Barley (Hordeum vulgare L. cv. Martin) plants grown in solution culture, were exposed to increasing cadmium (Cd) concentration (0, 5, 10, 25, 50, and 100 μM) for a duration of 12 days. The sequence of important biochemical steps of nitrate (NO3) assimilation were studied in roots and shoots as a function of external Cd concentration. Cadmium uptake in roots and shoots increased gradually with Cd concentration in the medium. This Cd accumulation lowered substantially root and shoot biomass. The nitrate reductase (NR, EC 1.6.6.1) and nitrite reductase (NiR, EC 1.6.6.4) activities declined under Cd stress. Concurrently, tissue NO3 contents and xylem sap NO3 concentration were also decreased in Cd‐treated plants. These results suggest that Cd could exert an inhibitory effect on the assimilatory NO3 reducing system (NR and NiR) through a restriction of NO3 availability in the tissues. We therefore examined, in short‐term experiments (12 h), the impact of Cd on NO3 uptake and the two reductases in nitrogen (N)‐starved plants that were pretreated or not with Cd. It was found that Cd induced inhibition of both NO3 uptake and activities of NR and NiR, during NO3 induction period. The possible mechanisms of Cd action on NO3 uptake are proposed. Further, in Cd‐grown plants, the glutamine synthetase (GS, EC 6.3.1.2) showed a decreasing activity both in shoots and roots. However, increasing external Cd concentration resulted in a marked enhancement of glutamate dehydrogenase (NADH‐GDH, EC 1.4.1.2) activity, coupled with elevated levels of ammonium (NH4 in tissues. On the other hand, the total protein content in Cd‐treated plants declined with a progressive and substantial increase of protease activity in the tissues. These findings indicate that under Cd stress the usual pathway of NH4 assimilation (glutamine synthetase/glutamate synthase) can switch to an alternative one (glutamate dehydrogenase). The changes in all parameters investigated were concentration‐dependent and more marked in roots than shoots. The regulation of N absorption and assimilation by Cd in relation to growth and adaptation to stress conditions are discussed.  相似文献

Abstract

Root‐tip, 1‐cm of Sorghum bicolor (L.) Moench cv SC283, SC574, GP‐10, and Funk G522DR were exposed to calcium (45Ca2+) at pH 5.5 for 2‐hr in the presence of nitrate‐nitrogen (NO3?‐N) or ammonium‐nitrogen (NH4+‐N). Nitrate (0.1 mM) induced significantly increased 45Ca uptake in Funk G522DR, SC283, and GP‐10 while 0.01 mM NO3 ?‐N induced significantly increased 45Ca'uptake in SC574, but 45Ca absorption was significantly decreased at 1 mM NO3—N. In the presence of the NH4+ ion, 45Ca uptake was increased up to 8X that of the NH4 +‐N untreated roots. When ammonium chloride (NH4CI) was used, the Cl? tended to induce an increased 45Ca uptake. Studies on the effects of salinity and nitrogen (N) fertilization on ionic balance, biomass, and organic N production of annual ryegrass (Lolium multiflorum Lam.) were conducted. Plants grown in sand were irrigated with nutrient solution with an electrical conductivity of 2 or 11.2 dS#lbm‐1, and N in the form of sodium nitrate (NaNO3), ammonium nitrate (NH4NO3), or ammonium sulfate [(NH4)2SO4] ranging from 0.5 to 9.0 mM. Salinity increased the concentration of total inorganic cations (C) in plants and specifically sodium (Na) by more than 3‐fold higher in plants grown at high salinity as compared with plants at low salinity. Sodium (Na) concentration in roots was higher than in shoots irrespective of the salinity level, suggesting a restriction of Na transport from roots to shoots. The concentration of total inorganic anions (A) increased with salinity and when plants were supplied with nitrate (NO3), salinity increased the concentrations of NO3 and chloride (Cl) in plants. Increasing salinity and N concentration in the growth medium increased organic anions concentration in plants, estimated as the difference between C and A. The effect of different N sources on C‐A followed the order: NH4NO3 > NO3 > ammonium (NH4). The base of organic anions and inorganic ions with salinity contributed significantly to the osmotic potential of plants shoots and roots. Changes in C affected N and organic acids metabolism in plants, since C were highly correlated (p=0.0001) with C‐A and organic N (Norg) concentrations regardless of the salinity level or N source in the nutrient solutions. A high and positive linear dependency was found between Norg and C‐A in plants grown at high and low salinity levels and different N sources, pointing out the close relationship between Norg and organic anions on metabolism under these conditions. The amount of biomass produced was correlated positively with organic anion concentration in plants exposed to different salinity levels. Plant biomass increased with N concentration in the nutrient solution regardless of the salinity level applied. Biomass accumulation decreased while Norg concentration increased with salinity. Organic N content remained unaffected in plants exposed to salinity when grown in N less than 9.0 mM.  相似文献

