Cross-pollination was performed with soft-X-irradiated hyuga-natsu pollens (1000 Gy) for ‘Tosa-Buntan’ pummelo (Citrus grandis (L.) Osbeck). This resulted in the transformation of large and complete seeds into small and empty ones (practically seedless). Although fruit set, fruit retention, total soluble solids content (TSS) and titratable acidity of the juice were not affected, decrement in the fruit size was observed. Two weeks after the pollination, endosperm cell division with free nuclei began in both the non-irradiated and irradiated pollen treatment conditions. Seven weeks after pollination, endosperm division with the cell wall occurred in the non-irradiated pollen treatment conditions; however, the endosperm development ceased in most ovules that underwent the irradiated pollen treatment, and the ovules remained in their free nuclear stage. The delayed degeneration of the ovules, following successful fertilization and commencement of endosperm cell division, allow these seedless fruits to be categorized as pseudo-parthenocarpic.  相似文献

The experiments were carried out between 2003 and 2005 in the Province of Amasya. The first objective of the study was to determine the best pollinizer for the cultivar ‘0900 Ziraat’ which is the most popular sweet cherry cultivar (Prunus avium L.) in Turkey. The ‘0900 Ziraat’ was used as a female parent and ‘Türko?lu’, ‘Köro?lu’, ‘Kargayüre?i’, ‘Hac? Ali’, ‘Starks Gold’ and ‘Geçkiraz’ were used as pollinizer cultivars. In 2003, for the hand cross pollination combinations, fruit set was between 31.25% and 42.80%. In 2004, fruit set was reduced by spring frost and was between 9.69% and 15.24%. Additionally, the blooming periods of the experimental cultivars, fruit set of open pollination and hand self pollination were examined. The fruit set ratios of all tested cultivars under open pollination conditions varied between 26.73% and 53.49% in 2003 and 4.38% and 38.35% in 2004. ‘Geçkiraz’ had the highest fruit set owing to its late flowering date. For open pollination, fruit set values of ‘0900 Ziraat’ were lower than hand cross pollination with other cultivars in 2003 when the results of both treatments are compared. On the contrary, fruit set was higher than hand cross pollination in 2004. In the present study, pollen germination values varied greatly among year, cultivar and sucrose concentration. Viability percentages were generally high, not less than 66%. According to phenological observations, partial or complete overlapping of the flowering period of ‘0900 Ziraat’ and its possible pollinizer cultivars were observed in 2003 and 2005. But in 2004, the flowering period overlapped for only ‘0900 Ziraat’ and ‘Geçkiraz’ cultivars. When blooming times are taken into consideration, ‘Geçkiraz’ was found to be the best pollinizer cultivar.  相似文献

An apple orchard consisting of a single cultivar under the condition of natural pollination must have suitable pollinizers such as Crab apples to ensure stable fruit production. We selected ‘Maypole’ and ‘Dolgo’ as pollinizers for the cultivar ‘Fuji’, and investigated the rate of fruit and seeds in ‘Fuji’ fruits produced by pollen of the pollinizers. We developed a method for tracing pollen flow based on the leaf color of progeny and S-RNase allele of ‘Maypole’, and on Simple Sequence Repeat (SSR) analyses of ‘Maypole’ and ‘Dolgo’. These were powerful tools for determining the distance insects (mainly Osmia cornifrons) carry pollen from the pollinizers to ‘Fuji’. Although the fruit set of ‘Fuji’ apples was not reduced with increasing distance between ‘Fuji’ apple trees and pollinizers (probably due to pollen flow from other commercial cultivars planted outside the area), the rate of ‘Fuji’ apple fruit produced by the pollen of the pollinizers decreased with increasing distance. The rate of fruit produced by the pollinizers was 84% and 77% when ‘Fuji’ was 2.5 m from ‘Maypole’ and ‘Dolgo’, respectively, and 71% and 64% when ‘Fuji’ was 5 m from ‘Maypole’ and ‘Dolgo’, respectively, but was reduced to 47% and 39% when ‘Fuji’ was 10 m from ‘Maypole’ and ‘Dolgo’, respectively. However, the spacing could cause reduced fruit size, and require extra fruit thinning for producing large fruits. It could also cause reduction of the yield for increasing the planting area of pollinizers. We recommend that pollinizers should be planted not more than 10 m from ‘Fuji’.  相似文献

