In order to clarify the functional relationship between thyroid, adrenal and gonadal hormones, hypothyroidism was induced by administration of thiuoracil in adult male and female rats, and the effects of hypothyroidism on the adrenal and the gonadal axes were investigated in the present study. 1. The functional relationship between thyroid and adrenal hormones: Adrenal weights and corticosterone were lowered, whereas the secretion of ACTH, corticotrophin-releasing hormone (CRH) and arginine vasopressin (AVP) increased in hypothyroid rats compared to euthyroid rats. These results indicate that hypothyroidism causes adrenal dysfunction directly and results in hypersecretion of CRH and AVP from the hypothalamus. 2. The functional relationship between thyroid and gonadal hormones: The pituitary response to LHRH was lowered, whereas the testicular response to hCG was not changed in hypothyroid rats. Hypothyroidism suppressed copulatory behavior in male rats. These results suggest that hypothyroidism probably causes dysfunction in gonadal axis at the hypothalamic-pituitary level in male rats. In adult female rats, hypothyroidism inhibited the follicular development accompanied estradiol secretion, whereas plasma concentrations of progesterone and prolactin (PRL) increased in hypothyroid female rats. Hypothyroidism significantly increased the pituitary content of vasoactive intestinal peptide (VIP) though it did not affect dopamine synthesis. These results suggest that hypothyroidism increases pituitary content of VIP and this increased level of VIP likely affects PRL secretion in a paracrine or autocrine manner. In female rats, inhibition of gonadal function in hypothyroid rats mediated by hyperprolactinemia in addition to hypersecretion of endogenous CRH.  相似文献

Thyroid hormones permit the annual reproductive transition of seasonal breeders. Although, precise function of thyroid hormones in seasonal breeding is not well understood. In the present study, we examined effects of hypothyroidism on the hypothalamus-pituitary-gonadal axis in adult male golden hamsters after transition of the short-day photoperiod (SD; 8 h light: 16 h dark) condition. We confirmed that hypothyroid, which had been induced by administration of thiouracil in drinking water for 4 weeks, did not have direct effects on testes in male hamsters under the long-day photoperiod. Plasma concentrations of free T3 and T4 decreased 15 weeks after transition of SD condition. Plasma concentrations of testosterone in the hypothyroid group decreased earlier than in the control group after the transition from LD to SD. In animals treated with testosterone after castration, plasma concentrations of LH in the hypothyroid group decreased earlier than in the control group after the transition of SD. On the other hand, pituitary response to GnRH for LH release did not change in castrated hamsters as a result of hypothyroidism. These results suggest that thyroid hormones act the hypothalamus and might be required to maintain GnRH secretion in male golden hamsters.  相似文献

Phenobarbital can interfere with the thyroid axis in human beings and rats by accelerating hepatic thyroxine metabolism because of enzyme induction. In human beings, it also can interfere with the low-dose dexamethasone suppression test (LDDST) used to assess adrenal function by accelerating dexamethasone metabolism. This effect can cause a lack of suppression of pituitary ACTH and subsequent adrenal cortisol release after dexamethasone administration. The effects of phenobarbital on the thyroid axis, the adrenal axis, and adrenal function tests were prospectively investigated in 12 normal, adult dogs. Phenobarbital was administered at 5 mg per kilogram of body weight (range, 4.8–6.6 mg/kg) PO q12h for 29 weeks, resulting in therapeutic serum concentrations (20–40 μg/mL). Serum total thyroxine (TT4), free thyroxine (FT4) by equilibrium dialysis, total triiodothyronine (TT3), thyrotropin (TSH), and cholesterol were determined before and during phenobarbital treatment. LDDST, ACTH stimulation tests, and ultra-sonographic evaluation of the adrenal glands were performed before and during treatment. TT4 and FT4 decreased significantly ( P ≤ .05), TT3 had minimal fluctuation, TSH had only a delayed compensatory increase, and cholesterol increased during phenobarbital treatment. The delayed increase in TSH, despite persistent hypothyroxinemia, suggests that accelerated hepatic thyroxine elimination may not be the only effect of phenobarbital on the thyroid axis. There was no significant effect of phenobarbital on either of the adrenal function tests. With the methods employed, we did not find any effects of the drug on the hormonal equilibrium of the adrenal axis.  相似文献

