Background:

Oral vanadyl sulfate (vanadium) induces normoglycemia, proliferates beta cells and prevents pancreatic islet atrophy in streptozotocin-induced diabetic rats. Soteriological method is used to quantitate the proliferative effects of vanadium on beta-cell numbers and islet volumes of normal and diabetic rats.

Methods:

Adult male Sprague-Dawley rats were made diabetic with intravenous streptozotocin injection (40 mg/kg). Normal and diabetic rats were divided into four groups. While control normal and diabetic (CD) groups used water, vanadium-treated normal (VTN) and diabetic (VTD) groups used solutions containing vanadyl sulfate (0.5-1 mg/mL, VOSO4+5H2O). Tail blood samples were used to measure blood glucose (BG) and plasma insulin. Two months after treatment, rats were sacrificed, pancreata prepared, and stereology method was used to quantitatively evaluate total beta cell numbers (TBCN) and total islet volumes (TISVOL).

Results:

Normoglycemia persisted in VTN with significantly decreased plasma insulin (0.190.08 vs. 0.970.27 ng/dL, P<0.002). The respective high BG (53249 vs. 14446 mg/dL, P<0.0001) and reduced plasma insulin (0.260.15 vs. 0.540.19 ng/dL, P<0.002) seen in CD were reversed in VTD during vanadium treatment or withdrawal. While the induction of diabetes, compared to their control, significantly decreased TISVOL (1.90.2 vs. 3.030.6 mm3, P<0.003) and TBCN (0.990.1 vs. 3.20.2 x 106, P<0.003), vanadium treatment significantly increased TISVOL (2.90.8 and 4.071.0 mm3, P<0.003) and TBCN (1.50.3 and 3.80.6 x 106, P<0.03).

Conclusion:

Two-month oral vanadium therapy in STZ-diabetic rats ameliorated hyperglycemia by partially restoring plasma insulin. This action was through proliferative actions of vanadium in preventing islet atrophy by increasing beta-cell numbers.Key Words: Vanadium, Pancreas, Islet volumes, Rats  相似文献

Background: Data shows vanadium protects pancreatic beta cells (BC) from diabetic animals. Whether this effect is direct or through the relief of glucose toxicity is not clear. This study evaluated the potential effect of oral vanadyl sulfate (vanadium) on glycemic status and pancreatic BC of normal and diabetic rats. Methods: Rats were divided into five groups of normal and diabetic. Diabetes was induced with streptozocin (40 mg/kg, i.v.). Normal rats used water (CN) or vanadium (1 mg/ml VOSO4, VTN). Diabetic rats used water (CD), water plus daily neutral protamine Hagedorn insulin injection (80 U/kg, ITD) or vanadium (VTD). Blood samples were taken for blood glucose (BG, mg/dL) and insulin (ng/dL) measurements. After two months, the pancreata of sacrificed rats were prepared for islet staining. Results: Pre-treated normal BG was 88 ± 2, and diabetic BG was 395 ± 9. The final BG in CD, VTD, and ITD was 509 ± 22, 138 ± 14, and 141 ± 14, respectively. Insulin in VTN (0.75 ± 0.01) and VTD (0.78 ± 0.01) was similar, higher than CD (0.51 ± 0.07) but lower than CN (2.51 ± 0.02). VTN islets compared to CN had larger size and denser central core insulin immunoreactivity with plentiful BC. CD and ITD islets were atrophied and had scattered insulin immunoreactivity spots and low BC mass. VTD islets were almost similar to CN. Conclusion: Besides insulin-like activity, vanadium protected pancreatic islet BC, and the relief of glucose toxicity happening with vanadium had a little role in this action. The aim of this study was to evaluate the effects of essential oil extracted from aerial parts of Artemisia sieberi in normal and alloxan induced diabetic rats. Fifty rats were divided into five groups of 10 each. Group I normal rats received 1 mL day(-1) of dimethyl sulfoxide (control); group II normal rats received a single dose (80 mg kg(-1) b.wt.) of essential oil extract of Artemisia sieberi; group III diabetic rats received 1 mL day-of dimethyl sulfoxide; group IV diabetic rats received the oil extract (80 mg kg(-1) b.wt.); group V diabetic rats received metformin (14.2 mg kg(-1) b.wt.). All treatments were orally administered once a day for six weeks. Changes in blood glucose concentration, body weight and food and water intake were measured and the data obtained were compared with that of metformin. The essential oil extract significantly (p < 0.05) lowered blood glucose level as well as food and water intake in diabetic rats accompanied by an increase in body weight gain with no apparent side effect when compared with untreated diabetic rats. These effects were found to be closely similar to that of metformin, a common antidiabetic drug. On other hand, no apparent improvement on body weight gain in diabetic rats treated with metformin. In addition, for all parameters measured, the oil extract showed no effect in normal rats. In conclusion, the essential oil of Artemisia sieberi exhibited antidiabetic activity in alloxan-induced diabetic rats. Present findings support the possible use of the essential oil of Artemisia sieberi as a remedy for diabetes mellitus in humans.  相似文献

