采用ELISA检测和室内生物测定方法,测定6个转Bt基因棉花品种(系)不同生育期叶片中的Bt毒蛋白含量及其对各世代棉铃虫的抗性水平。转Bt基因棉花叶片中的Bt毒蛋白含量总体上随着棉花生育期的推进而逐渐下降,6个品种(系)叶片的Bt毒蛋白含量子叶期最高,达1 210.03~1 733.15 ng/g,与子叶期相比,三叶期、七叶期、盛蕾期、花铃期、吐絮期Bt毒蛋白含量减少幅度为2.5%~96.0%。6个品种(系)对第4代棉铃虫幼虫抗虫性较低,毒杀效果仅为19.0%~41.3%。幼虫的校正死亡率逐代下降,各棉花品种对第4代幼虫的校正死亡率仅为12.0%~36.2%。  相似文献

为研究新疆南部植棉区转Bt基因棉花Bt杀虫蛋白时空表达规律,本文利用ELISA技术于2009年和2010年对南疆种植的转Bt基因棉花‘中棉所41’、‘中棉所43’、‘236’、‘K9030’、‘T3’、‘中棉所44’、‘中棉所45’、‘K7’和‘SGK321’各生育期中不同器官中的Bt杀虫蛋白含量进行测定。结果发现:在时间表达上,大多数转Bt基因棉花叶片的Bt杀虫蛋白含量随着棉花发育进程而逐渐下降,在子叶期含量最高,至蕾期或吐絮期达到最低点。并且不同品种间Bt杀虫蛋白含量差异显著。在空间表达上,在花铃期,棉铃和花苞叶的Bt杀虫蛋白含量最低,大多数品种表现为叶片>棉蕾>花瓣(花柱)>棉铃和花苞叶。  相似文献

 为检测几丁质酶的表达,制备稻瘟病菌的一假定几丁质酶多克隆抗体,探索其可能运用。将稻瘟病菌(Magnaporthe oryzae) 几丁质酶基因克隆入融合表达载体pET\|32a,并在大肠杆菌Escherichia coli BL21(DE3)中进行诱导表达。表达菌株经0.5 mmol/L IPTG诱导4~6 h后,用Ni2+\|NTA亲和柱纯化蛋白,得到可溶的几丁质酶\|His融合蛋白。以该融合蛋白免疫新西兰雄兔,制备多克隆抗体。ELISA分析表明该抗体效价达1∶204 800,特异性良好。用该抗体ELISA检测了稻瘟病菌4个不同田间菌株中,不同的菌株表达量不同。对稻瘟病菌侵染水稻3、5和7d后的病叶进行抗原Western验证,结果表明,稻瘟病菌不同的侵染时间其几丁质酶表达量有明显差异。 几丁质酶表达的差异是否与菌株间致病型等生物学和生理学差异有关,目前正在进一步研究。  相似文献

室内研究了Cry1Ab蛋白在转Bt水稻茎和叶片中的表达及在害虫稻纵卷叶螟(Cnaphalocrocis medinalis Gue-nee)幼虫体内的积累动态。结果表明,Cry1Ab蛋白在水稻各部位中均有表达。不同生育期及稻株不同部位中Cry1Ab蛋白的表达量有差异。总的趋势是生育前期的表达量较生育后期高。在时间上,主茎中的蛋白在分蘖期最高,成熟期最低,而叶片中蛋白的表达在整个生育期比较稳定。稻纵卷叶螟幼虫取食转Bt基因水稻后,在其体内及粪便中均能检测到Cry1Ab蛋白。其中取食6h前,虫体内的Cry1Ab蛋白含量高于粪便中的;到12 h时,粪便中Cry1Ab蛋白含量高于虫体内,至24 h时达显著水平(P0.05),直到48 h,粪便中的Cry1Ab蛋白含量一直维持较高水平。  相似文献

