采用扫描电镜观察玫烟色拟青霉(Paecilomyces fumosoroseus)分生孢子在小菜蛾(Plutella xylostella)幼虫体表的萌发和侵染过程及玫烟色拟青霉孢子粉、孢子悬浮液和孢子培养液对小菜蛾幼虫的致病力。结果表明:小菜蛾幼虫体表嵴状突起结构区附着的孢子较多, 刺状突起结构区附着的孢子较少;刺状突起结构区附着的孢子萌发较早, 多数孢子在接菌后16 h开始萌发, 而嵴状突起结构区附着的孢子多数在20 h开始萌发, 在平缓结构区孢子萌发最迟。在嵴状突起结构区和刺状突起结构区, 多数孢子萌发后产生芽管, 直接以芽管侵入幼虫体壁, 少数形成附着胞侵染寄主。此外玫烟色拟青霉3种不同接菌方法对小菜蛾幼虫的侵染速率和致病力大小明显不同, 其中孢子培养液比孢子悬浮液和孢子粉对小菜蛾幼虫的侵染速率快, 接菌44 h虫体表现出明显的感病症状, 致死中时为3.18 d;孢子悬浮液和孢子粉处理的小菜蛾幼虫, 60 h虫体出现感病症状, 致死中时分别为4.20 d和4.34 d;而且致病力大小也存在明显差异, 接孢子粉的处理明显高于孢子悬浮液和孢子培养液, 小菜蛾幼虫6 d的死亡率达76.2%, 而其他两种处理小菜蛾幼虫6 d的死亡率仅为59.8%和58.5%。本研究结果表明, 小菜蛾幼虫的体表结构影响玫烟色拟青霉孢子在寄主体表的附着、萌发和侵入, 玫烟色拟青霉孢子粉对小菜蛾幼虫的致病力最强, 明显高于孢子悬浮液和孢子培养液。  相似文献

为探讨航天诱变对淡紫拟青霉的生物学效应的影响,利用淡紫拟青霉山东菌株搭载"神舟八号"宇宙飞船进行诱变处理。对筛选得到的30个淡紫拟青霉航天诱变菌株进行了形态、色素、菌丝及孢子形态、菌落生长速度、菌丝干重和产孢量等生物学检测,并测定了各诱变菌株对南方根结线虫卵和松材线虫卵的寄生率。结果表明,淡紫拟青霉航天诱变菌株各项生物学特性与原始菌株存在分化。航天诱变后的菌株菌落形态、色素和菌丝结构与原始菌株差异明显;菌落生长速度、菌丝干重和产孢量的负突变率均高于正突变率;菌株对松材线虫卵的致病力较低,寄生率最高的菌株Sd-m-20的寄生率仅为18.0%,菌株对根结线虫卵致病力较高,菌株Sd-m-9,Sd-m-11,Sd-m-16,Sd-m-22和Sd-m-26对根结线虫卵的寄生率分别提高了14.48%、14.90%、12.35%、7.66%和17.45%,显著高于原始菌株的寄生率(78.3%)。航天处理对淡紫拟青霉诱变效果显著,其优良突变菌株可能用于淡紫拟青霉对南方根结线虫的生物防治。  相似文献

温度对淡紫拟青霉NH-PL-03菌株培养特性及毒力的影响   总被引:2,自引:0,他引:2  
试验测定了淡紫拟青霉NH-PL-03菌株在不同温度条件(15~35℃)下培养过程中生长特性(OD值、pH值、菌丝干重、孢子量、基质消耗)的变化趋势及对线虫的致死率,系统研究温度对淡紫拟青霉生长的影响,结果表明:温度不仅影响菌体的生长,且影响次生代谢物的毒力。在15~30℃范围内,菌体生长、孢子生成及基质消耗(C、N)速率随温度升高而升高;25℃为菌体生长的最合适温度,培养第5d进入快速增长期,第9d菌丝干重达到1.08g,第10d菌液孢子量达到401.2万个/mL;30℃时总体毒力最高,第7d培养液对茎线虫的致死率达到64.60%,其次是25℃和20℃,最高致死率分别达到56.17%和37.21%;15℃最高致死率仅为24.71%。说明在一定范围内,温度上升有利于毒力产物生成。  相似文献