Urtica dioica L. (Stinging nettle) has already been known for a long time as a medicinal plant in the world. This histopathological and morphometrical study was conducted to determine the effects of the hydroalcoholic extract of Urtica dioica leaves on testis of streptozotocin-induced diabetic rats. Eighteen male Wistar rats were allocated to equally normal, diabetic and treatment groups. Hyperglycemia was induced by Streptozotocin (80 mg kg(-1)) in animals of diabetic and treatment groups. One week after STZ injection (80 mg kg(-1)), the rats of treatment group received the extract of U. dioica (100 mg/kg/day) IP for 28 days. After 5 weeks of study, all the rats were sacrificed and testes were removed and fixed in bouin and after tissue processing stained with H and E technique. Tubular cell disintegration, sertoli and spermatogonia cell vacuolization and decrease in sperm concentration in seminiferous tubules were seen in diabetic and treatment groups group in comparison with control. External Seminiferous Tubular Diameter (STD) and Seminiferous Epithelial Height (SEH) significantly reduced (p < 0.05) in the diabetic rats compared with controls and these parameters in the treatment group were similar to diabetics animals. This study showed that hydroalcoholic extract of Urtica dioica leaves, after induction of diabetes; has no treatment effect on seminiferous tubules alterations in streptozotocin-induced diabetic rats.  相似文献

Background: The leaves of Eucalyptus globulus (eucalyptus) are used for treatment of diabetes mellitus in traditional medicine. The aim of this study was to evaluate the effects of eucalyptus in treatment of established systemic infection with Candida albicans in normal and streptozotocin-induced diabetic rats. Methods: Sixty normoglycemic male Wistar rats, weighing 200-250 g, were selected and randomly divided into six groups (n= 10): normal control, control + C. albicans, control + eucalyptus + C. albicans, diabetic control, diabetic + C. albicans, diabetic + eucalyptus + C. albicans. Diabetes was induced after a single intraperitoneal injection of streptozotocin (60 mg/kg body weight) and eucalyptus was added to the diet (62.5 g/kg) and drinking water (2.5 g/L) of treated animals for 4 weeks. The concerned groups were inoculated with C. albicans 15 days after diabetes induction. At the end of one month experiment, fasted rats were killed by cervical decapitation. Blood was collected from neck vein for estimation of glucose. C. albicans concentrations were estimated in liver and kidneys using serial dilution culture of tissue homogenates. Results: Eucalyptus administration significantly improved the hyperglycemia, polydipsia, polyphagia, and it also compensated weight loss of diabetic rats (P<0.05). Moreover, eucalyptus caused a significant reduction in C. albicans concentration in liver and kidney homogenates (P<0.01). Conclusion: The results revealed that eucalyptus improves Candidia infection in normal and diabetic rats that in some ways validates the traditional use of this plant in treatment of diabetic patients. Background:

Overexpression of lectin-like low density lipoprotein (LOX-1) receptor plays an important role in hyperglycemia-induced vascular complications such as atherosclerosis. Based on the beneficial effects of exercise on preventing cardiovascular complications of diabetes, we aimed to examine the protective effects of aerobic exercise on expression of LOX-1 receptor and production of free radicals in the heart of diabetic rats.