1985年在广西隆安县从未施过Bt菌剂的柑桔园中喷洒Bt菌剂(Bt5a5b).1986年对该园的土壤和柑桔叶片取样分析,结果表明,施菌区土壤及叶片的Bt(H5a5b)检出率均高于不施菌剂的对照区。1987年检测果园的鳞翅目害虫,施菌区活虫的Bt检出率为51.4%,其中Bt(H5a5b) 占60.2%,死虫的Bt检出率为81.3%,其中Bt(H5a5b)占67.1%。对照区活虫Bt检出率为20.5%,其中Bt(H5a5b)占34.3%,死虫的Bt检出率为35.7%,其中Bt(H5a5b)占10%。说明施用菌剂两年后,果园中仍残留一定量的Bt,对害虫有致死的作用。  相似文献

使用柠檬酸三钠还原法制备胶体金颗粒,在pH值7.6,蛋白用量10~15μg/mL条件下,制备形成4种葫芦科植物病毒多克隆抗体的稳定胶体金蛋白复合物;设计了双向复合试纸条,使用微定量喷头在试纸条两侧的硝酸纤维素膜上分别喷2条病毒检测线和1条质控线,组装制成免疫层析检测试纸条。结果表明经过条件优化后制备的双向复合试纸条特异性好,可在10min内同时检测4种葫芦科植物病毒,且对不同病毒阳性材料的检测灵敏度可达到稀释103倍以上。  相似文献

 根据已报道的瓜类褪绿黄化病毒(Cucurbit chlorotic yellows virus, CCYV)外壳蛋白(CP)基因的核苷酸序列设计引物,利用RT-PCR方法从感病甜瓜中扩增得到长度为753 bp的CP基因,将其克隆到原核表达载体pGEX-4T-3上,获得重组载体pGexCP,在大肠杆菌BL21菌株中诱导表达,SDS-PAGE分析表明,经IPTG诱导,CP基因在大肠杆菌BL21中得到了高效表达。以表达的蛋白为抗原,免疫家兔,制备了CCYV外壳蛋白的特异性抗血清。ACP-ELISA检测结果表明,当CP蛋白浓度为2.5 μg/mL时,血清效价高达5.12×105。Western blot分析表明制备的抗体检测CCYV侵染的甜瓜叶片得到特异性的条带,表明该抗体的特异性较好。ACP-ELISA检测结果表明,制备的抗血清可用于田间甜瓜病样的检测。本试验为CCYV的快速检测提供了重要的研究材料。  相似文献

 将番茄斑萎病毒(Tomato spotted wilt virus,TSWV)运动蛋白NSm基因克隆到原核表达载体pET-32a (+) 中,利用大肠杆菌系统表达NSm蛋白,以Ni2+-NTA agarose亲和柱纯化该蛋白与His组氨酸的融合蛋白,免疫家兔制备融合蛋白多克隆抗体。Western blot检测显示,该抗体与NSm重组蛋白以及普通烟病叶中的NSm蛋白具有特异性反应。采用该抗体对TSWV感染的普通烟叶片低温超薄切片进行免疫胶体金标记,透射电镜观察发现特异性金颗粒主要定位在病叶细胞的胞间连丝中,而健康烟草叶片组织中没有金颗粒特异性标记。结果表明,该抗体可用于研究NSm在细胞中的定位。  相似文献

为明确转cry1Ab基因玉米植株残体中Cry1Ab杀虫蛋白在不同条件下的降解动态,选取转cry1Ab基因玉米MON810和Bt11,采用ELISA方法测定4种条件下秸秆中Cry1Ab杀虫蛋白的残留量。结果表明,在不同条件、不同取样时间,2种玉米秸秆中Cry1Ab杀虫蛋白的残留量差异显著;整株秸秆中的Cry1Ab杀虫蛋白降解速度比粉碎后的慢;秸秆粉碎埋入土壤后播种冬小麦,玉米秸秆中Cry1Ab杀虫蛋白降解速度最快,且可完全降解;在不同条件下,MON810玉米秸秆中Cry1Ab杀虫蛋白的降解速度比Bt11玉米慢,MON810和Bt11玉米秸秆中Cry1Ab杀虫蛋白的DT50分别为10.2~207.8、13.6~124.0 d,DT90分别为185.1~368.3、45.2~224.0 d。研究表明,在不同条件下MON810和Bt11玉米秸秆中Cry1Ab杀虫蛋白的降解速度不同,依次为秸秆粉碎后埋在土壤后种植冬小麦秸秆粉碎埋在玉米田土壤中秸秆粉碎后放在地表整株秸秆放在地表。  相似文献