The objective of this study was to investigate the changes in secretion of inhibin and cellular localization of the inhibin alpha and inhibin/activin (beta(A) and beta(B)) subunits in male Japanese quail from 1 to 7 weeks after hatching. The post-hatch profile of plasma luteinizing hormone (LH), immunoreactive (ir) inhibin and testosterone were measured by radioimmunoassay. Testes were immunostained by the avidin-biotin-peroxidase complex method (ABC) using polyclonal antisera raised against inhibin alpha, inhibin/activin beta(A) and inhibin/activin beta(B) from one week of age to sexual maturity. Testicular weight increased gradually until 4 weeks and abruptly increased from 5 weeks of age onwards. The plasma concentrations of LH and ir-inhibin increased significantly at 5 weeks of age, and the plasma concentration of testosterone increased significantly at 6 weeks of age. Pituitary contents of LH showed a steady increase until 6 weeks of age and then abruptly increased at 7 weeks of age. Coincident to the increase in plasma testosterone, the testicular contents of testosterone significantly increased from 5 weeks through sexual maturity. Immunohistochemically, localization of the inhibin/activin alpha, beta(A) and beta(B) subunits was found in the Sertoli and Leydig cells at all ages of development from one week of age to sexual maturity. These results suggest that Sertoli and Leydig cells are the major source of inhibin secretion during development in male Japanese quail.  相似文献

1. Simultaneous changes of the width of the cloacal opening and plasma luteinising hormone (LH), follicle stimulating hormone (FSH), 17β-oestradiol, progesterone, prolactin, thyroxine (T4) and triiodothyronine (T3) during photo-induced ovarian growth and regression were measured in commercially bred Japanese quail from a heavy body weight line.

2. Somatically mature female Japanese quail were transferred from short days (light:dark 8L:16D) at 10°C to long days (16L:8D) at 20°C, and sexually mature female Japanese quail were transferred from long to short days. All variables were measured at transfer and every five days thereafter (except for a measurement at 12 instead of 10?d) for 35?d.

3. Transfer from short to long days caused significant increases in LH, FSH, 17β-oestradiol, ovary weights and oviduct weights after five days, and in the cloacal opening after 12?d. T3 decreased after five days, whereas no significant changes were observed in T4 concentrations. Progesterone and prolactin both decreased after 25 long days.

4. The transfer of quail from long to short days caused significant decreases in LH, FSH, 17β-oestradiol, progesterone, prolactin, ovary and oviduct weights after 12?d and an increase in T3. There was no significant change in T4 concentrations. The cloacal opening decreased after 25 short days.

5. These results are the first to show simultaneous changes in gonadotrophins, sex steroids, thyroid hormones and prolactin during photo-induced gonadal growth and regression in female Japanese quail.  相似文献

Hypothalamic-releasing factors regulate the secretion of anterior pituitary hormones. The anterior pituitary gland secretes the same six hormones as found in mammals: FSH, LH, prolactin, GH (somatotropic hormone), ACTH, and TSH, plus the melanotropic hormone. The endocrine hormones of the avian posterior pituitary gland concerned with reproduction are mesotocin and AVT. The pineal gland, through the secretion of the hormone melatonin, modulates the periodic autonomic functions of the central nervous system. The ovary produces estrogens, progestogens, and androgenic compounds. The testes produce testosterones and progesterone. The thyroid glands produce two hormones, T4 and T3. The avian adrenal glands produce corticosterone and aldosterone. The bursa of Fabricius is considered an endocrine organ since it is involved in the production of humoral factors. The male reproductive system undergoes hormonal changes associated with puberty, the breeding season, and molt. Some avian species undergo a type of disintegration and seasonal reconstruction of the testis and epididymis. The relationship of the ovarian follicular hormones and the plasma hormones varies depending on the stage of the reproductive cycle and the seasonal photostimulation. Female birds may conceive in the absence of a mate as a result of the fertile period phenomena. The blood chemistry of laying birds is different from that seen in nonlaying hens. Domestication has had a definite influence on the hormone cycles of some avian species. This may lead to certain reproductive problems.  相似文献

