characterssuch as capitulum and disc diameters, and number of ligulate flowers. SinceAster can be vegetatively propagated, selection of variants in R0can provide interesting material for breeding purposes.  相似文献

Summary Somaclonal variation, which is a welcome source of genetic variation for crop breeding, is unwanted when direct regenerants have to be used in tissue culture mass propagation (eg. in many forest trees), or in the regeneration of genetically transformed plants. Random amplified polymorphic DNA (RAPD) was used to analyse somatic embryos and plants regenerated from embryogenic cell lines in Norway spruce, Picea abies (L.) Karst. RAPD facilitated the identification of clones, as material from the same cell lines shared identical patterns of amplified fragments, whereas regenerants from different cell lines were easily distinguishable by their respective patterns. For comparisons with explant donor genotypes, cell lines were initiated from cotyledons. Some of the seedlings that had parts of their cotyledons removed were grown on as control plants. Somatic embryos regenerated from cotyledon cell lines showed no aberrations in RAPD banding patterns with respect to donor plants. Summary Callus induction and green plant regeneration at high frequencies from an interspefic hybrid, Oryza sativa L. x O. rufipogon Griff. has been achieved by simply coordinating the growth regulators in the induction medium. The study was conducted with two different basal media (Potato-2 and N6) and seven different combinations of growth regulators 2,4-D, NAA and kinetin. Synergistic effects of the two auxins in enhanced anther response to callus induction and subsequent green plant regeneration were observed in both media. The highest frequency of callus induction was obtained on Potato-2 medium supplemented with 1 mg/12,4-D, 2 mg/l NAA and 1 mg/l kinetin. The same combination of growth regulators which yielded higher frequencies of callus induction also induced higher mean number of calli per anther. Although the calli formed on N6 medium showed high regenerability, there was a concomitant increase in the number of albinos among the regenerants. The auxins in the induction media had considerable influence on the regeneration capacity of the calli. The regeneration frequencies were higher from calli formed in the presence of both auxins in the induction media. The levels of growth regulator combinations seem to influence the green plant regeneration especially for calli induced on Potato-2 medium. Among the pollen grain derived plants the majority were either haploids or double haploids and very few were chromosomal variants.  相似文献

对3个辣椒花药培养再生株DH-R2群体的主要农艺性状,包括株高、株幅、首花节位、叶色、单果质量、商品果横径、商品果纵径、果形指数、老熟果色等进行了较详细的测量和统计分析。结果表明:辣椒花药培养再生株群体的农艺性状存在基因型偏性;再生株群体的单果质量、商品果纵径、果形指数这3个性状都表现为低于供体,株高、株幅、首花节位、商品果横径这4个性状都表现为近似于供体;通过单倍体培养途径可以得到不同颜色层次的老熟果色的辣椒纯系,证明了该途径是多基因控制性状育种的有效途径;对耐TMV和CMV的亲本与感病亲本的杂种进行花药培养,获得了兼具TMV和CMV抗性的株系,证明了单倍体培养途径可以创造新的超亲抗病种质。  相似文献