Methods:

Four groups of rats were used: (n = 5 per group): sedentary normal, trained normal, sedentary diabetes and trained diabetes. Diabetes was induced by a single intraperitoneal injection of streptozotocin (50 mg/kg). The exercise protocol was consisted of swimming 30 min/day, 5 days/week for eight weeks. Plasma glucose was evaluated at initiation, weeks 4 and 8 of experiment. At the end of experiment, rats were sacrificed and the heart was removed for determination of nitrate, malondialdehyde, and LOX-1 gene expression.

Results:

In normal non-diabetic rats, the blood glucose level was <150 mg/dl; however, the induction of diabetes resulted in levels more than >400 mg/dl. Gene expression of LOX-1 was increased in the heart of diabetic rats. Exercise reduced the gene expression of this protein in diabetic states without reducing the blood glucose. Finally, swimming exercise decreased the malondialdehyde and nitrate levels in heart tissue both in control and diabetic rats.

Conclusion:

Swimming exercise reduces heart expression of the LOX-1 receptor in accompany with reduction of free radicals production. Since these parameters are important in generation of diabetic complications, swimming exercise is a good candidate for reducing these complications. Streptozotocin destroys the beta-cells of pancreas by generation of reactive oxygen species and vitamin E has documented antioxidant properties. To evaluate the preventive effect of vitamin E on induction of diabetes by streptozotocin and effect of oral vitamin E consumption on carbohydrate and lipid metabolism, forty male Wistar rats divided randomly to control, E1, E2 and E3 groups. The diet of E1, E2 and E3 groups were supplemented with 1, 2 and 4 g kg(-1) of vitamin E, respectively. Four days later all rats were made diabetic by IP injection of 45 mg kg-' streptozotocin and blood glucose was measured 72 h later to determine the severity of blood glucose elevation. Glycosylated hemoglobin, triglyceride, total cholesterol and HDL-c were measured and LDL-c and VIDL-c calculated in plasma of 6 diabetic rats with glucose more than 200 mg dL(-1) in each groups 21 days after streptozotocin injection. Vitamin E had no effect on diabetes induction by streptozotocin, but elevation of glycosylated Hb and reduction of LDL-c in group E3 were significant. Vitamin E also increased HDL-c although it was not statistically significant. We suggest that oral vitamin E consumption may have some beneficial effect on the correction of lipid metabolism disorders of diabetes, although it may worsen carbohydrate metabolism in mild diabetes.  相似文献

The present study is aimed to assess the therapeutic potential of sulfonylurea drug glimepiride in comparison with Nerium oleander plant extract on insulin, glucose levels and some liver enzymes activities in streptozotocin-induced diabetic rats. Rats were rendered diabetic by intraperitoneal injection of a single dose of 50 mg kg(-1) body weight streptozotocin. Rats with serum glucose levels > 200 mg dL(-1) were subdivided into three sub-groups: the first sub-group were remained without treatment and considered as diabetics. The second and third subgroups were orally administered 0.1 mg kg(-1) body weight/day glimepiride and 250 mg kg(-1) body weight/day Nerium oleander, respectively for 4 weeks. Streptozotocin-induced diabetic rats showed hypoinsulinemia and hyperglycemia compared to controls. Strong negative correlation (r = -0.8) was found between serum insulin and glucose levels in diabetic rats. This correlation was +0.4 and -0.3 in glimepiride and Nerium olender-treated rats, respectively implying that glimepiride and plant extract improved insulin and glucose levels with the former was more efficient. The activities of serum aspartate aminotransferase, alanine aminotransferase and alkaline phosphatase were significantly increased in streptozotocin-induced diabetic rats compared to controls. Treatment of diabetic rats with glimepiride or Nerium oleander extract also improved liver enzymes activities.  相似文献