Effects of suspension media used for spray pollination on pollen grain viability were investigated in Japanese pear (Pyrus pyrifolia (Burm.f.) Nakai ‘Kosui’). The suspension media tested in this study consisted of pectin methylesterase (PME) and polygalacturonase (PG) combined with either 0.1% agar or 0.04% xanthangum (XG). We also evaluated the influences of spray pollination on fruit set and fruit quality. Pollen grain viability, as measured by germination and pollen tube growth rates, was maintained in media supplemented with 0.1 mg L−1 PME or 0.1 mg L−1 PG. The level of fruit set after spray pollination using media containing PME or PG in combination with either 0.1% agar or 0.04% XG was almost the same as the level after artificial pollination by hand (hand pollination). The media containing XG combined with either PME or PG seemed to show better results for pollen grain viability and fruit set, although the results were variable from year to year. With regard to fruit size, shape and other parameters for fruit quality, spray pollination and hand pollination gave comparable results, irrespective of the medium composition. The time required for spray pollination was less than half of that required for hand pollination, and the amount of pollen grain required for spray pollination was one-third or less than the amount required for hand pollination. Thus, spray pollination may be a time- and labor-saving pollination system for the cultivation of Japanese pear.  相似文献

An improved in vitro pollen germination assay was developed to assess the viability of stored Hedychium pollen. The effect of polyethylene glycol (PEG) (10, 15, and 20%, w/v) on pollen germination and tube growth was evaluated for Hedychium longicornutum and two commercial Hedychium cultivars, ‘Orange Brush’ and ‘Filigree’. Overall, the inclusion of PEG 4000 in the medium improved both pollen germination and tube growth for the three different genotypes tested and the results varied depending on genotype. In vitro germination was used to assess the viability of Hedychium pollen stored up to two months. Pollen nucleus status was determined for four Hedychium cultivars, ‘Orange Brush’, ‘Anne Bishop’, ‘Filigree’, and ‘Daniel Weeks’. Pollens of ‘Orange Brush’, ‘Anne Bishop’, and ‘Daniel Weeks’ were found to be binucleate but ‘Filigree’ was shown to possess both binucleate and trinucleate pollens. High pollen:ovule ratio values were obtained in several Hedychium taxa. The results obtained on the nuclear pollen status and pollen:ovule ratios will further our understanding of the pollination biology and help clarify the taxonomy and phylogeny of Hedychium species.  相似文献

‘Success’ or ‘Red Splendor’ plants propagated by shoot tip culture or embryogenesis had higher pollen germination than those propagated by cutting. All clones of ‘Pepride’, ‘Peter Star’ and ‘Nobel Star’ propagated by cutting or by embryogenesis or shoot tip culture did not have viable pollen. ‘Peter Star’ mutants through mutagenesis could recover pollen viability. When both parents were propagated by tissue culture, fruit set increased compared to when one or both parents were propagated by cuttings. Furthermore, the number of the harvested seeds doubled per pollination set, when the tissue culture parents were used instead of cutting-propagation plants.  相似文献

The fact that rainy weather negatively affects production in almond is well-known by growers. However, as far as we know, no previous study has been carried out in this species to determine if the rain is able to wash off pollen grains from the stigma surface and consequently affect fertilization efficiency. To answer this question, flowers of the self-compatible almond cultivar ‘Antoñeta’ were emasculated on branches of trees in the field, pollinated by hand with pollen of the cultivar ‘Marcona’, and sprayed with water to simulate the rain effect at 4, 8, 24 and 48 h after pollination. A control for which no treatment was applied was also included in the assay. Seven days after spraying the flowers, the number of germinated pollen grains on the stigma was recorded in 10 pistils for each treatment by fluorescent microscopic observation. In addition, for each treatment the initial and final fruit set were determined at 30 and 60 days after pollination, respectively. The following year additional assays were performed in the field and in the laboratory in order to complete the study with earlier treatment times. Thus, an assay of rain simulation in the field at 0 and 2 h, and other consisting on immersing a branch with flowers and shaking it in a water container in the laboratory at 0, 30, 60 and 120 min after pollination were performed. One sample more was immersed, shaken, then pollinated and again immersed and shaken. The results from field assays showed that adhesion of pollen grains to the stigma was very quick and strong, and the treatments did not significantly affect pollen–stigma adhesion and subsequent fruit set. On the other hand, laboratory results showed that all washing treatments decreased the number of germinated pollen grains on the stigma, mainly when the flowers were immersed before pollination. In conclusion, the results showed that the simulated rain was not able to completely wash off pollen grains from the stigma surface, although it seems to affect adhesion in forthcoming pollinations.  相似文献