Uptake and assimilation of inorganic N in young rice plants has been studied with labelled N (N-15). Depletion of the plants' carbohydrate content, obtained by a preceding dark period, resulted in a drastic reduction of NH4 +-N uptake. Plants exposed to low light intensity showed diminishing NH4 +-N uptake rates as compared with plants exposed to full light intensity, the latter showing constant NH4 +-N uptake rates during the whole experimental period. The percentage of labelled insoluble N in total labelled N was not significantly affected by a preceding dark period, whereas the low light intensity resulted in a lower proportion of insoluble N in roots and shoots. The incorporation of labelled N into the insoluble fraction (proteins, nucleic acids) was higher in plants fed with NH4 +-N than in those fed with NO3 -.

The uptake of NH4 +-N was not significantly affected by NO3 -, whereas the NO3- uptake rate was considerably reduced in the presence of NH4 +-N. Low energy status of plants affected the nitrate uptake more than the uptake of NH4 +-N. The results show that uptake and assimilation of inorganic N depend much on the energetic status of plants. Nitrate uptake and assimilation is more sensitive to low energy conditions than NH4 +-N.  相似文献

The beneficial effect of sewage sludge in crop production has been demonstrated, but there is concern regarding its contribution to nitrate (NO3) leaching. The objectives of this study were to compare nitrogen (N) rates of sewage sludge and ammonium nitrate (NH4NO3) on soil profile (0–180 cm), inorganic N [ammonium nitrate (NH4‐N) and nitrate nitrogen (NO3‐N)] accumulation, yield, and N uptake in winter wheat (Triticum aestivum L.). One field experiment was established in 1993 that evaluated six N rates (0 to 540 kg·ha‐1·yr‐1) as dry anaerobically digested sewage sludge and ammonium nitrate. Lime application in 1993 (4.48 Mg ha‐1) with 540 kg N ha‐1·yr‐1 was also evaluated. A laboratory incubation study was included to simulate N mineralization from sewage sludge applied at rates of 45, 180, and 540 kg N ha‐1·yr‐1. Treatments did not affect surface soil (0–30 cm) pH, organic carbon (C), and total N following the first (1994) and second (1995) harvest. Soil profile inorganic N accumulation increased when ≥270 kg N ha‐1 was applied as ammonium nitrate. Less soil profile inorganic N accumulation was detected when lime was applied. In general, wheat yields and N uptake increased linearly with applied N as sewage sludge, while wheat yields and N uptake peaked at 270 kg N ha‐1 when N was applied as ammonium nitrate. Lime did not affect yields or N uptake. Fertilizer N immobilization was expected to be high at this site where wheat was produced for the first time in over 10 years (previously in native bermudagrass). Estimated N use efficiency using sewage sludge in grain production was 20% (average of two harvests) compared to ammonium nitrate. Estimated plant N recovery was 17% for sewage sludge and 27% for ammonium nitrate.  相似文献

Uptake of the two forms of nitrogen NO3 ? and NH4 + and nitrate reduction have been studied in two varieties of Moroccan wheat seedlings, cvs. Sais and Jouda, cultivated under controlled conditions in a hydroponic medium. The available form of nitrogen in the medium had a direct effect on the rate of nitrogen absorption. A brief nitrogen starvation increased the root length and stimulated accumulation of soluble carbohydrates, especially in the root tissues. A resupply of nitrogen stimulated ammonium and nitrate uptake and assimilation with higher mobilization of the soluble carbohydrates previously accumulated in root seedlings. It is suggested that root nitrogen and carbohydrate status might both regulate nitrate uptake and reduction in Moroccan wheat seedlings.