为获得生物学性状良好且对粘虫具有高毒力的球孢白僵菌菌株,本研究以8株球孢白僵菌(Beauveria bassiana)菌株为材料,根据菌株产孢量、萌发率和生长速率差异筛选优势菌株,然后测定优势菌株孢子悬浮液对粘虫3龄幼虫的致死率,以及3龄幼虫感染高毒力白僵菌后体内抗氧化酶和解毒酶活性的变化。结果表明,8株球孢白僵菌菌株生长特性各项指标均存在显著差异,菌株Bb314、Bb345、Bb412和Bb378的生长速率、产孢量和孢子萌发率高于其他4个菌株。菌株Bb314孢子悬浮液对粘虫3龄幼虫致死率最高,达69%。Bb314侵染粘虫后,感染前期虫体酚氧化酶(PO)和过氧化物酶(POD)活性显著提高,于36 h达峰值;过氧化氢酶(CAT)活性在感染初期(24 h)时低于对照,处理48 h后达到峰值,并显著高于对照;谷胱甘肽-S-转移酶(GSTs)和羧酸酯酶(CarE)活性分别在处理36 h和48 h后达到高峰,并显著高于对照。以上结果表明,菌株Bb314可能通过影响粘虫幼体内抗氧化酶和解毒酶活性的动态平衡,进而影响其生理代谢。本研究为开发粘虫生防菌的潜力菌株提供了参考。  相似文献

热水处理对桃果采后青霉病及自然腐烂的控制   总被引:4,自引:1,他引:3  
为了寻找一种能够代替化学杀菌剂控制桃果采后病害的方法,研究了热水处理对桃果采后青霉病的控制及对桃果贮藏品质的影响,探讨了热水处理对扩展青霉孢子萌发的影响.在测定不同温度、不同时间热水处理对桃果青霉病抑菌效果的基础上,选择54℃、2 min的条件对桃果进行热水处理,处理后的桃果放在20℃下贮藏7 d或在4℃下贮藏30 d再转移到20℃下贮藏7 d,观察桃果的腐烂情况,并对桃果的贮藏品质指标进行检测.试验结果表明:热水处理能够有效地起到控制桃果采后病害的作用,热水处理对桃果的失重率、硬度、总可溶性固形物含量、抗坏血酸含量、可滴定酸度等水果品质没有显著的不利影响.热水处理可以显著抑制扩展青霉孢子的萌发及芽管的延长.因此,54℃、2 min热水处理是一种能代替化学杀菌剂对桃果采后病害进行控制的有效方法.  相似文献

为研究经不同温度驯化的寄主昆虫对昆虫病原线虫生物学特性的影响,开展了室内生测试验,调查了2种斯氏属线虫对不同温度驯化的5龄脐橙螟幼虫的致病力、侵入力和繁殖力及寄主体内繁殖出线虫的冷冻存活率。结果表明,线虫Steinernema carpocapsae对经10℃、15℃、20℃和25℃驯化7 d的5龄脐橙螟幼虫的致死率没有显著影响,但线虫S. feltiae对经10℃驯化的5龄脐橙螟幼虫致死率为96. 7%,显著高于25℃处理81. 4%;线虫S. carpocapsae和S. feltiae对经10℃处理的5龄脐橙螟幼虫的侵入率分别为49. 3%和39. 7%,均显著高于25℃处理(26%和24%);随着驯化温度的降低,2种线虫从体表出现的寄主数量占总寄主数量的比率呈升高的趋势,线虫S. feltiae的升高趋势更明显; 2种线虫在经10℃驯化的5龄脐橙螟幼虫体内繁殖的产量均显著高于25℃处理,并且随驯化温度的降低S. feltiae产量呈上升的趋势较S. carpocapsae更明显;低温驯化寄主昆虫可提高其体内寄生的2种线虫的冷冻存活能力。10℃驯化7 d的5龄脐橙螟幼虫有助于提高S. feltiae线虫的致病力、侵入力、繁殖速度和产量及其冷冻存活力,提高程度较S. carpocapsae更明显。  相似文献