Background: Diabetes is a major public health problem. The development of new therapies that are able to improve glycemia management and even to cure diabetes is of great interest. In this study, protective effects of sodium tungstate against STZ-induced beta-cell damages were investigated. Methods: Sixty rats were divided into six groups: control, diabetic, sodium tungstate treated diabetic rats from one week before STZ injection (TDB), food-restricted diabetic (FRD), tungstate treated control, sodium tungstate treated diabetic rats from one week after STZ administration (TDA). We evaluated serum insulin, glucose and glucose tolerance; liver glycogen content, glucokinase (GK) activity; blood and pancreas antioxidant power, lipid peroxidation; and fuchsin-aldehyde histochemical staining of beta-cells. Results: Blood glucose levels of TDB group were lower than other diabetic groups (P<0.01). Blood insulin levels of all diabetic groups were lower than controls (P<0.01). Glucose intolerance improved in TDB animals. Blood and pancreas antioxidant power, liver glycogen contents and GK activities and granulated beta cells increased in TDB rats in comparison with other diabetic groups (P<0.01). Likewise, lipid peroxidation decreased significantly in TDB rats (P<0.01). Conclusions: Results suggested that sodium tungstate if administrated before STZ injection improves glycemic state by a direct effect on pancreatic beta-cells and preserves them by reducing the activity of these cells at the time of STZ injection, reducing STZ-induced oxidative stress, reducing insulin secretion, or all of the above mentioned.  相似文献

Background:

Hypertension is considered an independent risk factor for cardiovascular mortality in diabetic patients. The present study was designed to determine the role of gamma amino butyric acid B (GABAB) receptor and L-arginine (L-Arg) in GABA-induced vasorelaxation in normal and streptozotocin-induced diabetic rat vessels.

Methods:

Diabetes was induced by a single i.p. injection of streptozotocin (STZ, 60 mg/kg). Eight weeks later, superior mesenteric arteries of all groups were isolated and perfused according to the McGregor method.

Results:

Baseline perfusion pressure of STZ diabetic rats was significantly higher than non-diabetic rats in both intact and denuded endothelium. In the presence of faclofen, a selective GABAB receptor blocker, GABA-induced relaxation in intact and denuded endothelium mesenteric beds of STZ diabetic rats was suppressed, but this response in non-diabetic rats was not suppressed. Our results showed that in the presence of L-Arg, a nitric oxide precursor, GABA induced vasorelaxation in both diabetic and non-diabetic vessels.

Conclusion:

From the results of this study, it may be concluded that the vasorelaxatory effect of GABA in diabetic vessel is mediated by the GABAB receptor and nitric oxide, but it seems that in non-diabetic vessel GABAB receptor does not play any role in GABA-induced vasorelaxation, but nitric oxide induced GABA relaxation in non-diabetic vessel. Bitter gourd (Momordica charantia), a commonly consumed vegetable is used as an adjunct in the management of diabetes mellitus. A study was carried out to examine the effect of edible portion of bitter gourd at 10% level in the diet in streptozotocin induced diabetic rats. To evaluate the glycaemic control of bitter gourd during diabetes, diet intake, gain in body weight, water intake, urine sugar, urine volume, glomerular filtration rate and fasting blood glucose profiles were monitored. Water consumption, urine volume and urine sugar were significantly higher in diabetic controls compared to normal rats and bitter gourd feeding alleviated this rise during diabetes by about 30%. Renal hypertrophy was higher in diabetic controls and bitter gourd supplementation, partially, but effectively prevented it (38%) during diabetes. Increased glomerular filtration rate in diabetes was significantly reduced (27%) by bitter gourd. An amelioration of about 30% in fasting blood glucose was observed with bitter gourd feeding in diabetic rats. These results clearly provided experimental evidence that dried bitter gourd powder in the diet at 10% level improved diabetic status signifying its beneficial effect during diabetes.  相似文献