The caryophyllaceae is a family of flowering plants commonly known as the pink or carnation family. Members of this family are widely grown as ornamental plants. The species Dianthus chinensis, Dianthus barbatus, and Dianthus superbus include notable examples of such ornamental varieties. Intra- and inter-specific hybridization is important for the breeding of improved varieties and, therefore, it is desirable to optimize the efficiency of cross-pollination procedures within this family. Here, we conducted intra-specific pollination using four lines of D. chinensis, and inter-specific pollination between D. chinensis, D. barbatus and D. superbus genotypes. For each of these crosses we recorded information of pollen viability, stigma development stage at time of pollination, pollen germination on the stigma surface and pollen tube elongation within the stigma, and final seed set. Pollen viability levels were determined at the start of anther dehiscence, which occurred on day 1 of flower opening. The genotypes D. chinensis var. ‘Bz2’, D. barbatus var. ‘Xb’, and D. superbus wild species ‘Qm’ all displayed good levels of pollen viability (i.e. between 68% and 75%); D. chinensis vars. ‘H58III’, ‘H68I’ and ‘H6II’ showed lower levels of pollen viability (i.e. between 32% and 51%). In each Dianthus line, the stigma organs were observed to undergo morphological changes during the 1–7 d following flower opening. We have characterized these changes as five distinct development stages, namely, ‘column’ (day 1), ‘Y-like’ (day 2), ‘Y-shaped with slight curve’ (days 3–4), ‘Y-shaped horn-like’ (days 5–6), and ‘wilting’ (day 7). Pollen was artificially applied to stigma organs at each of these development stages and aniline blue staining was used to follow pollen tube germination on the stigma surface and pollen tube elongation down the length of the stigma. This showed that the ‘Y-shaped slightly curved’ stigma stage (3–4 d) was optimal for pollination. The ‘Y-shaped horn-like’ stage (5–6 d) was also receptive to pollination. By contrast, the earlier ‘Y-shaped’ (2 d) and ‘column’ (1 d) stages, and also the later ‘wilting’ (7 d) stage, were far less receptive. Parallel field experiments were conducted in which seed set was measured following the artificial pollination of flowers at different development stages. These data also indicated that the optimal time for pollination was during 3–4 d of flower opening. This conclusion held true for both intra- and inter-specific crosses of the various Dianthus lines. Thus, stigma receptivity in Dianthus does not become optimal until 3–4 d after flower opening. Since anther dehiscence occurs at 1 d (i.e. when stigma are at the unreceptive ‘column’ stage), this suggests that Dianthus employs a delayed self-fertilization strategy. We also found a significant effect of genotype on pollen tube growth and seed-set values. The highest numbers of seed per flower were achieved when the D. superbus wild species ‘Qm’ was the female parent in inter-specific crosses. Thus, genotype and stigma development are identified as key factors that determine the success of cross-pollination in Dianthus lines.  相似文献

The ‘Mopanshi’ persimmon is a seedless, astringent parthenocarpic cultivar that does not produce male flowers. After pollination using four non-astringent cultivars (‘Zenjimaru’, ‘Nishimurawase’, ‘Okugosho’ and ‘Hanagosho’), seeds were produced to different degrees. ‘Mopanshi’ fruits pollinated with ‘Zenjimaru’ produced far more seeds than those pollinated with the other three cultivars. The ratio of abnormal seeds obtained from the fruits pollinated with ‘Hanagosho’ was higher than that obtained from the fruits pollinated with the other three cultivars. Most embryos degenerated in the early to late stages of seed development. Immature embryos were cultured in a modified MS medium (half of NO3 in MS medium + 0.4 μM BA + 0.1 μM IBA) with the greatest success (52–80%) from embryos taken from fruits 60–80 days after pollination. Seedlings failed to initiate radicles so they were transferred to dark culture conditions for 8 days or to a rooting media that contained 3% sucrose and 1% Chinese ink. The seedlings on the medium darkened with ink rooted at greater than 90.83% compared to 75.83% for dark cultured seedlings. This study demonstrated that ‘Mopanshi’ persimmon could be used as a female parent in crosses, but embryos needed to be moved to tissue culture conditions to continue to develop and form plantlets.  相似文献