绿霉和酸腐是柑橘果实采后主要的病害,生产上迫切需要化学杀菌剂的替代方法。该研究对毛霉诱导脐橙果皮产生的具有抗病活性的红色物质(Citru Peel Red-substance Extract,CPRE)进行部分分离纯化,评价了CPRE对指状青霉和酸腐菌的抑菌活性,分析活性成分对两病原真菌菌丝细胞壁与膜功能的影响。结果表明,在50~200μg/mL的质量浓度下,CPRE对指状青霉和酸腐菌的孢子萌发,芽管伸长,菌丝生长有强烈的抑制作用,并且呈现剂量效应,最低抑菌浓度均为200μg/mL。进一步研究表明,与空白对照比较,50、100μg/mL抗菌活性成分处理明显改变指状青霉和酸腐菌菌丝细胞壁中几丁质分布,降低了细胞壁中几丁质含量、促进了质外体碱性磷酸酶的释放,表明CPRE严重损伤两致病真菌菌丝的细胞壁功能;CPRE处理还明显损伤两病菌菌丝的细胞膜通透性(P0.05),刺激菌丝膜外电导率显著上升(P0.05),加剧细胞内蛋白和核酸的丢失,降低了菌丝细胞膜总脂质含量,影响了细胞膜的正常功能。上述结果表明,红色抗菌成分在防控柑橘采后绿霉病和酸腐病中有潜在应用前景。  相似文献

Penicillium expansum是果蔬采后主要的病原真菌,对果蔬的贮藏具有极大的危害,寻找有效的防治方法具有重要的意义.本研究对苹果(Malus domestica)青霉病菌进行了分离及rDNA-ITS法鉴定,确定为P.expansum;利用不同浓度的外源一氧化氮(NO)处理病菌,通过孢子萌发率、芽管长度、菌丝扩展速度等指标以及苹果接种实验,研究了外源NO与P.expansum发育及致病力的关系,并通过进行胞内活性氧水平、蛋白质羰基化、丙二醛(MDA)及ATP含量的检测,对NO的作用机理进行了初步的探索.结果表明,外源NO可以显著抑制P.expansum的发育,且浓度越高抑制作用越强,其抑菌机理可能是通过诱导P.expansum胞内活性氧积累引起氧化损伤.本研究为果蔬采后病原菌的防治提供了新的思路.  相似文献

根寄生杂草瓜列当(Orobanche aegyptiaca)严重危害番茄(Solanum lycopersicum)等多种经济作物的产量和品质。如何有效防除仍是当今瓜列当研究重点之一。真菌是列当的生防因子之一,但目前对农作物无致病性的列当生防真菌的研究尚少。本研究通过培养皿试验研究1株灰黄青霉(Penicillium griseofulvum,CF3)的无细胞发酵滤液对瓜列当种子萌发和发芽管生长的影响,通过盆栽试验研究CF3粉状制剂对瓜列当的防除效果及对寄主番茄生长和根区土壤微生物的影响。结果表明:1)培养皿试验中,CF3发酵液抑制了瓜列当种子萌发和发芽管生长。其中,在放有瓜列当种子与番茄幼苗的培养皿中,加入CF3发酵液后培养6 d,瓜列当种子的萌发均被完全抑制;添加CF3发酵液与霍格兰德营养液体积比为1∶2、1∶4、1∶6和1∶8的混合液培养8 d后,瓜列当种子的萌发率与对照相比分别减少80.26%、70.26%、68.10%和47.51%。CF3发酵液原液、10倍稀释液和100倍稀释液处理后使瓜列当发芽管长度与对照相比分别缩短100.00%、68.84%和19.24%。2)盆栽试验中,CF3菌剂抑制了瓜列当的出土和单株瓜列当的生长,并使番茄增产。施加1.0 g·kg-1 CF3菌剂130 d后,瓜列当的出土数量、出土率和单株瓜列当干重分别降低76.19%、85.30%和28.48%,番茄果实鲜重增加51.57%。此外,灰黄青霉菌剂还调整了番茄根区土壤的微生物区系结构,使施加菌剂130 d后番茄根区土壤中除接入CF3外真菌数量与对照相比降低75.60%,细菌与真菌的数量之比增加117.57%。平均来看,CF3使番茄根区土壤中除CF3外真菌数量降低42.81%,放线菌总数增加84.15%。本研究表明,灰黄青霉CF3具有防除番茄上寄生瓜列当的能力,适宜作为瓜列当的生防真菌。  相似文献