The seedlings raised with the nitrate supply accumulated nitrate in the vacuole as an osmoticum. Shoots are the preferential site for such accumulation. The accumulation of mineral nitrogen was absent in the seedlings cultivated with ammonium supply but was compensated by accumulation of soluble organic nitrogen.  相似文献

Phosphorus uptake is often enhanced by ammonium compared to nitrate nitrogen nutrition of plants. A decrease of pH at the soil-root interface is generally assumed as the cause. However, an alteration of root growth and the mobilization of P by processes other than net release of protons induced by the source of nitrogen may also be considered. To study these alternatives a pot experiment was conducted with maize using a fossil Oxisol high in Fe/Al-P with low soil solution P concentration. Three levels of phosphate (0, 50, 200 mg P kg?1) in combination with either ammonium or nitrate nitrogen (100 mg N kg?1) were applied. Plants were harvested 7 and 21 d after sowing, P uptake measured and root and shoot growth determined. To assess the importance of factors involved in the P transfer from soil into plants, calculations were made using a model of Barber and Claassen. In the treatments with no and low P supply NH4-N compared to NO3-N nutrition increased the growth of the plants by 25 % and their shoot P content by 38 % while their root growth increased by 6 % only. The rhizosphere pH decreased in the NH4-N treatments by 0.1 to 0.6 units as compared to the bulk soil while in the NO3-N treatments it increased by 0.1 to 0.5 units. These pH changes had a minor influence on P uptake only, as was demonstrated by artificially altering the soil pH to 4.7 and 6.3 respectively. At the same rhizosphere pH, however, P influx was doubled by the application of NH4-compared to NO3-N. It is concluded that in this soil the enhancement of P uptake of maize plants after ammonium application cannot be attributed to the acidification of the rhizosphere but to effects mobilizing soil phosphate or increasing P uptake efficiency of roots. Model calculation showed that these effects accounted for 53 % of the P influx per unit root length in the NO3-N and 72 % in the NH4-N supplied plants if no P was applied. 以芥菜为研究对象, 采用盆栽试验, 探讨了不同用量的5种氮肥对污染农田土壤中镉(Cd)在土壤–根系–地上部迁移累积的影响。结果表明: 5种氮肥均促进了芥菜根系对Cd的吸收, 且根系Cd含量随施氮量的增加而增加; 但根系吸收转运Cd的能力随氮肥施用量的增加呈先降后增的变化趋势。在≤200 mg(N)·kg-1(土)的施氮水平下, CO(NH2)2和Ca(NO3)2处理能显著降低芥菜地上部Cd含量, 降低幅度分别为13%~29%和24%~30%。在施氮量相同的条件下, NH4Cl和(NH4)2SO4显著降低了土壤pH, 增加了土壤DTPA-Cd含量, 促进了芥菜对Cd的吸收。本试验条件下, 200 mg(N)·kg-1(土)的CO(NH2)2在增加芥菜产量和降低芥菜地上部Cd含量等方面优于其他氮肥处理。  相似文献

利用控制条件下的溶液培养方法,研究了增硝营养(NH4+∶NO3-比例为100∶0和50∶50)对4种不同的基因型水稻(常规籼稻、常规粳稻、杂交籼稻、杂交粳稻)苗期生长和氮素吸收同化的影响。结果表明,增NO3-营养可以增加水稻叶片的光合速率,促进水稻对氮素的吸收,提高氮素利用率,进而促进水稻生长;不同基因型水稻在增NO3-营养下氮积累量增幅不同主要是由于其生物量增幅不同,而整株氮素含量增幅差异不大;NO3-的存在可增强谷氨酰胺合成酶和硝酸还原酶的活力,促进水稻对NH4+和NO3-的同化利用,从而增加了氮素在植株地上部的积累同化;籼稻与粳稻相比,杂交粳稻与杂交籼稻相比,前者在氮素吸收利用上均表现出更为明显的优势。  相似文献