Parsley is one of the medicinal herbs used by diabetics in Turkey. It has been reported to reduce blood glucose levels. In this study the effects of feeding parsley on diabetes-induced free radical mediated injury in rat aorta and heart tissues were investigated. Swiss albino rats were divided into six groups: Control, diabetic, parsley, diabetic + parsley, glibornurid, and diabetic + glibornurid. Rats were subjected to i.p. streptozotocin (STZ, 65 mg/kg) to induce diabetes. On the fourteenth day of the study, either parsley (2 g/kg) or glibornurid (5 mg/kg) were given for 28 days to the diabetic rats. Aorta and heart tissue lipid peroxidation and glutathione levels as well as blood glucose levels were determined. The results of the present study indicate that lipid peroxidation was increased and glutathione levels were decreased in both aorta and heart tissue of the diabetic rats. However, treatment of the diabetic rats with either parsley or glibornurid reversed the effects of diabetes on blood glucose, and tissue lipid peroxidation and glutathione levels.  相似文献

BACKGROUND: Teucrium polium is an analgesic, antidiabetic and antilipeidemic herbal medicament. The aim of this survey was to evaluate the effect of aqueous extract T. polium on liver enzymes linked to liver dysfunction, serum lipids and glucose, in diabetic male rats. METHODS: A total of 20 Sprague-Dawly male rats became diabetic by intraperitoneal injection of streptozotocin (60 mg/kg). the animals were divided randomly into two groups. Experimental group was fed Teucrium polium (50 mg/kg) for a month but control group was received the same volume of distilled water. Liver enzymes, biochemical parameters (cholesterol, triglyceride, low density lipoprotein, alanine transaminase, aspartae transaminase) and glucose were measured by kinetic (Enzymatic) and colorimetric methods. Data obtained were analyzed and mean values were compared by paired student's t-test. The results were expressed as mean +/- SD. Significant differences were set at P < 0.05. RESULTS: Our results showed that in test group, serum glucose values decreased significantly (P < 0.05), but cholesterol, triglyceride, low density lipoprotein, alanine transaminase and aspartae transaminase increased significantly after use of T. polium (P < 0.05). This parameters value did not show any changes in control group. CONCLUSION: Although the aqueous extract of Teucrium polium has strong hypoglycemic properties in experimental animals, but because of some hepatotoxic effects, it is not suitable to use it in human as an antidiabetic agent.  相似文献

观察豆豉对早期动脉粥样硬化大鼠主动脉平滑肌细胞凋亡及相关凋亡基因突变型p53和Fas蛋白表达的调节作用,以探讨其机制.采用高脂饲料喂养法建立大鼠早期动脉粥样硬化模型,同时连续10周灌胃给予豆豉提取物进行干预.末次给药后处死动物,光镜下观察血管平滑肌细胞的形态,透射电镜观察超微结构,采用流式细胞术检测细胞凋亡率以及凋亡相关基因突变型p53和Fas蛋白的表达.模型对照组主动脉平滑肌细胞的凋亡率及增值指数(FI)明显高于正常对照组(P<0.05),053蛋白表达上调;豆豉干预组(20 g生药·kg-1,dO g生药·kg-1)大鼠主动脉平滑肌细胞的凋亡率明显高于模型对照组(P<0.05),增殖指数明显低于模型对照组(P<0.05),Fas蛋白表达上调(P<0.05),p53蛋白表达下调(P<0.05),光镜及透射电镜可见主动脉损伤明显较模型对照组改善.豆豉可调节早期动脉粥样硬化大鼠血管平滑肌细胞凋亡与增殖的平衡,其机制可能与调节突变型p53和Fas蛋白的表达有关.  相似文献