Seed inoculation is frequently essential for annual Medicago establishment in Mediterranean dryland farming systems. As post-planting soil inoculation is often practiced when seed inoculation fails, the effect of delayed inoculation was investigated. Roots of Medicago rigidula (L.). All selection 716, and Medicago rotata Boiss. selection 2132, were initially pre-exposed to Rhizobium meliloti strains. Subsequently, roots were exposed to a secondary inoculum after 6, 48 and 168 h to simulate delayed inoculation. and subsequent establishment of others strains in the nodules were investigated. Combinations of highly effective and host compatible ICARDA strains (M15 and M53) and effective-ineffective strains (M15 and M28) were used to evaluate proportional nodulation responses. Plants were harvested after 6 weeks of growth. Nodules were assessed for distribution in the root system and for occupancy based on their differential resistance to kanamycin and streptomycin, and, in the case of ICARDA M28, on nodule characteristics. The strain ICARDA M15 was a better competitor than ICARDA M53 when applied in equal density in M. rotata, at zero time. While forming nodules with M. rigidula, ICARDA M53 was equally competitive under the same conditions. With ICARDA M15 as the primary inoculum, and ICARDA M53 delayed for 6, 48 and 168 h, the incidence of ICARDA M15 nodules increased on M. rigidula from 43% (at zero time) to 86, 94, and 97% and, on M. rotata, from 78% (at zero time) to 88%, 95%, and 98% for the three time delays, respectively. Pre-exposure of 2-day old M. rigidula seedlings to the ineffective strain ICARDA M28 as the primary inoculant was followed by significant nodule number increases (P 0.01) as compared with ICARDA M15. Nonetheless, when ICARDA M15 was the primary inoculum, M28 was able to produce about 35% of the nodules when applied at the 6 and 48 h time delays. However, with ICARDA M28 as the primary inoculum, followed by ICARDA M15, nodule occupancy of ICARDA M28 in M. rigidula increased from 40 (at zero time) to 75%, 80%, and 95% for the three time delays, respectively. The percentage of total nodulation by M28, applied at the three delay times, was markedly lower (10%, 5% and 1%, respectively) when M. rotata was pre-exposed to ICARDA M15. This suggested a host preference for ICARDA M15, even if applied as a late inoculum. Results indicate that the early events in the nodulation process of annual medics coupled with host-specificity factors are perhaps the most critical for competition among R. meliloti strains for nodule formation.  相似文献

为探讨农药剂型对烟粉虱的毒力差异,分别以甘蓝和黄瓜叶片作为生物测定载体,比较了啶虫脒乳油(EC)、微乳剂(ME)、可溶性液剂(SL)、可溶性粉剂(SP)及可湿性粉剂(WP)等5种剂型对烟粉虱成虫的毒力,并通过对植物叶片临界表面张力、啶虫脒药液表面张力、动态接触角及叶片持液量的测定,分析了不同啶虫脒剂型间毒力差异形成的原因。结果表明:以甘蓝叶片为生物测定载体,不同啶虫脒剂型间对烟粉虱的毒力差异明显;以黄瓜叶片为生物测定载体,不同剂型间毒力差异不明显。啶虫脒SL、SP、WP在两种生物测定载体间的毒力差异大于EC和ME。甘蓝和黄瓜叶片的临界表面张力值分别为30.73 mN.m 1和57.91~63.30 mN.m 1。啶虫脒有效成分浓度大于7.81 mg.L 1时,啶虫脒EC和ME溶液表面张力即小于甘蓝和黄瓜叶片的临界表面张力;啶虫脒有效成分浓度大于500 mg.L 1时,啶虫脒SL、SP、WP溶液的表面张力才小于甘蓝叶片的临界表面张力。啶虫脒ME溶液液滴接触两种植物叶片的瞬间(0 s),液滴与叶片间的接触角就明显小于90°。甘蓝叶片对低浓度的啶虫脒ME和EC溶液的持液量高于其他3种剂型,黄瓜叶片对啶虫脒各剂型溶液的持液量无明显差异。植物叶片表面性质和不同剂型溶液表面张力的差异是导致剂型间产生毒力变化的重要原因。  相似文献