This study was conducted to obtain information on the properties of the nitrate (NO3 ) uptake system in Vicia faba. The results showed that in plants grown in the absence of NO3 , they cannot absorb it upon initial exposure to this anion. When this plant makes contact with environmental NO3 , NO3 uptake by the roots is progressively induced. However, in NO3 ‐grown plants, the uptake system is yet induced so they had the ability to take up NO3 at high rates from the beginning. The Michaelis’ constant (K,M) and maximum uptake rate (Vmax) were estimated from measurements of NO3 depletion in the uptake medium. The apparent KM value for net NO3 uptake in induced V. faba plants was 60 μM and the corresponding Vmax 0.44 umol NO3 (g FW)‐1 h‐1. This last value was low compared with that of other species, while KM is in the range for other higher plants. From the results of our experiments at low and high NO3 concentrations, we discuss the possibility of the existence of two NO3 uptake systems, one of a high affinity (functioning at low NO3 concentrations) and another of low affinity (operative at high NO3 level in the medium).  相似文献

Exposing 12‐day‐old soybean plants to 0.2 ppm nitrogen dioxide (NO2) for four weeks increased the nitrite concentration and acidity, and decreased the Leghemoglobin (LHb) concentration and the nitrogenase activity of root nodules. The supply of 1 mol.m‐3 nitrate to the roots intensified the nitrite accumulation, decreased the acidity of the nodules, and alleviated the inhibition of nitrogenase activity by NO2 fumigation. These results suggested that the inhibition of nitrogen (N2) fixation by N fertilizer supply might relate to the acid‐alkali balance in nodules.  相似文献

ABSTRACT

A pot experiment was conducted to study the influence of four nitrogen (N) fertilizer forms [Urea; calcium nitrate, Ca(NO3)2; ammonium sulfate, (NH4)2SO4; and organic N] on growth, photosynthesis, and yield of rice under two cadmium (Cd) levels (0 and 100 mg Cd kg?1 soil). Cadmium addition significantly reduced photosynthetic rate, and the reduction varied with N fertilizer form, with ammonium (NH4 +)-N and urea treated plants having more reduction. Nitrogen form had a distinct effect on SPAD value, and the effect was also dependent on Cd level and growth stage. Cadmium-stress significantly reduced flag leaf area, but for the second leaf, only the plants supplied with organic N showed the reduction. There was a significant difference in plant height among four N forms, with NH4 +- and nitrate (NO3 ?)-treated plants having the highest and lowest height, respectively. Cadmium stress caused significant reduction in grains per panicle and total plant weight, and the reduction varied with N form, with organic N treatment showing more reduction. There were significant differences among N forms in N and Cd concentrations of the plants subjected to Cd stress, with NH4 +-N treated plants having highest N and lowest Cd concentrations and NO3 ?-treated plants having lowest N and highest Cd uptake. The results showed that the inhibition of Cd stress on growth and yield formation of rice is closely related to N fertilizer form.  相似文献

Bean plants (Phaseolus vulgaris L.) were cultured for 10 or 18 days on phosphate sufficient (+P) or phosphate deficient (‐P) nutrient medium. Nitrate and phosphate distribution between shoot and root, nitrate uptake, and nitrate reductase activity (NR activity, in vivo and in vitro) in root and leaves was estimated. The decrease in Pi concentration in leaves and roots led to decreased rate of NO3 uptake and increased NO3 accumulation in roots, accompanied by alterations in NO3 distribution between shoot and roots. Nitrate reductase activity estimated in vitro was twice higher than estimated in vivo and both in +P and ‐P plants was lower in the roots than in the shoots. The decrease of NR activity in ‐P plants was more pronounced in the roots and after 2 weeks of phosphate starvation it was about 40% lower as compared with the control. The depression in nitrate uptake may be the result of feedback inhibition due to accumulation of nitrate in the roots. The increased NO3 concentration in root tissue may be explained by decreased NR activity and lower transport of nitrate from roots to shoot.  相似文献