The effect of selection for high stress response on adrenocortical function was examined by measuring the corticosterone response of adrenocortical cells isolated from random-bred Japanese quail and quail selected for high serum corticosterone response to immobilization (high-stress). Highly enriched adrenocortical cells were incubated with various concentrations of ACTH1-24 (ACTH), 8-bromo-cyclic AMP (8Br-cAMP) and pregnenolone for 2 hr. Corticosterone production was measured by radioimmunoassay. Basal corticosterone production values by cells from random-bred and high-stress birds were not different. In contrast, the average maximal ACTH- and 8Br-cAMP-induced corticosterone production by cells from high-stress quail was 89% greater than that of cells from random-bred quail. However, the average pregnenolone-supported corticosterone production by cells from high-stress birds was 34% less than that of cells from random-bred birds. Thus, the data suggest that although random-bred quail cells had a greater potential capacity for corticosterone production, high-stress quail cells had a greater ability to couple ACTH, ACTH-transmembrane-signaling factors and subsequent second messengers with the available steroidogenic enzyme pool. The magnitude of the differences in function between cells from high-stress and random-bred birds was greater for female cells compared to male cells. In addition to differences in cellular function, there were also differences in adrenal and relative adrenal weights between random-bred and high-stress quail. The average, adrenal and relative (mg/100 g body weight) adrenal weights of high-stress quail were 14-16% greater than those of random-bred quail. It is concluded that the enhanced serum corticosterone response of the high-stress quail line is, in part, due to an increase in relative adrenal weight and an increase in adrenocortical cell responsiveness to ACTH.  相似文献

Fecal corticosterone metabolites and plasma corticosterone in Japanese quail selected for low- or high-plasma corticosterone responses to brief mechanical restraint (low- and high-stress lines), and in a line of unselected quail, were measured in this study. No line differences were observed in baseline plasma corticosterone concentrations, but fecal corticosterone metabolite concentrations and daily fecal corticosterone metabolite production were 20% higher in quail of the high-stress line than in unselected or low-stress quail for males and females living together in group cages (P < 0.05). No differences were observed between lines in corticosterone metabolite concentrations and production for male birds in individual cages. Baseline plasma corticosterone concentrations, fecal corticosterone metabolite concentrations, and production appeared to be higher for males and females in group cages compared with males in individual cages. This difference might have been because of greater corticosterone secretion by male quail living in mixed sex groups than living individually. Correlations between baseline plasma corticosterone concentrations and fecal corticosterone metabolite concentrations in low-stress and high-stress quail, and for all birds combined, were r = 0.521 (P = 0.038), r = 0.604 (P = 0.013), and r = 0.431 (P = 0.002), respectively. The low- and high-stress lines that have been selected for low- and high-corticosterone responses differ in other characteristics, including growth and reproductive performance, and the current results are consistent with the assumption that these other differences are a consequence of greater daily corticosterone secretion in quail of the high-stress line.  相似文献

In relevance to osmoregulatory and reproductive functions, activity of hypothalamic neurosecretory neurons may also vary seasonally. The current study was performed to determine annual changes in ir-AVT neurons of hypothalamus and adrenal gland function. We examined changes in ir-AVT neuron by immunohistochemical method and plasma testosterone was measured by enzyme immunoassay. The steroidogenic interrenal activity was studied by histological and biochemical methods. Birds were sampled in February (quiescent), April (recrudescent), June (breeding) and November (regressive). A significant and gradual increase in the number of ir-AVT neurons was observed from quiescent to breeding phase which decreased during regressive phase of annual gonadal cycle. The gradual increase in ir-AVT neurons along with annual gonadal activity of quail were accompanied by increase in plasma levels of testosterone. These results indicate a functional interaction between sex steroid and AVT synthesizing neurons. Adrenal activity (as judged by weight, ascorbic acid content, cortical cord width and cortico-medullary ratio) was also maximum during breeding phase. It is thus postulated that domesticated quail when exposed to natural day length (NDL), exhibits seasonal/annual cyclicity in vastocinergic activity and adrenal function which may be due to difference in sex steroid hormone.  相似文献

In a previous study, we found that 3-methyl-4-nitrophenol (PNMC), a component of diesel exhaust particles and also a degradation product of the insecticide fenitrothion, exhibits reproductive toxicity in the adult male Japanese quail. The present study investigated the toxicity of PNMC in the female Japanese quail and its ability to influence reproduction in immature females. The quail (21-day-old) were injected intramuscularly (im) with PNMC at doses 0.1, 1 or 10 mg/kg body weight daily for 3 days. There was no significant difference in body growth between the PNMC-administered and control birds. However, the weights of the oviducts were significantly lower in the PNMC-treated birds at all doses. Furthermore, the plasma concentrations of luteinizing hormone (LH) and estradiol-17 beta were significantly decreased with 1 and 10 mg/kg of PNMC. These findings suggest that PNMC might influence the hypothalamo-pituitary-gonadal axis with decreasing in secretion of GnRH, LH and ovarian steroid hormones and subsequently disturb growth of the reproductive organs of immature female quail. This study indicates that PNMC induces reproductive toxicity at the central level and disrupts reproductive function in the immature female quail.  相似文献

4. Transferring quail from long to short days caused significant decreases in LH and FSH after 5?d, and decreases in T, TW and CGA after 10?d. T4 decreased after 5?d whilst T3 increased significantly by day 15. PRL decreased significantly after 10?d then rose before declining again. The testicular regression rate was k?=?0·0582.

