A new cytotoxic thiodepsipeptide, verrucosamide (1), was isolated along with the known, related cyclic peptide thiocoraline, from the extract of a marine-derived actinomycete, a Verrucosispora sp., our strain CNX-026. The new peptide, which is composed of two rare seven-membered 1,4-thiazepane rings, was elucidated by a combination of spectral methods and the absolute configuration was determined by a single X-ray diffraction study. Verrucosamide (1) showed moderate cytotoxicity and selectivity in the NCI 60 cell line bioassay. The most susceptible cell lines were MDA-MB-468 breast carcinoma with an LD50 of 1.26 µM, and COLO 205 colon adenocarcinoma with an LD50 of 1.4 µM. Also isolated along with verrucosamide were three small 3-hydroxy(alkoxy)-quinaldic acid derivatives that appear to be products of the same biosynthetic pathway.  相似文献

The recent creation of a veterinary clinical pathology biologic variation website has highlighted the need to provide recommendations for future studies of biologic variation in animals in order to help standardize and improve the quality of published information and to facilitate review and selection of publications as standard references. The following recommendations are provided in the format and order commonly found in veterinary publications. A checklist is provided to aid in planning, implementing, and evaluating veterinary studies on biologic variation (Appendix  S1 ). These recommendations provide a valuable resource for clinicians, laboratorians, and researchers interested in conducting studies of biologic variation and in determining the quality of studies of biologic variation in veterinary laboratory testing.  相似文献

The use of wrapped forage bales with high dry‐matter (DM) content implies risk of fungal growth inside the wrapping, and impaired feed quality. Since fungi may be unevenly distributed in bales, the method of sampling can influence the outcome of the analysis. Three common sampling methods for detection of fungi in wrapped forage were compared: direct plating of visible mouldy patches on bale surfaces (Method I), direct plating of pieces of foliage from drilled core samples (Method II) and quantitative analysis dilution series of drilled core samples (Method III). All samples were cultured on two media at two temperatures. Samples were collected from 124 farms in Sweden and Norway. Using Methods I, II and III, fungi were detected on 52%, 77% and 56% of the farms respectively. Fifty‐two fungal species were identified using a combination of culturing and molecular methods. The most predominant species were Arthrinium spp., found on 55% of the farms, followed by Penicillium roqueforti at 48% of the farms. Incubation at 25°C resulted in higher numbers of all genera, except Aspergillus, compared to incubation at 37°C. The different methods of sampling and culturing did not identify the same fungal species within the same bale. Analysing one bale per farm resulted in detection of less than half the number of species compared to analysing three bales. Of the sampling and culturing methods compared, direct plating of drilled core samples cultivated at malt extract agar at 25°C performed best in terms of qualitative analysis of fungi species.  相似文献

Infectious pancreatic necrosis (IPN) is an important restraint to production of salmonids in aquaculture globally. In order to implement efficacious mitigation strategies for control of this disease, it is important to understand infection routes under current production systems. IPN virus has been shown to be transmitted vertically in Rainbow trout, from broodstock to fingerlings in hatcheries, and there is circumstantial evidence suggesting that vertical transmission can also occur in Atlantic salmon, in addition to horizontal transmission between grow‐out fish in farms. In this study, we show that the smolt carries infection with IPN from hatchery to the marine farm. We do this by comparing sequences from fish groups taken both in hatcheries and on corresponding marine grow‐out farms. We use statistical analysis to prove that sequences obtained from the same fish group in both hatchery and marine farm are more similar than sequences obtained from random fish groups on hatcheries and marine farms.  相似文献