经室内筛选获得了棉铃虫对Bt杀虫剂、Bt毒蛋白和转Bt基因棉的抗性种群。利用RAPD技术 ,成功地扩增得到 105条多态性条带 ,经过聚类分析发现 ,棉铃虫对Bt产生抗性后在基因水平发生了变异。RAPD技术不仅可以用来鉴定棉铃虫对Bt是否产生抗性 ,而且可以区分不同的Bt抗性种群  相似文献

cry8 Ca2基因是本实验室自行分离克隆的一种新型苏云金芽孢杆菌(Bacillus thuringiensis,Bt)杀虫晶体蛋白基因,其表达130 kDa蛋白对铜绿异丽金龟(Anomala corpulenta)和黄褐异丽金龟(A.exoleta)幼虫具有较高的活性。本研究对Cry8Ca2蛋白原毒素及胰凝乳蛋白酶消化片断的纯化条件及活性进行研究,最适的消化条件为质量比1∶20,37℃,消化1 h。通过阴离子交换层析得到纯的活性毒素蛋白。用原毒素与纯化的毒素对铜绿丽金龟幼虫进行生测,Cry8Ca2原毒素蛋白的LC50为0.47μg/g土,纯化的毒素的LC50为0.08μg/g土,毒素的杀虫活性可以达到原毒素的6倍。  相似文献

With the scheduled phasing out of methyl bromide, there is an urgent need for alternatives. We evaluated the efficacy of propargyl bromide as a potential replacement for methyl bromide for the control of barnyardgrass (Echinochloa crus-galli) and Fusarium oxysporum in an Arlington sandy loam, a Carsitas loamy sand and a Florida muck soil. Soil was mixed with barnyardgrass seeds or F oxysporum colonized on millet seeds, and treated with propargyl bromide at a range of concentrations. The mortality of the fungi and weed seeds was determined after 24 h of exposure at 30 degrees C. The concentrations required to inhibit 50% barnyard seed germination (LC50) were 2.8, 2.4 and 48.5 micrograms g-1 in the sandy loam, loamy sand and muck soil, respectively. In contrast, the LC50 values for F oxysporum were 11.2, 10.8 and 182.1 micrograms g-1 in the sandy loam, loamy sand and muck soil, respectively. The low efficacy of propargyl bromide in the muck soil was a result of the rapid degradation and high adsorption of the compound in the soil. The degradation half-life (t1/2) was only 7 h in the muck soil at an initial concentration of 6.8 micrograms g-1, compared to 60 and 67 h in the sandy loam and loamy sand, respectively. The adsorption coefficients (Kd) were 0.96, 0.87 and 5.6 cm3 g-1 in the sandy loam, loamy sand and muck soil, respectively. These results suggest that registration agencies should consider site-specific properties in recommending application rates for propargyl bromide.  相似文献

暗黑鳃金龟Holotrichia parallela是一种重要的农业害虫,其成虫和幼虫均能对多种作物造成严重危害。为筛选对暗黑鳃金龟成虫具有毒杀活性的Bt蛋白,本研究首先对暗黑鳃金龟成虫的生活习性进行观察,用浸泡蛋白的榆树叶饲喂成虫,建立了Bt蛋白对暗黑鳃金龟成虫的生物学活性测定方法;制备了6种Bt Cry8类杀虫晶体全长蛋白和胰蛋白酶消化蛋白,对暗黑鳃金龟成虫进行了杀虫活性测定。初筛结果发现Cry8Ga1原毒素和消化蛋白均对暗黑鳃金龟成虫具有较高毒杀活性,其在浓度1.0 mg/mL时对暗黑鳃金龟成虫的7 d校正死亡率分别为100%和55%。为更进一步确定Cry8Ga1蛋白的杀虫活性,进行了致死中浓度测定,7 d生测结果显示,Cry8Ga1原毒素的LC50为0.387 mg/mL (95%置信区间为0.108~0.746 mg/mL),Cry8Ga1蛋白经胰蛋白酶消化后的LC50为0.702 mg/mL (95%置信区间为0.150~2.664 mg/mL)。取食Cry8Ga1原毒素蛋白及其消化蛋白后的暗黑鳃金龟成虫中肠细胞产生显著的病理学变化。本研究筛选到对暗黑鳃金龟成虫有毒杀作用的Bt蛋白,研究结果将为金龟子的生物防治开辟新的途径。  相似文献