5. The rates of photo-induced testicular development and regression in a strain of large Japanese quail did not differ from rates reported for other strains of quail. CGA was a better indicator of TW than plasma T concentrations during growth and regression. The role of PRL in photo-induced reproductive cycles in male Japanese quail remains to be determined.

6. The photoperiod-induced changes in gonad size and hormone concentrations, together provide valuable information that can be used in future studies of the endocrinology and neuroendocrinology of photoperiodism in birds.  相似文献

The present study investigates the effects of temporal synergism of 5-hydroxytryptophan (5-HTP) and l-dihydroxyphenylalanine (l-DOPA) and pineal function (pinealectomy and melatonin administration) on gonadal response and nitric oxide (NO) activity in Japanese quail. Three-week-old male quails were divided into 9 groups, as follows: (I) control (treated with normal saline); (II) 8-h and (III) 12-h (receiving l-DOPA after the administration of 5-HTP [5 mg/100 g body weight] at the interval of 8 and 12 h, respectively, over a period of 13 days); (IV) SO (sham operated); (V) Px (pinealectomized); (VI) Veh (vehicle treated control); (VII) Mel (melatonin 25 μg/100 g body weight for 45 days); (VIII) 12-h + Veh; and (IX) 12-h + Mel (same as in (III) but further treated with the vehicle or melatonin, respectively, for 32 days). Body weight and cloacal gland volume were recorded weekly, whereas spermatogenesis, plasma testosterone concentration, and total nitrite and nitrate concentration in plasma, hypothalamus, and testes were measured at the termination of the study. Results indicate that compared to the controls, 8-h, Mel, and 12-h + Mel treatments suppressed whereas 12-h and Px stimulated the reproductive system. Further, a significant increase in plasma testosterone, total nitrite, and nitrate in the 12-h and Px groups and a decrease in these concentrations in 8-h, Mel and 12-h + Mel quail were observed. It is evident that 5-HTP and l-DOPA given at the interval of 8 h, as well as melatonin, are potent inhibitors of gonadal development, and the gonado stimulatory effect of the 12-h temporal relationship of the neurotransmitter precursors is masked by the inhibitory effect of melatonin. It is concluded that modulation of the daily phase relationship of neural oscillations and pineal function alters gonadal function and NO activity. Further, NO exhibits a parallel relationship with gonadal function in Japanese quail; this relationship appears to be mediated through NO activity, suggesting a causal relationship, although the mechanism between the 2 systems remains a topic of discussion.  相似文献

The present study was conducted to determine the effects of supplemental dietary l-carnitine at different levels on semen traits, reproductive parameters, and testicular histology in male Japanese quail breeders. Forty-five 5-wk-old male Japanese quail breeders were fed the same basal diet that was supplemented with 0 (control), 250, or 500 mg of l-carnitine/kg of diet. There were no significant effects of dietary l-carnitine supplementation at different levels on BW, feed intake, testes weight, fertility rate, hatchability rate of set and fertile eggs, and malonaldehyde production (μg/mL of semen) of male Japanese quail breeders. However, the supplementation of dietary l-carnitine at levels of 250 or 500 mg/kg to a basal diet significantly increased sperm viability and decreased multinucleated giant cells per testes in mature male Japanese quail breeders. Additional studies are required to explore the antioxidant role that l-carnitine has in Japanese quail breeders.  相似文献