探讨茶黄素-3,3'-O-双没食子酸酯(TFDG)对糖尿病大鼠血管内皮损伤及炎症反应的保护作用及相关机制。以高脂饲料喂养加链脲佐菌素注射制造糖尿病大鼠,将其分为模型组(CON)、5 mg·kg-1和10 mg·kg-1 TFDG干预组(TFDG5和TFDG10),另取正常大鼠为对照组(NC),分组处理8周,监测体重和空腹血糖变化,观察大鼠腹主动脉病理形态学变化,ELISA检测血浆IL-6、IL-1β和TNF-α水平,Western blot检测蛋白表达水平。结果表明,与CON组相比,TFDG干预对体重和血糖无显著影响,病理切片观察显示TFDG干预组大鼠腹主动脉组织损伤较CON组有所改善,同时TFDG干预组大鼠血浆炎症因子水平显著降低,血浆NO水平显著升高,腹主动脉组织MDA水平降低。进一步研究显示,TFDG下调了糖尿病大鼠腹主动脉组织NLRP3、caspase-1和IL-1β的表达,抑制NLRP3炎症通路激活。TFDG能够有效保护糖尿病大鼠血管内皮损伤并抑制炎症反应,其机制可能是通过下调NLRP3炎症通路实现的。  相似文献

The aim of the present investigation was to examine the effect of Flax Seeds (FS) and Date Palm Leaves (DPL) extracts on the concentration of serum glucose and lipids in alloxan diabetic rats. Rats were divided into six groups, normal control rats (without treatment with either FS or DPL), normal control rats treated with either FS or DPL extract for four weeks, diabetic control rats and diabetic rats treated with either FS or DPL extract for four weeks. The concentration of glucose in diabetic rats treated with FS (D+FS) for four weeks was significantly decreased from 17.20 +/- 2.33 to 8.14 +/- 0.54 mmol L(-1) (p < 0.001). Serum total cholesterol concentration (TC) of diabetic rats treated with either FS or DPL extract showed significant decrease by 40 and 31% respectively in the fourth week. Also, serum low density lipoprotein cholesterol (LDL-C) concentration was significantly decreased after two weeks of treatment (p < 0.001 and p < 0.01, respectively). However, no significant changes were obtained in serum high density lipoprotein cholesterol (HDL-C) concentration. Results of the present study showed that FS extract has a hypoglycaemic effect against alloxan diabetic rats. Present findings also showed that treatment of alloxan diabetic rats with either FS or DPL extract significantly decreased serum concentration of TC and LDL-C. Present findings suggest that both of FS and DPL extracts could have a protective effect against diabetes complications as well as against hyperlipidemia through improvement of lipid profile.  相似文献

The potential of dietary potato peel (PP) powder in ameliorating oxidative stress (OS) and hyperglycemia was investigated in streptozotocin (STZ)-induced diabetic rats. In a 4-week feeding trial, incorporation of potato peel powder (5 and 10%) in the diet of diabetic rats was found to significantly reduce the plasma glucose level and also reduce drastically the polyuria of STZ diabetic rats. The total food intake was significantly reduced in the diabetic rats fed 10% PP powder compared to the control diabetic rats. However, the body weight gain over 28 days was nearly four times greater in PP powder supplemented diabetic rats (both at 5 and 10%) compared to the control diabetic rats. PP powder in the diet also decreased the elevated activities of serum transaminases (ALT and AST) and nearly normalized the hepatic MDA and GSH levels as well as the activities of specific antioxidant enzymes in liver of diabetic rats. The result of these studies clearly establishes the modulatory propensity of PP against diabetes induced alterations. Considering that potato peels are discarded as waste and not effectively utilized, these results suggest the possibility that PP waste could be effectively used as an ingredient in health and functional food to ameliorate certain disease states such as diabetes.  相似文献