Sorption, mineralization and mobility of glyphosate were studied in six substrates: the five types of gravel most frequently used as surfacing in Denmark and a sandy agricultural soil from Simmelkaer that served as a reference soil. Cumulative mineralization of [methyl-14C]glyphosate in batch studies was highest in coarse gravel, amounting to 14% after 4 days at 30 degrees C and 32% after 31 days. Mineralization was slowest in the sandy reference soil, amounting to only 2% after 31 days. The adsorption coefficient (Kd) of glyphosate in gravel ranged from 62 to 164 litre kg(-1), while that in the sandy reference soil was 410 litre kg(-1). The results indicate that the relatively low Kd in gravel allows a relatively high rate of glyphosate mineralization by the biomass. When Kd is high, in contrast, mineralization is slow. Lowering the temperature to 10 degrees C decreased mineralization by 50% in one of two gravels. The leaching of glyphosate was screened in simple columns of gravel or soil in which precipitation events (20 mm over a 2-h period) were simulated on three occasions, starting either immediately after or 2 days after application of glyphosate. [14C]Glyphosate was applied as a tracer mixed with the commercial product Roundup Garden at the recommended rate of 2.4 kg glyphosate ha(-1), equivalent to 1 microg g(-1) soil. The highest concentration of [14C] compounds (expressed in terms of glyphosate concentration) in leachate from the columns exceeded 1300 microg litre(-1), and was detected in rounded gravel after the first rain event. No glyphosate was detected in leachate from the sandy reference soil.  相似文献

Although glyphosate (N‐(phosphonomethyl)glycine) is one of the most frequently used herbicides, few controlled transport experiments in undisturbed soils have been carried out to date. The aim of this work was to study the influence of the sorption coefficient, soil‐glyphosate contact time, pH, phosphorus concentration and colloid‐facilitated transport on the transport of [14C]glyphosate in undisturbed top‐soil columns (20 cm height × 20 cm diameter) of a sandy loam soil and a sandy soil. Batch sorption experiments showed strong Freundlich‐type sorption to both soil materials. The mobility of glyphosate in the soil columns was strongly governed by macropore flow. Consequently, amounts of glyphosate leached from the macroporous sandy loam soil were 50–150 times larger than from the sandy soil. Leaching rates from the sandy soil were not affected by soil‐glyphosate contact time, whereas a contact time of 96 h strongly reduced the leaching rates from the sandy loam soil. The role of pH and phosphorus concentration in solution was relatively unimportant with respect to total glyphosate leaching. The contribution of colloid‐facilitated transport was <1 to 27% for the sandy loam and <1 to 52% for the sandy soil, depending on soil treatment. The risk for glyphosate leaching from the top‐soils seems to be limited to conditions where pronounced macropore flow occurs shortly after application. © 2000 Society of Chemical Industry  相似文献

Biochar addition can improve the physical and hydraulic characteristics of sandy soil. This study investigated the effects of biochar on water holding capacity and water movement in sandy soil under drip irrigation. By indoor simulation experiments, the effects of biochar application at five levels (0%, 1%, 2%, 4%and 6%) on the soil water retention curve, infiltration characteristics of drip irrigation and water distribution were tested and analyzed. The results showed thatbiochar addition rate was positively correlated with water holding capacity of sandy soil and soil available water. Within the same infiltration time, with an increasing amount of added biochar, the diffusion distance of the horizontal wetting front (HWF) tended to decrease, but the infiltration distance of vertical wetting front (VWF) initially declined and then rose. The features of wetted bodies changed from "broad-shallow" to "narrow-deep" type. The relationship between the transport distance of HWF and VWF and the infiltration time was described by a power function. At the same distance from the point source, the larger was the amount of added biochar, the higher was the soil water content. Biochar had a great influence on the water content of the layer with biochar (0-200mm) and had some effects at 200-250mm without biochar; but had less influence on the soil water content deeper than 250mm. For the application rate of biochar of 4%, most water was retained within 0-250mm soil layer. However, when biochar application amount was high (6%), it would be helpful for water infiltration. During the improvement of sandy soil, biochar application rate of 4% in the plow layer had the best effect.  相似文献