BACKGROUND: Differentiation between hypothyroidism and nonthyroidal illness in dogs poses specific problems, because plasma total thyroxine (TT4) concentrations are often low in nonthyroidal illness, and plasma thyroid stimulating hormone (TSH) concentrations are frequently not high in primary hypothyroidism. HYPOTHESIS: The serum concentrations of the common basal biochemical variables (TT4, freeT4 [fT4], and TSH) overlap between dogs with hypothyroidism and dogs with nonthyroidal illness, but, with stimulation tests and quantitative measurement of thyroidal 99mTcO4(-) uptake, differentiation will be possible. ANIMALS: In 30 dogs with low plasma TT4 concentration, the final diagnosis was based upon histopathologic examination of thyroid tissue obtained by biopsy. Fourteen dogs had primary hypothyroidism, and 13 dogs had nonthyroidal illness. Two dogs had secondary hypothyroidism, and 1 dog had metastatic thyroid cancer. METHODS: The diagnostic value was assessed for (1) plasma concentrations of TT4, fT4, and TSH; (2) TSH-stimulation test; (3) plasma TSH concentration after stimulation with TSH-releasing hormone (TRH); (4) occurrence of thyroglobulin antibodies (TgAbs); and (5) thyroidal 99mTcO4(-) uptake. RESULTS: Plasma concentrations of TT4, fT4, TSH, and the hormone pairs TT4/TSH and fT4/TSH overlapped in the 2 groups, whereas, with TgAbs, there was 1 false-negative result. Results of the TSH- and TRH-stimulation tests did not meet earlier established diagnostic criteria, overlapped, or both. With a quantitative measurement of thyroidal 99mTcO4(-) uptake, there was no overlap between dogs with primary hypothyroidism and dogs with nonthyroidal illness. CONCLUSIONS AND CLINICAL IMPORTANCE: The results of this study confirm earlier observations that, in dogs, accurate biochemical diagnosis of primary hypothyroidism poses specific problems. Previous studies, in which the TSH-stimulation test was used as the "gold standard" for the diagnosis of hypothyroidism may have suffered from misclassification. Quantitative measurement of thyroidal 99mTcO- uptake has the highest discriminatory power with regard to the differentiation between primary hypothyroidism and nonthyroidal illness.  相似文献

Effect of coccidiosis on reproductive maturation of male Japanese quail   总被引:1,自引:0,他引:1  
The effects of coccidiosis on reproductive development of male Japanese quail were examined. Male Japanese quail were exposed to high (5 x 10(5) sporulated oocysts/quail) or low (5 x 10(3) sporulated oocysts/quail) doses of Eimeria uzura at 16 or 30 days of age and sampled at 37 days. Quail given high doses of coccidia had reduced testes weight and lowered circulating concentrations of androgen compared with control males. Low doses of coccidia did not affect testes weight but did result in elevated plasma androgen levels. There were no differences in average testes weights by 51 days; however, plasma androgen was still reduced in most groups. To study the effects of coccidiosis on egg production, males exposed to high doses of coccidia at 16 (16H) or 30 (30H) days of age were mated with control females, and control males were mated with control or 16H females. The onset of laying was delayed for 5 days in the control male: 16H female group. During the first week of production, eggs from females bred to 30H males had lower fertility and hatchability than those bred to control or 16H males. By the third week of production, levels of fertility were similar. Apparently, exposure of quails to coccidiosis before sexual maturation might result in long-term effects on later reproductive capability.  相似文献

本实验对内蒙古二狼山白山羊遗传性甲状腺肿(Inherited goitre,IHG)杂合子羊进行了甲状腺吸~(131)碘率测定;对IHG纯合子羊和杂合子羊进行了血清总T_3(TT_3)、总T_4(TT_4)、反T_3(rT_3)、游离甲状腺激素指数(FTI)以及促甲状腺激素(TSH)等甲状腺功能参数测定.结果表明,IHG杂合子羊的甲状腺吸~(131)碘率及吸~(131)碘速率降低(P<0.01);IHG纯合子羊和杂合子羊的血清TT_3、TT_4、FT_3I及FT_4I均显著降低(P<0.05或P<0.01),而TSH显著升高(P<0.01).据此认为,这些甲状腺功能参数(特别是TT_3和FT_3I)的改变,是山羊IHG的生化表型特征,可用于IHG基因群体中杂合子携带羊的检出.  相似文献

The purpose of the present study was to investigate commercially available ELISA methods designed for the determination of total and unbound thyroxine (TT(4) and FT(4)) and total triiodothyronine (TT(3)) in human serum for their usefulness in evaluating thyroid function in dogs when data describing the biological variation were included in the characterization of the assays. The TT(3) analysis was evaluated with intraassay coefficients of variation (CV%) ranging between 12% and 20%, and interassay CV% ranging between 5 and 17% at naturally occurring TT(3) concentrations. At concentrations around the limit of detection (0.27 nmol/l) CV% was considerably higher (99%). The analysis exhibited a satisfying accuracy since the recovery of added TT(3) was not different from unity and since parallelism between the dose-response curve and plasma dilutions could be verified. Determination of TT(4), TT(3) and FT(4) in eight normal dogs during 4 weeks resulted in a significant variation between dogs and between weeks in the individual animals (p < 0.01 in all cases). From the inter- and intraindividual CV%, quality goals for the maximally allowed analytical variation could be computed to be 8.4, 10.0, and 10.1% for individual testing of animals, and 12.0, 12.9, and 15.8% for screening for diseased animals in healthy populations for TT(4), TT(3) and FT(4), respectively. A comparison between quality goals derived from the inter- and intraindividual CV% and the measured analytical CV% (4.0, 17.3, and 6.7%, respectively) evidenced that TT(4) and FT(4) analyses fulfilled the requirements for analytical precision, whereas the TT(3) analysis could not be accepted as an effective tool for the evaluation of thyroid function in dogs due to too high analytical variation.  相似文献