In this work, pea (Pisum sativum) plants exposed to increasing cadmium and copper concentrations were tested for heavy metals accumulation in flowers and for ‘in vivo’ pollen germination. Based on the Cd and Cu accumulation amounts in the flowers, an evaluation of the same metals effects on ‘in vitro’ pollen germination was achieved. Moreover, the effects of both metals on fruits number and weight and on seed set and yield at individual plant level were examined. While cadmium concentrations did not affect ‘in vivo’ pollen germination, only higher copper concentrations rendered a significant reduction. This is in contrast with the clear negative effect on pollen germination in vitro and might be explained by the different dynamic and bioavailability of both metals. A clear effect of Cd and Cu was observed on two important yield components ie, fruit weight and seed set. Although results obtained herein cannot give a clear cut relationship between the effect of Ca and Cu on reproductive development and its consequences on yields, they represent emerging results on the potential consequences of metals contamination on reproductive development in plants.  相似文献

Fruit traits and molecular markers (RAPD) were used to survey genetic similarities and inheritance pattern of offspring, derived from self- and open pollination of pomegranate cv. Malase-Tourshe-Saveh (MTS) as well as progenies derived from cross between ‘MTS’ and ‘Narm-Daneh’ (ND) a soft-seed cultivar in the present study. Clustering based on fruit traits, divided the accessions firstly into two major groups, sweets and sour or sweet–sours. Members of sub-clusters were different in fruit characters, and seed softness also was affecting the grouping of individuals. Offspring of ‘MTS’ × ’ND’ showed somehow seed softness with varying degrees compared to other individuals. In RAPD analysis, by using 26 polymorphic primers, 325 fragments were produced from 39 individuals, among which 70 reproducible, polymorphic bands were scored for data analysis. NTsys and Bootstrap software were used for RAPD data analysis and respective dendrogram depicted by Jaccard's similarity coefficients using UPGMA method. The similarity coefficients were ranging from 0.2 to 0.94 and the lowest was obtained between pollen parent (ND cv.) and a genotype from open pollination of MTS cultivar. The highest similarity coefficient (0.94) was observed between two offspring both from self-pollination of MTS cultivar. In corresponding dendrogram, pollen parent was separated from other genotypes and laid individually in a sub-cluster, while the mother plant clustered with its progenies. In addition, some marker–trait associations were observed, especially for seed softness.  相似文献

Prevailing ambient temperature during the reproductive phase is one of important factors for seed and fruit set in different plant species. In mango (Mangifera indica L.), natural low temperatures during flowering induced seedless fruits. Here the sexual reproduction process of ‘Tainong 1’ mango at low temperatures (diurnal maximum temperature < 20 °C) was studied. For comparison, we also examined this process at “normal” temperatures (diurnal maximum ranging from 25 to 30 °C, diurnal average temperature > 20 °C). Results showed: natural low temperatures significantly affected pistil and male gametophyte development, resulting in pollen grains with low viability. Meiotic chromosomal irregularities, including univalents, multivalents, laggards, bridges and micronuclei were detected at higher incidences and significantly greater proportions of nucleolus fragmentation and dissolution were detected when temperatures were low. Pollen tube growth was retarded under low temperature stress, either in vivo or in vitro. The virtual absence of sexual reproduction of ‘Tainong 1’ mango at low temperatures appears to be due largely to slow growth of pollen tube in vivo and to a low rate of successful fertilization.  相似文献