田间试验对比研究了树脂包衣尿素和普通尿素在灌漠土和风沙土两种土壤上种植玉米的肥料投入产出效益和产量差异。结果表明:树脂包衣尿素在灌漠土和风沙土两种土壤的玉米产量效应上均有良好的表现,与等氮量的普通尿素相比,灌漠土玉米增产6.4%,风沙土玉米增产15.4%。而在灌漠土和风沙土上分别施用是常规氮肥用量50%和67%的包衣尿素,则可获得与普通尿素相等的产量。灌漠土上施用树脂包衣尿素的经济效益好于普通尿素。风沙土上施用等氮量包衣尿素时,经济效益最好,施用67%包衣尿素时,经济效益略低。灌漠土与风沙土两个试验点比较,风沙土试验点不但施氮量比灌漠土高出127.5 kg/hm2,而且其产量还略低于灌漠土试验点。  相似文献

根据前期研究获得的樱桃病毒A北京分离物(Cherry virus Aisolate Beijing,CVA-BJ)外壳蛋白基因序列设计引物,将此基因克隆到原核表达载体pET-28a后转化大肠杆菌BL21(DE3),以终浓度为1 mmol/L的IPTG进行诱导表达。Ni珠吸附法纯化表达产物后免疫家兔制备抗血清。间接ELISA测得抗血清效价为1∶2048。以纯化后的蛋白和樱桃叶片总蛋白为检测材料,Western blot分析表明抗血清具有高度特异性。间接ELISA方法检测樱桃病叶,结果呈阳性,与RT-PCR检测结果一致。表明制备的抗血清可用于感病樱桃样品中CVA的检测。  相似文献

辣椒轻斑驳病毒(Pepper mild mottle virus,PMMoV)属于烟草花叶病毒属,是辣椒的重要病毒种类之一。将PMMoV的外壳蛋白(cp)基因克隆至原核表达载体pET32a(+)中,转化大肠杆菌BL21(DE3),经IPTG诱导,重组PMMoV CP蛋白以可溶形式表达,纯化后获得分子量约35 kDa的融合CP蛋白。以该重组蛋白为抗原免疫新西兰兔制备了PMMoV CP特异性抗血清。Western blot检测结果表明该抗血清与重组CP蛋白发生强烈的免疫反应,间接酶联免疫吸附测定(ID-ELISA)结果显示该抗血清针对重组CP蛋白的效价达1∶25 600,针对PMMoV病汁液的效价达1∶4 000,检测灵敏度为1∶3 200,且该抗血清不与PMMoV同属或不同属的其它6种病毒汁液发生免疫反应,具有较好的特异性。利用该抗血清对42份田间辣椒病样进行检测,阳性率达38%,与进口商品化试剂盒检测结果一致,说明该抗血清可用于PMMoV的ELISA诊断。  相似文献

The seed potatoes used in these experiments had been grown in a slightly acid pleistocene sandy soil or in a marine, holocene sandy loam. They were free of sclerotia ofR. solani or lightly or moderately speckled with them. Seed potatoes from the sandy soil produced plants that suffered less fromRhizoctonia than plants from seed potatoes that had been grown on the marine sandy loam. Similarly harvested tubers had, in a non-conducive soil and in conducive soils with a (very) low inoculum density ofR. solani, fewer sclerotia when they came from seed potatoes grown in an acid sand. In each soil, the degree of infestation of the crop not only depended on the severity of infection of the seed potatoes, but also on their origin. With regard to sclerotia production on tubers, three types of soil were distinguished: suppressive, conducive with a high, and conducive with a very low inoculum density ofR. solani. The differences in infestation and in the amounts of sclerotia on tubers between the crop grown from seed potatoes from the sandy soil and that from seed potatoes from the marine sandy loam soil, is attributed to a richer load of antagonists on the former and possibly to a larger proportion of saprophyticRhizoctonia strains among their sclerotia. The antagonists seem to be inhabitants of the subterranean parts of the plant and to function independently of the soil. This implies possibilities for their use in biological control.  相似文献