Sea urchins produce high‐energy, membrane‐bound fecal pellets that contain residual nutrients and large quantities of microbiota. These egesta are readily consumed by the shrimp, Litopenaeus vannamei. Egesta of the sea urchin, Lytechinus variegatus, were evaluated as a feed supplement or total replacement for a commercial shrimp diet. Shrimp were stocked at 0.49 g ± 0.06 g initial body weight and housed individually in 2.8‐L tanks in a commercial recirculating zebrafish system. Shrimp were assigned to one of six diets: commercial shrimp feed, reference sea urchin feed, collected dried sea urchin egesta, collected wet sea urchin egesta, half ration of shrimp feed and half collected wet sea urchin egesta, and egesta naturally produced by two sea urchins in polyculture. Equivalent dry matter amounts of each diet were proffered to shrimp in each treatment twice daily, except for those that had complete access to natural egesta excreted by sea urchins in polyculture. Sea urchins were proffered a reference sea urchin feed at 2% body weight daily. After 27 days, shrimp proffered collected dried or wet egesta did not differ significantly in percent weight gain and showed the lowest weight gain. The percent weight gain of shrimp fed the commercial shrimp diet did not differ significantly from that of the shrimp fed half commercial shrimp diet and half egesta. The highest weight gain was recorded for those shrimp that consumed the untouched egesta produced by sea urchins in polyculture. These data suggest that consumed egesta have noteworthy nutritional value and therefore would be beneficial to the culture of extractive species in an integrated multitrophic aquaculture system.  相似文献

ABSTRACT

The global interest in growing perennial grain crops such as intermediate wheatgrass (Thinopyrum intermedium) (Kernza) for production of food and feed is increasing. Intercropping Kernza with legumes may be a sustainable way of supplying nitrogen to soil and associated intercrop. We determined the competitive interactions between intercropped Kernza (K) and alfalfa (Medicago sativa L.) (A) under three inorganic nitrogen (N) rates N0, N1, N2 (0, 200, 400 kg ha?1) and five species relative frequencies (SRF) (100%K:0%A, 75%K:25%A, 50%K:50%A, 25%K:75%A and 0% K:100%A) in mixed intercrops (MI) in a greenhouse pot experiment. After 11 weeks of growth. Kernza dry matter yield (DM) and N accumulated (NACC) were low, but alfalfa DM and NACC high at 0 kg N ha?1. 200 and 400 kg N ha?1 fertiliser application increased the competitive ability (CA) of Kernza and reduced the CA of alfalfa. SRF had large impacts on alfalfa DM, NACC and NFIX only at 0 kg N ha?1 fertiliser, and insignificant impacts on Kernza at all N fertiliser levels, indicating that adjustment of SRF may not be an effective way to modulate the interspecific competition of Kernza. Further research on the other factors that influence the interspecific competition are warranted.  相似文献

A captive adult female bottlenose dolphin presented with stillbirth. The placenta appeared oedematous. No other gross lesions were evident in the placenta or the stillborn calf. Histopathology revealed mild multifocal placentitis and foetal encephalitis. Brucella sp. was isolated from lung, liver, spleen and kidney. Sequence and phylogenetic analysis demonstrated this organism to be most similar to Brucella ceti sequence type (ST) 27. Brucella sp. DNA was detected in formalin-fixed paraffin-embedded placenta and brain by real-time PCR using primers targeting the IS711 gene. Immunohistochemical staining revealed Brucella sp. antigen in placental inflammation. This is the first report of isolation of Brucella sp. from a marine mammal in the Southern Hemisphere and the first report of marine Brucella-associated disease in Australia.  相似文献

A multivariate model was developed and used to estimate genetic parameters of body weight (BW) at 1–6 weeks of age of broilers raised in a commercial environment. The development of model was based on the predictive ability of breeding values evaluated from a cross-validation procedure that relied on half-sib correlation. The multivariate model accounted for heterogeneous variances between sexes through standardization applied to male and female BWs differently. It was found that the direct additive genetic, permanent environmental maternal and residual variances for BW increased drastically as broilers aged. The drastic increase in variances over weeks of age was mainly due to scaling effects. The ratio of the permanent environmental maternal variance to phenotypic variance decreased gradually with increasing age. Heritability of BW traits ranged from 0.28 to 0.33 at different weeks of age. The direct genetic effects on consecutive weekly BWs had high genetic correlations (0.85–0.99), but the genetic correlations between early and late BWs were low (0.32–0.57). The difference in variance components between sexes increased with increasing age. In conclusion, the permanent environmental maternal effect on broiler chicken BW decreased with increasing age from weeks 1 to 6. Potential bias of the model that considered identical variances for sexes could be reduced when heterogeneous variances between sexes are accounted for in the model.  相似文献