Novel in vitro techniques have the potential to aid conventional breeding programs. Somaclonal variation is considered to be a useful source of variation and has been demonstrated to be feasible in crop species like wheat, rice and maize. A study was taken up at Tissue Culture Laboratory, University of Agricultural Sciences Dharwad, India, for callus induction, maintenance and regeneration of two well established sorghum varieties, M35-1 and A-1 to study somaclonal variation for both qualitative and quantitative characters. The frequency of callus induction was 90 and 83.3 percent in M35-1 and A-1, respectively, on MS medium with 2 mg l-l of 2,4-D. On higher levels of sucrose (60 gl-l), M35-1 showed good response for both regeneration and rooting, while for variety A-1 BA(0.5 mg l-l) and NAA (0.2 mg l-l) had to be supplemented for satisfactory levels of regeneration (73.3%) and rooting respectively. The regenerated plants (SC1) were selfed to obtain the seeds for the next generation. In M35-1, variations were observed for chlorophyll, phyllotaxy and midrib structure, while male sterile and branched phenotypes occurred in A-1 families. Combined analysis of variances showed that there was significant difference between and within families for most of the quantitative characters in both cultivars except for the character, fourth leaf breadth in the cultivar A-1. The variation, accompanied by a positive shift from the mean is an indication of the response to selection for any specific character. Occurrence of productive variants among the somaclones of established cultivars like M35-1 and A-1 indicates the possibility of their improvement through somaclonal variation. In the present study, embryogenic calli of sugarcane variety BL4 were induced using 2,4-dichlorophenoxyacetic acid (2,4-D) and kinetin in different concentrations and combinations. In contrast to earlier studies, embryogenic callus sectors were identified and isolated microscopically within 1–2 weeks. Subsequently, 51 media formulations were used for regeneration of proliferated embryogenic callus, using MS medium supplemented with three different cytokinins [kinetin, 6-Benzylamino purine (BAP), and thidiazuron (TDZ)] and auxins (IAA/NAA and IBA) in different combination and concentrations. After acclimatization, the genomic DNA of regenerated plants was studied to explore the insertion polymorphism of transposable elements in order to ascertain the variation among somaclones. Though low concentration of kinetin with 2,4-D was found supportive to embryogenic callus development, the highest number of regenerated plantlets was observed using BAP (1 μM), however the plantlets had very low fresh weight (2.2 g). Conversely, TDZ alone supported a significant increase in the number of plantlets regenerated (38–40) with higher fresh weight. The somaclones generated during this study showed considerable positional polymorphism of activator-like transposable elements possibly due to the stress associated with in vitro culture. This study provides a procedure to produce regenerated sugarcane plants from embryogenic callus in a relatively short time.  相似文献

在石刁柏花药培养过程中,2,4-D在引起愈伤组织细胞染色体倍性变异方面起主导作用,高浓度的2,4-D虽然能提高愈伤组织中单倍体细胞的频率,但同时又加剧其多倍化。不同激素配比的分化培养基在诱使再生植株染色体倍性变异方面存在着明显差异。在附加1.0ppmBA和0.2ppmIBA的培养基上,再生植株茎尖单倍体细胞频率最高,达31.17%。比较愈伤组织和再生植株的染色体倍性变异幅度,发现后者明显地大于前者。  相似文献

Five populations of lemon plants [Citrus limon (L.) Burm] obtained from undeveloped ovules through different tissue culture procedures were examined for the presence of somaclonal and irradiation-induced genetic variation. Tested groups were: (1) nucellar seedlings; (2) organogenic, regenerated via adventitious buds from nucellar seedling internodes; (3) embryogenic population, regenerated from non-irradiated nucellar callus via somatic embryogenesis; (4) embryogenic population, regenerated from irradiated nucellar callus via somatic embryogenesis; and (5) protoplast-derived, regenerated via somatic embryogenesis. Genomic DNA samples from 360 plants (72 from each group) were screened for polymorphism among RAPD fingerprints amplified by 10 decamer primers. Among all tested plants, genetic variation was detected only within the group of plants recovered from irradiated embryogenic calli. Out of 72 plants from that group, three had RAPD fingerprints different from the rest of the population, and fourth plant was found to be cytochimeric, consisting of diploid and tetraploid cells as revealed by flow cytometry. In all other populations of regenerated plants, we did not come across any plants with changed ploidy level.  相似文献

Summary Plant regeneration from cultured immature inflorescence segments of Eleusine coracana was obtained by direct shoot development and somatic embryogenesis. Direct development of shoots from cultured inflorescence segments occurred on MS medium supplemented with 2,4-D in combination with zeatin. Inflorescences with well developed spikelets differentiated at a low frequency (<5%) from callus cultures initiated on media supplemented with 2,4-D in combination with zeatin or coconut water or picloram + kinetin. Somatic embryogenesis was also induced in callus cultures growing on MS + picloram + kinetin at the end of four passages. Supplementation of the media with different concentrations of sucrose showed 3% sucrose as the best concentration for plant differentiation from somatic embryos. The majority of the regenerated plants showed the diploid chromosome constitution in their root tips.