Direct observation of washed conidia of Cylindrocladium scoparium on non-sterile soils, air dried and rewetted immediately before deposition of conidia, indicated that peak germination (33–58%) occurred after 24 h incubation at 26°C. Peak germination on continually moist soils was lower (18–26%) than on rewetted soils. Lysis of germ tubes and germinating conidia on continually moist soils at 26°C was evident with 48 h. Conidia did not germinate on continually moist soils at 6°C and lysis did not become apparent until 168 h. Conidia germinated at a high level (93–99%) in axenic culture in the absence of exogenous C and N sources. The inhibition of conidial germination on soils may be attributed, in part, to the presence of soil volatiles. Germination of conidia placed on washed agar disks and exposed to volatiles from four soils ranged from 51 to 86% of the no-soil controls. Addition of carbon (13 ng C per conidium as glucose) and nitrogen (65 pg N ng?1 C as NH4C1) nullified the inhibitory effect of the soil volatiles. Germinability assayed on a selective medium at 26°C of conidia in artificially infested soils (approximately 104 conidia g?1 soil) decreased progressively during incubation at 26°C from 1 week to 4 months. No germinable conidia were recovered from artificially infested soils after 2 months incubation at 6°C. Conidia of C. floridanum and C. crotalariae responded similarly to C. scoparium in many assays.  相似文献

为研究假蒟对螺旋粉虱的作用效果及对体内酶活力的影响,在实验室采用叶片浸渍法测定了不同浓度的假蒟石油醚萃取物对螺旋粉虱成虫、若虫和蛹的生物活性,并用生化方法测定了成虫体内几种代谢酶的活性变化。结果显示,假蒟石油醚萃取物对螺旋粉虱具有良好的生物活性,且随着处理时间的延长,活性增强,其活性大小为成虫若虫蛹。处理168 h时,假蒟石油醚萃取物对螺旋粉虱若虫的LC50值(0.932 mg.mL 1)仅为处理24 h时LC50值(2.207 mg.mL 1)的42.23%。处理72 h时,假蒟石油醚萃取物对螺旋粉虱成虫和若虫的LC50值(0.430 mg.mL 1和1.303 mg.mL 1)分别为蛹(1.894 mg.mL 1)的22.70%和68.79%。经假蒟石油醚萃取物处理24 h后,螺旋粉虱成虫体内的乙酰胆碱酯酶(AchE)、羧酸酯酶(CarE)、谷胱甘肽-S-转移酶(GSTs)等代谢酶均受到一定程度的抑制。10 mg.mL 1的假蒟石油醚萃取物对3种酶的抑制率分别为82.53%、82.79%、52.26%,但低浓度的假蒟石油醚萃取物对螺旋粉虱成虫CarE、GSTs两种酶的活性反而有诱导现象,其中对GSTs诱导效应较为明显。由此表明,假蒟石油醚萃取物对螺旋粉虱有明显的毒杀作用。  相似文献