A hypothesis has been presented and tested that bicarbonate (HCO3) and nitrate (NO3) are the most important anions inducing iron (Fe) chlorosis because these anions increase the pH of leaf apoplast which in turn depresses ferric‐iron [Fe(III]) reduction, and hence, the uptake of Fe into the symplasm. Experiments with young sunflower (Helianthus annuus) plants showed that nutrition with NO3 as the sole nitrogen (N) source induced chlorosis whereas ammonium nitrate (NH4NO3) did not. Monohydrogen carbonate (bicarbonate) also favoured the development of chlorosis. The degree of chlorosis was not related to the Fe concentration in the leaves. Both anion species, NO3 and HCO3, increased the pH of the leaf apoplast which was measured by means of the fluorescence dye 5‐carboxyfluorescein. A highly significant negative correlation between leaf apoplast pH and chlorophyll concentration in the leaves (r = ‐0.97) was found. Ferric‐Fe reduction in the apoplast—measured by means of ferrocene—provided evidence that a low leaf apoplast pH, obtained with ammonium (NH4) supply, favoured the reduction of Fe(III) as compared with a higher leaf apoplast pH obtained with NO3 supply. Under conditions of limited nutrient supply, plant nutrient uptake is controlled by the external concentration of the ions. Limited information exists about the whole-plant regulation of nutrient uptake when the supply is adequate. To study the relationship between growth rate and carbon dioxide (CO2) assimilation with nutrient uptake, growth chamber experiments were conducted with temperatures ranging from 10 to 35°C at medium (600 µmol m?2 s?1) and high (1200 µmol m?2 s?1) light intensities. Nutrient solution samples were collected every 24 hours and the concentration of ions was analyzed by Inductively coupled plasma -atomic emission spectroscopy (ICP-AES) and nitrate and ammonium (NO3?/NH4+) conductivity. Leaf photo-synthesis was measured using a closed gas exchange system and the total amount of CO2 assimilated was calculated from dry weight increases. The daily absorption of NO3?, Total nitrogen (N), dihydrogen phosphate (H2PO4?) and potassium (K+) responded linearly to plant growth, while ammonium (NH4+) and sulfate (SO42?) uptake showed a curvilinear response. All the ions studied showed a curvilinear relation with CO2 assimilation.  相似文献

The effects of nitrogen (N) forms (ammonium- or nitrate-N) on plant growth under salinity stress [150 mmol sodium chloride (NaCl)] were studied in hydroponically cultured cotton. Net fluxes of sodium (Na+), ammonium (NH4+), and nitrate (NO3?) were also determined using the Non-Invasive Micro-Test Technology. Plant growth was impaired under salinity stress, but nitrate-fed plants were less sensitive to salinity than ammonium-fed plants due mainly to superior root growth by the nitrate-fed plants. The root length, root surface area, root volume, and root viability of seedlings treated with NO3-N were greater than those treated with NH4-N with or without salinity stress. Under salinity stress, the Na+ content of seedlings treated with NO3-N was lower than that in seedlings treated with NH4-N owing to higher root Na+ efflux. A lower net NO3? efflux was observed in roots of nitrate-fed plants relative to the net NH4+ efflux from roots of ammonium-fed plants. This resulted in much more nitrogen accumulation in different tissues, especially in leaves, thereby enhancing photosynthesis in nitrate-fed plants under salinity stress. Nitrate-N is superior to ammonium-N based on nitrogen uptake and cotton growth under salinity stress.  相似文献

Maize plants, grown for 7 and 21 days on a nutrient solution with NO3 as the sole nitrogen source showed a clear diurnal pattern with respect to the in vivo NRA. Especially in roots dark/light fluctuations of the enzyme activity were high. Also in NO3 uptake, OH efflux and endogenous content of water soluble carbohydrates a diurnal variation was found. The plant age did not significantly affect the daily rhythm.

Because day/night changes of the in vivo root NRA and nitrate uptake were proportional, the relative content of reduced N in the xylem sap of the plants was constant during a day/night interval. At both day 7 and day 21 about 40–50% of the N was transported via the xylem as amino N. As a result of non‐synchronous variation of the specific root and shoot NRA, root reduction capacity showed a great within‐day variation. It varied between 20 and 40% of the whole plant reduction capacity. Since the ratio N‐organic to N‐total in the xylem sap was about 0.5, cycling of organic nitrogen was very likely in these maize plants.  相似文献