This study aimed to describe the association between incidence of boar taint and pubertal changes in gonadal hormones, size of reproductive organs and aggressive behaviour in entire male pigs. In total, 111 entire male pigs were included in the study. Sampling was performed first at 90 kg live weight (LW) and, then, at 115 kg LW. Variables measured were skatole and androstenone levels in plasma and fat, testosterone and oestrone sulphate in plasma, free oestrone in fat, weight of testes and length of bulbourethral glands. Aggressive interactions between pigs were registered when a limited amount of feed was provided to the pigs prior to routine feeding. The number of initiated interactions (attacks) and the difference between number of initiated and received interactions (relative attacks) were calculated for each pig. Multivariate analysis revealed that gonadal hormones and reproductive organ size influenced prevalence of boar taint, accounting for 30% of the variation in skatole levels in fat and for 37% of the variation in androstenone levels in fat. These relations were independent of aggression levels in entire male pigs. Skatole levels were influenced by the levels of oestrone sulphate in plasma and free oestrone in fat, but not levels of plasma testosterone. Pigs with testes weight below 565 g and a bulbourethral gland length <90 mm did not produce high amounts of skatole; therefore, these values can be used as a threshold level to detect pig carcasses with low skatole levels. High androstenone levels could not be predicted by measuring reproductive organ sizes. Several nonthyroidal illnesses in euthyroid dogs can affect the results of thyroid function testing, making interpretation of the results more difficult with an increased risk of overdiagnosing hypothyroidism. The purpose of this study was to evaluate the effect of chronic, moderate to severe, osteoarthritis on canine thyroid function. Ninety-six, healthy, client-owned dogs, 65 of which were suffering from moderate to severe osteoarthritis and 31 euthyroid dogs without any physical evidence of osteoarthritis, were used in this study. Blood samples were collected to evaluate serum basal total thyroxine (TT4), free thyroxine (FT4), and thyrotropin (TSHc) concentrations. Basal serum TT4 concentration was not affected by osteoarthritis in dogs. Mild, but statistically significant, differences were noticed in FT4 and TSHc concentrations among the 2 groups. However, this had limited clinical relevance, since virtually all values were within their reference range, and no dogs would have been misdiagnosed as hypothyroid. Therefore, based on the results of our study, osteoarthritis does not need to be considered a factor influencing thyroid function evaluation in dogs.  相似文献

Obesity and weight loss have been shown to alter thyroid hormone homeostasis in humans. In dogs, obesity is the most common nutritional problem encountered and weight loss is the cornerstone of its treatment. Therefore, it is important to clarify how obesity and weight loss can affect thyroid function test results in that species. The objectives of this study were to compare thyroid function in obese dogs and in lean dogs and to explore the effects of caloric restriction and weight loss on thyroid hormone serum concentrations in obese dogs. In the first experiment, 12 healthy lean beagles and 12 obese beagles were compared. Thyroid function was evaluated by measuring serum concentrations of total thyroxine (TT4), free thyroxine (FT4), total triiodothyronine (TT3), thyrotropin (TSH), and reverse triiodothyronine (rT3) as well as a TSH stimulation test using 75 microg i.v. of recombinant human TSH. In the second experiment, eight obese beagles were fed an energy-restricted diet [average 63% maintenance energy requirement (MER)] until optimal weight was obtained. Blood samples for determination of TT4, FT4, TT3, TSH and rT3, were taken at the start and then weekly during weight loss. Only TT3 and TT4 serum concentrations were significantly higher in obese dogs as compared to lean dogs. In the second experiment, weight loss resulted in a significant decrease in TT3 and TSH serum concentrations. Thus obesity and energy restriction significantly alter thyroid homeostasis in dogs, but the observed changes are unlikely to affect interpretation of thyroid function test results in clinics.  相似文献