Weanling male, Wistar rats (40 per diet) were maintained on 15 diets which provided 4 graded trypsin inhibitor (TI) levels (93, 215, 337, and 577 mg/100g diet) repeated at 3 levels of protein (10%, 20%, and 30%). Raw and heated (toasted) soy flour provided 10% protein in the diet, while casein was added to increase the protein level to 20% and 30%. Three diets containing only casein as the protein source, at 10%, 20%, and 30% were included. Histologic changes occurring in the pancreas were evaluated in approximately 5 rats from each diet at 3 month intervals, beginning at 6 months, for 22 months. Criteria for the diagnosis of the observed histologic changes in the pancreas are discussed. Ultrastructure of the endocrine cells in the gastrointestinal mucosa was also examined.Pancreatic acinar cell density, as determined morphometrically, was reduced throughout the study in rats fed the 577 mg TI/100 g diet, indicating hypertrophy at the cellular level. Nodular hyperplasia was observed in the first sacrifice group at 6 months. Incidence of the lesion was positively related to both time of exposure and level of dietary TI. Several studies support the health-promoting benefits of lupins, particularly lupin proteins. It has been demonstrated that Lupinus albus gamma conglutin (Cγ) protein lowered blood glucose levels; thus, Cγ showed promise as a new anti-diabetic compound for type 2 diabetes (T2D) treatment. The aim of this study was to evaluate the effect of Cγ on Ins-1 gene expression and on pancreatic insulin content in streptozotocin-mediated diabetic rats. Cγ was isolated from Lupinus albus seeds. Its identification was confirmed with polyacrylamide gel electrophoresis under native and denaturing conditions. We used streptozotocin (STZ) to induce T2D on the 5th day of life of newborn male Wistar rats (n5-STZ). After 20 weeks post-induction, these animals (glycemia?>?200 mg/dL) were randomly assigned to three groups that received the following one-week treatments: vehicle, 0.90 %?w/v NaCl (n5 STZ-Ctrl); glibenclamide, 10 mg/kg (n5 STZ-Glib); or Cγ, 120 mg/kg (n5 STZ-Cγ). Glucose and insulin levels were measured before and after treatment. Ins-1 gene expression was quantified using real time polymerase chain reaction and the pancreatic insulin content was evaluated with immunohistochemistry. Post-treatment, the n5 STZ-Cγ and n5 STZ-Glib groups showed reductions in glucose, increments in serum insulin, and increases in Ins-1 gene expression and beta cell insulin content compared to the n5 STZ-Ctrl group. The results showed that Cγ had beneficial effects on Ins-1 gene expression and pancreatic insulin content. These biological effects of Cγ strengthen its promising potential as a nutraceutical and/or new agent for controlling hyperglycemia.  相似文献

This study investigated the effects of chitosan oligosaccharide (COS) on glucose metabolism and hepatic steatosis in a high-fat (HF) diet/streptozotocin-induced diabetic rat model. Male Wistar rats were divided into: (1) normal control (NC group), (2) HF diet (HF group), (3) streptozotocin (STZ)-induced diabetes with HF diet (DF group), and DF group supplemented with (4) 0.5% COS (D0.5F group), (5) 1% COS (D1F group), and (6) 5% COS (D5F group) for 4 weeks. COS supplementation significantly decreased the plasma glucose, BUN, creatinine, uric acid, triglyceride (TG), and total cholesterol (TC) levels, and hepatic glucose-6-phosphatase activity, and significantly increased hepatic hexokinase activity and glycogen content in diabetic rats; but the increased hepatic TG and TC levels could not be significantly decreased by COS supplementation. Supplementation of COS increased superoxide dismutase activity and decreased lipid peroxidation products in the diabetic rat livers. COS supplementation significantly increased phosphorylated AMP-activated protein kinase (AMPK) protein expression, and attenuated protein expression of hepatic phosphoenolpyruvate carboxykinase (PEPCK) and phosphorylated p38 and renal sodium-glucose cotransporter-2 (SGLT2) in diabetic rats. These results suggest that COS may possess a potential for alleviating abnormal glucose metabolism in diabetic rats through the inhibition of hepatic gluconeogenesis and lipid peroxidation and renal SGLT2 expression.  相似文献