采用溶液培养方法探讨了莴笋、菠菜、小白菜和大青菜 4种蔬菜作物对硝、铵态氮的相对吸收能力以及这两种氮源对它们生长发育的影响。结果表明 ,单独供给NO3-N ,4种作物均生长发育良好 ;供给NO3--N +NH4+-N(NO3-∶NH4+=1∶1) ,生长量均有所下降 ,而单独供给NH4+-N时 ,生长量则大幅度下降。莴笋单独供给NO3--N时 ,其吸氮量显著高于供给NO3--N +NH4+-N的处理 ,大青菜、菠菜供给NO3--N +NH4+-N与单独供给NO3--N相比吸氮量大体相当 ;小白菜同时供应NO3--N +NH4+N时吸氮量最高 ,供给NO3--N时次之 ,供给NH4+-N时显著降低。供给NH4+-N时 4种作物吸氮量均比其它氮源显著降低。 4种作物对NO3--N与NH4+-N的吸收具有明显的偏向性。供给等氮量铵、硝态氮 (NO3--N +NH4+-N处理 )时 ,菠菜、小白菜吸收的NO3-N显著多于NH4+-N ,表现出喜硝性 ,莴笋则与此相反 ,表现出喜铵性 ;而大青菜对两种形态氮素的吸收量相差不多 ,表现出兼性吸收的特点。但上述偏向性具有阶段特点 ,即喜硝作物可能在某一阶段表现出喜铵性状  相似文献

Abstract

The changes in availability and uptake of boron (B) by M.26 apple rootstocks as affected by applications of different forms and rates of nitrogen (N) were examined. The study was carried out in a greenhouse using soil with low contents of organic matter, clay, calcium carbonate, NH4‐oxalate soluble aluminum (Al) and iron (Fe), NH2OH·HCl extractable manganese (Mn), poor cation exchange capacity and low pH. Soil N application was in the form of urea, calcium nitrate, ammonium sulphate, or ammonium nitrate at rates of 0, 17, 34, and 51 mg N kg?1. After 1, 3, and 5 days of N application, soil B fractions were determined: B in soil solution, B specifically and non‐specifically adsorbed on soil surfaces, B occluded in Mn oxyhydroxides, and B occluded in crystalline Al and Fe oxides. The results showed that N as calcium nitrate and ammonium nitrate increased B both in soil solution and non‐specifically adsorbed on soil surface and decreased B concentration on Al and Fe oxides. This indicates that N‐NO3 inhibited B sorption on Fe and Al oxides. Maximum B desorption from Fe and Al oxides was obtained within one day after N‐NO3 was supplied. Nitrogen application as calcium nitrate and ammonium nitrate increased availability and uptake of B by plant roots. Thus, it was concluded that apple trees planted on coarse‐textured soils where risk of B deficiency is high, calcium nitrate or ammonium nitrates would be appropriately to apply to keep B more available.  相似文献

ABSTRACT

The interactions between salinity and different nitrogen (N) sources nitrate (NO3 ?), ammonium (NH4 +), and NO3 ? + NH4 + were investigated on Indian mustard (Brassica juncea cv. RH30). Treatments were added to observe the combined effect of two salinity levels (8 and 12 ds m? 1) and three nitrogen sources (NO3 ?, NH4 +, and NO3 ? + NH4 +) on different growth parameters and mineral composition in different plant parts, i.e., leaves, stem, and root. Salinity has been known to affect the uptake and assimilation of various essential nutrients required for normal growth and development. Different growth parameters, i.e., leaf area, dry weight of different plant parts, absolute growth rate (AGR), relative growth rate (RGR), and net assimilation rate (NAR) declined markedly by salinity at pre-flowering and flowering stages. All growth indices were less sensitive to salinity (12 d s m? 1) with the nitrate form of nitrogen. It is pertinent mention that a high dose (120 kg ha? 1) of nitrogen in ammonium form NH4 +, acted synergistically with salinity in inhibiting growth. Plants fed with combined nitrogen (NO3 ? + NH4 +) had an edge over individual forms in ameliorating the adverse effects of salinity on growth and yield. Under salt stress, different nutrient elements such as N, phosphorus (P), potassium (K+), and magnesium (Mg2 +) were decreased in different plant parts (leaves, stem, and root). The maximum and minimum reduction was observed with ammoniacal and combined form of nitrogen, respectively, while the reverse was true of calcium (Ca2 +), sodium (Na+), chloride (Cl?), and sulfate (SO4 2?) at harvest. Nitrogen application (120 Kg ha? 1) in combined form had been found to maintain highest concentrations of N, P, Mg2 +, and Ca2 + along with reduced concentrations of Na+, Cl?, and SO4 2 ?. However, reverse was true with ammoniacal form of nitrogen.  相似文献