Female pistachio (Pistacia vera) trees were hand pollinated with pollen from three different male genotypes 0,1,2,3 and 4 d after anthesis. Results were measured by mature fruit set and by fluorescence microscopy of pollen germination and tube growth in the pistil. Maximum pollen tube growth and fruit set of split nuts was achieved following pollination within two days of anthesis. Three day old pistils supported pollen tube growth, but fruit set was low. Four day old pistils supported little pollen tube growth or fruit set. Significant differences were also apparent between pollen parents, with low pollen tube growth and fruit set following interspecific pollination with P. atlantica pollen. All pollen tubes were observed to penetrate chalazogamously, and parthenocarpic production of blank fruits occurred in both unpollinated and pollinated treatments. The results indicate that for optimum fruit production of pistachio, pollen transfer must be achieved within 2 d of anthesis and pollination by P. atlantica should be avoided.  相似文献

Cherimoya (Annona cherimola (Mill.)) is a subtropical fruit tree, which is cultivated in a good range of subtropical regions. In most of these areas the crop relies on hand pollination. However, following this practice, erratic fruit set is often produced, which could be related to problems in pollen handling. Indeed, very little is known of the time that the pollen remains viable and on which is the best stage to collect the anthers or pollen from the flower. The aim of this work is to evaluate pollen germinability prior and after anther dehiscence and also how the age of pollen affects pollen vigor, understood as speed of germination. Pollen samples at different times following anther dehiscence were germinated in vitro and in vivo. Pollen up to 90 min following dehiscence performed as well as freshly dehisced pollen. However, the pollen taken 120 min following dehiscence, showed a clear reduction in vigor and germinated much slower in vivo. To overcome this short pollen germinability, pollen was taken from anthers 30 and 5 h prior to natural anther dehiscence and compared with pollen taken at anther dehiscence and 20 h later. However, a reduction in germination rate was obtained in pollen taken prior to anther dehiscence. The narrow stage at which pollen can be collected together to its ephemeral germinability explains erratic results obtained following hand pollination in this crop and these results provide the clues for an adequate pollen handling.  相似文献

Species of barberry (Berberis vulgaris L. var. asperma) is cultivated in arid and semi arid areas of Iran (South Khorasan province). It is widely used as a food additive. Fruits of this species are seedless, while wild type barberries produce seeds in the same area. In this study, we investigated the mechanism of seedlessness in seedless barberry by pollen viability test, field pollination experiments and microscopic observation of pollen tube growth in pistil and ovule development. For comparison, we also examined ovule development in wild type barberry (B. crataegina DC). In seedless barberry pollen germination was about 54%. Seedless barberry produced 20% seeded fruits when pollinated with pollen of wild type barberry. There was a sharp decrease in fruit set in emasculated unpollinated flowers of seedless barberry. In seedless barberry, a large number of pollen grains (about 370) were observed on stigma of each flower at 12 h after balloon stage (ABS). Most of them germinated and penetrated intracellular area of stigma surface, but no pollen tube reached ovary. In seedless barberry, many ovules did not have any embryo sac or had a very small incomplete embryo sac. In addition, unfused polar nuclei were clearly recognized in some cases at 14 days after full bloom (AFB). However, in wild type, double fertilization was accompanied by disappearance of polar nuclei. In seeded barberry, the cellularized endosperm became apparent at seven days AFB. At 21 days AFB, all ovules of seedless barberry were degenerated, while at the same time in wild type, one or two ovules of each flower were normal and were developing into complete seeds. Results showed that self-incompatibility has a main role in seedlessness of seedless barberry. However, the high frequency of abnormal ovules and single fertilization can be considered as two other reasons of seedlessness. Apple (Malus x domestica Borkh.) shows gametophytic self incompatibility and requires cross pollination by suitable pollinizers. Gradual decline in natural pollinators and insufficient proportion of pollinizers have been observed in the apple orchards over the years across the North Western Himalayan region. This situation resulted in considerable pollination problem in the region. The present study was undertaken (during the years 2006–2010) to examine the effect of top working methods and time as well as type of pollinizers on different growth parameters for efficient pollination management. Chip budding performed in fall, late winter and summer gave significantly higher success rate (90.0, 89.8 and 80.9%, respectively). Other methods viz., side rind graft (85.6%) and cleft graft (71.2%) during February–March, and T-bud (71.7%) during July–August also gave better success. The highest shoot numbers (4.1) and length (92.8 cm) were recorded when chip budding was performed during September–October and February–March, respectively. In 4th year, chip budded scion branches of all the pollinizers recorded significantly higher number of spurs/m twig length with a range of 22.8 (December–January) to 24.7 (September–October). Significantly the highest bloom density (22.1) was recorded on chip budded (February–March) branches of pollinizers in 4th year. Fruit set on top worked trees of ‘Oregon Spur’ was only in the range of 18.0–20.1% in 1st year and reached to the range of 35.6–41.5% in 4th year. Seed numbers/fruit also increased to the range of 7.3–7.9 in 4th year from 2.9 to 4.6 in 1st year. All these factors resulted in better yield efficiency (2.6–3.8) of ‘Oregon Spur’ in 4th year. ‘Manchurian’ crab produced significantly higher shoot length of 86.5–87.2 cm and 146.1–149.2 cm in 1st and 4th years, respectively. However, ‘Stark Spur’ produced the highest number of spurs/m twig length of 28.9 when grafted/chip budded during December–January or February–March, and 27.1 when chip/T-budded during September–October or July–August. Top working with ‘Manchurian’ crab also resulted in significantly higher fruit set (44.2–45.4%) and yield efficiency (3.6–3.8) on ‘Oregon Spur’ in 4th year. This was followed by ‘Stark Spur’ as pollinizer (37.1–38.2% and 2.9–3.1, respectively). Highly significant positive correlation of shoot numbers, spur density and bloom density of pollinizers with fruit set and yield efficiency of ‘Oregon Spur’ were observed. ‘Manchurian’ crab was found to be very efficient as pollinizer, followed by ‘Stark Spur’ for spur type ‘Oregon Spur’ apple cultivar on the basis of higher bloom density and fruit set parameters.  相似文献