采用硅胶干燥法, 将两种落叶松种子含水量由自然含水量降至5%以下, 测定不同含水量种子老化前后的发芽势、发芽率、发芽指数、活力指数、相对电导率、脱氢酶和异柠檬酸裂解酶活性。结果表明: 长白落叶松种子含水量降至3.02%时, 种子各发芽指标在老化前后保持在较高水平, 且老化前的活力指数及老化后的发芽率、发芽指数、活力指数显著高于自然干燥种子; 在此含水量下, 老化后种子脱氢酶和异柠檬酸裂解酶活性显著高于自然干燥种子, 相对电导率比后者低, 说明长白落叶松种子以含水量为3.02%时储藏较为适宜。兴安落叶松种子含水量为2.63%时, 各发芽指标均高于自然干燥种子, 且老化前除发芽势外各发芽指标及老化后各发芽指标差异显著; 此含水量下, 老化后种子脱氢酶和异柠檬酸裂解酶活性显著高于自然干燥种子, 相对电导率比后者低, 比其他含水量种子表现出更强的抗老化能力, 说明兴安落叶松种子较适宜在2.63%含水量下储藏。  相似文献

Effects of compost products, enriched or not-enriched with a strain of the mycoparasitic fungus Trichoderma atroviride, on the survival of the collembolan Protaphorura armata and the viability of fungal conidia after the transit through the springtail gut were investigated. The effect of compost enriched with Ca-Lignosulphonate (Ca-Ls), a low cost by-product of the acid sulphite pulping process, with lignin-like structure, on P. armata was also evaluated. All compost products enriched or not with the mycoparasitic fungus or Ca-Ls did not affect P. armata survival. No statistical differences were found in animal survival for different types of product or in enriched and not-enriched products. In addition to adults, live juveniles were also observed in all compost products. The gut content of animals, collected at the end of the survival test from compost enriched with T. atroviride, was examined under the light microscope, and in a few cases observations revealed the presence of some T. atroviride conidia. Subsequent tests carried out to study the viability of conidia after the transit through the springtail gut showed that colonies of the fungus developed from all faecal pellets produced by adult and juveniles specimens of P. armata previously fed on conidia of T. atroviride. These results suggest compatibility between Collembola and Trichoderma or Ca-Ls in the composts.  相似文献

The determinants of saprotrophic or predatory modes of the nematophagous fungus Arthrobotrys oligospora were investigated in soil microcosms and on solid nutrient media. A sterilized soil amended with 1% w/w alfalfa meal (C:N=32) and inoculated with conidia of A. oligospora, showed lower mycelium biomass and higher specific rate of conidia production in the presence of the bacterivorous nematode Caenorhabditis elegans than in its absence. As few as 10 nematodes g−1 soil were sufficient to enhance spore formation by the vegetative mycelium. Given that the fungus was not limited by available carbon and nitrogen, this indicates that nematodes provide essential growth factors regulating the development of A. oligospora. Carbon mineralisation by A. oligospora, measured as the rate of CO2 production, was found to be 25–35% lower in the presence of 20–60 C. elegans g−1 soil compared to soil without nematodes. This showed that A. oligospora had lower saprotrophic activity in the predaceous phase. Trap formation and nematophagous activity of A. oligospora were observed only where conidia were inoculated on nutrient poor medium (water agar), on low-nitrogen medium (Yeast Carbon Base agar) or on medium containing no amino-acids or vitamins (Czapek-Dox agar). A. oligospora did not form trapping structures when grown on nutrient-rich media containing three amino-acids (l-histidine monohydrocloride, dl-methionine and dl-tryptophan) and vitamins (biotin, calcium pantothenate, folic acid, inositol, niacin, p-aminobenzoic acid, pyridoxine hydrochloride, riboflavine, thiamine hydrochloride). It is concluded that predaceous behaviour of A. oligospora can be regulated either by nitrogen sources or by physiologically active compounds (amino-acids or vitamins) present in nematodes.  相似文献

为了明确胶质芽孢杆菌和苜蓿根瘤菌在矿区排土场复垦中的协同作用,以矿区排土场土壤作为基质进行盆栽试验,设置两菌单接种处理、双接种处理及对照,其中胶质芽孢杆菌和苜蓿根瘤菌用量分别为1.50%和0.25%,测定紫花苜蓿产量指标、叶片生理指标、土壤速效养分含量和土壤微生物指标。结果表明,接种胶质芽孢杆菌和苜蓿根瘤菌的各处理均能显著提高紫花苜蓿的产量指标,其中以双接种处理效果最优,将生物量和结瘤量分别比对照显著提高110.27%和124.32%;在生理指标方面,双接种处理效果低于单接种处理,未表现出协同作用;在土壤养分和微生物性质方面,单接种胶质芽孢杆菌仅显著提高土壤速效钾含量,单接种苜蓿根瘤菌对大部分指标均有显著效果,而双接种处理效果最佳,土壤碱解氮、速效钾含量比对照分别显著提高94.14%和84.55%,土壤微生物指标MBC含量、呼吸强度、蔗糖酶、脲酶、磷酸酶、过氧化氢酶活性比对照分别显著提高204.02%,65.86%,212.32%,91.87%,30.57%,51.87%。双接种处理对紫花苜蓿产量指标、养分和微生物指标表现出显著的协同作用。因此,1.50%用量胶质芽孢杆菌和0.25%用量苜蓿根瘤菌双接种能够有效提高排土场土壤的复垦效果,具有协同作用,可以作为矿区排土场复垦的高效方法。  相似文献

Summary Seedlings of rice (IR42 and IR50) were aseptically dipped into Azospirillum lipoferum strain 34H suspension under dark, and the presence of bacteria on the differentiating regions of rice roots was observed by scanning electron microscopy. The bacterium did not colonize the root tips of IR42, while it colonized this region in the case of IR50, within 24 h after inoculation. In the early stages, most of the bacteria were embedded in the ruptured mucigel below the root cap cells of IR42. Mucigel was hardly detectable in IR50. While the root hair primordia of IR50 were colonized heavily with the bacterium within 24 h, the root hairs of IR42 were colonized 48 and 72 h after inoculation. 为研究寄主植物对茄二十八星瓢虫药剂敏感性及体内酶活力的影响,采用浸渍法分别测定了取食茄子、马铃薯、番茄和龙葵的茄二十八星瓢虫幼虫(以下简称幼虫)对雷公藤和曼陀罗提取物的敏感性,用生化方法测定了雷公藤和曼陀罗提取物对幼虫体内乙酰胆碱酯酶(AChE)和主要解毒酶活性的影响。结果表明,雷公藤提取物对取食茄子、马铃薯、番茄和龙葵的幼虫LC50值分别为1.407 9 mg.L-1、1.595 8 mg.L-1、1.464 7 mg.L-1和1.109 7 mg.L-1,相对毒力指数为78.82、69.54、75.76和100;曼陀罗提取物对取食茄子、马铃薯、番茄和龙葵的幼虫LC50值分别为0.641 7 mg.L-1、0.610 3 mg.L-1、0.758 0 mg.L-1和0.488 3 mg.L-1,相对毒力指数为76.09、80.01、64.42和100;取食龙葵的幼虫对2种植物提取物的敏感性显著高于取食茄子、马铃薯和番茄的幼虫。取食4种寄主植物的幼虫体内靶标酶AChE和主要解毒酶谷胱甘肽-S-转移酶(GST)、羧酸酯酶(CarE)、多功能氧化酶(MFO)活力顺序均为茄子>马铃薯>番茄>龙葵。其中,取食番茄和龙葵的幼虫体内AChE和GST活力显著低于取食茄子和马铃薯的幼虫,取食马铃薯、番茄和龙葵的幼虫CarE活力显著低于取食茄子的幼虫,取食4种寄主植物的幼虫MFO活力差异达极显著水平。雷公藤和曼陀罗提取物对幼虫体内AChE和GST、CarE和MFO活性均有抑制作用。其中,对取食马铃薯幼虫的AChE活力抑制作用最强,其次为取食茄子和番茄的幼虫,对取食龙葵的幼虫抑制作用最低。雷公藤和曼陀罗提取物对取食龙葵幼虫CarE活力的抑制作用最强,而对取食马铃薯的幼虫MFO活力的抑制作用最强。寄主植物中的次生物质对解毒酶的诱导或抑制可能是引起药剂敏感性差异的主要原因。  相似文献