 以‘富士’× 新疆红肉苹果杂交F1群体中的2个绵肉株系‘绵肉1号’、‘绵肉2号’和2个脆肉株系‘脆肉1号’、‘脆肉2号’不同发育时期的果实为试材,测定其果实硬度、脆度、乙烯释放量和细胞壁降解酶活性,分析探讨它们之间的相关性。结果表明:①2个脆肉株系各个时期的果实硬度和脆度均显著高于2个绵肉株系;②在幼果期和果实膨大期的乙烯释放量均小,绵肉株系与脆肉株系间差异不明显,在花后130 d的成熟期,2个绵肉株系的乙烯释放量是2个脆肉株系的10倍;③2个绵肉株系果实的果胶酶和β–半乳糖苷酶活性绝大部分发育时期高于2个脆肉株系,而α-L–阿拉伯呋喃糖苷酶、β–木糖苷酶、淀粉酶和脂氧合酶的活性在发育前期差异较小,到发育后期2个绵肉株系显著高于2个脆肉株系;④相关分析显示,绵肉脆肉特性与多聚半乳糖醛酸酶、α-L–阿拉伯呋喃糖苷酶及淀粉酶活性密切相关。  相似文献

Mal secco (caused by Phoma tracheiphila (Petri) Kantsch. and Gik.) is the most destructive fungal disease of lemon plantations worldwide and seedless lemons would be preferred by most consumers. Five dosage levels, 0, 3, 5, 7, and 9 kiloradian (krad), of cobalt (60Co) gamma irradiation were applied to budstick of ‘Kutdiken’ lemon (Citrus limon (L.) Burm. f.) clone KT-2A. Mutations were stabilized in three vegetative generations. Three hundred fifty-eight and 478 M1V3 (mutation one and vegetation three) plants were evaluated for seed number and mal secco tolerance in the field and the greenhouse, respectively. LD50 was around 5 krad gamma irradiation for ‘Kutdiken’ lemon. The seed number varied from 0 to 34 per fruit. The level of mal secco tolerance also varied significantly among the plants from 1.0 (no symptom) to 4.3 (high level of disease occurance). The stable seedless and mal secco tolerant plants were obtained from 5 and 7 krad irradiation: the three mutants from 5 krad irradiation gave more lemon-like fruits, while 7 krad irradiation caused altered tree morphology and early maturation of fruits. This study shows considerable potential for lemon cultivar improvement aiming to obtain seedless and mal secco tolerant lemons.  相似文献
